Description
Fremgangsmåte ved fremstilling
av cefalosporinderivater
Foreliggende oppfinnelse angår en fremgangsmåte ved fremstilling av nye 7-(halogenmethylaryl)-acetamidocefalosporinderivater som er nyttige som antibiotiske midler.
I nederlandsk patentansøkning nr. 6.916.151 beskrives forbindelser med den følgende generelle formel hvor substituentene betegnet med R og R° eventuelt kan være substituert med en halogen-' alkylgruppe:
hvor R o er eventuelt substituert thienyl , diazolyl, triazolyl, tetrazolyl, thiazolyl, thiadiazolyl, thiatriazoly1, oxazolyl, oxa - diazolyl, pyridyl, diazinyl, benzothiazolyl, benzimidazolyl, benzoxazolyl eller benzotriazolyl; RJ er en eventuelt substituert diazolyl, triazolyl, tetrazolyl, thiazolyl, thiadiazolyl, thiatria-zolyl, oxazolyl, oxadiazolyl, benzimidazolyl, benzoxazolyl, tria-zolopyridyl, diazinyl eller purinyl; R er hydrogen eller en alkoholrest ; og Q er en binding eller svovel. . Alle forbindelsene i ovenstående nederlandske patentansøkning er substituert i 3-stillingen på cefalosporinringen med en methylen-thiosubstituent. Denne ansøkning beskriver ikke eller antyder ikke forbindelsene som fremstilles ifølge foreliggende oppfinnelse.
Forbindelser med den følgende generelle formel I er nyttige som antibiotiske midler:
hvor Aryl er fenyl eller 2-thienyl; X er klor, brom eller fluor;
Y er hydrogen, klor, brom, rettkjedet eller forgrenet lavere alkyl med 1-4 carbonatomer, lavere alkoxy med 1-4 carbonatomer eller -CI-^X hvor X er brom, klor eller fluor, med det forbehold at når Aryl er 2-thienyl, er Y hydrogen; Z er en binding, oxygen eller svovel med det forbehold at når Aryl er 2-thienyl, er Z en binding; W er hydrogen, meth3'l , amino, hydroxy, carboxy eller sulfo med det forbehold at når Z er oxygen eller svovel, er W ikke hydroxy; R er hydrogen eller et farmasøytisk godtagbare ikke-giftig kation eller en anionladning-, og R<1>er hydrogen eller acetoxy.
De ikke-giftige syreaddisjonssalter av forbindelsene hvor
W er amino, som mineralsyreddisjonssalter, f.eks. hydrogenklorid, hydrogenbromid, hydrogenjodid, sulfat, sulfamat og fosfat, og organiske syreaddisjonssalter, f.eks. maleat, acetat, eit rat, oxalat, succinat, benzoat, tartrat, fumarat, malat, mandelat og ascorbat , er også innbefattet i f remgarigsmåteforbindelsene ifølge oppfinnelsen.
Også innen rammen av oppfinnelsen er fremstillingen^av ikke-giftige farmasøytisk godtagbare salter av forbindelsene med formel I hvor W er carboxy eller sulfo og forbindelser hvor R er hydrogen. Illustrerende farmasøytisk godtagbare salter av disse syrederivater er primære, sekundære eller tertiære aminer, f.eks. cyclohexylamin, ethylamin og pyridin.
De farmasøytisk godtagbare kationer som kan være tilstede som substituenten R i forbindelsene med formel I innbefatter alkalimetallioner, f.eks. natriumion, kaliumion, calciumion såvel som ammonium, og organiske aminkationer, f.eks. laverealkylammon-iumgrupper som triethylammonium, og N-ethylpiperidin.
Eksempler på rettkjedede eller forgrenede la vere-a Ikyl •-grupper som Y kan være i forbindelsene med formel I, er methyl,
. ethyl, n-propyl, isopropyl, n-butyl og t-butyl.
Eksempler på lavere-alkoxygrupper som Y kan være i forbindelse med formel I, er methoxy, ethoxy, propoxy og butoxy.
Når gruppen Aryl i forbindelsene med formel I er fenyl, kan hver av substituentene -CH^X og Y individuelt være bundet til en hvilken som helst av stillingene 2 til 6 på fenylringen.. De fore-trukne stillinger for gruppen - CU^- X er ortho- og pa ra-st Ulingene på fenylgruppen.
Når arylgruppen i forbindelsene med formel I er thienyl, er
den bundet til gruppen
i 2-stillingen av thienylgruppen.
Substituenten -CH2-X er bundet i 4~eller 5-stillingen på thienyl- . gruppen.
