Description
Foreliggende oppfinnelse vedrører nye 1,3-tiazolidin-4-yl-karboksylsyrederivater med den generelle formel I
hvor
R betyr hydrogen, (C-^-C^) alkyl, aralkyl, fenyl, fenyl substituert med et halogenatom eller en metoksygruppe, formyl, acyl, trimetylsilyl;
R<1>betyr hydrogen, et farmasøytisk akseptabelt uorganisk eller organisk kation, (C^C^)alkyl, 2,2,2-trikloretyl, acetonyl, benzyl, benzyl substituert med nitro eller metoksy, fenyl, nitrofenyl, benzhydryl eller trimetylsilyl;
R' kan også være en rest som kan gi metabolsk aktivering
in vivo valgt fra acetoksymetyl, pivaloyloksymetyl, ftalidyl, benzoyloksymetyl, 5-indanyl, en gruppe med formel
eller en gruppe med formel hvor R'' står for (C^-C^) alkyl, (C5~C6)cykloalkyl eller aryl; A betyr hydrogen, halogen, N,, OH, NH0; eller et kvartærnært N-atom, spesielt
(og i dette tilfellet er R' en nega-
tiv ladning); eller en O-CO-Nh^gruppe; eller en gruppe med formel OR'' ', O-COR''', NH-CO-R'"' eller SR''1, hvor R''' er (C^-C^)alkyl, aryl, substituert aryl eller en heterocyklisk gruppe som kan ha lavere alkylgrupper;
n kan være null eller 1.
Karakteristiske betydninger for resten R ved siden av de allerede ovenfor illustrerte er metyl, etyl, isopropyl, benzyl, trityl, acetyl, propionyl, trifluoracetyl, benzoyl, benzoyl substituert med en til tre hydroksygrupper, metyl, metoksy, halogen, amino og nitrogrupper, benzyloksykarbonyl, tert.-butoksykarbony1 eller en rest avledet fra en naturlig aminosyre.
Typiske, men ikke begrensende eksempler på uorganiske eller organiske kationer som er angitt gjennom resten R<1>er natrium, kalium, kalsium og magnesiumkationer; kationer avledet fra organiske baser såsom f.eks. dibenzylamin, N,N-dibenzyletylen-diamin, glukamin, N-metylglukamin, heksametylentetramin, 2-amino-2-hydroksymety1-1,3-propandiol, arginin, lysin, prolin, karnitin; eller aluminium, sink eller sølvkationer.
Typiske betydninger for R''' ved siden av de ovenfor allerede illustrerte er metyl, etyl, propyl, butyl, isobutyl; fenyl eller benzyl; imidazol-2-yl, 1, 2 , 3^-triazol-5-yl, l-metyl-1,2, 3,4-tetrazol-5-yl, tiazol-2-yl, 1,3,4-tiadiazol-2-yl, 5-metyl-1,3,4-tiadiazol-2-y1, 1,2,3,4-tiatriazol-5-yl, oksazol-5-yl, 3-metylisoksazol-5-yl, 1,2,4-oksadiazol-5^yl, pyrazinyl. På grunn av det asymmetriske karbonatomet i 4-stilling i tia-zolidinringen kan forbindelsen med formel I foreligge i en D- og L-konfigurasjon eller som en racemisk forbindelse. Følgelig omfatter oppfinnelsen alle disse muligheter, selv om hele L-kohfigurasjonen er den foretrukne. . r,n videre gjenstand for oppfinnelsen er også saltene av forbindelsene med formel I, hvori nitrogenatomet i tiazolidin-kjernen har basisk karakter, med farmasøytisk akseptable sy-rer, f.eks. sitronsyre, askorbinsyre, maleinsyre, eddiksyre, saltsyre, salpetersyre, hydrogenbromid og svovelsyre.
En annen gjenstand for oppfinnelsen er de farmasøytiske bland-inger for human eller veterinærmedisinsk bruk, eller pesti-cide formuleringer for jordbruksformål som inneholder som aktiv bestanddel en eller flere av forbindelsene med formel I ovenfor eller et farmasøytisk akseptabelt syreaddisjonssalt derav.
