Description
Den foreliggende oppfinnelse er rettet på neurobeskyttende (antiischemiske og eksitatoriske aminosyrereseptor-blokkerende 3R<*>4S<*>3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kroman-4,7-diol; enantiomere derav; farmasøytiske akseptable salter derav og en farmasøytisk blanding. Disse forbindelsene kan anvendes for behandlinger av hodesmerter, slag eller CNS degenerative sykdommer så som Alzheimers sykdom, Huntingtons sykdom, Parkinsons sykdom og andre tilstander som forbedres ved blokkering av N-metyl-D-aspartinsyre (NMDA) reseptoren.
De eksitatoriske aminosyrer er en viktig gruppe neurotransmittere som bevirker eksitatorisk neurotransmisjon i sentralnervesystemet. Glutaminsyre og aspartinsyre er to endogene ligander som aktiverer eksitatoriske aminosyre (EAA)-reseptorer. Det er to typer av EAA-reseptorer, ionotrope og metabotrope, som adskiller seg i sin form av signaltransduksjonen. Det er minst tre adskilte ionotrope EAA-reseptorer karakterisert ved den selektive agonist som aktiverer hver type: NMDA, (N-metyl-D-aspartinsyre), AMPA (2-amino-3-(5-metyl-3-hydroksyisoksazol-4-yl)propansyre), og kaininsyre-reseptorer. De ionotrope EAA-reseptorer er knyttet til ionekanaler som er gjennomtrengelige for natrium, og i tilfelle NMDA-reseptorer, kalsium. Metabotrope-reseptorer knyttet til fosfoinositid-hydrolyse med et membran tilknyttet G-protein aktiveres av quisqualsyre, ibotensyre og (1S,3R)-1-aminocyklo-pentan-1,3-dikarboksylsyre.
NMDA-reseptoren er et makromolekylært kompleks som består av flere adskilte bindingsseter som avgrenser en ionekanal som er gjennomtrengelig for natrium og kalsiumioner. Hansen og Krogsgaard-Larsen, Med.Res.Rev., 10, 55-94
(1990). Der er bindingsseter for glutaminsyre, glysin og polyaminer, og et sete inne i ionekanalen hvor forbindelse så som fencyklidin (PCP) utøver sine antagonist-virkninger.
Kompetitive NMDA-antagonister er forbindelser som blokkerer NMDA-reseptoren ved å samvirke med glutamatbindingssetet. En spesiell forbindelsesevne til kompetitiv til å binde til NMDA glutamat-reseptoren kan bestemmes ved å bruke en radioligandbindingsmåling. Se Murphy et. al. British J. Pharmacol. 95, 932-938

Agonister for neurotransmisjonen på NMDA-reseptorer er anvendelige terapeutiske midler for behandling av neurologiske forstyrrelser. US-patent nr. 4.902.695 er rettet på en serie kompetitive NMDA-antagonister som er anvendelige for behandling av neurologiske forstyrrelser innbefattet epilepsi, slag, angst, cerebral ischemi, muskelkramper og neurodegenerative forstyrrelser så som Alzheimers sykdom og Huntingtons sykdom. US-patent nr. 4.968.878 er rettet på en andre serie kompetitive NMDA-reseptor antagonister som er anvendelige for behandling av lignende neurologiske forstyrrelser og neurodegenerative forstyrrelser. US-patent nr. 5.192.751 beskriver en fremgangsmåte for behandling av urininkontinens hos et pattedyr som omfatter administrering av en effektiv mengde av en kompetitiv NMDA-antagonist.
NMDA-antagonister er også anvendelige terapeutiske midler med antikrampe, angstløsende, muskelrelaksant og antipsykotisk aktivitet, J. Lehmann, The NMDA Receptor, Drugs of the Future 14, nr. 11, s. 1059 (1989). NMDA-antagonister er også rapportert å være effektive for behandling av migrene (Can. J. Neurol. Sei. 19(4), s. 487, 1992); drogeavhengighet (Science, 251, s. 85, 1991); og neuropsykiatriske forstyrrelser i forbindelse med AIDS (PIPS 11A s. 1, 1990).

