Claims (13)

1. Способ идентификации последовательности целевого полинуклеотида, предусматривающий
(i) приведение в контакт целевого полинуклеотида с полимеразным ферментом и одним из нуклеотидов А, Т(U), G и С в условиях, подходящих для прохождения полимеразной реакции,
(ii) измерение времени, затраченного полимеразой на то, чтобы она связалась и затем отделилась от целевого полинуклеотида для того, чтобы таким образом определить, включила ли полимераза нуклеотид в целевой полинуклеотид,
(iii) необязательно повторение этапов (i) и (ii) с дополнительными нуклеотидами для того, чтобы таким образом идентифицировать последовательность целевого полинуклеотида.
2. Способ идентификации мутации в целевом полинуклеотиде, включающий в себя этапы, на которых
(i) целевой полинуклеотид приводят в контакт с полимеразным ферментом и одним из нуклеотидов А, Т(U), G и С в условиях, подходящих для прохождения полимеразной реакции, и
(ii) измеряют время, затраченное полимеразой на связывание и последующее отделение от целевого полинуклеотида, чтобы таким образом идентифицировать, включила ли полимераза нуклеотид в целевой полинуклеотид, и исходя из природной последовательности мишени определить, существует ли мутация.
3. Способ по п.1 или 2, в котором этапы (i)-(ii) выполняют по очереди с каждым из различных нуклеотидов до тех пор, пока не обнаружат включение.
4. Способ по п.1 или 2, в котором целевой полинуклеотид иммобилизован на материале подложки.
5. Способ по п.1 или 2, в котором множество целевых полинуклеотидов иммобилизованы на материале подложки.
6. Способ по п.1 или 2, в котором этап (ii) выполняют путем измерения приложенного излучения.
7. Способ по п.1 или 2, в котором этап (ii) выполняют путем измерения комбинационного рассеяния.
8. Способ по п.1 или 2, в котором этап (ii) выполняют путем приложения поверхностной электромагнитной волны.
9. Способ по п.8, в котором поверхностная электромагнитная волна представляет собой поверхностную плазменную волну.
10. Способ по п.1 или 2, в котором обнаружение выполняют путем измерения поверхностной электромагнитной волны.
11. Способ по п.1 или 2, в котором полимераза содержит детектируемую метку, прикрепленную к ней.
12. Способ по п.11, в котором метка представляет собой флуорофор.
13. Способ по п.11 в той части, в которой он зависит от любого из пп.1-6, в котором полимераза дополнительно содержит метку в виде донора энергии или метку в виде акцептора энергии и в котором этап (ii) выполняют путем измерения переноса энергии между флуорофором и донором или акцептором энергии.
