"Fremgangsmåte for fremstilling
av 5- m- tolyloksyuracil"
En rekke 5-substituerte fenoksyuraciler er beskrevet i•kjemilitteraturen. ,5-fenoksyuracil ble først fremstilt av'
T.B. Johnson og H.H. Guest som beskrevet i Am. Chem. J., 42,.
271 (1909). Den samme forbindelse ble også fremstilt av B. R. Baker, J.L. Kelley som beskrevet i J. Med. Chem., Y3, 461
(1970) og ble undersøkt som en inhibitor for.FUDR-fosforylase
og funnet å ha dårlig aktivitet. De samme forfattere beskrev
i en senere publikasjon i J. Med. Chem., 14, 812 (1971) en undersøkelse av den samme forbindelse som en inhibitor for tymidin-fosforylase, og den ble funnet å ha dårlig aktivitet.
En rekke 5-substituerte fenoksyuraciler omfattende usubstituert fenoksy-, m-fluorfenoksy-, o-tolyloksy- og p-tolyloksyuracil ble syntetisert for eancer-kjemoterapeutisk undersøkelse av Chun-Nien Chang, Shih-An Yang, Tien-Tu Wang og Yen Hu i Yao-Hsueh Pao 10, 600>1963 (ActaPharmaceutica Sinica 10, 600
med sammendrag i CA 60: 14504b). Ved innledende biologisk under-søkelse fant disse forfattere at m-fluorfenoksyuracil hemmet veksten av sarcoma 180 i mus.
En rekke 5-substituerte fenoksytiouraciler er i US-patent 2.697.708 og britisk patent 706.253 angitt å være aktive mot
.Neurovaccinia-infeksjon. R.L. Thompson, M. Price, S.A. Minton, JR., E.A. Falco og G.H. Hitchings i J. Immunol., 67, 483 (1951)
4 beskriver beskyttelse av mus mot Vaccinia-virus ved administrering av 5-substituerte fenoksytiouraciler. E.A. Falco, P.B. Russeli og G.H. Hitchings i J. Am. Chem. Soc, 73^4466 (1951) beskriver fremstilling av noen 5-substituerte fenoksy-2-tiouraciler, innbefattet fremstilling av 5-m-tolyloksy-2-tiouracil.
I US-patent 3.154.551 beskrives fremstilling av 2,4-di-hydroksy-5-arylmerkaptppyrimidiner via den generelle metode for kondensering av en tiofenol med 5-bromuracil. Britisk patent 951.431 beskriver lignende forbindelser, men ingen av disse patenter beskriver 5-(m-metyltiofenoksy)uracil eller 5-(p-metyltio-fenoksy)uracil. B. Roth og G.H. Hitchings i J. Org. Chem., 26, 2771 (1961) angir at forfatterne ikke var i stand til å frem-stille 5-(p-metylfenoksy)-uracil ved å omsette 5-bromuracil med p-kresol.
Britisk patent 971.307, som angår 5-anilinopyrimidiner, beskriver de fysikalske egenskaper for 5-(m-metylanilino)uracil som ble undersøkt som et antibakterielt og antimetabolisk middel. Tilnærmet det samme er beskrevet i US-patent 3.238.308. En senere publikasjon av F.R. Geins, A. Perrotta og G. H. Hitchings i-J. Med. Chem., 9_, 108 (1966) angår omtrentlig det samme. M. R. Atkinson, G. Shaw og G. Sugowaz i J. Chem. Soc., 3207 (1957) beskriver fremstilling av en rekke 5-substituerte fenylsulfonyl-uracil-forbindelser som en del av en antimetabolitt-undersøkelse.
T. Zsolnai i Biochem. Pharmacol. ,-11., 995 (1962) beskriver fremstilling av p-klor- og p-metoksy-fenylhydrazinouracil for undersøkelse som fungistatiske midler.