Ifenprodil er en racemisk såkalt dl-erytroforbindelse med den relative stereokiemiske formel
som markedsføres som et hypotensivt middel, en anvendelse det deler med flere nære beslektede analoger; Carron et al., US-patent 3.509.164; Carron et al., Drug. Res., v. 21, s. 1992-1999 (1971). Mer nylig er ifenprodil vist å ha antiischemisk og eksitatorisk aminosyrereseptor-blokkerende aktivitet; Gotti et al., J. Pharm. Exp. Therap., v. 247, s. 1211-21 (1988); Carter et al., loe. eit. s. 1222-32 (1988). Se også publisert europeisk patentsøknad 322.361 og fransk patent 2546166. Et mål som foreliggende oppfinnelse når har vært å finne forbindelser som har slik neurobeskyttende virkning i stor grad, men samtidig har nedsatt eller ingen signifikant hypotensiv virkning.
Visse strukturelt beslektede 1-fenyl-3(4-aryl-4-acyloksypiperidino)-1-propanoler er også rapportert å være anvendelige som analgetika, US-patent 3.294.804; og 1-[4-amino- og hydroksy-alkyl)fenyl]-2-(4-hydroksy-4-tolylpiperazino)-1-alkanoler og alkanoler er rapportert å ha analgetisk, antihypertensiv, psykotropisk eller antiinflammatorisk aktivitet, japansk Kokai 53-02,474 (CA 89:43498y; Derwent Abs. 14858A) og 53-59.675 (CA 89:146938w; Derwent Abs. 48671A).
SAMMENFATNING AV OPPFINNELSEN
Foreliggende oppfinnelse vedrører følgelig forbindelse, kjennetegnet ved at den er 3R<*>4S<*>3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kroman-4,7-diol, dens optiske isomer og farmasøytiske akseptable salter.
Oppfinnelsen er følgelig rettet på neuobeskyttende kromanolforbindelser med formel (I) som har eksepsjonell oral aktivitet. Optiske isomere samt farmasøytisk akseptable salter av forbindelse (I) er også gjenstand for denne oppfinnelsen.
Den foreliggende oppfinnelse er videre rettet på farmasøytiske blandinger omfattende en forbindelse med formel (I) og en farmasøytisk akseptabel bærer.
Sykdommer eller tilstander som reagerer på behandling med en forbindelse med formel (I) innbefatter hodesmerter, ryggmargsmerter, slag og mulitiinfarkt dementia.
Andre sykdommer eller tilstander som reagerer på behandling ved en forbindelse med formel (I) innbefattet Alzheimers sykdom, Huntingtons sykdom, Parkinsons sykdom, epilepsi og amytrop lateral sclerose.
Ytterligere sykdommer eller tilstander som reagerer på behandling med en forbindelse med formel (I) omfatter smerte, AIDS dementia, psykotiske tilstander, drogeavhengighet, migrene, hypoglykemi og anxiolyttiske tilstander.
Enda en annen sykdom eller tilstand som reagerer på behandling med forbindelse med formel (I) er urininkontinens.
Enda en annen sykdom eller tilstand som reagerer på behandling med forbindelse med formel (I) er et ischemisk anfall som stammer fra CNS-kirurgi, hjerteåpningskirurgi eller annen fremgangsmåte hvor under funksjonen av det kardiovaskulære system berøres.
DETALJERT BESKRIVELSE AV OPPFINNELSEN
De neurobeskyttende kromanoler med formel (I) fremstilles lett som beskrevet i europeisk patentsøknad 441506, som heri inntas som henvisning.
Forbindelser ifølge den foreliggende oppfinnelse fremstilles lett ved å omsette et hensiktsmessig beskyttet 7-hydroksykromanon, f.eks. 7-benzyloksykromanon, med brom i et reaksjonsinert løsningsmiddel under dannelse av 7-benzyloksy-3,3-dibromkromanon.
Dibromforbindelsen omsettes deretter med to molekvivalenter 4(4-fluorfenyl)-4-hydroksy-piperidin under dannelse av 7-benzyloksy-3[4-(-fluorfenyl)-4-hydroksy-piperidin-1-yl]kromenon. Reaksjonen utføres generelt i nærvær av en base, vanligvis en tertiær base i et reaksjonsinert løsningsmiddel så som etanol eller acetonitril. Om ønsket kan reaksjonen katalyseres på opp til en molekvivalent av et et jodidsalt. Temperaturen er ikke kritisk, men bør ikke være så høy at den fører til uønsket spaltning. En temperatur i området romtemperatur til 100°C er i alminnelighet tilfredsstillende.
