Description
Foreliggende oppfinnelse angår nye aminosyrederivater, fremgangsmåter for fremstilling av disse og deres anvendelse som nye terapeutiske midler.
Nærmere bestemt angår oppfinnelsen D-aminosyrederivater som er substrater for enzymet D-aminosyreoxydase, fremgangsmåter for fremstilling av disse og deres anvendelse som terapeutiske midler.
I særdeleshet angår oppfinnelsen D-aminosyrederivater som er substrater for enzymet D-aminosyreoxydase, hvilke derivater har formel: hvori
Rer H, CH3, CH3CH2, CH3CH2CH2, CH3 (CH2) 2CH2, (CH^<C>H,
CH^CH2CH/HOCH2 / CH^CH2SCH2CH2/CH^SCf^f CH-^SCH2CH2f C6H5CH2, 4-hydroxy-C6H5CH2, 3,4-dihydroxy-CgH3CH2og
R-^betegner des-aminoresten av et terapeutisk middel som bærer et aktivt primært amin; og farmasøytisk akseptable salter derav.
Det skal understrekes at med et unntak må amino-syredelen representert i formel (I) i hvert tilfelle være i D-enantiomorf form, slik som avbildet ved den sort opptrukne binding til symbolet R. Det ene unntak er aminosyren glycin som selvsagt ikke utviser noe assymetrisk senter.
Det skal ennvidere bemerkes at for at forbindelsen skal være et egnet substrat for enzymet D-aminosyreoxydase, må a-aminonitrogenet være i form av et sekundært amin for å danne et aktivt primært amin av det terapeutiske middel som dannes. Det kan ikke representere et tertiært amin. Dette kan illustreres skjematisk som følger: Oppfinnelsen angår således forbindelser omfattende (a) en primær aminoterapeutisk del eller reaktivt derivat derav, og (b) en D-aminosyredel av formel
eller et reaktivt derivat derav, hvilke forbindelser er i stand til in vivo enzymatisk splitting når de bringes i kon-takt med enzymet D-aminosyreoxydasen. En slik splitting ved setet av D-aminosyreoxydase vil danne det primære amin-bærende terapeutiske ntiddel som i de fleste tilfeller mer effektivt vil være tilgjengelig for å behandle de sykdomstil-stander for hvilke det terapeutiske middel er kjent å være virksomt. Eksempelvis vil forbindelsen N-[ (E)-3-f luor-2-(3-trifluormethylfeny1)-2-propeny1]glycin fremstilt via de reaktive derivater av glycin og det antidepressive middel (E)-2-(3-trifluormethylfenyl)-3-fluorallylamin utvise en anti-depressiv effekt i mattedyr takket være det faktum at den er et substrat for D-aminosyreoxydase.
Oppfinnelsen omfatter således D-aminosyresubstrater av mange forslkjellige forbindelser valgt fra et stort antall terapeutiske midler. Generelt har forbindelsene av formel (I) en forhøyet, men lignende terapeutisk aktivitet enn den som er kjent for de aminobærende terapeutiske midler per se.
To klasser av primære amino-bærende terapeutiske midler tjener imidlertid ikke som substrater for D-aminosyreoxydase. For det første vil terapeutiske midler med én terapeutisk ring eller en aromatisk heterocyklisk gruppe tilstøtende til det primære amin ikke virke som D-aminosyresubstrater . For det andre vil terapeutiske mdiler som har en carboxyl- eller carbalkoxylgruppe i a-stilling til det primære amin ikke tjene som D-aminosyresubstrater.
Eksempler på primære amino-bærende terapeutiske midler som ikke danner D-aminosyresubstrater omfatter:
a-monofluormethylornithin a-difluormethylornithin
Generelt er de foretrukne D-aminosyrederivater av formel (I) hvori R betegnet en rest av glycin (R er H), alanin (R er -CH3), valin [R er (CH3)2CH-], methionin (R er CH3SCH2CH2-), og methylcystein (R er CH3SCH2-), hvor resten av glycin, alanin og valin er de mest foretrukne.