Det fremgår av foregående beskrivelse at forbindelsene som fremstilles ifølge oppfinnelsen, er halogenmethylerte fenylsubsti-tuerte cefalosporinderivater med den generelle formell
eller er halogenmethylerte thienylsubstituerte cefalosporinderivater med formelen:
I ovenstående formler II og III er de forskjellige substituenter Y, CH2X, Z, W, R og R1 som ovenfor angitt.
Forbindelsene med formel I fremstilles ved å koble 7-aminocefalosporansyre eller et derivat derav med formelen: hvor R er hydrogen eller et farmasøytisk godtagbart kation, og R^er som ovenfor angitt,' med en syre med formelen: eller en funksjonell ekvivalent derav, hvor Aryl, Y, Grl^ K, Z og W er som ovenfor angitt. Når substituenten W i formel V er en amino-gruppe, anvendes passende blokkerende grupper, f.eks. t-butoxycarbonyl eller carbobenzyloxy, for å beskytte aminofunksjonen. Slike blokkerende grupper fjernes efter koblingsreaksjonen ved kjente fremgangsmåter, f.eks. som beskrevet i US patent nr. 3.657-232.
Funksjonelle ekvivalenter av syrene med formel V innbefatter syrehalogenidene f.eks. syrekloridet; syreanhydrider innbefattende blandede anhydrider med f.eks. alkylfosforsyrer; lavere alifatiske monoestere av carbonsyre; eller alkyl- eller a ryi-sulfonsyrer. Dessuten kan syreazidet eller en aktiv ester eller thioester, f.eks. med p-nitrofenol, 2,4-dinitrofenol eller thioeddiksyre, anvendes, eller den frie syre med formel V kan kobles med 7-amino-cefalosporansyrederivatet med formel IV efter først å ha omsatt
syren med N,N<1->dimethylklorforiniumklorid eller ved anvendelse av et carbodiimidreagens, f.eks. N,N'-diisopropylcarbodiimid, N,N'-
dicyclohexylca rbodiimid eller N-cyclohexyl-N ' - (2-morfolinoethy1) •-carbodiimid.
Koblingsreaksjonen utføres i alminnelighet i nærvær av et oppløsningsmiddel. Passende oppløsningsmidler innbefatter ethylacetat, aceton, dioxan, acetonitril, kloroform, ethylenklorid, tetrahydrofuran og dimethylformamid. Som hydrofile oppløsnings-midler anvendes blandinger av disse oppløsningsmidler med vann, hvilket også er egnet ved ovenstående reaksjon. Koblingsreaksjonen utføres i alminnelighet i nærvær av en base, f.eks. et alkalisk bicarbonat. Reaksjonstemperaturen kan variere fra -10° til 100°C, og reaksjonstiden kan variere fra ca. 1/2 time til 10 timer. Cefalosporinproduktene isoleres ved konvensjonelle metoder.
Forbindelsene med formel I kan også fremstilles ved å forene et modifisert polystyren inneholdende nitrofenol- eller hydroxy-succinimid-grupper med en syre med formel V og blande den således dannede aktiverte syre med en forbindelse med formel IV ved fremgangsmåten beskrevet i kanadisk patent nr. 892.580, ved å anvende en forbindelse med formel IV istedenfor penicillansyrederivatene beskrevet i patentet.
Saltformene av forbindelsene med formel I hvor R.er et farma-søytisk godtagbarf kat ion, fremstilles på kjent vis og kan dannes in situ eller ved å omsette den tilsvarende syre med en base, f.eks. natriumbicarbonat eller triethylamin.
De individuelle optiske isomerer av forbindelsene med formel I hvor W er methyl, amino, hydroxy, carboxy eller sulfo, omfattes også av fremgangsmåteforbindelsene ifølge oppfinnelsen.
Forbindelsene med formel IV, dvs. 7-aminocefalosporansyre og 7-arainodesacetox<y>cefalosporansyre og salter derav, er kommer-sielt tilgjengelige eller kan fåes fra cefalosporin c ved kjente metoder.
De halogenmethylerte aromatiske syrer med formel V fremstilles ved direkte halogenmethylering av syren med formelen:
hvor Aryl, Y, Z og W er som ovenfor angitt. Når substituenten W
i forbindelsene med formel VI er amino, beskyttes aminogruppen med
en passende blokkerende gruppe, f.eks, en acylgruppe som acetyl eller trifluoracetyl, før halogenmethyleringsreaksjonen. Efter av-slutning av halogenmethyleringsreaksjonen kan den blokkerende gruppe fjernes ved syrehydrolyse ved kjente metoder.