Kromenonet som oppnås som beskrevet ovenfor reduseres til kromanol ved å bruke vanlig hydride reduksjonsmidler, f.eks. NaBH4, i et reaksjonsinert løsningsmiddel så som metanol eller etanol, i alminnelighet ved romtemperatur eller noe høyere temperatur.
I et sluttrinn fjernes beskyttelsesgruppen, f.eks. benzyloksy, ved vanlig metoder, f.eks. katalytisk hydrogenering under dannelse av forbindelsen (I).
Om ønsket kan forbindelsen (I) overføres i et farmasøytisk akseptabelt salt.
Den ovennevnte henvisning til «farmasøytisk akseptable salter» viser til vanlig syreaddisjonssalter og kationsalter. Således inneholder forbindelsene med formel (I) en aminogruppe som er basisk, og således er i stand til å danne slike salter. Slike salter omfatter, men er ikke begrenset til, slike med Hel, HBr, HNO3, H2S04, H3PO4, CH3SO3H, p_-CH3C6H4S03H, CH3CO2H, glukonsyre, vinsyre, maleinsyre og ravsyre. De fremstilles generelt ved vanlige metoder, f.eks. ved å kombinere forbindelsen med formel (I) med minst en molekvivalent av syren i et egnet løsningsmiddel. Forbindelsen med formel (I) inneholder en fenolisk hydroksygruppe og er også i stand til å danne kationsalter (f.eks. Na, K og lignende); uttrykket «farmasøytiske akseptable salter» er også ment å omfatte slike salter. Disse salter fremstilles også ved vanlige metoder, f.eks. ved å kombinere en fenolisk forbindelse med formel (I) med en molekvivalent av NaOH eller KOH i et egnet løsningsmiddel.
Forbindelsene med formelen (I) inneholder to asymmetriske karboner - svarende til to racemater og fire optiske aktive forbindelser. En av disse racemater er den ovenfor nevnte cis-isomer, og den andre trans-isomeren. I den foreliggende oppfinnelse dominerer cis-isomeren og er den foretrukne form. Hver av disse racemater kan oppløses i et par enantiomere gjennom diastereomere syreaddisjonssalter med en optisk aktiv syre. Alternativt overføres den racemiske alkohol i tilsvarende diastereomere estere eller uretaner dannet med en optisk aktiv syre eller isocyanat. Slike kovalent bundne derivater er egnet for en rekke separasjons-metoder (f.eks. kromatografi, omkrystallisering osv.). Slike diastereomere estere dannes fra alkoholen, og den optiske aktive syre ved standardmetoder, generelt slike som medfører aktivering av syren, f.eks. som syrekloridet, som et blandet anhydrid med et alkylklorformiat, eller med et dehydratiserende koblingsmiddel så som dicykloheksylkarbodiimid. Så snart de resulterende diastereomere estere er adskilt, f.eks. ved kromatografiske metoder, hydrolyseres de ved vanlige metoder, f.eks. vandig syre eller vandig base, og gir de enantiomere, optiske aktive alkohol-forbindelser med formel (I). Foretrukne forbindelser ifølge den foreliggende oppfinnelse er (+) cis og (+) cis-isomerene. En spesielt foretrukket forbindelse er (+) (3R, 4S)-3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kroman-4,7-diol tartrat etanolat hydratet.
Utgangsmaterialene og reagensene som kreves for syntesen av forbindelsene ifølge den foreliggende oppfinnelse er lett tilgjengelige, enten i handelen eller ifølge litteraturmetoder, eller ved metoder eksemplifisert i mellomprodukter nedenunder.
