Abstract

Un metodo incluye hacer fluir un reactivo de incorporacion a traves de un sistema de gestion de reactivos y una celda de flujo de un instrumento. La celda de flujo tiene un primer polinucleotido colocado en su interior. El reactivo de incorporacion anade una primera base a una secuencia de bases. La secuencia de bases incluye un segundo polinucleotido complementario al primer polinucleotido. Se captura una imagen de una senal de identificacion que emana de la primera base despues de que la primera base se haya anadido al segundo polinucleotido. Se hace fluir un reactivo de escision a traves del sistema de gestion de reactivos y la celda de flujo para eliminar un primer terminador de la primera base con el fin de permitir que una base posterior en la secuencia de bases se anada al segundo polinucleotido. Se hace fluir un reactivo tampon a traves del sistema de gestion de reactivos y la celda de flujo en una pluralidad de ciclos de direcciones de flujo consecutivas hacia adelante y hacia atras.
