For å bestemme jern i serum ved frigjørelse av det bundne jern gjennom tilsetning av et proteindenaturerings- middel, reduksjon av det frigjorte Jern til Fe2', tilsetning av en fargereagens- oppløsning og fotometrisk måling av fargekomplekset som dannes, tilsettes en blanding av et denatureringsmiddel som inneholder urea eller et ureaderivat, og en fettalkoholpolyglykoleter til prøveoppløsningen.