De foreliggende forbindelser med ovennevnte formel (I) har selektiv neurobeskyttende aktivitet, basert på sin antiischemiske aktivitet og evne til å blokkere eksitatoriske aminosyre-reseptorer, og har samtidig nedsatt eller ingen signifikant hypotensiv aktivitet. Dn antiischemiske aktivitet av de foreliggende oppfinnelser bestemmes ifølge en eller flere metoder som er beskrevet i detalj tidligere av Gotti et al., og Carter et al., nevnt ovenfor, eller ved lignende metoder. For begynnelsene ifølge foreliggende oppfinnelses evne til å blokkere eksitatoriske aminosyre-reseptorer kan demonstreres ved deres evne til å blokkere N-metyl-D-aspartinsyre-indusert (NMDA) forhøyelser av cGMBP i neonatale rottecerebellums ifølge den følgende fremgangsmåte. Cerebellums fra ti 8-14 dager gamle Wistarrotter skjæres raskt ut og plasseres i 4°C Krebs/bicarbonatbuffer, pH 7,4 og hakkes så opp i 0,5 mm x 0,5 mm snitt ved bruk av en Mcllvain vevshakker (The Nickle Laboratory Engineering Col, Gomshall, Surrey, England). De resulterende stykker av cerebellum overføres til 100 ml Krebs/bikarbonatbuffer ved 37°C som ekvilibreres kontinuerlig med 95:5 O2/CO2. Cerebellumstykkene inkuberes på en slik måte i 90 minutter med tre skifte av buffer. Bufferen dekanteres deretter, vevet sentrifugeres (1 min., 3200 opm.) og vevet gjennomslemmes i 20 ml Krebs/bikarbonatbuffer. Deretter fjernes 250^ alikvoter (ca. 2 mg) og plasseres i 1,5 ml mikrofugerør. Til disse rørene settes 10 \ i\ av forbindelsen som undersøkes fra en stamløsning, etterfulgt etter en ti minutters inkuberingsperiode, av 10 ^l 2,5 mM løsning NMDA for å starte reaksjonen. NMDA sluttkonsentrasjon er 100 ^M. Kontroller er ikke tilsatt NMDA. Rørene inkuberes i ett minutt ved 37°C i ert rystevannbad og deretter tilsettes 750 |il av en 50 mM Tris-CI, 5 mM EDTA-løsning for å stoppe reaksjonen. Rørene plasseres straks i et kokende vannbad i 5 minutter. Innholdet i hvert rør ultralydbehandles deretter i 15 sekunder ved bruk av et probe ultralydapparat satt på styrkenivå tre. Ti mikroliter fjernes, proteinet bestemmes ved metoden til Lowry, Anal. Biochem. 100: 201-220 (1979). Rørene sentrifugeres deretter (5 min., 10.000 xg), 100 |xl av supernatanten fjernes og nivået av cyklisk GMP (cGMP) måles ved bruk av New England Nuclear (Boston, Massachusetts) cGMBP RIA måling ifølge metoden til leverandøren. Dataene er rapportert som pmol cGMP dannet pr. mg. protein. Uønsket hypotensiv aktivitetet bestemmes også ved kjente metoder, f.eks. ifølge metodene til Carron et al., også sitert ovenfor.
En alternativ og foretrukken fremgangsmåte for evaluering av neurobeskyttende aktivitet er metoden til Ismail A. Shalaby, et al., J. Pharm, and Experimental Therapeuics, 260, (1992) som herved inngår som henvisning og beskrives nedenunder.
Cellekultur
Sytten dagers fetale (CD, Charles River Avlslaboratorier, Inc., Wilington, MA) hippokampale celler dyrkes på PRIMARIA kulturplater (Falcon Co., Lincoln Park, NJ) i 2 til 3 uker i serum inneholdende kulturmedium (minimum essensiell medium med ikke-essensielle aminosyrer inneholder 2 mM glutamin, 21 mM glukose, penicillin/ streptomycin (5000 enheter hver), 10% fetalt bovinserum (dag 1-7) og 10% heste-serum (dag 1-21) (Choi et al., 1987). Cellene plates enten på 96-brønners mikro-titerplater på en densitet på 80.000 celler pr. brønn eller på 24-brønners kulturplater ved en densitet på 250.000 celler pr. brønn. Kulturer ble dyrket ved 37°C i en fuktet CO2 vevs kulturinkubator innholdende 5% CC*2-95% luft. Formering av ikke-neurale celler ble kontrollert ved tilsetning av 20 fiM uridin og 20 ^iM 5-fluor-2-deoksyuridin (Sigma Chemical Co., St Louis, MO) fra dag 6 til 8 dyrking (Martin et al., 1990). Dyrkingsmedier ble skiftet hver annen til tredje dag med friskt stammedium.
