Description
Foreliggende oppfinnelse vedrører alkaloider med camptothecin-grunnstruktur isolert fra Mappia foetida eller frem-stilt ved semisyntese fra disse alkaloider.
Mappia foetida, som er en plante som vokser på det indiske subkontinent, er kjent for å i sine forskjellige deler, hovedsaklig i frøene, inneholde camptothecin, mappicin og foetidin I og II (EP-A-685481).
I "Journal of Medicinal Chemistry", 1979, vol. 22, nr. 3, beskrives camptothecinderivater og deres fremstilling.
Alkaloidene ifølge oppfinnelsen er særpregede ved følgende generelle formel:
hvori R er et hydrogenatom eller en metoksygruppe; Rx er hydroksy; en OM-gruppe hvori M er et alkalikation, en Cx-C6-alkoksygruppe; en fenoksygruppe eventuelt substituert
med halogenatomer, C^-Cg-alkyl, C^-Cj-alkoksy, nitro, cyano, Ci-Cj-haloalkylgrupper, amino, Ci-Cg-monoalkylamino eller C2_12-dialkylaminogruppe hvori alkylgruppen eventuelt er substituert med aminogrupper eller en arylaminogruppe; R2
er en C^-Cg-alkylgruppe eller en gruppe med formelen COR3, hvor R3 er Ci-Cg-alkyl eller fenyl eller benzyl, eventuelt substituert med halogenatomer, C^-Cg-alkyl, Ci-Cg-alkoksy, nitro, cyano, C1-C3-haloalkylgrupper, med det forbehold at R, Ri og R2 ikke samtidig er henholdsvis H, NHCH(CH3)2 og COCH3/COCsH11.
Ytterligere trekk fremgår av kravene 2-8.
Forbindelsene 1 hvor R betyr hydrogen eller metoksy, ^ betyr hydroksy eller en OM-gruppe (M = natrium eller kalium) og R2 betyr acetyl, kan isoleres fra Mappia foetida under ekstrahering av den kunstig tørkede vegetabilske biomasse ved en temperatur som ikke er høyere enn 50°C, for-trinnsvis ved 35°C, først med alifatiske ketoner eller alifatiske estere og deretter med alifatiske alkoholer. Under disse operative betingelsene kan 17-acetylderivatene av camptothecinsyre og 9-metoksycamptothecinsyre ekstraheres med høyt utbytte. Selv om Mappia foetida er blitt omfattende undersøkt som en camptothecin-selektiv kilde, ble disse nevnte alkaloider ikke identifisert, sannsynligvis grunnet deres spaltning til camptothecin under ekstraherin-gen når det brukes uegnede oppløsningsmidler. I nærvær av alifatiske alkoholer kan disse alkaloidene lett omdannes til camptothecin, selv ved ekstrahering ved naturlig pH-verdi.
Den samme gruppe forbindelser kan fremstilles ved selektiv acetylering av C17-hydroksylet av camptothecin i alkalisk medium.
De dannede forbindelsene kan i sin tur brukes som utgangsstoffer ved fremstilling av andre forbindelser med formel 1 hvor R2 betyr noe annet enn acetyl og/eller Rx betyr en alkoksy-, fenoksy- eller aminogruppe som definert ovenfor, eller ved fremstilling av Foetidin I og II. Med denne hensikt kan konvensjonelle fremgangsmåter ved fremstilling av estere eller amider brukes, f.eks. omsetning av forbindelser 1 hvor Rx betyr en OM-gruppe, med alkylhalogenider såsom etyl- eller benzylbromacetat, for fremstilling av estere, eller omsetning av forbindelser 1 hvor Rx betyr OH, med amin og dicykloheksylkarbodiimid for fremstilling av amider.
Forbindelsene 1 har cytotoksisk aktivitet mot svulstcelle-linjer. F.eks. viser Tabell 1 den cytotoksiske aktivitet mot en colon-karsinomcellelinje (HCT116) og mot den samme cellelinje som i tillegg er resistent mot de fleste kjemo-terapeutiske midler (HCT116/VM46) . Resultatene viser at en forbindelse ifølge oppfinnelsen er mer aktiv enn camptothecin.