Det er nå overraskende funnet at forbindelsene med formel I ifølge oppfinnelsen er meget bestandige overfor penicillina- ser når de prøves ifølge den metode som er beskrevet av GROVE et al., Assay methods of antibiotics, A Laboratory Manual, Med. Encyclop. Inc., 1955, 7, eller i henhold til LORIAN V., Antibiotics and chemotherapeutic agents in clinical and laboratory practice, CC. Thomas Publ., 1966, 242. De har også vist seg å være aktive mot forskjellige patogene organismer, f.eks. gram-positive og gram-negative, aerobe og anaerobe bakterier, innebefattet (3-laktamase produserende stammer.
Spesielt er forbindelsene med formel I aktive mot gram-posi-'tive bakterier såsom f.eks. Staphylococcus aureus, Diplococ-cus pneumoniae, Streptococcus pyogenes, Streptococcus fae-calis, Bacillus subtilis og Sarcina lutea, samt gramnegative bakterier såsom f.eks. Escherichia coli, Proteus mirabilis, Shigella sonnei,Klebsiella pneumoniae, Pseudomonas aerugi-nosa, Haemophilus influenzae og Salmonella typhimurium.
Forbindelsene méd formel I utviser også en bemerkelsesverdig aktivitet mot forskjellige patogene organismer som er ansvar-lige for infeksjoner på planter, f.eks. Pseudomonas syringae, som er ansvarlig for bacteriosis hos citrusfrukter og syriner; Pseudomonas phaseolicola (fettsyke på bønner).; Pseudomonas aliicola (bakterielt angrep på løk); Pseudomonas savastandi (skabb på oliventrær); Pseudomonas marginata (skabb på sabel-lilje); Xantomonas phaseoli (bakteriell meldugg på bønner); Pectobacterium carotovorum (stump på poteter og bakterielle angrep på iris rhizomes); Erwinia amilovora (necrose på eple-grener); Erwinia carotovora (bakterielt angrep på gulerøtter); Corynebacterium f laccum^-faciens (bakterielt angrep på bønner) ; Penicillum sp. (soppangrep på blomsterløk og knoller).
Som et representativt men ikke begrensende eksempel angis at 7- (N-pivaloyl-1,3-tiazolidin-4-yl) -karboksamido-^cef alosporan-syre (forbindelse med formel I hvori R = (CH-^^C-CO, R' = H, A= O-CO-CH^, n = null) har en antibakteriell aktivitet (mot gram-positive bakterier) tilsvarende også noen ganger bedre enn cephaloridin og cephaleksin. n = null)
er mot gram-positive bakterier minst like aktiv som cephaleksin, men gir ingen resistens, mens cephaleksin in-duserer resistens på S. Aureus stammer i en dose på 25 mcg/ml.
Følgelig kan de nye forbindelser som er en av gjenstandene for foreliggende oppfinnelse gis til varmblodige dyr innebefattet mennesker av en oral, parenteral eller topisk vei for å bekjempe septikjemi, meningitt, endocarditt, infeksjoner i luftveier, fordøyelseskanal eller urinveiskanal og på huden, øre-,nese-,halsinfeksjoner; 'infeksjoner av ben; endoabdominale<p>g endopleuriale infeksjoner. De -kan også anvendes for å gjøre huden aseptisk før injeksjoner eller kirurgiske inngrep, samt for desinfeksjon av kirurgiske in-strumenter.
De egnede farmasøytiske doseringsformer for oral administrer-ing kan f.eks. være tabletter, kapsler, piller, sukkerover-trukne tabletter, syrups, suspensjoner, dråper, eliksirer- og granulater. Farmasøytiske doseringsformer som er egnet for topisk bruk er hovedsakelig salver, kremer, linimenter, collyria og lotions.
Alle de ovenfor nevnte farmasøytiske formuleringer fremstilles som tidligere kjent og inneholder sammen med den aktive bestanddel smøremiddel, fortynningsmiddel, eksipient og søtnirigsmidler samt vanlig brukte farmasøytisk akseptable tilsetningsmidler.