De NH2~kærenc^e te ra<p>e uti ske midler kan være anti-hypertensiva, vasodilaterende dopaminagonister, anorectiske midler, anti-migrenemidler, anti-depressiver, anti-bacterielle midler, anti-fungale midler, anti-virale midler og anti-protozoale midler, diuretica, anti-epileptica, cytotoksiske midler, angstdempende midler, sentralt virkende antihypertensive midler, Angiotensin-omdannende enzym (ACE) inhibitorer anvendbare som anti-hypertensive midler og lignende .
De foretrukne anti-hypertensive midler av formel (I) er D-aminosyreoxydase-substrater av formel: hvori R-delén er som definert for formel I og ' er representert ved
Foretrukne anorectiske midler av formel (I) er E-aminosyreoxydase-substrater av formel 2 hvori R-delen er som definert for formel (I) og R-^ ' er des-aminorestene av slike forbindelser som: a-methyl-4-(trifluormethyl)fenethylamin,
a-methyl-3-(trifluormethyl)fenethylamin,
3,3-difluorampetamin,
2-amino-l-fenylpropanol (norisoefedrin),
2-(1-aminopropyl)-2-indanol (indanazolin),
2-klor-a,a-dimethylfenethylamin (klortermin),
4-klor-a,a-dimethylfenethylamino (klorfentermin),
a,a-dimethylfenethylamin (fentermin),
2-amino-l-(2<1->methylfenyl)propan (ortetamin),
2-fenyl-3-methyl-3-butylamin (pentorex).
Foretrukne sentralt virkende forbindelser av formel (I) som er anvendbare for behandling hypertensjon er D-aminosyreoxydase-substrater hvori R-^-terapeutiske midler er des-aminorestene av a-methy1-dopamin og a-methy1-norepine-f rin.
Antallet av terapeutiske midler som er funnet å være anvendbare ved behandling av mental depresjon representerer en meget stor klasse av forbindelser. Disse forbindelser som generelt angis som anti-depressiva, kan karakteriseres som (1) MAO inhibitorer, både irreversible og konkurrerende inhibitorer, (2) "tricykliske forbindelser", som er antatt å interferere med opptaket av norepinefrin eller serotonin av nervecellen og (3) en blandet gruppe av forbindelser som ikke faller inn under noen av de foregående generelle klasser. De foretrukne anti-depressive midler av formel (I) er de D-aminosyreoxydase-substrater hvori R-^-delen representerer des-aminorestene av forbindelser slik som: 4-dimethylamino-a-2-dimethylfenethylamin, tranylcypromin, etryptamin, forbindelser av strukturformler: og deres steriske isomerer, hvori hver av fenyldelene kan være 1, 2 eller 3 substituenter valgt fra gruppen: lavere alkyl, alkoxy, hydroxy, halogen eller CF^; og (CH2)n kan være en alkylendel med 1-11 carbonatomer.
Det er antatt at et hvilket som helst av de tricykliske anti-depressiva som for tiden er i bruk idag hvor disse inneholder en -NH2~del (i stedet for i deres eksiste-rende sekundære eller tertiære aminform) ville være egnet som R-^des-aminorester og ville være anvendbare substrater for D-aminosyreoxydase. Således vil des-aminoforbindelser av anti-depressive tricykliske forbindelser av formel:
hvori den stiplede linje er en fakultativ dobbeltbinding, gruppen (CH2)nav formel (a) er en rettkjedet eller forgrenet lavere alkylgruppe inneholdende minst 3 carbonatomer for å separere de to nitrogenatomer, og hvori for formel (b) X er nitrogen, Y er oxygen og n er 2; eller X er carbon, Y er CH2og n er én, være anvendbare.
Når det gjelder Angiotensin-omdannende enzym (ZCE) inhibitorer som er anvendbare som antihypertensive midler, foretrekkes det å anvende des-aminorestene av forbindelser av formel: r r ..
R2er (CH2)4NH2 og
R3er H eller lavere alkyl (C1-4).