Forbindelsene 1 kan derfor brukes som aktiv ingrediens i farmasøytiske svulstmotvirkende sammensetninger blandet med egnede bærerstoffer, f.eks. injiserbare fysiologiske opp-løsninger. Doseringen kan variere innen vide grenser (5 til 500 mg/dag), men i prinsippet vil den være ca. 10 mg alka-loid pr. dag.
De følgende eksempler skal ytterligere illustrere oppfinnelsen.
Eksempel 1
Isolering av 17- acetyl- camptothecinsyre og 17- acetyl- 9-metoksy- camptothecinsyre
3 kg Mappia foetida-frø ble ekstrahert tre ganger med tørt aceton (3 x 3 1) ved romtemperatur. De sammenslåtte eks-trakter ble konsentrert til tørrhet for å gi 580 g av en voksaktig masse som inneholdt camptothecin, 9-metoksy-camptothecin og en liten mengde 17-acetyl-camptothecinsyre. Det vegetabilske materiale fra acetonekstraheringen ble ekstrahert på nytt gjentatte ganger med metanol (3 x 3 1) ved 10°C, og etter konsentrasjon av ekstraktene ved en lav temperatur erholdt man 200 g av et tørt residuum, som ble suspendert ill vann og ekstrahert tre ganger med 500 ml n-butanol. De sammenslåtte butanolekstraktene ble inndampet til tørrhet under vakuum ved en temperatur som ikke var høyere enn 30°C. Man fikk 28,9 g av en alkaloidf raks jon med høyt innhold av en blanding av 17-acetyl-camptothecinsyre og 9-metoksy-17-acetyl-camptothecinsyre, og denne ble kro-matografert med revers fase gjennom en RP18-kolonne under eluering med metanol/vann og metanol for å danne tre frak-sjoner som bestod av henholdsvis kumaroylagmatin og campto-thecinsyrer. Denne fraksjon ble renset ytterligere over kiselgel for å gi 3,8 g 17-acetyl-camptothecinsyre med de følgende spektroskopiske og kjemisk-fysikalske egenskaper: smp.: 258°C, Ctc = +63,4 (c = 0,05, H20) ; ^-NMR (DMSO-d6) 8: 0,85 (t, 3H, H-18) , 1,95 (m+s, 5H, H-19+COCH3) , 5,20 (s, 2H, H-17), 5,40, 60 (q, JM = 10,6 Hz, H-5), 7,65-8,65 (m, 6H, arom.).
Mengden 9-metoksy-17-acetyl-camptothecinsyre er en femtedel av mengden av den ovennevnte syre, og denne syren har de følgende kjemisk-fysikalske egenskaper: smp.: 208°C, = 56,4 (c = 0,05 H20) .
Eksempel 2
Fremstilling av 17- acetyl- camptothecinsyre fra camptothecin 1 g camptothecin ble suspendert i 30 ml vann, 340 mg NaOH ble satt til, og det hele ble omrørt ved 40°C i to timer, eller i hvert tilfelle inntil det forelå fullstendig opp-løsning; vann ble fjernet under vakuum og residuet ble tatt opp i 20 ml DMF under en sterk reaksjon. Til oppløsningen tilsatte man gradvis 600 mg eddiksyreanhydrid, og det hele ble omrørt i ca. 2 timer. Oppløsningsmidlet ble fjernet under vakuum, og residuet ble fordelt i en 5:6:4 kloroform/metanol /vann-blanding. Metanolfasen ble inndampet til tørrhet, og residuet ble krystallisert for å gi 17-acetyl-camptothecinsyre med samme egenskaper som hva som ble angitt i Eksempel 1.
Eksempel 3
17- Acetylcamptothecin- 21- metylester
17-Acetylcamptothecin (100 mg, 0,25 mmol) ble oppløst i tørt DMF (8 ml), og tørt kaliumkarbonat (68 mg, 0,49 mmol) og jodmetan (69 mg, 0,49 mmol) ble tilsatt dertil under omrøring ved romtemperatur i 20 timer. Reaksjonsblandingen ble filtrert og vasket med kloroform (5 ml). Filtratene ble fortynnet med kloroform (10 ml) og vasket med vann (5 ml x 3). Den organiske fase ble tørket over tørt natriumsulfat. Etter filtreringen ble oppløsningsmidlet fjernet under vakuum, og residuet (170 mg) ble underkastet flash-kromato-grafi (CHC13; CH3OH = 9:1). Tittelforbindelsen ble erholdt (45 mg, utbytte: 45%) i form av et fast stoff.