I US-patent 3.922.345 som angår pyrimidoner med bronkodilator- og antiulcer-aktivitet, kreves beskyttelse for 5-(m-metylfenoksy)-2-(1H)-pyrimidinon og fremgangsmåte for be-handling av peptisk ulcer ved at det til en pasient som lider av dette, administreres med en daglig dose av den.ovennevnte forbindelse i en mengde på ca. 0,60 til 21 mg pr. kg kroppsvekt.
I patentet beskrives også anvendelse av 4-hydroksy-5-(m-metyl-fenoksy)-pyrimidin som bronkodilator.
Vi har nu funnet at i rotter,.aper og mennesker omdannes 5-(m-metylfenoksy)-2-(1H)-pyrimidinon raskt og effektivt metabolisk til 5-m-tolyloksyuracil. Litteraturen angir at 5-substituerte uraciler så som 5-fluoruracil omdannes metabolisk til 5,6-dihydro-uraciler som gjennomgår ringåpning- med påfølgende tap av det C^-pyrimidin-karbonatornet som karbondioksyd. Vi har nu funnet at 5-m-tolyloksyuracil uventet ikke gjennomgår denne metaboliske nedbrytning i rotter og aper. Videre har vi funnet at 5^m-tolyloksyuracil har'en meget lengere plasma-halveringstid i mennesker enn hva 5-(m-metylfenoksy)-2-(1H)-pyrimidinon har.
Ifølge oppfinnelsen tilveiebringes en fremgangsmåte for fremstilling av forbindelsen med formelen:
Forbindelsen kan fremstilles i tilnærmet ren form og kan f.eks. anvendes i enhetsdoseform i en mengde fra ca. 50 - 600 mg i et farmasøytisk godtagbart fortynningsmiddel. Forbindelsen kan også eventuelt anvendes som et farmasøytisk godtagbart salt. For be-handling av peptisk ulcer kan forbindelsen 5-m-tolyloksyuracil, som også er kjent som 5-m-metylfenoksyuracil, administreres til
en pasient i en effektiv mengde.
Den nye forbindelse kan eksistere i enten keto- eller enol-form, og det skal forstås at oppfinnelsen omfatter fremstilling av begge former. Oppfinnelsen omfatter også fremstilling av farmasøytisk godtagbare, ugiftige metallsalter av forbindelsen,
så som natrium- og kaliumsaltene og ammonium- og substituerte ammoniumsa1ter.
Utgangsmaterialene som anvendes for fremstilling av 5-m-tolyloksyuracil, er kommersielt tilgjengelige materialer så som m-kresol og en ester av en ot-halogen-eddiksyre så som brom- eller klor-metyl- eller -ety1-acetat. m-kresolen kondenseres med en passende base så som et alkalimetallhydroksyd eller -alkoksyd, f.eks. natrium- eller kaliumhydroksyd eller natriummetoksyd eller -etoksyd i et passende oppløsningsmiddel så som en lavere alkohol, f.eks. metanol eller etanol. Det resulterende m-kresolat omsettes med en a-halogen-eddiksyreester så som klor- eller brom-, -metyl-eller -etyl-acetat for å danne en m-tolyloksy-eddiksyreester så som metyl- eller etyl-m-tolyloksyacetat. Esteren kan renses ved destillasjon eller den kan anvendes uten videre rensning.
Metyl- eller etyl-m-tolyloksyacetatet i et egnet opp-løsningsmiddel så som dietyleter, benzen, toluen eller tetrahydro-furan i nærvær av en sterk base så som kalium-tert.-butoksyd eller natriumhydrid kondenseres med en maursyreester så som metyl- eller etylformiat for å gi alkalimetallsaltet av en a-formyl-m-tolyloksy-eddiksyreester så som natrium- eller kalium-a-formyl-m-tolyloksy-metyl- eller -etyl-acetat. Saltet kan isoleres ved filtrering eller kan anvendes uten ytterligere rensning.
