Description
Foreliggende oppfinnelse angår en fremgangsmåte for fremstilling av formel I forbindelsen [2-((8,9)-diokso-2,6-diazabicyklo[5.2.0]-non-l(7)-en-2-yl)etyl]fosfonsyre, en NMDA antagonist nyttig som en antikonvulsant og neuroprotektant i situasjoner som medfører overskudd av frigjøring av eksitatoriske aminosyrer.
Formel I forbindelsen og en fremgangsmåte for dens fremstilling er beskrevet i US 5,168,103.1 den beskrevne prosess blir 3-aminopropyl karbaminsyrefenylmetylester omsatt med 2-oksoetylfosforsyredialkyl ester og natriumcyanoborhydrid for å oppnå mellomproduktet [3-[[2-(dialkoksyfosfinyl)etyl]amino]propyl]karbaminsyrefenylmetylester (a) i 36% utbytte. Omsetning av (a) med et 3,4-dialkoksy (eller 3,4-diarylalkoksy)-cyklobut-3-en-l,2-dion så som 3,4-dietoksycyklobut-3-en-l,2-dion gir 3-[[2-dialkoksyfosforyl)etyl]-( 2-alkoksy-3,4-diokso-l-cyklobuten-l-yl)-amino]propyl-karbaminsyrefenylmetylester (b) i 89% utbytte. Fjerning av benzyloksykarbonylbeskyttelsesgruppen og cyklisering av (b) til [2-((8,9)-diokso-2,6diazabicyklo[5.2.0]-non-l(7)-en-2-yl)etyl]fosfonsyredialkyl ester (c) oppnåes i 62% utbytte ved behandling av (b) med 10% palladium på karbon og 1,4-cykloheksadien. Behandling av den bicykliske diester (c) med bromtrimetylsilan gir [2-((8,9)-diokso-2,6-diazabicyklo-[5.2.0]-non-l(7)-en-2-yI)etyl]fosfonsyre (I) i 78% utbytte. Det totale utbytte for sekvensen av reaksjoner er 15,5%.
I prosessene ifølge foreliggende oppfinnelse som illustrert i Trinn II i Skjema I nedenfor, blir en 3-aminopropylkarbaminsyredialkylester (IV) i stedet omsatt med et dialkylvinylfosfonat (V) for å oppnå N-[3-(t-butyloksy-karbonylamino)propyl]-2-aminoetyl-fosfonsyredialkylester (VI). Videre forbedringer tilveiebragt ved den foreliggende oppfinnelse i forhold til fremgangsmåten fra US 5,168,103 som er angitt nedenfor, øker det totale utbytte til å gi I i 38,8 %.
Et aspekt ved foreliggende oppfinnelse er en fremgangsmåte for fremstilling av [2-((8,9)-diokso-2,6-diazabicyklo[5.2.0]non-l(7)-en-2-yl)etyl]fosfonsyre som omfatter de følgende trinn: a) omsetning av en vinylfosfonat di-Ci-C6 alkylester med 3-aminopropyl-karbaminsyre-l,l-dimetyletylester, hvilket gir en di-Ci-C6alkylester av N-[3-(t-butyloksykarbonylamino)propyl]-2-aminoetylfosfonsyre; b) omsetning av et 3,4-di-Ci-C4alkoksycyklobut-3-en-l,2-dion med en di-Ci-C6 alkylester av N-[3-(t-butyloksykarbonylamino )propyl]-2-aminoetylfosfonsyre fra trinn a, hvilket gir 3-[[2-(di-Ci-C6alkoksyfosforyl)etyl]-(2-C|-C6alkoksy-3,4-diokso-l-cyklobuten-1 -yl)amino]propyl-karbaminsyre-tert-butyl ester; c) avbeskyttelse av 3-aminogruppen i produktet fra trinn (b); d) cyklisering av produktet fra trinn (c) for å danne det bicykliske mellomprodukt [2-((8,9)-di-okso-2,6-diazabicyklo[5.2.0] non-l(7)-en-2-yl)etyl]fosfonsyre-di-Ci-C6
alkylester; og
e) omdannelse av di-Ci-C6 alkylesteren fra trinn (c) til fosfonsyreproduktet.
Den foretrukne vinylfosfonat-di-Ci-C6 alkylester er dimetylvinylfosfonat eller er
dietylvinylfosfonat, hvorav dimetylvinylfosfonat er mest foretrukket.