Ved å følge den samme fremgangsmåten, men ved bruk av etyl-bromacetat eller t-butylbromacetat istedenfor jodmetan, erholdt man den tilsvarende etylester (a) eller t-butyl-ester (b): (a) 'H-NMR (CDC13) 8: 1,10 (t, J=7,5 Hz, 3H, H-18), 1,30 (t, J=7,5 Hz, 3H, CH3) , 2,10 (s, 3H, OCOCH3) , 2,30-2,55 (m, 2H, H-19), 4,25 (q, J=7,5 Hz, 2H, CH2) , 4,70 (q, JM=15 Hz, 2H, OCOCH2CO) , 5,32 (s, 2H, H-5), 5,52 (s, 2H, H-17), 7,6 (m, 5H, arom).
(b) 'H-NMR (CDC13) 8: 1,10 (t, J=7,5 Hz, 3H, H-18), 1,46
(s, 9H, C(CH3)3), 2,10 (s, 3H, OCOCH3) , 2,35-2,52 (m, 2H, H-19), 4,60 (q, JM=15 Hz, 2H, OCOCH2CO) , 5,30 (s, 2H, H-5), 5,52 (s, 2H, H-17), 7,58-8,38 (m, 6H, arom).
Eksempel 4
17 - Deace tyl - camptothecinsyre, 21 - es ter
Forbindelse b (60 mg, 0,11 mmol) ble oppløst i tørr kloroform (2 ml). Jodtrimetylsilan (33 mg; 0,17 mmol) ble tilsatt ved 0°C under en nitrogenatmosfære under omrøring ved 0°C i 1 time og ved romtemperatur i 1 time. Reaksjonsblandingen ble helt i en 5% NaHC03-oppløsning (5 ml) . Den vandige fase ble vasket gjentatte ganger med kloroform inntil kloroformfasen ble fargeløs. Den vandige fase ble nøytrali-sert med en 2,5% HCl-oppløsning ved 0°C inntil pH-verdien nådde 7, og ekstrahert med butanol (5 ml x 6) . Butanol-fasene ble slått sammen og inndampet under vakuum for å gi et residuum (51 mg) , som ble underkastet f lash-kromatograf i gjennom kiselgel under eluering med kloroform/metanol for å gi tittelforbindelsen (11 mg).

Claims (8)
1. Forbindelse karakterisert ved formel (1):
hvori R er et hydrogenatom eller en metoksygruppe; Rj er hydroksy; en OM-gruppe hvori M er et alkalikation, en C^-C6-alkoksygruppe; en fenoksygruppe eventuelt substituert med halogenatomer, Ci-Cg-alkyl, Ci-Cg-alkoksy, nitro, cyano, Ci-Cj-haloalkylgrupper, amino, C^-Cj-monoalkylamino eller C2_12-dialkylaminogruppe hvori alkylgruppen eventuelt er substituert med aminogrupper eller en arylaminogruppe; R2 er en Ci-Cj-alkylgruppe eller en gruppe med formelen COR3, hvor R3 er C^-C^-alkyl eller fenyl eller benzyl, eventuelt substituert med halogenatomer, C^-Cg-alkyl, C^-Cs-alkoksy, nitro, cyano, C^-C^-haloalkylgrupper, med det forbehold at R, Rx og R2 ikke samtidig er henholdsvis H, NHCH(CH3)2 og COC^/COCgH^.
2. Forbindelse med formel (1) ifølge krav 1, karakterisert ved at H betyr et alkalikation utvalgt fra natrium og kalium.

7. Fremgangsmåte ved fremstilling av en forbindelse ifølge krav 1 omfattende å forestre eller amidere en forbindelse ifølge krav 3 på kjent måte.
8. Farmasøytisk blanding som inneholder en forbindelse ifølge et av kravene 1-3 som aktiv ingrediens, for fremstilling av et cytotoksisk antisvulst - legemiddel.