Når det gjelder vekstaktiverende midler, foretrekkes det å anvende des-aminorestene av peptider slik som for eksempel histidin-D-tryptofan-alanin-tryptofan-D-fenylalanin-lysin.
Når det gjelder analgesia kan på ligende måte R-^betegne des-aminorestene av et peptid slik som for eksempel tyrosin-glycin-glycin-fenylalanin-leucin (leucin-enkefalin) eller tyrosin-glycin-glycin-fenylalanin-methionin (methionin-enkefalin).
Når det gjelder diuretica kan symbolet R^ av formel (I) betegne des-aminorestene av forskjellige diuretica såfremt det diuretiske middel ikke inneholder et primært amin tilstøtende til en aryl- eller aromatisk-heterocyklisk ring, eller en a-carboxylsyre eller ester tilstøtende til det primære amin. Således utviser derivater av dopamin og beslek-tede forbindelser i tillegg til deres andre egenskaper, en diuretisk effekt ved renal vasodilatering og natriumutskil-lelse. Følgelig utgjør deres des-aminorester anvendbare substrater ved utøvelse av oppfinnelsen.
Eksempler på foretrukne anti-migreneforbindelser er de D-amino-syreoxydase-substrater hvori symbolet R-^av formel (I) er en des-aminorest av
Eksempler på antibiotiske forbindelser av formel (I) som er anvendbare som terapeutiske midler er de D-amino-syreoxydase-subs trater hvori R^-delen representerer des-aminorester av aminoglycosider slik som gentamicin, kanamycin, amikacin, paramonycin, tobramycin og antimalariamidlet primaqine.
Generelt kan forbindelsene fremstilles ved anvendelse av analoge kjemiske reaksjoner kjent innen faget.
Slike prosesser kan hensiktsmessig karakteriseres som (1) N-alkyleringen av D-aminosyreestere., (2) N-alkyleringen av N-beskyttede D-aminosyreestere, (3) N-alkyleringen av aminderivater, (4) reduktiv alkylering av D-aminosyren og dets estere og (5) reduktiv alkylering av det primære amin. Gan-ske naturlig er disse prosesser ikke de eneste prosesser som er tilgjengelige, men de representerer de foretrukne synte-tiske metoder for fremstilling av forbindelsene av formel (I). Valget av hvilke som helst av disse prosesser avhenger av
de vanlige parametre som generelt er kjent for fagmannen så-velsom tilgjengeligheten av de nødvendige reaktanter.
Generelt kan N-alkyleringen av D-aminosyreestere illustreres som følger:

I den ovenfor angitte reaksjon er R og R-^som definert for formel (I), symbolet X er en hvilken som helst egnet forlatende gruppe slik som halogen, f.eks. Cl, Br eller jod, tosylat, mesylat og lignende; og symbolet R4er hydrogen eller en rettkjedet eller forgrenet lavere alkylgruppe
Ved utførelse av den foregående reaksjon omsettes ekvivalente mengder av reaktanter anvendt som D-formen av aminosyren (IIII) i et organisk løsningsmiddel slik som dimethoxyethan, dimethylformamid, dimethylsulfoxyd, tetrahydrofuran og lignende. Reaksjonen utføres i nærvær av en ekvivalent av en base slik som triethylamin eller dets funk-sjonelle ekvivalenter ved temperaturer varierende fra 20 til 180° C i tidsrom på fra 1 til 4 8 timer. Ved endt reaksjon hydrolyseres den N-alkylerte ester til den tilsvarende syre under anvendelse av standard sure eller basiske betingelser, f.eks. LiOH i en vann/alkoholblanding eller kokning under tilbakeløpskjøling med vandig HBr eller HC1.
N-alkyleringen av N-beskyttet D-aminosyreestere kan illustreres som følger:

I den ovenfor angitte reaksjon er symbolene R, R-^, R^og X som tidligere definert, og symbolet R,- representerer en aminbeskyttende gruppe slik som
0 0 0 ■i ii ii C6H5C-, CgH5CH2C- eller RgOC-, hvori Rg er en lavere alkylgruppe hvor t-butylgruppen er en særlig foretrukket gruppe.