De halogenmethylerte derivater av forbindelsene med formel VI fåes ved flere fremgangsmåter. Eksempelvis ved omsetning av en forbindelse med formel VI med paraformaldehyd i nærvær av en Lewis - syre som ZnCl,-,, AlCl^jSnCl^eller CISO^H i et oppløsningsmiddel som petrolether, kloroform, carbontetraklorid eller benzen, ved en temperatur i området fra -10° til 100°C i løpet av hvilken tid hydrogenkloridgass eller hydrogenbromidgass bobles inn i reaksjonsblandingen, fåes forbindelser med formel V.
Omsetningen av en syre med formel VI med 34 -.38%-iQformalin i konsentrert saltsyre ved temperaturer fra -10° til 100°C, i løpet av hvilken tid hydrogenkloridgass eller hydrogenbromidgass bobles gjennom reaksjonsblandingen, gir også forbindelser med formel V. "''
Dessuten fåes ved omsetning av en syre med formel VI med trioxan i eddiksyre eller fosforsyre ved temperaturer fra -10° til 100°C, i løpet av hvilken tid hydrogenbromid- eller hydrogenkloridgass bobles gjennom reaksjonsblandingen, forbindelser med formel V. Eller omsetningen av en syre med formel VI i nærvær av en Lewis-syre som de ovenfor beskrevne, med klormethylether ved temperaturer
fra -10° til 100°C, eller ved omsetning av syren i eddiksyre eller konsentrert svovelsyre med diklormethylether i nærvær av zinkklorid, vil gi forbindelser med formel V.
Forbindelsene med formel V hvor W er carboxy, og Aryl er fenyl, fåes fortrinnsvis ved å behandle en tilsvarende diethylester med formel VI med 40%-ig formalin i nærvær av vannfritt zinkklorid i benzen ved ca. 50°C i løpet av hvilken tid hydrogenklorid- eller hydrogenbromidgass bobles inn i reaks jonsblandingen, fulgt av syrehydrolyse.
Forbindelser med formel V hvor W er sulfo, kan fåes ved halogenmethyleringsreaksjonene beskrevet ovenfor under anvendelse av en syre med formel VI hvor W er sulfo eller methylesteren derav i hvilket sistnevnte tilfelle den dannede klormethylerte forbindelse overføres til den frie sulfosyre ved syrehydrolyse.
I halogenmethyleringen av forbindelser med formel VI hvor W er hydroxy, kan det være .fordelaktig å beskytte hydroxygruppen før halogenmethyleringen som beskrevet av B. Reichert et al., Pharmazie 5, 10 (1950).
Forbindelsene med formel VI er lett tilgjengelige eller kan fremstilles ved alminnelig kjente metoder.
De nye forbindelser som fremstilles ifølge oppfinnelsen, er nyttige som antibiotiske midler som vist ved deres aktivitet mot grampositive og gramnegative bakterier in vitro og in vivo, og fungi. Fremgangsmåteforbindelsene er særlig nyttige ved at de har en lengre varighet av aktivitet enn mange velkjente cefalosporinforbindelser. Dessuten har, som det er vel kjent, cefalosporinforbindelser i alminnelighet en ubehagelig smak. Forbindelsene som fremstilles ifølge oppfinnelsen, har en markert forbedret smak.
Fremgangsmåteforbindelsene kan administreres. på lignende måte som mange velkjente cefalosporinforbindelser, f.eks. cefalexin, cefalothin eller cefaloglycin. De kan administreres alene eller i form av farmasøytiske preparater, enten oralt, parenteralt eller lokalt til varmblodige dyr, dvs. fugler eller pattedyr, f.eks.
dyr av katteslekten, dyr av hundeslekten, klovdyr eller hovdyr,
og mennesker. For oral administrasjon kan forbindelsene administreres i form av tabletter, kapsler eller piller eller i form av eliksirer eller suspensjoner. For parenteral administrasjon anvendes de best i form av en steril, vandig oppløsning som kan inneholde andre oppløste stoffer, f. eks. nok natriumklorid eller glucose til å gjøre oppløsningen isotonisk. For lokal administrasjon kan forbindelsene inkorporeres i kremer eller salver.
Illustrerende eksempler på bakterier mot hvilke fremgangs-måtef orbindelsene er aktive er "Staphylococcus aureus, Salmonella schottmuelleri, Klebsiella pneumoniae, Diplococcus pneumonia og Streptococcus pyogenes.
Forbindelsene som fremstilles ifølge oppfinnelsen- med den generelle formel II, er halogenmethylerte fenylacetamido-, halogenmethylerte fenoxyacetamido- og halogenmethylerte fenylthioacet-amido-cefalosporinderivater hvori fenylringen kan være substituert med rettkjedet eller forgrenet alkyl med 1-4 carbonatomer,
alkoxy med 1-4 carbonatomer, klor, brom eller -CH2X hvor X er klor, brom eller fluor. Når Y er hydrogen i forbindelsene med formel II, kan fenylringen også være substituert med hydroxy eller trifluormethyl.