P.g.a. sine phytofarmasøytiske egenskaper kan forbindelsene ifølge oppfinnelsen anvendes i forskjellige administrerings-former, som et eksempel vandige løsninger som inneholder eller ikke inneholder ytterligere tilsetningsmidler såsom f.eks. talkum eller leire, pulvere, innebefattet atomiserte preparater; sprays, både flytende og faste; suppositorier innebefattet formuleringer med langsom avgivning; granulater innebefattet formuleringer med langsom avgivning; former absor-bert på inerte materialer eller ionebytterharpikser; kapsler og mikroinnkapslede preparater. Alle disse administrerings-former er velkjente for en fagmann. Følgelig kan forbindelsene ifølge oppfinnelsen også anvendes for preservering av næringsmidler såsom f.eks. sitrusfrukter eller poteter, etter-som de, i motsetning til mange vanlige anvendte substanser, f.eks. bis-fenyl, er virksomme antibiotiske midler med meget lav toksisitet. Forbindelsene ifølge oppfinnelsen kan fremstilles ad forskjellige veier. Således kan.f.eks. amino-gruppen i en forbindelse med formel II hvor R', A og n er som ovenfor angitt, acyleres med et egnet N-acyleringsreagens avledet fra en syre med formel III
hvor R er som forut angitt. Beskyttelsesgruppene, hvis slike foreligger, må være lette å fjerne under milde betingelser. Et typisk eksempel på en beskyttelsesgruppe er ben-zyloksykarbonylresten. Slike acyleringsreaksjoner er velkjente på området penicilliner og cephalosporinér (se f.eks. Flynn, Cephalosporins and penicyllins, Academic Press, 1972). Også utgangsmaterialene med formel II og III er kjente for en fagmann. Valget av ac_yleringsreagenser vil åpenbart av-henge av den kjemiske natur og substituentene R og R', ifølge de vanlige teknikker og prinsipper.
De N-acylerende reaktanter som er avledet fra forbindelsen med formel III ovenfor er f.eks. halogenidene eller slike som oppnås gjennom omsetning av III med karbodiimidet eller aci-det; fortrinnsvis anvendes et anhydrid og med fordel et
blandet anhydrid fremstilt in situ ved omsetning med etyl-eller isobutylklorkarbonat, eller pivaloylklorid.
Valget av løsningsmiddel og betingelser for reaksjoner, hvilket hovedsakelig avhenger av det valgte N-acyleringsmid-del, er et familiært spørsmål for fagmannen. Som et eksempel kan reaksjonen, hvis acyleringsm iddelet er et blandet an*-hydrid, utføres i et organisk løsningsmiddel valgt blant etylacetat, dimetylformamid, dietyleter, tetrahydrofuran, dioksan, metylenklorid eller■analoge inerte løsningsmidler; temperaturen kan ligge mellom ca. -40°C og romtemperatur og fortrinnsvis ligger den mellom ca. -20 og -30°C.
En videre fordelaktig fremgangsmåte for fremstilling av forbindelsene med formel I er å omsette forbindelsene med formel II og III ovenfor med silisiumtetraklorid ifølge itali-ensk patentansøkning nr. 29445 A/77. Forbindelsene kan iso-leres ved hjelp av velkjente teknikker innenfor området penicilliner og cephalosporiner såsom f.eks. gjennom krystalli-sering, liofylisering eller spraytørking. En egnet isoler-ingsmetode er absorpsjon på kjemisk inert eller aktivert materiale, som f.eks. ionebytterharpikser, og denne fremgangsmåte kan være spesielt anvendelig når det erholdte derivat må brukes som for for dyr eller phytofarmasøytika.
De følgende eksempler er gitt i illustrerende hensikt men
er ikke på noen måte ment å begrense oppfinnelsens omfang.
7-amino^cefalosporansyre vil i det følgende bli angitt med forkortelsen 7-ACA; 7-amino^desacetoksycephalosporansyre med forkortelsen 7-ADCA.
EKSEMPEL 1
7-( 1, 3- tiazolidin— 4- yl)- karboksamido- desacetoksycephalo-sporansyre natriumsalt (forbindelse med formel I hvori R =
H; R<1>= Na; A = H; n = 0)
25,0 gram 7-ADCA oppslemmet i 185 ml metylenklorid avkjølt til -40°C ble under kraftig røring tilsatt 13,0 gram n-pro-pylamin idet man passet på at temperaturen ble holdt på. ca. -2<5>°C. Etter 30 minutter ved denne temperatur ble 26 g trimetylklorsilan langsomt helt inn i reaksjonskolben, og ved slutten av tilsetningen, var temperaturen mellom -5°C og 0°C. Etter røring i 30 minutter og kjøling "til^10°C ble 18,5 g N,N-dimetylanilin og 87,5 g hydroklorid av 7,3-tiazolidin-4-y1-karboksylsyreklorid raskt tilsatt, hvorunder temperaturen fikk stige til omgivelsestemperatur. Den resulterende blanding ble rørt i 2 timer ved romtemperatur, tilsatt 500 ml vann, rørt 30 minutter for fullstendig å fjerne trimetylsi-lylgruppen og forsiktig bragt til pH 7-7,5 med vandig 10% natriumhydroksyd.