Trinn (b) blir fortrinnsvis utført i vannfri metanol eller etanol ved omgivelsestemperaturer (25-30°C). Trinn (b), (c) og (d) blir fortrinnsvis kjørt in situ. Trinn (c) blir fortrinnsvis utført i metylenklorid ved -5 °C til 25 °C. Trinn (e) blir fortrinnsvis utført i metylenklorid eller acetonitril ved ca. 20 °C . Det foretrukne 3,4-di-C|-C4alkoksycyklobut-3-en-l,2-dion i trinn (b) er 3,4.dietoksycyklobut-3-en-l,2-dion.

En forbindelse som er en N-[3-(t-butyloksy-karbonylamino)propyl]-2-aminoetylfosfonsyre-di-Ci-C6 alkyl ester; en forbindelse som er N-[3-(t-butyloksy-karbonylamino)propyl]-2-aminoetyl-fosfonsyre-dimetylester eller N-[3-(t-butyloksy-karbonylamino)propyl]-2-amino-metylfosfonsyre-dietylester; en forbindelse som er en 3-[[2-(di-Ci-C6alkoksy-fosforyl)etyl]-(2-Ci-C4 alkoksy-3,4-dioksocyklobut-1 -enyl)amino]propylkarbaminsyre-tert-butylester: og en forbindelse som er 3-[[2-(dietoksy-fosforyl )etyl]-(2-etoksy-3 ,4-diokso-cyklobut-l-enyl)amino]propylkarbaminsyre-tert-butylester eller er 3-[[2-(dietoksy-fos-foryl)etyl]-(2-metoksy-3,4-dioksocyklobut-l-enyl)amino]propylkarbaminsyre-tert-butyl ester.
Fremgangsmåten ifølge foreliggende oppfinnelse er illustrert i Skjema I nedenfor, hvor "alkyl" er representert ved etylgruppen og "alkoksy" er representert ved etoksygruppen og er videre beskrevet som følger: I Trinn 1 blir 1,3-diaminopropan (II) omsatt med et di-t-butylkarbonat (III), hvilket gir 3-aminopropylkarbaminsyre-l,l-dimetyletylester (IV).
I Trinn 2 blir karbaminsyreesteren IV omsatt med et di-Ci-C6alkylvinylfosfonat (V), hvilket gir N-[3-(t-butyloksy-karbonylamino)propyl]-2-aminoetylfosfonsyre-di-Ci-Cealkyl ester (VI). I det illustrerte tilfellet hvor V er dietylvinylfosfosfonat, gir dette trinnet et 80% utbytte. Imidlertid gir dimetylvinylfosfonat sammenliknbare utbytter og er foretrukket kommersielt fordi det er lettere tilgjengelig i større mengder.
I Trinn 3 blir aminofosfonsyre-di-C|-C6alkylesteren (VI) omsatt med et 3,4-di-Ci-C4alkoksycyklobut-3-en-1.2-dion (VII), hvilket gir en 3-[[2-(di-C i-C6-alkoksyfosforyl)etyll-(2-C i -C4alkoksy-3,4-diokso-1 -cyklobuten-1 -yl)amino]propyl-karbaminsyre-l,l-dimetyletylester (VIII). I det illustrerte tilfellet hvor alkyl angir etyl i begge tilfeller, gir dette trinnet et 96% utbytte. Dette trinnet blir fortrinnsvis utført i vannfri metanol eller etanol ved omgivelsestemperaturer.
I Trinn 4 gir avbeskyttelse av (VIII) i trifluoreddiksyre fulgt av cyklisering med
trietylamin som cykliseringsmiddel [2-((8,9)-diokso-2,6-diazabicyklo[5.2.0]non-l(7)-en-2-yl)etyl]-fosfonsyre-di-C i-C6alkylester (IX). I det illustrerte tilfellet er utbyttet av produktet IX 58% utbytte. Avbeskyttelsen blir fortrinnsvis utført i metylenklorid ved -5°C til 25°C . Cykliseringen blir fortrinnsvis utført i metylenklorid eller acetonitril, idet metylenklorid er spesielt foretrukket, ved ca. 20 °C.