Forbindelsene som fremstilles ifølge oppfinnelsen med formel III, er halogenmethylerte 2-thienylacetamido-cefalosporinderivater. Forbindelsene med formel III kan også være substituert på thienylringen med substituentene betegnet med Y i forbindelsene med formel II.
En foret rukken gruppe av forbindelser som fremstilles ifølge oppfinnelsen, har formelen:
hvor W"'" er hydrogen, methyl, amino eller hydroxy, og X, Y, Z, R og R"'" er som ovenfor angitt .
En mere foretrukken gruppe av forbindelser som fremstilles ifølge oppfinnelsen, er de med den generelle formel VII hvor Y er hydrogen eller alkoxy med 1-4 carbonatomer, og X, W"*" , R og R"*" er som ovenfor angitt.
En enda mere foretrukken gruppe av forbindelser som fremstilles ifølge oppfinnelsen, er de med formel VII hvor Y er hydrogen eller alkoxy med 1-4 carbonatomer; Z er en binding; og X, W1, R og R ' er som ovenfor angitt.
En annen foretrukken gruppe av forbindelser som fremstilles ifølge oppfinnelsen, har formelen:
hvor W<2>er hydrogen eller methyl; og X, R og R1 er som ovenfor angitt .
De følgende eksempler illustrerer oppfinnelsen ytterligere.
Eksempel 1
p- klormethylfenylacetylklorid
(A) Ved en temperatur på fra -10° til 0°C bobles hydrogenklorid-
gass gjennom en omrørt blanding av 102 g fenyleddiksyre, 67,5 g paraformaldehyd. og 67,5 Q zinkklorid i 1000 ml petrolether i 1 time. Omrøringen fortsettes i ca. 1 time ved værelsetemperatur, hvorefter blandingen kokes under tilbakeløp i ca. 2 timer i løpet av hvilken tid hydrogenkloridgass bobles.inn i blandingen. Reaksjonsblandingen tilsettes 1000 ml methylenklorid og lOOO ml vann. Den organiske fase fraskilles, og vannfasen ekstraheres to ganger med methylenklorid. De forenede organiske faser forenes og ekstraheres fire ganger med en mettet natriumbicarbonatoppløsning. Den organiske nøytrale fase tørres over vannfritt natriumsulfat, filtreres og oppløsningsmidlet fjernes under vakuum, hvilket gir et nøytralt biprodukt som er videre identifisert i eksempel 4 nedenfor. Den basiske vannfase fraskilles og syres med kold konsentrert saltsyre til pH 2-3, ekstraheres så tre ganger med methylenklorid.. Methylenkloridfraksjonen tørres over vannfritt natriumsulfat, filtreres og oppløsningsmidlet fordampes. Det gjenværende oljeaktige sure produkt kromatograferes på silicagel under•anvendelse av benzen og benzen-aceton som elueringsmiddel, hvilket gir p-klormethylfenyleddiksyre som omkrystalliseres fra varm kloroform,
smp. 147 - 149°C. (B) En blanding av 1 g p-klormethylfenyleddiksyre og 6 ml thionylklorid omrøres ved værelsetemperatur i 25 timer, hvorefter overskudd av thionylklorid fjernes under vakuum, hvilket gir p-klormethylf enylacetylklorid, smp. 35 - 50°C.
Eksempel 2
3-[(acetyloxy)-methyl]~7-[[2-[4-(klormethyl)-fenyl]-acetyl]-amino]- 8 - oxo - 5 - thia - l - azabicyclo -" 4 - 2 . 0 j - oet - 2 - en - 2 - carboxy 1 syre
En blanding av 1 g 7-aminocefalosporansyre og 1 g p-klormethylf enylacetylklor id i 45 ml ethylacetat kokes under tilbakeløp i ca. 2 timer, hvorefter oppløsningsmidlet fjernes under vakuum,
hvilket gir et gulbruntamorft produkt som kromatograferes på silicagel under anvendelse av benzen-aceton som elueringsmiddel,
og man får 3~[(acetyloxy)-methyl]-7-[[2-[4-(klormethyl)-fenyl]-acetyl]-amino]-8-oxo-5-thia-1-azabicyclo-[4•2.0]-oet-2-en-2~carboxylsyre, smp. 164 - l65°C (spaltn.).