Den lavere organiske fase ble skilt fra, den øvre organiske fase ble ekstrahert med 100 ml metyl-isobutylketon og 100 ml dietyleter, behandlet med avfargingskull (forut vasket med saltsyre og vann) og filtrert under vakuum på Dicalite 438 . Den derved erholdte løsning ble lyofilisert hvorved man fikk et hvitt pulver. I.R. og N.M.R. analyse er i overensstemmelse med den ønskede struktur. T.L.C. (Tynnsjiktkromato-grafi) analyse viste at forbindelsen var i det vesentlige ren.
EKSEMPEL 2
7-( l, 3- tiazolidin- 4- yl)- karboksamido- cephalosporansyre nat-riumsalt (forbindelse med formel I hvor R=H; R<1>= Na; A
-0-COCH3; n = 0)
29,5 gram 1, 3-tiazolidin-4-^y 1-karboksylsyre blé oppslemmet i 200 ml aceton og under røring ble 24,2 g trietylamin tilsatt. Blandingen ble så avkjølt til -20°C og ved denne temperatur tilsatt en løsning av 24,0 g pivaloylklorid i 40 ml
aceton. Etter at denne temperatur ble holdt i 3 0 minutter, ble en forut fremstilt løsning av 54,4 g 7-ACA og 24,2 g trietylamin i 400 ml kloroform, avkjølt til -10°C helt i reaksjonskolben og det hele fikk reagere 30 minutter ved -10° C, 1 time ved 0°C og 3 timer uten noen utvendig kjøling. Løsningsmiddelet ble dampet av under vakuum ved en temperatur ikke over 40°C, og den erholdte rest ble tatt opp i 200 ml vann som inneholdt 10% natriumhydrogenkarbonat. Den derved erholdte blanding ble rørt kraftig og bragt til pH 1-1,5 med vandig 15% saltsyre, først ekstrahert med 100 ml dietyleter som ble kastet, og deretter med 300 ml etylacetat. Etylacetatekstraktet ble vasket 2 ganger med 100 ml (2 x 100 ml) vann, tørket over natriumsulfat, deretter ble løsnings-middelet fordampet i vakuum ved en temperatur ikke over 4 0°C. Man fikk en rest som ble behandlet med 200 ml vann, forsiktig gjort nøytral med vandig 10% natriumhydroksyd, uløselige materialer ble filtrert fra denne vandige løsning og filtratet ble lyofilisert og ga således det ønskede produkt. I.R. og N.M.R. analyse er i overensstemmelse med den ønskede struktur. TLC-analyse [Kiselgel F 254 plate (Merck).; elueringssystem: n-butanol/eddiksyre/vann = 30/10/10 (v/v);
synliggjøring: UV-lys (X = 254 nm) og joddamper] viste at forbindelsen var i det vesentlige ren.
EKSEMPEL 3
7-( l, 3- tiazolidin- 4- yl) - karboksamido-^- cephalosporansyre heksa-metylentetraaminsalt (forbindelse med formel I hvor R = H;
R' = heksametylentetraaminkationet; A = -O-COCH^; n = 0)
1000 gram av forbindelsen som erholdtes som beskrevet i eksempel 1 før behandling med vann og natriumhydroksyd ble oppslemmet i 5 liter vann og den resulterende suspensjon ble under røring tilsatt en omtrent ekvimolar mengde av heksa-mety lentetraamin , hvorved pH hadde en verdi på 7-7,5.. Den resulterende løsning ble filtrert fra uløselig stoff, og
det erholdte filtrat ble spraytørket hvilket ga et hvitt, vannløselig pulver som var egnet for veterinær bruk, enten som sådant eller i blanding med integrert for eller drikke-.
vann, for profylakse eller behandling av infeksjoner som for-årsakes av patogene organismer som er sensitive overfor anti-biotikumet .