Alkalimetallsaltet av en a-formyl-m-tolyloksy-eddiksyreester i et egnet oppløsningsmiddel så som en lavtkokende alkohol, kondenseres med tiourinstoff i nærvær av varierende mengder base, så som et alkalimetallhydroksyd eller -alkoksyd. Det resulterende produkt, alkalimetallsaltet av 5-(m-tolyloksy)-2-tiouracil, behandles med en minerålsyre så som.saltsyre eller svovelsyre for å utfelle 5-(m-tolyloksy)-2-tiouracil. Dette materiale kan renses ved utfelning fra fortynnet natriumhydroksyd med en minerålsyre så som saltsyre eller eddiksyre eller ved krystal-lisering fra et egnet oppløsningsmiddel.
5-(m-tolyloksy)-2-tiouracil behandles i henhold til oppfinnelsen med én ekvivalent mengde eller et overskudd av. en a-halogen-eddiksyre så som kloreddiksyre, og oppvarmes i en vandig minerålsyre. Det resulterende hydrolyseprodukt 5-(m-tolyloksy)uracil isoleres ved filtrering og. kan renses ved oppslemning med et oppløsningsmiddel så som vann, aceton eller en lavere alkohol så som metanol eller etanol.
Betegnelsen peptisk ulcer har historisk vært anvendt
for å beskrive en sykdom som kjennetegnes ved sårdannelse i den øvre del av mave-tarmkanalen, og omfatter den sykdom som kjennetegnes ved sårdannelse på selve mavesekken, vanligvis kalt mave--sår, såvel som den sykdom som kjennetegnes ved sårdannelse på tolvfingertarmen, vanligvis kalt duodenal ulcer. For fagfolk som kjenner, til den gastrointestinale litteratur, er det nu kjent at disse to sykdommer er forskjellige. Allikevel anvendes her betegnelsen peptisk ulcer i historisk betydning.
Pasienter som lider av mavesår, lider som gruppe ikke av for kraftig utskillelse av saltsyre i maven. Sykdommens årsak antas generelt å være en ubalanse mellom de "aggressive" krefter i maven så som syre, pepsin og gallesure salter, og de "defensive" krefter i maven som består av mavens slimlag og en slimhinne-barriere i maven som hindrer skadelig diffusjon av saltsyre fra
4 mavesekken tilbake gjennom mavens slimhinne.
Legemidlet "Carbenoxolone-Na" er kjent for å beskytte dyr mot eksperimentelt fremkalt sårdannelse i maven, og er hos menneske-pasienter som lider av mavesår, funnet ved tallrike kliniske forsøk å være et effektivt middel mot mavesår. 5-m-tolyloksyuracil, i likhet med "Carbenoxolone-Na'! , beskytter rotter mot sårdannelse. Den nye forbindelse 5-m-tolyloksyuracil kan derfor ventes å være klinisk effektiv som et middel mot mavesår hos mennesker.
US-patent 3.922.345 beskriver en fremgangsmåte for be-, handling av peptisk ulcer, omfattende at det til en pasient som lider av slik ulcer, administreres daglig en dose på ca. 0,60
til 21 mg pr., kg kroppsvekt av 5- (m-metylfenoksy)-2- (1H)-. pyrimidinon. Vi har funnet at i rotter, aper og mennesker omdannes 5-(m-metylfenoksy)-2-(1H)-pyrimidinon raskt og effektivt metabolisk til 5-m-tolyloksyuracil. Videre har vi funnet, i motsetning til hva som tidligere er angitt i litteraturen, at andre mulige metaboliske modifikasjoner av 5-(m-metylfenoksy)-2-(1H)-pyrimidinon ikke forekommer i noen påviselig grad. Vi har ikke kunnet påvise stoffskifteprodukter som skyldes 1) fenoksyring-hydroksylering, 2) oksydasjon av m-metylgruppen eller 3) eter-spaltning.