Eksempel 3
7"[ [2-[ 4-(klormethyl) -f enyl ] -acetyl] -amino]-3 -methyl-8-oxo-5-thia - l- azabicyclo-[ 4 . 2 .0"j-oct -2-en-2-carboxylsyre
Ved fremgangsmåten i eksempel 2, men ved å anvende 1 g 7- aminodesacetoxycefalosporansyre istedenfor 7-aminocefalosporansyre, fåes 7-[[2-[4-(klormethyl)-fenyl]-acetyl]-amino]-3-methyl-8-oxo-5-thia-1-azabicyclo-[4.2.o]-oet-2-en-2-carboxylsyre med smeltepunkt145-146°C.
Eksempel 4
o- hydroxymethylf enyleddiksyrelacton
Det nøytrale biprodukt erholdt i eksempel 1 renses ved sub-limasjon under vakuum (0,05 mm Hg ved 80°C) , hvorved man får o-hydroxymethylfenyleddiksyrelacton med smeltepunkt 82°C.
Eksempel 5
0- brommethylfenylacetylklorid
Til en oppløsning av 5 ml iseddik mettet med hydrogenbromidgass tilsettes ved 0°C en oppløsning av 0,55 g o-hydroxymethyl-fenyleddiksyrelacton i 2 ml iseddik. Blandingen omrøres ved værelsetemperatur i 2 timer og kokes så under tilbakeløp i 1 time i løpet av hvilken tid hydrogenbromidgass bobles inn i blandingen. Overskuddet av lacton og oppløsningsmiddel fjernes under høyvakuum ved værelsetemperatur. Det erholdte oljeaktige residuum tritureres tre ganger med hexan, hvorved man får o-brommethylfenyleddiksyre med smeltepunkt 110°C.
En oppløsning av 0,18 g o-bromrnethy lf enyleddiksy re i overskudd av thionylklorid omrøres ved værelsetemperatur i 18 timer, hvorefter det uomsatte thionylklorid fjernes under høyvakuum, og man får o-brommethylfenylacetylklorid som et oljeaktig residuum.
Eksempel 6
7-[[2-[2-(brommethyl)-fenyl]-acetyl]-amino]-3-methyl-8-oxo-5-thia-1- azabicyclo-[ 4.2.0]-oet-2-en-2-carboxylsyre
En blanding av 0,175 g o-brommethylfenylacetylklorid og 0,175 g 7-aminodesacetoxycefalosporansyre i 40 ml ethyiacetat kokes under tilbakeløp i 50 minutter, hvorefter oppløsningsmidlet fjernes under vakuum, hvorved man får et oljeaktig residuum som kromatograferes på silicagel under anvendelse av benzen og benzen-aceton som elueringsmiddel, hvilket gir 7-[[2-[2-(brommethyl)-fenyl ]-acetyl ]-amino]-3-methyl-8-oxo-5-thia ;-l-azabicyclo-[4.2.0 ]-oet-2-en-2-carboxylsyre med smeltepunkt l45°C (spaltn.).
Eksempel 7 3-[(acetyloxy)-methyl]-7-[[2-[2-(brommethyl)-fenyl]-acetyl]-amino]-8-oxo-5-thia-1-azabicyclo-[4.2,0]-oet-2-en-2-carboxylsyre
Når i fremgangsmåten i eksempel 6 7-aminocefalosporansyre anvendes istedenfor 7~aminodesacetoxycefalosporansyre, fåes 3~
[(acetyloxy)-methyl]-7-[[2-[2-(brommethyl)-fenyl]-acetyl]-amino]-3 - oxo-5-thia-1-azabicyclo-[4.2.0]-oet-2-en-2-carboxylsyre.
Eksempel 8
o- klormethyl- p- methoxymandelsyreklorid
En oppløsning av 1,1 g 2-klormethyl-4-methoxymandelsyre, erholdt ved fremgangsmåten beskrevet av B. Reichert et al., Pharmazie 5, 10 (1950), i 25 ml thionylklorid omrøres ved værelsetemperatur i ca. 16 timer, hvorefter overskuddet av thionylklorid fjernes under høyvakuum, hvorved man får o-klormethyl-p-methoxymandelsyreklorid som en olje.
E ksempel 9
3-[ (acetyloxy)-methyl ]-7-[ [2-[2-(klormethyl) -4-methoxyf enyl.]-2-hydroxyacetyl]-amino ] -8~oxo-5~thxa-1-azabicyclo-[4.2.0]-oet-2-en - 2- ca rboxylsyre
En blanding av 1 g 7-aminocefalosporansyre og 1,2 g 2-klormethyl-4-methoxymandelsyreklorid i 200 ml ethylacetat kokes under tilbakeløp i 50 minutter, hvorefter oppløsningsmidlet fjernes under høyvakuum. Det erholdte produkt kromatograferes på silicagel under anvendelse av benzen-aceton. som elueringsmiddel. Det erholdte produkt tritureres med ether, hvorved man får 3-[(acetyloxy)-methyl ] ~ 7~[ [ 2-[2-(klormethyl) -4-methoxyf enyl ] -2-hydroxyacetyl ] --amino]-8-oxo-5-thia-1-azabicyclo-[4.2.0 ]-oct-2-en-2-carboxylsyre med smeltepunkt ].4o - l42°C (spaltn.).