Det erholdte pulver kan med fordel også anvendes i formuleringer for phytofarmasøytika, alene eller sammen med vanlige anvendte tilsetningsmidler• Den følgende blanding som må fremstilles før bruk viste seg å være spesielt effektiv: forbindelse ifølge eksempel 3: 1 del; såpe: 1 del; petroleum: 1 del; vann: 100 deler. Anvendes ved nebulisering. EKSEMPEL 4 ( 7-( l, 3- tiazolidin- 4- yl)- karboksamido)-( 3-( 5- metyl- l, 3, 4-tiadiazol- 2- y 1- tio ). mety 1) - 3 - cephem- 4 - karboksyl sy re (forbindelse med formel I hvor R = H; R' = H;
En suspensjon av 0,1 mol av forbindelsen fra eksempel 2 i 1000 ml 0,1 M fosfatpuffer (pH 6,4) ble i små porsjoner under røring tilsatt 0,11 mol 2-mety1-5-merkapto-l,3,4-tiadiazol og 0,15 mol NaHCO^idet man passet på at boblingen ikke ble for kraftig. Den erholdte blanding ble oppvarmet til 50-60 C, holdt på denne temperaturen i 6 timer, i hvertfall inntil den acetylerte forbindelse var forsvunnet, deretter ble den avkjølt og vasket to ganger med 200 ml (2 x 200 ml) metyl-isobutylketon. Etter at pH. forsiktig var bragt til 2,5-3,5 fikk man en felling som etter noen timer ble filtrert, vasket med vann og tørket ved en temperatur ikke over 4 0°C. I.R.
og N.M.R. analyse er i overendsstemmelse med den ønskede struktur. T.L.C. analyse (Kiselgel F 254 plate; elueringssystem: n-butanol/eddiksyre/vann = 30/10/10 (v/v); synlig-gjøring: UV-lys (X-=254 nm) og jodazid] viste at forbindelsen var i det vesentlige ren.
EKSEMPEL 5
( 7-( 1, 3- tiazolidin- 4- yl)- karboksamido)-( 3-( 5- metyl- l, 3, 4-tiadiazol- 4- yl- tio) metyl)- 3- cephem- 4- karboksylsyre N- metyl-glukaminsalt (forbindelse med formel I hvor R = H; R' =

n = 0) 1 mol av forbindelsen fra eksempel 4 ble oppslemmet i 1000 ml vann og under kraftig røring og kontinuerlig kontroll av pH tilsatt 1 mol N-metylglukamin i porsjoner, ihvertfall inntil en pH verdi på 7-7,5. Den erholdte løsningen ble filtrert på et steriliserings- og de-pyrogeneringsfilter, filtratet ble lyofilisert ved kjent teknikk hvorved man fikk tittelforbindelsen som et sterilt og pyrogenfritt pulver for intramuskulær injeksjon i veterinærpraksis.. EKSEMPEL 6 3-( pyridiniometyl)- 7-( l, 3- tia2olidin- 4- yl)- karboksamido- 3-cephem-4-karboksylat (forbindelse med formel I hvor R = H; R1 = negative ladning; A
n = O)
En løsning av 180 g av forbindelsen fra eksempel 2 i 500 ml vann ble under kraftig røring tilsatt 100 ml pyridin og deretter 100 g kaliumtiocyanat i små porsjoner. Den resulterende blanding ble bragt til pH 6,5 med 85% fosforsyre og oppvarmet under røring i 6 timer ved 60°C. Etter kjøling til romtemperatur ble reaksjonsblandingen behandlet med Amberlite LA-1® (som det tilsvarende acetat) og oppslemmet
i 25% metyl isobutylketon. Tre behandlinger med 1 liter hver gang ble utført. Den således behandlede vandige løsning ble igjen ekstrahert 3 ganger med 200 ml metyl-isobutylketon (3
x 200 ml) og ble til slutt lyofilisert hvorved man fikk et
hovedsakelig hvitt pulver. I.R. og N.M.R. analyse er i overensstemmelse med den ønskede struktur. TLC analyse [Kiselgel F-254 plate; elueringssystem: etylacétat/eddiksyre = 60/20 (v/v), mettet med vann; synliggjøring: U.V.-lys = 254 nm) og joddamper] viste at forbindelsen var hovedsakelig ren.