Glutamat toksisitet
Kulturene ble bestemt med hensyn til glutamat toksisitet 2 til 3 uker fra initial plating. Kulturmediene ble fjernet og kulturene renset to ganger med CSS (Choi et al., 1987) (i millimolar): NaCI, 120; Kcl, 5,4; MgCI2, 0,8: CaCI2,1,8: glukose, 15; og 4-(2-hydroksyetyl)-1-piperazinetansulfonsyre, 25 mM (pH 7,4). Kulturene ble deretter i 15 min. (37°C) utsatt for forskjellige konsentrasjoner av glutamat. Etter denne inkuberingen ble kulturene renset 3 ganger med glutamatfri CSS og to ganger med friskt kulturmedium uten serum. Kulturene ble deretter inkubert i 20 til 24 timer i serumfritt kulturmedium. Droger ble tilsatt 2 min. før og under 15 minutters eksponeringen for glutamat. I noen eksperimenter ble droger tilsatt på forskjellige tidspunkter etter glutamateksponeringen og i de følgende 20 til 24 timer.
Cellelevedyktighet ble rutinemessig målt 20 til 24 timer etter eksitotoksinen eksponeringen ved å måle aktiviteten til cytosolenzymet LDH (Koh og Choi, 1987; Wroblewski og LaDue, 1955). LDH-aktiviteten ble bestemt fra dyrkningsmediet til hver av de 96 brønner i mikrotiterplatene. En 50 |il prøve på mediet ble så satt til et likt volum natriumfosfatbuffer (0,1 M, pH 7,4) inneholdende 1,32 mM natriumpyrovat og 2,9 mM NADH. 340 nm absorbansen til den totale reaksjonsblanding for hver av de 96 brønner ble kontrollert hvert 5. sek. i 2 min. med en automatisert spektrofoto-metrisk mikrotiterplateavleser (Molecular Devices; Menlo Park, CA). Absorbans-graden ble beregnet automatisk ved bruk av et IBM SOFTmax program (versjon 1.01; Molecular Devices) og ble brukt som indeks for LDH-aktivitet.
Morfologisk bestemmelse av neuronal levedyktighet ble bestemt ved å bruke fasekontrastmikroskopi. 96-brønners kulturplatene tillot ikke gode fasekontrastbilder, så celler dyrket på 24-brønners plater ble brukt for dette formål. Kvantitativt var begge kulturplater like sensitive for glutamattoksisitet og viste 2- til 3-gangers økning i LDH aktivitet 241 etter eksponering for 0,1 til 1,0 mM glutamat.
Reagenser
DTG ble kjøpt fra Aldrich Chemical Company (Milwaukee Wl) og haloperidol fra Research Biochemicals Inc. (Natick, MA). Spermin ble kjøpt fra Sigma Chemical Co. (St. Louis, MO). Heste- og fetalt bovinserum ble kjøpt fra Hyclone (Logan, UT). Kulturmedium, glutamin og penicillin/streptomycin ble kjøpt fra Gibco Co. (Grand Island, NY).
Dataanalyse
Neurotoksisitet ble kvantifisert ved måling av aktiviteten av LDH som var tilstede i kulturmediet 20 til 24 timer etter glutamateksponering. Våre begynnelses-eksperimenter fastslo publiserte rapporter som viste at den økende LDH-aktivitet i kulturmediene korrelerer med destruksjon og degenerering av neuroner (Koh og Choi, 1987). Fordi aktuelle nivåer av LDH varierte fra forskjellige kulturer, ble data rutinemessig uttrykt i forhold til bufferbehandlende søsterbrønner i den samme kulturplate. For å oppnå en indeks for LDH aktivitet fra glutamat og drogebehandle-kulturer ble LDH verdiene fra kontrollkulturen trukket fra sådanne for behandlings-gruppene. Data for drogebehandlinger ble uttrykt som en prosentdel av økningen i LDH indusert med 1 mM glutamat (eller NMDA) for hvert eksperiment. Konsentrasjoner av NMDA antagonister ble krevet for å reversere 50% av LDH økningen indusert med eksitotoksinet (IC50) ble beregnet ved å bruke log-probitanalyse fra det sammenslåtte resultater av tre uavhengige eksperimenter. Forskjellige behandlings-grupper ble sammenlignet ved bruk av en «two-tailed t test».