Eksempel 10
7-[[2-[2-(klormethyl)-4-methoxyfenyl]-2-hydroxyacetyl]-amino]-3-methyl- 8- oxo- 5- thia- 1- azabicyclo-[4.2.o]-oet-2-en-2-carboxylsyre
Når i fremgangsmåten i eksempel 9 197-aminodesacetoxycefalosporansyre anvendes istedenfor 7-aminocefalosporansyre, fåes 7-[[2-[2-(klormethyl)-4-methoxyfenyl]-2-hydroxyacetyl]-amino]-3-methyl-8-oxo-5-thia-1-azabicyclo-[4.2.0]-oet-2-en-2-ca rboxylsyre.
Eksempel
2, 4- bis-( klormethyl)- fenoxyacetylklorid
(A) I en blanding av 18 g fenoxyeddiksyre, 80 ml 36 - 38%-ig formalin og 100 ml konsentrert saltsyre innbobles hydrogenkloridgass i 1 time ved værelsetemperatur, og derpå i 10 minutter ved 75°C. Temperaturen på blandingen holdes ved 75°C i 1,5 timer. Til reaksjonsblandingen tilsettes 1000 ml methylenklorid og 150 ml vann, og blandingen omrøres. Det organiske skikt fraskilles, tørres over vannfritt natriumsulfat, filtreres og inndampes til et oljeaktig residuum under vakuum ved 35°C. Det oljeaktige residuum kromatograferes på silicagel under anvendelse av benzen og benzen-aceton som elueringsmiddel, hvilket gir 2,4-bis-(klormethyl)-fenoxyeddiksyre som omkrystalliseres fra kloroform-hexan. Smeltepunkt 110 - 111°C. (B) En oppløsning av 2 g 2,4-bis-(klormethyl)-fenoxyeddiksyre i 15 ml thionylklorid omrøres ved værelsetemperatur i ca. 16 timer
hvorefter overskudd av thionylklorid fjernes under høyvakuum ved værelsetemperatur og man får 2,4~bis-(klormethyl)-fenoxyacetylklorid som et halvfast stoff.
Eksempel 12
3-[(acetyloxy)-methyl]-7-[[2-[2,4-bis-(klormethyl) —fenoxy]-acetyl ]-amino]-8-oxo-5-thia-1-azabicyclo-[4.2.0]-oct-2-en-2-ca rboxylsyre
En blanding av 0,25 g 2,4-bis-(klormethyl)-fenoxyacetylklorid og 0,25 g 7-aminocefalosporansyre i 150 ml ethylacetat kokes under tilbakeløp i 50 minutter, hvorefter oppløsningsmidlet fjernes under høyvakuum, hvilket gir et oljeaktig produkt som tritureres med ether. Det erholdte produkt kromatograferes på silicagel under anvendelse av benzen og benzen-aceton som elueringsmiddel, hvorved man får 3-[(acetyloxy)-methyl]- 7 ~[[2-[2,4~bis-(klormethyl)-f enoxy ] -acetyl ] -amino ] -8 -oxo-5-thia -1 -azabicyclo-[4 .2 .o] -oet -2-en-2-carboxylsyre. Smeltepunkt 137 - 138°C.
Eksempel 13
5- klormethyl- 2- thienylacetylklorid
2-thiofencarboxylsyre behandles i en oppløsning av kloroform med klormethylether i nærvær av 0,9 - 2,2 ekvivalenter aluminium-klorid, hvilket gir 5-klormethy1-2-thienylcarboxylsyre. Behandling av den erholdte syre med overskudd av thionylklorid ved værelsetemperatur i ca. 16 timer gir syrekloridet som omsettes med diazo-
met han for å få det tilsvarende diazoketon. En methanoloppløsning av diazoketonet bestråles under nitrogen i ca. 1 time med en høy-trykks kvikksølvlampe under anvendelse av et kvartsfilter. Methyl-5-klormethyl-2-thienylacetatet erholdes ved opparbeidelse og kolonnekromatografi på silicagel. Acetatet hydrolyseres ved behandling med en 1:1 blanding av eddiksyre og konsentrert saltsyre ved værelsetemperatur over natten, hvorved man får 5-klormethyl-2-thienyleddiksyre.