Ved innførelse av den foregående reaksjon behand-les den N-beskyttede aminosyre (V) med en ekvivalent av en sterk base i et aprotisk løsningsmiddel slik som tetrahydrofuran, dimethylformamid og lignende. Basen skal være i stand til å trekke hydrogenet fra den N-beskyttede aminfunk-sjon, og kan for eksempel være natriumhydrid, kaliumhydrid, lithiumdiisopropylamid og lignende. Én eller to ekvivalenter av reaktanten (II) tilsettes til anionet, og den resulterende blanding omrøres i et tidsrom på fra 1 til 2 timer ved en temperatur varierende fra -7 til 100° C. N-alkyl-derivåtet (VI) erholdt på denne måte kan renses via silica-gelkroraatografi eller ved anvendelse av krystalliseringsteknikker som er lett tilgjengelige for fagmannen innen faget. De ønskede forbindelser (I) erholdes via splitting av este-* ren, og de N-beskyttende grupper for anvendelse av standard betingelser er kjent innen faget.
N-alkyleringen av aminderivater kan illustreres som følger:

Når symbolet R2er hydrogen, kan den foregående reaksjon utføres i et inert organisk løsningsmiddel slik som dimethoxyethan, dimethylformamid, dimethylsulfoxyd, tetrahydrofuran etc. Ekvivalente mengder av de egnede syrederi-vater (VIII) omsettes med det N-beskyttede amin( VII) i nærvær av en ekvivalent av en base slik som triethylamin, i et tidsrom på fra 1 til 24 timer ved temperaturer varierende fra 0 til 100° C.
De resulterende N-alkylerte-D-aminosyreesterderi-vater kan renses under anvendelse av silicagelkromatografi eller krystalliseringsteknikker kjent innen faget. De ønskede D-aminosyresubstrater (I) kan lett erholdes ved splitting av de tilsvarende estere (IX) under anvendelse av de tidligere beskrevne prosedyrer. Når symbolet R^ representerer en N-beskyttende gruppe, anvendes prosessen ifølge Reaksjonsskjerna B.
Den reduktive alkylering av D-aminosyre og dets estere kan illustreres ved følgende reaksjonsskjema:
Reaksjonsskjerna D I den ovenfor angitte reaksjon er symbolene R, R-^, R. som tidligere definert og den strukturelle gruppe representerer en ekvivalent del for symbolet R^ . Således representerer ketonet (X)
det tilsvarende ketoderivat av det des-aminobærende terapeutiske middel.
Ved utførelse av den reduktive alkylering anvendes betingelser som er velkjent innen faget, se R.F. Borsch et al., JACS 93, 2897 (1971). Således omsettes carbonylderivå-tet (X) og D-aminosyren eller dets ester (III) i nærvær av et reduksjonsmiddel slik som hydrogen, og en egnet kataly-sator. Egnede katalysatorer innbefatter Pd/C, Pt02/ BI^Na, BH^NaCH og lignende. I de tilfeller hvori borhydridene anvendes, er det viktig å regulere pH på reaksjonsblandingen til en pH på 6 - 8. Den reduktive alkylering forløper som et enkelt trinn. Således isoleres aldri det hypotetiske imin-mellomprodukt (XI) men forløper direkte til det tilsvarende amin (XII). Avestring av (XII) eller dets ekvivalent struktur (IV) utføres som tidligere beskrevet.
Den reduktive alkylering av et primært amin kan illustreres som følger:
Reaksjonsskjerna E
I den ovenfor angitte reaksjon er symbolene R, R-^og<R>^som tidligere definert. Generelt er reaksjonsbetin-gelsene de samme som beskrevet for Reaksjonsskjerna D. Imidlertid erholdes aminosyrederivatene (IVa) som racemiske blandinger. Aminosyrederivatene (IVa) kan oppløses til de ønskede D-enantiomerer av de tilsvarende D-aminosyrederivater (IV) under anvendelse av standard oppløsningsteknikker som er velkjente innen faget. Avestring av (IV) forløper deretter på den tidligere beskrevne måte.