I Trinn 5 blir fosfonsyredietylesteren (IX) behandlet med bromtrimetylsilan, hvilket gir forbindelse I i 87% utbytte. Det totale utbytte for fremstilling av [2-((8,9)-diokso-2,6-diazabicyklo[5.2.0]-non-l(7)-en-2-yl)etyl]fosfonsyre (I) i henhold til fremgangsmåten ifølge foreliggende oppfinnelse er 38,8%, en betydelig forbedring i forhold til 15,5% utbyttet beskrevet i US 5,168,103.
Trinn 2, 3 og 4 blir fortrinnsvis utført in situ.
I reaksjonssekvensen ovenfor betyr betegnelsen "alkyl" en C1-C6 alkyl, lineær eller forgrenet, og betegnelsen "alkoksy" betyr en C1-C6 alkoksygruppe, bortsett fra med hensyn til de skvarate forbindelser VII hvor alkyl betyr metyl, etyl eller butyl. Utbyttene referer til spesifikke tilfeller hvor "alkyl er etyl og "alkoksy" er etoksy.
Fordeler ved fremgangsmåten ifølge foreliggende oppfinnelse er mange. Først blir 1,3-propandiamin mono-beskyttet i utmerket utbytte med di-t-butyldikarbonat, en reaksjon som er mindre farlig enn ved anvendelse av benzylklorformiat, for å fremstille en benzyloksy-karbamatbeskyttelsesgruppe. For det andre blir det monobeskyttete 1,3-propandiamin omsatt med dietylvinylfosfonat i utmerket utbytte uten anvendelse av ytterligere reagenser så som natriumcyanoborhydrid og syre, som hver ikke er uten fare, som var nødvendig ved anvendelse av dietyl-2-oksoetylfosfonat som tidligere beskrevet. For det tredje blir t-butyloksy-karbonylgruppen lett fjernet med syre, mens fjerning av benzyloksykarbonylgruppen krevet anvendelse av palladiumkatalysator og en hydrogenkilde.
Fremgangsmåten ifølge foreliggende oppfinnelse er beskrevet i det følgende skjema 1.1 dette skjema blir "alkyl" representert ved etylgruppen og "alkoksy" er representert ved etoksygruppen. De følgende spesifikke eksempler er inntatt for å illustrere fremgangsmåten ifølge foreliggende oppfinnelse.
Eksempel 1
3-(t-Butyloksykarbonylamino)propanamin
Til en løsning av 1,3-diaminopropan (500 g, 6,75 mol; Aldrich D2, 360-2) i tetrahydrofuran (1,6 1) ved fra -3°C til + 2°C med omrøring ble det tilsatt dråpevis en løsning av di-t-butyldikarbonat (300 g, 1,375 mol; Aldrich 19, 913-3) i tetrahydrofuran (800 ml) over et tidsrom på 8 timer. Den resulterende hvite suspensjon ble rørt ved 0°C og latt langsomt oppvarmes til omgivelsestemperatur natten over. Reaksjonsblandingen ble konsentrert i vakuum og residuet ble tatt opp i en blanding av etylacetat og mettet natriumkloridløsning. Det vandige sjiktet ble ekstrahert med etylacetat. De samlede ekstrakter ble vasket én gang med mettet natriumklorid, tørket over magnesiumsulfat og filtrert, hvilket gir en olje (~ 245 g, kvantitativt). Oljen ble suspendert i vann (200 ml) og avkjølt til 0°C til - 5°C . En vandig løsning av bromkresolgrønt (3 ml 0,04% i vann; Aldrich 31, 870-1) ble tilsatt, hvilket resulterer i en blå løsning. Med kraftig omrøring ble (IN saltsyre (1,4 1) tilsatt dråpevis, hvilket gir en lyseblå til grønn/gul farget løsning (pH ~ 5-6, pH-papir). Denne suspensjonen ble ekstrahert med metylenklorid (2 x 300 ml), og det vandige sjiktet gjort basisk med 2,5 N natriumhydroksyd til pH ~ 12. Det basiske vandige sjikt ble ekstrahert med metylenklorid (5 x 300 ml), og det organiske laget ble vasket med mettet natriumkloridløsning (2 x 100 ml), tørket over vannfritt kaliumkarbonatpulver, filtrert gjennom Celite og konsentrert, hvilket gir tittelforbindelsen som svakt blålig olje som krystalliserer ved henstand (165 g, 69%) (meget hygroskopiske faste stoffer). NMR (CDCI3,400 MHz): 1,43 (s, 9H), 1,61 (p, 2H), 1,59 (s, 2H), 2,76 (t, 2H), 3,20 (q, 2H). 4,95 (br, 1H).