Slike selektive neurobeskyttende antiischemiske og eksitatoriske aminosyre-blokkeringsaktiviteter viser den verdifulle anvendelighet av foreliggende forbindelser ved behandlig av degenerative CNS (sentralnervesystem) forstyrrelse så som slag og Alzheimers sykdom, Parkinsons sykdom og Huntingtons sykdom, uten signifikant potensiale for samtidig uønsket fall i blodtrykk. Ved den systemiske behandling av slike sykdommer med neurobeskyttende mengde av forbindelser med formel (I) er dosen typisk fra ca. 0,02 til 10 mg/kg/dag (1-500 mg/dag hos et typisk menneske som veier 50 kg) i enkle eller oppdelte doser uavhengig av administreringsveien. Selvfølgelig kan, avhengig av den enkelte forbindelse og den nøyaktige art av den individuelle sykdom, dose utenfor dette område foreskrives av den behandlende lege. Den orale administreringsvei foretrekkes i alminnelighet. Hvis pasienten imidlertid ikke er i stand til å svelge, eller oral absorpsjon på annen måte er nedsatt, vil den foretrukne administreringsvei være parenteral (i.m., i.v.) eller topisk.
Forbindelsene ifølge den foreliggende oppfinnelse kan administreres i alminnelighet i form av farmasøytiske blandinger omfattende minst en av forbindelsene med formel (I), sammen med farmasøytisk akseptabel bærer eller fortynnings-middel. Slike blandinger formulerer generelt på en vanlig måte ved bruk av faste eller flytende bærere eller fortynningsmidler som passer formen for ønsket administrering; for oral administrering i form av tabletter, harde eller myke gelatinkapsler, suspensjoner, granulater, pulvere eller lignende; for parenteral administrering i form av injiserbare løsninger eller suspensjoner eller lignende; for topisk administrering i form av løsninger, lotioner, kremer, salver og lignende.
Oral administrering foretrekkes, og oral biotilgjengelighet er en viktig parameter ved valg av forbindelser som er spesielt anvendelige som NMDA antagonister. Oral biotilgjengelighet kan evalueres ved metoden som er angitt av Schmidt og Bubser (Pharmacol. Biochem. Behavior, 1989, 32, 621) som måler en oral dose av forsøksforbindelsens evne til å reversere katalepsi indusert med haloperidol.
Sprague-Dawley hanrotter (Interfauna, Tutlingen, F.R.G.) fikk intraperitoneale (IP) injeksjoner av 0,5 mg/kg haloperidol (injiserbar løsning fra ampuller, Jansen, Neuss, (F.R.G.)) for å indusere en moderat grad av katalepsi. Samtidig ble enten en forbindelse med formel (I) i en inert bærer eller den inerte bærer administrert oralt. Løsningen for oral administrering ble fremstilt ved å oppløse forbindelsen i 0,3% vandig vinsyre. Fast vinsyre ble tilsatt for å bringe pH til 3,5. Tretti minutter etter behandling ble katalepsigraden målt hos hver rotte. Tre etablerte tester kan utføres i den følgende rekkefølge:
1) Plassering av begge forben på en horisontal stang 9 cm over flaten.
2) Plassering av et forben på et podium (3 cm høyt).
3) Henging på et vertikalt trådnett.
Tidsrommet fra plassering av potene inntil den første bevegelse av en av disse poter (descent latens) ble målt (i minst 180 sek.).
Forskjellene mellom grupper ble analysert ved å bruke den to-halede Mann-Whitney U-test. En p-verdi <0,05 ble ansett å vise en signifikant forskjell mellom grupper.
Vi fant uventet at forbindelsen med formel (I) er signifikant mer effektiv i en oral dose enn andre forbindelser som er beskrevet i europeisk patentsøknad 441506
Den foreliggende oppfinnelse er illustrert ved de følgende eksempler.