Alternativt kan de racemiske aminosyrederivater (IVa) avestres under dannelse av racemiske aminosyreoxydase-substrater av (I), som deretter kan oppløses under dannelse av de ønskede D-aminosyreoxydase-substrater (I) ifølge oppfinnelsen .
De etterfølgende eksempler illustrerer de foregående generelle reaksjoner. Ved anvendelse av den egnede . reaksjon kan hvilke som helst av forbindelsene av formel (I) lett erholdes når de egnede derivater av forbindelser R-|.NH2og H-N-C-COOH anvendes.

Eksempel 1
N-( 3, 4- dihydroxyfenethyl) glycin
En løsning av 1,12 g N-tert-butoxycarbonylderivat tilgjengelig fra 3-3 ,4-dimethoxy f enethylamin og di-tert-butyldicarbonat i 15 ml THF ble langsomt tilsatt til kald (ca. -70° c) LDA (fremstilt fra 0,40 g diisopropylamin og 2,7 ml 1,5M n-butyllithium) i IB mlTHF. Etter 30 minutter ble en løsning av 0,72 g tert-butylbromacetat i 5 ml THF tilsatt, kjølebadet ble fjernet og reaksjonsblandingen ble omrørt over natten. Løsningsmidlet ble fordampet og produktet ble deretter isolert ved ethereks tråksjon som en lys oransje olje (1,10 g) . Kromatografi (silica; 40 % ether i lett petroleum) ga hovedsakelig rent beskyttet aminosyre (0,66 g) som en farveløs olje.
En del av dette produkt (0,50 g) i 47 % vandig HBr (2 0 ml) ble kokt under tilbakeløpskjøling i 5 timer og ble deretter fordampet til tørrhet. Det faste residuum (0,29 g) ble oppløst i 5 ml EtOH og blé behandlet med et overskudd av propylenoxyd (0,18 g), og det resulterende bunnfall ble oppsamlet og tørket under dannelse av N-(3,4-dihydroxyfenethyl)-glycin som et gråaktig pulver (0,20 g), sm.p'. 245 - 256° C (spaltning).
Eksempel 2
N-[( E)- 3- fluor- 2-( 3- trifluormethylfeny1)- 2- propenyl] glycin
0,96 g N-tert-butoxycarbonylderivat 2_ fremstilt fra (E ) -2-(3-trifluormethylfenyl)-3-fluorallylamin og di-tert-butyldicarbonat på vanlig måte, ble oppløst i 20 ml dimethylformamid og ble avkjølt til ca. -10° C. En disper-sjon av natriumhydrid i mineralolje (0,14 g, ca. 55 % NaH) ble tilsatt, og etter omrøring i 2 0 minutter ble en løsning av 0,55 g tert-butylbromacetat i lm DMF dråpevis tilsatt. Omrøring ble fortsatt i 1 time ved -10° C hvoretter vannet
ble tilsatt og produktet ble isolert ved etherekstraksjon. Det urene produkt ble renset ved silicakromatografi (under anvendelse av 20 % ether i lett petroleum som elueringsmid-del) hvorpå den beskyttede aminosyre ble erholdt som en farve-løs olje (0,63 g, 48 % utbytte).
Denne substans ble oppløst i 20 ml tri fluoreddik-syre ved romtemperatur, ble deretter fordampet til tørrhet etter 1 time. Residuet ble tatt opp i 5 ml CH3OH og ble behandlet med 1 ml propylenoxyd ved -20° C over natten. Det resulterende bunnfall ble oppsamlet og tørket under dannelse av det ønskede glycinderivat (0,12 g, 30 % utbytte) som farveløse nåler med sm.p. 199 - 201° C (spaltning).