Eksempel 2
N-[3-(t-butyloksykarbonylamino)propyl]-2-aminoetylfosfonsyre-dietylester
Til en oppløsning av 3-(t-butyloksykarbonylaminopropanamin (77 g, 0,44 mol) i metanol (500 ml) ble det satt dietylvinylfosfonat 97% (75 g, 0,44 mol; Aldrich 11, 613-0) under nitrogen og holdt i et vannbad ved ~ 20°C i 48 timer. Reaksjonsblandingen ble konsentrert i vakuum og residuet (~ 160 g) ble satt på et sjikt av "Florisil" (3" x 6") og eluert med metylenklorid/heksan 1:1, metylenklorid og til slutt 10% metanol/metylenklorid, hvilket gir tittelforbindelsen som en fargeløs olje (121 g, 80%).
NMR (CDC13,400 MHz): 1,32 (t, 6H), 1,43 (s. 9H). 1,65 (t. 2H). 1,80 (br, 1H),
1,97 (dt,2H), 2,67 (t, 2H). 2,85 (dt, 2H), 3,20 (q, 2H). 4,09 (m, 2H), 5,08 (br,lH).
Eksempel 3
3,4[[2-(dietoksy-fosforyl)-etyl]-(2-etoksy-3,4-dioksocyklobut-l-eny l)amino] propy lkarbaminsyre-tert-butyl ester
Til en løsning av 3,4-dietoksy-3-cyklobuten-l,2-dion (45 g, 0,265 mol; Aldrich 31.077-8) i absolutt etanol (1,2 1) under nitrogen ble det satt dråpevis en oppløsning av N-[3-(t-butyloksykarbonylamino)propyl]-2-aminoetylfosfonsyredietylester (80 g, 0,24 mol) i absolutt etanol (600 ml), og reaksjonsblandingen ble rørt ved omgivelsestemperatur 15 timer. Reaksjonsblandingen ble konsentrert i vakuum og residuet ble satt på et sjikt av silikagel (6" x 4") og eluert først med en blanding av metylenklorid/heksan 1:1 for å fjerne overskudd av 3,4-dietoksy-3-cyklobuten-l,2-dion og til slutt med 10% metanol/metylenklorid, hvilket gir tittelforbindelsen etter inndampning som en viskøs olje (107 g, 96%). NMR: (CDCI3, 400 MHz): 1,34 (t, 6H), 1,43 (s, 9H), 1,46 (t, 3 H), 1,80 (m, 2 H), 2,12 (m, 2H), 3,14 (m, 2H), 3,49 (t, 1H), 3,66 (m, 1H), 3,73 (t, 1H), 3,90 (m, 1H), 4,10 (m, 4H), 4,74 (m, 4 H), 5,05 (br, H).
Eksempel 4
[2-(8,9-diokso-2,6diazabicyklo[5.2.0]non-l (7)-en-2-yl)etyl]fosfonsyre-dietylester
En løsning av N-[3-(t-butyloksy-karbonylamino)propyl]-N-[4-etoksy-2.3-diokso-cyklobut-l-en-l-yl]-2-aminoetylfosfonsyre-dietylester (100 g, 0,22 mol) i metylenklorid (600 ml) ble avkjølt i is og behandlet med trifluoreddiksyre (300 ml). Reaksjonsblandingen fikk stå og oppvarmes til omgivelsestemperatur natten over. Løsningen ble konsentrert i vakuum ved maks. 40°C og inndampet sammen med toluen (2 x 500 ml), hvilket gir en viskøs olje (159,5 g) som ble løst i absolutt etanol (1,5 1) og satt dråpevis over åtte timer til en oppløsning av trietylamin (350 ml) i etanol (1,5 1) og rørt i 8 timer ved romtemperatur. Reaksjonsblandingen ble konsentrert i vakuum til en olje som ble tatt opp i etylacetat (11). Forbindelsen krystalliserte og ble avkjølt i is, filtrert og vasket med etylacetat og til slutt heksan, hvilket ga tittelforbindelsen som en hvit forbindelse (40 g, 58%).