Når i fremgangsmåiten i eksempel 1 (B) 5-klormet.hyl--2-thienyl-eddiksyre anvendes istedenfor p-klormethyl-fenyleddiksyre, fåes 5~klormethy1-2-thienylacetylklorid.
Eksempel 14
3-[(acetyloxy)-methyl]-7-[[2-[5-(klormethyl)-2-thienyl]-acetyl]-amino]- 8- oxo- 5- thia- 1- azabicyclo-[ 4. 2. 0]- oet- 2- en- 2- carboxylsyre

Når i fremgangsmåten i eksempel 1 (A) acetylert fenylglycin anvendes istedenfor fenyleddiksyre med tilleggstrinnet syrehydrolyse for å fjerne den aminbeskyttende gruppe, fåes p-klormethylfenylglycin.

3- [(acetyloxy)-methyl]-7~[[2-[4~(klormethyl)-fenyl]-2-aminoacetyl]-amino]-8-oxo-5-thia-1-azabicyclo-[4.2;0]-oet-2-en-2-carboxylsyre
p-klormethylfenylglycin hvor aminogruppen er beskyttet med t-butoxycarbonyl, behandles med isobutylklorformiat i nærvær av triethylamin. Det således erholdte blandede anhydrid omsettes med 7-aminocefalosporansyre-triethylaminsa1t ved 0°C i ca. 4 timer. Det dannede produkt isoleres, og den aminobeskyttende gruppe fjernes ved syrehydrolyse, hvorved man får 3-[(acetyloxy)-methyl]-7-[ [ 2-[4-(klormet hyl) -f enyl ]-2-aminoacetyl ] -amino ]-8-oxo-5-thia -1-a za b icy cio - [ 4 .2 .0 ] -oet -2 -en -2 -ca rboxy lsy re .

o-klor-p-klormethylfenylacetylklorid.

Når i fremgangsmåten i eksempel 2 en ekvivalent mengde a-hydroxy-5-klormethyl-2-thienylacetylklorid eller o-klor-p-klormethylf enylacetylklorid anvendes istedenfor p-klormethylfenylacetylklorid, fåes de følgende produkter: 3-[(acetyloxy)-methyl]- 7 ~[[2-[5-(klormethyl)-2-thienyl]-2-hydroxy-acetyl]-amino]-8-oxo-5-thia-1-azabicyclo-[4.2.0]-oct-2-en-2-carboxylsyre,
3-[ (acetyloxy) -methyl ]-7-[ [ 2-[ 2 -klo r-5 -(klormethyl) -f enyl ] -acetyl]-amino]-8-oxo-5-thia-1-azabicyclo-[4.2.o]-oet-2-en-2-carboxylsyre.

Når i fremgangsmåten i eksempel 11 (A) en ekvivalent mengde av en av syrene angitt nedenfor anvendes istedenfor fenoxyeddiksyre, fåes det tilsvarende klormethylerte derivat angitt nedenfor:

Når i fremgangsmåten i eksempel 11 (B) o-klormethyl-p-iso-propylfenoxyeddiksyre, p-klormethyl-o-methyl-a-methylfenoxyeddik-syre , p-klormethyl-o-klorthioeddiksyre eller p-klormethyl-a-methyl-f enyleddiksy re anvendes istedenfor 2 ,4-bis-(klormethyl) -f enoxy-eddiksyre, fåes det tilsvarende syreklorid.

Når i fremgangsmåten i eksempel 12 en ekvivalent mengde av hver av syreklpridforbindelsene erholdt i eksempel 21 anvendes istedenfor 2,4-bis-(klormethyl)-fenoxyacetylklorid, fåes de følg-ende produkter: 3-[(acetyloxy)-methyl]-7-[[2-[2-klormethy1-4-isopropylfenoxy]-acetyl]-amino]-8-oxo-5-thia-1-azabicyclo-[4.2.0 J-oet-2-en-2 - carboxylsyre,
3-[(acetyloxy)-methyl]-7~[[2-[4-klormethyl-2-methylfenoxy]-acetyl]-amino]-8-oxo-5-thia-1-azabicyclo-[4.2.o]-oct-2-en-2-carboxylsyre, 3-[(acetyloxy)-methyl]- 7 ~[[2-[2-klor-4-klormethylfenoxy]-acetyl]-amino]-8-oxo-5-thia-1-azabicyclo-[4.2.0]-oet-2-en-2-carboxylsyre, 3-[(acetyloxy)-methyl]-7-[[2-[4-klormethylfenoxy]-acetyl]-amino]-8-oxo-5-thia-l-azabicyclo-[4.2.0]-oct-2-en-2-carboxylsyre.