I motsetning til hva som er angitt i litteraturen, har
vi nu funnet at 5-m-tolyloksyuracil har uvanlig metabolisk stabilitet. I litteraturen er det angitt at en 5-substituert uracil så som 5-fluoruraci1 gjennomgår en reduksjon av 5,6-dobbeltbindingen for å gi et 5,6-dihydrouracil som derefter gjennomgår ringåpning til et N-acetamido-3-alanin som derefter taper C-2-pyrimidin-karbonatomet som karbondioksyd, slik som illustrert i det følgende reaksjonsskjerna.
Ved undersøkelser på rotter og aper under, anvendelse av 5-m-metyl-fenoksy-2-(1H)-pyrimidinon inneholdende et radioaktivt merket karbonatom ved C2i pyrimidinringen, har vi vist at den metaboliske ustabilitet som 5-fluoruracil har, ikke forekommer med 5-m-tolyloksyuracil, eftérsom det ikke blir iakttatt noen nevneverdige mengder av radioaktivt C02fra endogent dannet 5-m-tolyloksyuracil.
Vi har også oppdaget ved undersøkelser på mennesker at 5-m-tolyloksyuracil har en meget lengere plasma-halveringstid og når høyere toppkonsentrasjoner enn hva tilfellet er med 5-(m-metylfenoksy)-2-(1H)-pyrimidinon. Hos mennesker, ved anvendelse av enkeltdoser på 400-700 mg 5-(m-metylfenoksy)-2-(1H)-pyrimidinon, har dette middel avtatt under påvisbar mengde i løpet av 2 timer efter administrering. I motsetning til dette nådde endogent dannet 5-m-tolyloksyuracil toppkonsentrasjoner høyere enn for utgangsmidlet 4 timer'efter administrering, og halveringstiden varierte fra 12,6 til 15,5 timer.
Ved våre undersøkelser i dyr og mennesker har vi aldri vært i stand til å isolere 5-m-tolyloksyuracil i tilnærmet ren form. Vi har derimot oppnådd prøver av urin og blodplasma fra dyr og mennesker, bestående av en rekke naturlige komponenter, hvorav en er identifisert som 5-m-tolyloksyuracil ved sammenligning av de fysikalske egenskaper med materialet fremstilt i tilnærmet ren form ved fremgangsmåten ifølge oppfinnelsen. I prøver av plasma fra mennesker behandlet med 5-m-tolyloksy-2(1H)-pyrimidinon har vi bare påvist toppkonsentrasjoner av 5-m-tolyloksyuracil ved den meget lave konsentrasjon på 40 yg/ml. Når det her er tale om "tilnærmet ren form", skal forstås en renhet på 95% eller mer.
For å undersøke antiulcer-aktiviteten for 5-m-tolyloksyuracil, ble stressfremkalt mavesår frembragt ved immobilisering av ikke-fastende hunhrotter liggende på ryggen og anbringelse av dem i et kjøleskap ved 12°C i 3 timer. Forsøksmidlene ble administrert 3 timer før stressperioden i kjøleskapet ble igangsatt. Ved slutten av stressperioden ble rottene avlivet, mavesekkene ble pumpet opp med 5 ml vann og tatt ut. De isolerte maver ble anbragt i en 4% vandig oppløsning av formaldehyd for å gjøre serosal-siden av den opp-pumpede mave stiv.. Claims (3)
1. Fremgangsmåte for fremstilling av 5-m-tolyloksyuracil, karakterisert ved at 5-(m-tolyloksy)-2-tiouråcil behandles med en a-halogen-eddiksyre, og produktet hydrolyseres i en vandig minerålsyre.
2. Fremgangsmåte som angitt i krav 1, karakterisert ved at det som a-halogen-eddiksyre anvendes kloreddiksyre.
3. Fremgangsmåte som angitt i krav 1 eller 2, karakterisert ved at det som vandig minerålsyre anvendes saltsyre.