NMR (CDCI3, 400 MHz): 1,34 (t, 6H), 2,06 (m, 2H). 2,20 (dt, 2H), 3,50 (m, 4H),
4,05 (m, 2H), 4,15 (m, 4H), 7,87 (br, 1H).

LC analyse (kolonne: Mikrosorb-MV C 18,150 x 4,6 mm; Elueringsmiddel: 30/70 MeOH/ 0.01 M NH4H2PO4 pH 4,7; Strømningshastighet: 1 ml/imin; UV detektor ved 210 nm; Analyse: Ber. for C13H21N2O5P: C, 49,36; H, 6,69; N, 8,85%.

Eksempel 5
[2-(8,9-diokso-2,6diazabicyklo[5.2.0]non-l (7)-en-2-yl)etyl]fosfonsyre
Under N2ble bromotrimetylsilan (83 ml, 96,3 g, 0,63 mol; Aldrich 19,440-9) tilsatt dråpevis ved rask hastighet til en løsning av [2-(8,9-diokso-2,6-diazabicyklo[5.2.0]non-l(7)-en-2-yl)etyl]fosfonsyre-dietylester (37,6 g, 0,12 mol) i metylenklorid (350 ml). Reaksjonsblandingen ble holdt i et vannbad på omtrent 20°C i 15 timer. Den klare løsning ble konsentrert i vakuum og den skumaktige rest ble tatt opp i aceton (600 ml) med kraftig risting, hvilket resulterer i en tynn suspensjon. Vann (50 ml, 2,78 mol) ble tilsatt, hvilket ga en gummiaktig felling som stivnet øyeblikkelig. Suspensjonen ble ristet kraftig i 10 minutter, filtrert og vasket med aceton, hvilket gir en gul, faststoff-forbindelse. De faste stoffene ble tatt opp i kokende vann (450 ml), og den varme løsning ble filtrert gjennom et foldefilterpapir for å fjerne en liten mengde av uløselig materiale. Den klare vandige løsning ble avkjølt i is og krystallisering begynte med én gang. Den tykke krystallinske masse ble fortynnet ved langsom tilsetning av aceton (800 ml) holdt kald i 1 time, filtrert og vasket med aceton og deretter heksan, hvilket gir tittelforbindelsen som et blekgult, fast stoff (20,2 g). En andre høsting fra moderluten (100% renhet ved LC) ga en ytterligere mengde (6,5 g) for et totalutbytte på 87%.

Eksempel 6
Fremstilling av 3-(t-butoksykarbonylamino)propanamin ("t-BOC-Propanamin")
En løsning av di-t-butyldikarbonat (0,50 kg, 2,29 mol) i metanol ble satt til et overskudd av (5 ekvivalenter) av 1,3-diaminopropan (0,83 kg, 11,2 mol) over et 4 timers tidsrom ved 25-30°C . Produktet 1,3-diaminopropan-t-butoksykarbonat ble filtrert fra og metanolen fjernet ved vakuumdestillering. Den gjenværende olje ble tatt opp i etylacetat, vasket med saltvann, vann tilsatt og pH regulert til 5,5. Sjiktet ble separert fra og den vandige fasen gjort sterkt alkalisk. Den alkaliske vandige løsning ble ekstrahert seks ganger med toluen. De samlede toluen- ekstrakter ble tørket over natriumsulfat og konsentrert, hvilket gir t-BOC-propanamin (0,365 kg) i et 77,4% utbytte, 11,5% totale urenheter.
Eksempel 7
Fremstilling av [2-(8,9-Diokso-2,6-diazabicyklo[5.2.0]non-l (7)-en-2-yl]fosfonsyre dimetylester ("Dimetylfosfonatester")
A. Dannelse av t- BOC- fosfonat- dimetylester
En løsning av t-BOC-propanamin, 98% (0,67 kg, 3,77 mol) og dimetylvinylfosfonat (0,59 kg, 4,12 mol) i vannfri metanol (2,7 1) ble rørt ved romtemperatur i 2 dager, hvilket ga en løsning av dimetylfosfonatester-produktet.