Eksempel 23
Når i fremgangsmåten i eksempel 1 (A) en ekvivalent mengde fenylmalonsyre-diethylester eller a-carboxy-2-thienyleddiksyre-diethylester anvendes istedenfor fenyleddiksyre, fåes p-klormethyl-malonsyre-diethylester og 4~og 5-klormethyl-a-carboxy~2-thienyl-eddiksyre-diethylester. Syrehydrolyse av produktene gir de tilsvarende syrer.
Eksempel 24 3-[(acetyloxy)-methyl]-7-[[2-[4~(klormethyl)-fenyl]-2-carboxy-acetyl]-aminoJ-8-oxo-5-thia-1-azabicyclo-[4-2.o]-oet-2-en-2-carboxy 1 syre 4 g thienylacetyl-nitrofenolpolymer, fremstilt i henhold til fremgangsmåten beskrevet i kanadisk patent nr. 895.580, med 1 mmol pr. g p-klormethylfenylmalonsyre suspenderes i ca. 8 timer i 20 ml tørr methylenkloridoppløsning inneholdende 1 mmol 7-aminocefalosporansyre -t r iethylammoniumsa It , som fremstilles fra 544 mg (1 mmol) 7-aminocefalosporansyre og 0,56 ml (1 mmol) triethylamin ved værelsetemperatur. Efter at bare spor av 7-aminocefalosporansyre er tilbake i oppløsningen, hvilket bestemmes ved tynnskikts-kromatogra.fi på cellulose i 70%-ig vandig propanol , frafiltreres polymeren og vaskes med 3 x 50 ml methylenklorid. De forenede filtrater inndampes , og residuet oppløses i 20 ml destillert vann/. Denne oppløsning syres til pH 2 ved tilsetning av 0,2 N saltsyre og ekstraheres to ganger med ethylacetat. Oppløsningen tørres over natriumsulfat og inndampes ved værelsetemperatur. Det gjenværende faste stoff tørres over natten over fosforpentoxyd under vakuum, hvorved man får 3~[(acetyloxy)-methyl]-7_L[2-[4-(klormethyl )-fenyl]-2-ca rboxyacetyl]-amino]-8-oxo-5-thia-1-azabicyclo-[4•2.o]-oet-2-en-2-carboxylsyre .

3-[(acetyloxy)-methyl]-7-[[2-[5-(klormethyl)-2-thienyl]-2-carboxy-acetyl]-amino]-8-oxo-5-thia-1-azabicyclo-[4.2.0]-oet-2-en-2-carboxylsyre
Når i fremgangsmåten i eksempel 24 en ekvivalent mengde 5-klormethyl-a-carboxy-2-thienyleddiksyre anvendes istedenfor p-klormethylfenylmalonsyre, fåes 3-[(acetyloxy)-methyl]-7-[[2-[5-(klormethyl)-2-thienyl]-2-carboxyacetyl]-amino]-8-oxo-5-thia-l-azabicyclo-[4.2.0]-oet-2-en-2-carboxylsyre.
Claims (1)
- Fremgangsmåte ved fremstilling av en forbindelse med formelen:hvor Aryl er f enyl eller 2-thienyl; X er klor,, brom eller fluor; Y er hydrogen, brom, klor, rettkjedet eller forgrenet laverealkyl med 1-4 carbonatomer, alkoxy med 1-4 carbonatomer eller -CI-^X hvor X er som ovenfor angitt, med det forbehold at når Aryl ■er 2-thienyl, er Y hydrogen; Z er en binding, oxygen eller svovel med det forbehold at når Aryl er 2-thienyl, er Z en bindingj og W er hydrogen, methyl, hydroxy, amino, carboxy eller sulfo med det forbehold at når Z er oxygen eller svovel, er W ikke hydroxy; R er hydrogen eller et farmasøytisk godtagbart ikke-giftig kation' , og R^ er hydrogen eller acetoxy, karakterisert ved at en 7-aminocefalosporansyre eller et derivat derav med formelen:.1 5 hvor R" er som ovenfor angitt, og R er hydrogen eller et farma-søytisk godtagbart ikke-giftig kation, veller et funksjonelt derivat derav, hvor Aryl, Y, X, Z og W er som ovenfor angitt, i nærvær av et oppløsningsmiddel som er ethylacetat, dioxan, acetonitril, kloroform, ethylenklorid, tetrahydrofuran eller dimethylformamid, og eventuelt i nærvær av et alkalisk bicarbonat, idet koblingsreaksjonen utføres ved temperaturer fra -10° til 100°C i fra 30 minutter til 10 timer, med det forbehold at når. W er amino, blokkeres aminogruppen med t-butoxycarbonyl eller carbobenzyloxy før koblingsreaksjonen.