EKSEMPEL 7
7-[( N- pivaloyl)- 1, 3- tiazolidin- 4- yl]- karboksamido- cephalosporansyre (forbindelse med formel I hvor R = (CH^)^C-CO;
R' = H; A = -0-C0CH3; n = 0)
10,0 gram N-pivaloy1-tiazolidinkarboksylsyre ble oppslemmet i 50 ml aceton, og deretter ble 4,65 g trietylamin tilsatt. Etter kjøling til -15°C ble suspensjonen tilsatt 5,5 g pivaloylklorid i 25 ml aceton idet man passet på at temperaturen ikke steg over -10°C. Etter ferdig tilsetning ble to dråper 4-metyl-morfolin tilsatt, og blandingen fikk stå ved -15°C
i 20 minutter, og deretter ble en forut fremstilt løsning av 12,5 g 7-ACA i 10 ml vann, 20 ml aceton og 5,0 g TEA av-kjølt til -10°C raskt helt i reaksjonskolben. Den resulterende blanding fikk reagere 4 timer, og kjølebadet ble fjernet etter to timer hvorved temperaturen fikk stige til -20°C, så ble 2,0 g Dicalite^ tilsatt og det "hele ble filtrert under vakuum. Man fikk en rest som ble vasket med 20 ml aceton og 5 ml vann, den erholdte løsning ble filtrert fra alt uløselig
stoff og inndampet til tørrhet i vakuum ved en temperatur
ikke over 40°C. Den erholdte rødlige olje ble oppslemmet i 100 ml vann, den vandige dispersjon ble to ganger ekstrahert med 50 ml (2 x 50 ml) dietyleter, eterekstraktene ble kastet og den vandige fase bragt til pH 1 ved hjelp av vandig 10% saltsyre. Den resulterende emulsjonen ble to ganger ekstra- ■ hert med 100 og 50 ml henholdsvis av etylacetat. De organiske faser ble slått sammen, vasket med 50 ml av. en mettet løs-ning natriumklorid, tørket over natriumsulfat og bragt til tørrhet i vakuum ved en temperatur ikke over 4 0°C. Man fikk et krystalinsk fast stoff som.ble filtrert, vasket med 10 ml dietyleter og tørket i luft. Utbytte: 10 g av titte.lforbind-elsen. Smp.: 148-50°C. TLC analyse ga en enhetlig flekk
med Rf verdi = 0,53 [Kiselgelplate F 254; elueringssystem: etylacetat/aceton/iseddik/vann = 5/2/1/1 (v/v); påvisning: joddamper]. Forbindelsens struktur ble bekreftet gjennom N.M.R. analyse i CDC13ved bruk av TMS (trimetylklorsilan) som intern standard.

10,0 gram N-pivaloyl-1,3-tiazolidin-4-yl-karboksylsyre ble oppslemmet i 50 ml aceton sammen med 4,65 g trietylamin, den resulterende oppslemming avkjølt til -15°C og deretter dråpe-vis tilsatt en løsning av 5,5 g pivaloylklorid i 25 ml aceton. 2 dråper 4-metylmorfolin ble også tilsatt. Etter 20 minutters henstand ved -15°C ble en forut fremstillet løs-ning av 15,5 g 7-amino-3-[(5-metyl-l,3,4-tiadiazol-2-yl-tio)-metyl]-ZN.<3->cephem-4-karboksylsyre i 10 ml vann og 20 ml aceton raskt helt i reaksjonskolben, hvorved temperaturen steg til 0°C og pH ble bragt til 8,5 ved hjelp av trietylamin. Den resulterende blanding fikk stå to timer ved 2°C og i løpet av 2 ytterligere timer fikk temperaturen stige til 20°C. Det dannede uløselige stoff ble fjernet ved fil-trering, vasket på filter med 10 ml aceton, de sammenslåtte acetonløsninger ble fortynnet med 100 ml vann og 2 ganger ekstrahert med 100 ml ( 2 x 100 ml) av en 1/1 (v/v) blanding av etylacetat/dietyleter. Den vandige fasen ble bragt til pH 3 ved hjelp av vandig 10% saltsyre, ekstrahert tre ganger med 50 ml (3 x 50 ml) av etylacetat, etylacetatfasen vasket med 50 ml av en vandig mettet løsning av NaCl, tørket over natriumsulfat og inndampet i vakuum ved en temperatur ikke over 40°C. Den erholdte rest ble tatt opp i 50 ml dietyleter hvorved man fikk et krystallinsk fast stoff som ble filtrert, vasket med 20 ml dietyleter og tørket i luft. Utbytte: 6,5 g av tittelforbindelsen. Smp.: 158-60°C.
TLC analyse ga en enhetlig flekk med Rf verdi = 0,49 [Kisel gel F 254 plate; elueringssystem: etylacetat/aceton/iseddik/ vann = 5/2/1/1 (v/v); påvisning = joddamper].
Forbindelsens struktur ble bekreftet gjennom N.M.R. analyse i CDC-13ved bruk av TMS (trimetylklorsilan) som intern standard.