B. Dannelse av t- BOC- fosfonatdimetvletvl- skvarat
Dimetylfosfonatester-produktløsningen i A ble deretter satt til en oppløsning av 3,4-dietoksy-3-cyklobuten-l,2-dion ("Etyl skvarat") (0,55 kg, 3,23 mol) i vannfri metanol (3,241) over en seks timers periode. Etter omrøring ved 0-5 °C natten over, ble reaksjonsblandingen konsentrert ved destillasjon. Toluen (11) tilsatt og destilleringen gjentatt til et sluttvolum på 1,6 1 3-[[2-(dietoksy-fosforyl)etyl](2-etoksy-3,4-dioksocyklobut-1 -enyl)amino]propylkarbaminsyre-tert-butylester ("t-BOC-fosfonatdimetyletyl-skvarat").
C. Dannelse av avbeskvttet fosfonatdimetvletvl- skvarat
Til produktløsningen av t-BOC-fosfonatdimetyletyl-skvarat fra B ble det satt toluen (5,01) ved 0-5°C, fulgt av trifluoreddiksyre (4,71 kg, 41,31 mol) over 0,25-5 timer, mens temperaturen ble holdt under 15°C . Etter røring i 4 timer ved romtemperatur ble reaksjonsblandingen konsentrert, hvilket ga det rå avbeskyttete fosfonatdimetyletyl-skvarat.
D. Dannelse av d imetvlfosfonatester
Vannfri metanol (4,5 IL) ble satt til reaksjonsproduktkonsentratet fra C, og den resulterende løsning ble tilsatt over et 6 timers tidsrom til en løsning av overskudd av trietylamin (2,9 kg, 28,66 mol) i vannfri metanol ved romtemperatur. Den resulterende blanding ble konsentrert, fulgt av tilsetning av etylacetat som utfelte tittelforbindelsen dimetylfosfonatester-produktet. Etter filtrering og vasking av kaken med kald etylacetat, ble et 50,4% utbytte av [2-(8,9-diokso-2,6-diazabicyklo[5.2.0-non-l(7)-en-2-yl )etyl]fosfonsyredietylester ("dimetyl- fosfonatester") (0,56 kg), 97,6% styrke, oppnådd, en forurensing på 1,05% og totale forurensinger på 1,76%.
Eksempel 8
Fremstilling og rensning av [2(8,9-diokso-2,6- diazabicyklo[5.2.0]non-l(7)-en-2-yl)etyl]fosfonsyre
Bromtrimetylsilan, 97,6% (0,55 kg, 3,59 mol) ble satt ved romtemperatur til en rørt suspensjon av dimetylfosfonatester (0,46 kg, 1,56 mol) i acetonitril (4,11). Den resulterende løsning blir deretter satt under røring til acetonitril-vann. Tittelfosfonsyren falt ut fra løsningen som kremfargete faste stoffer. Oppslemningen ble avkjølt til 0°C og produktet oppsamlet ved filtrering. Den våte kaken ble rørt i vann og 30% NaOH tilsatt til en pH på 13. En løsning ble dannet. Saltsyre ble tilsatt til en pH på 1,0. Tittelfosfonsyren falt ut fra løsningen som hvitfarget fast stoff. Produktet ble oppsamlet på en Buchnertrakt og vasket med iskaldt vann. Tittelfosfonsyren ble deretter renset ved omkrystallisering fra vann. Den våte kaken ble løst i 12 deler vann, filtrert gjennom papir på en Buchnertrakt og filtratet konsentrert. Etter avkjøling krystalliserte tittelfosfonsyren ut fra løsningen. Oppslemningen ble avkjølt til 0°C og oppsamlet på en Buchnertrakt. Kaken ble vasket med vann, tørket i en vakuumovn ved 65°C , hvilket gir tittelfosfonsyren, renset i et 86 % utbytte, 99,9 % styrke, en enkelt urenhet, 0,05 %, totale urenheter, 0,13 %.