Eksempel 3
N-[ 6, 7- dihydroxy- 2- tetralyl] glycin
En blanding av 2,06 g 6,7-dimethoxy-2-tetralon og 1.3 g tert-butylglycinat i 20 ml vannfri tetrahydrofuran ble omrørt ved romtemperatur i 4 timer med pulverformede molekylsiler nr. 4A. 0,6 g natriumcyanoborhydrid ble tilsatt i to porsjoner på 0,3 g ved 6 timers intervaller. Reaksjonsblandingen ble deretter filtrert og og filtratet
ble konsentrert i vakuum. Residuet ble deretter tatt opp i 50 ml diethylether. Den organiske fase ble vasket med vann, saltvann, ble tørket over MgSO^og inndampet i vakuum. Residuet ble underkastet kromatografi på silicagel under dannelse av N-tert-butyloxycarbonylmethy1-6,7-dimethoxy-2-aminotetralin som ble kokt under tilbakeløpskjøling i 47 % vandig hydrobromsyre i 5 timer. Det faste residuum erholdt ved konsentrert i vakuum ble oppløst i ethanol og behandlet med et overskudd av propylenoxyd under dannelse av tittel-forbindelsen.
Eksempel 4
N- [ 3-( 3- trif luormethylfenyl) - 2- propyl ] alanin
En blanding av 1,9 g a-methy 1-3-trifluormethyl-benzen-ethanamin, 2,09 g (S)-tert-butyl-2-brompropionat og 1.4 ml triethylamin i 20 ml dimethylformamid ble omrørt ved romtemperatur i 16 timer. Løsningen ble deretter konsentrert i vakuum og residuet ble ekstrahert med 50 ml diethylether. Den organiske fase ble vasket med vann, saltvann, ble tørket over magnesiumsulfat og ble konsentrert i vakuum. Residuet ble kromatografert på silicagel under dannelse av (R) -tert-butyl-N-[3-(3-trifluormethylfenyl)-2-propy1]alanat og ble hydrolysert til det ønskede produkt.
Bestemmelsen hvorvidt heller ikke en bestemt for-bindelse omfattet av den generelle ramme for oppfinnelsen vil være et egnet substrat for D-aminosyreoxydase kan foretas på basis av en enkel in vitro-test fastslås med standard laboratorie in vivo-teknikker. For in vitro-testing under anvendelse av kommersiell svinenyre D-amino-syreoxydase inkuberes forbindelsene under optimale betingelser for enzymatisk reaksjon, og hvorpå dannelsen av I^C^eller av det forventede primære amin deretter kvantitativt bestemmes som en funksjon av substratkonsentrasjon, inkuberingstid etc.
Generelt bestemmes dosen og administreringsmåten for de nye forbindelser ved standard vurderingsteknikker som er kjent for den bestemte klasse av forbindelser til hvilke det amino-bærende terapeutiske middel tilhører.

1. Fremgangsmåte for fremstilling av D-aminosyreoxydase -substrater av formel

CH3 CH2 CH-, -CH2 OH, CH3 CH2 SCH2 CH2~ , CH3 S <CH>2<->, CH3 SCH2 CH2~ , C6 H5 CH2~ , 4-OH-CgH4 CH2 -, 3,4-diOH-CgH3 C <H>2<-,> og R^ er des-aminoresten av et amino-bærende terapeutisk middel/karakterisert ved at den omfatter N-alkylering av et derivat av angitte terapeutiske middel av formel
hvori Ry betegner hy* RgS02 hvori Rg betegner lavere alkyl med 1-4 carbonatomer og Rg betegner aryl; med et D-syrederivat av formel
hvori X er halogen, tosylat eller mesyltat, og R^ er hydrogen eller en rettkjedet eller forgrenet lavere alkylgruppe med 1-6 carbonatomer, i nærvær av en base i et tidsrom på fra 1 til 2 4 timer ved en temperatur varierende fra 0 til 100° C, med etterfølgende fjerning av esteren og aminbeskyttende grupper under dannelse av D-aminosyreoxydase-substratet.