Claims (14)
1. Fremgangsmåte for fremstilling av [2-((8,9)-diokso-2,6-diazabicyklo[5.2.0]-non-l(7)-en-2-yl)etyl]fosfonsyre karakterisert ved de følgende trinn: a) omsetning av en vinylfosfonat di-Ci-C6 alkylester med 3-aminopropyl-karbaminsyre-l,l-dimetyletylester, hvilket gir en di-Ci-C6 alkylester av N-[3-(t-butyloksykarbonylamino)propyl]-2-aminoetylfosfonsyre; b) omsetning av et 3,4-di-C i -C4 alkoksycyklobut-3-en-1,2-dion med en di-C 1-C6 alkylester av N-[3-(t-butyloksykarbonylamino )propyl]-2-aminoetylfosfonsyre fra trinn a, hvilket gir 3-[[2-(di-Ci-C6alkoksyfosforyl)etyl]-(2-Ci-C6 alkoksy-3,4-diokso-l-cyklobuten-1 -yl)amino]propyl-karbaminsyre-tert-butyl ester; c) avbeskyttelse av 3-aminogruppen i produktet fra trinn (b); d) cyklisering av produktet fra trinn (c) for å danne det bicykliske mellomprodukt [2-((8,9)-di-okso-2,6-diazabicyklo[5.2.0] non-l(7)-en-2-yl)etyl]fosfonsyre-di-C|-C6 alkylester; og e) omdannelse av di-Ci-C6 alkylesteren fra trinn (c) til fosfonsyreproduktet.
2. Fremgangsmåte ifølge krav 1, karakterisert ved at reaksjonene i trinn (b), (c) og (d) blir utført in situ.
3. Fremgangsmåte ifølge krav 1 eller krav 2, karakterisert ved at reaksjonen i trinn (b) blir utført i vannfri metanol eller etanol ved omgivelsestemperatur.
4. Fremgangsmåte ifølge hvilket som helst av kravene 1 til 3,karakterisert ved at reaksjonen i trinn (c) blir utført i metylenklorid ved -5°C til 25°C.
5. Fremgangsmåte ifølge hvilket som helst av kravene 1 til 4, karakterisert ved at reaksjonen i trinn (e) blir utført i metylenklorid eller acetonitril ved ca. 20 °C.
6. Fremgangsmåte ifølge hvilket som helst av kravene 1 til 5, karakterisert ved at 3,4-di-Ci-C4alkoksycyklobut-3-en-l,2-dionet er 3,4-dietoksycyklobut-3-en-l,2-dion.
7. Fremgangsmåte for fremstilling av N-[3-(t-butyloksykarbonylamino)-propyl]-2-aminoetylfosfonsyre-di-Ci-C6alkylester, karakterisert ved at den omfatter omsetning av en vinylfosfonat-di-Ci-Cealkylester med 3-aminopropylkarbaminsyre-l,l-dimetyl-etylester.
8. Fremgangsmåte ifølge til hvilket som helst av kravene 1 til 7, karakterisert v e d at reaksjonen med vinylfosfonat-di-Cj-Cealkylesteren blir utført i vannfri metanol ved ca. 20°C.
9. Fremgangsmåte ifølge til hvilket som helst av kravene 1 til 8, karakterisert v e d at vinylfosfonat-di-Ci-C6 alkylesteren er dimetylvinylfosfonat eller dietylvinylfosfonat.
10. Fremgangsmåte ifølge hvilket som helst av kravene 1 til 8, karakterisert v e d at N-[3-(t-butyloksykarbonylamino)propyl]-2-aminoetylfosfonsyre-di-Ci-Cealkylesteren er N-[3-(t-butyloksykarbonylamino)propyl]-2-aminoetylfosfonsyre-dimetylester eller N-[3-(t-butyloksykarbonylamino)propyl]-2-aminoetylfosfonsyre-dietylester.
11. Forbindelse, karakterisert ved at den er en N-[3-(t-butyloksykarbonyl-aminopropyl]-2-aminoetylfosfonsyre-di-Ci-C6alkylester,.
12. Forbindelse, karakterisert ved at den er N-[3-(t-butyloksykarbonyl-amino)propyl]-2-aminoetyl-fosfonsyre-dimetylester eller N-[3-(t-butyloksykarbonyl-amino)propyl]-2-aminoetylfosfonsyre-dietylester.
13. Forbindelse, karakterisert ved at den er en 3-[[2-(di-C i-C6alkoksy-fosforyl)etyl]-(2-Ci-C6alkoksy-3,4-dioksocyklobut-l-enyl)amino]propylkarbaminsyre-tert-butylester, hvor alkoksy-gruppene er like eller forskjellige.
14. Forbindelse, karakterisert ved at den er 3-[[2-(dietoksy-fosforyl)etyl]-(2-etoksy-3,4-dioksocyklobut-1 -enyl)amino]propylkarbaminsyre-tert-butyl ester.