MELLOMPRODUKTER OG EKSEMPLER
Alle ikke-vandige reaksjoner ble kjørt under nitrogen for sikkerhets skyld og generelt for å maksimalisere utbytter. Alle løsningsmidler/fortynningsmidler ble tørket i følge publiserte standardmetoder eller kjøpt i en foruttørket form. Alle reaksjonssatser ble rørt enten magnetisk eller mekanisk. NMR spekteret ble tatt opp ved 250 eller 300 Mhz og er angitt i ppm. NMR løsningsmiddelet var CDCI3 med mindre annet er angitt. IR spekteret er rapportert i cm"<1>, generelt bare med angivelse av sterke signaler. De følgende forkortelser ble brukt; DMF for dimetyl-formamid, THF for tetrahydrofuran, HRMS for høyoppløsningsmassespektrum.

7-benzyloksykromanon
En blanding av 7-hydroksykromanon (10,0 g, 61 mmol), benzylbromid (7,6 ml, 64 mmol) og kaliumkarbonat (16,5 g, 120 mmol) i aceton (250 ml) ble forsiktig tilbakeløpskokt i 24 timer. Blandingen ble avkjølt og filtrert gjennom celitt. Filtratet ble inndampet og resten ble tatt opp i etylacetat. Den organiske løsningen ble vasket med vann og saltvann, tørket over kalsiumsulfat og inndampet til et brunt fast stoff. Omkrystallisering fra etanol gav 12,63 g (81%) av 7-benzyloksykromanon som brune krystaller som hadde smeltepunkt 99-102°C; NMR 5 7,85 (d, J=9 Hz, 1H); 7,44-7,32 (m, 5H), 6,66 (dd, J=2,4, 9 Hz, 1H), 6,49 (d, J=2,4 Hz, 1H), 5,09 (s, 2H), 4,52 (t, J=6,5 Hz, 2H), 2,76 (t, J=6,5 Hz, 2H). 7-benzyloksy-3,3-dibromkromanon
Til en oppslemming av 7-benzyloksykromanon (12,63 g, 49,7 mmol) i karbontetraklorid (200 ml) og etylacetat (100 ml) ble tilsatt en løsning av brom (5,38 ml, 104,4 mmol) i karbontetraklorid (100 ml) i løpet av 20 min. Den klare røde løsning ble rørt 10 min lenger, deretter ble hydrogenbromid drevet ut av løsningen med en nitrogenstrøm (15 min). Den organiske løsningen ble vasket med vandig natrium-bisulfit, vandig natriumbikarbonat og saltvann; deretter ble den tørket over kalsiumsulfat og inndampet til en olje som langsomt størknet. Omkrystallisering fra etanol gav 15,73 g (76%) av 7-benzyloksy-3,3-dibromkromanon som lyserøde krystaller som hadde smeltepunkt 89-90°C, NMR 6 796 (d, J=9 Hz, 1H); 7,43-7,36 (m, 5H), 6,79 (dd, J=2,4, 9 Hz, 1H), 6,57 (d, J=2,4 Hz, 1H), 5,12 (s, 2H), 4,71 (s, 2H). Eksempel 1
3RMS*3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kroman-4,7 -diol
En blanding av 7-benzyloksy-3,3-dibromkromanon (54,7 g, 133 mmol), 4-(4-fluorfenyl)-4-hydroksy-piperidin (52,06 g, 266 mmol), og trietylamin (38 ml, 270 mmol) i acetonitril (2,5 I) ble rørt 16 timer ved romtemperatur. En gul felling ble dannet og samlet, vasket godt med vann og eter og lufttørket. Utbyttet av 7-benzyloksy-3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kromanon var 55,4 g (93%) som var egnet for bruk uten videre rensing. En prøve omkrystallisert fra etanol/tetrahydrofuran hadde smeltepunkt 220-221°C; NMR DMSOd6 6 7,99 (d, J=9 Hz, 2H), 7,56-7,40 (m, 8H), 7,18-7,08 (m, 4H), 5,25 (s, 2H), 5,06 (s, 1H), 3,60 (brs, 1H), 3,55-3,35 (m, 1H, delvis skjult av vann fra NMR løsningsmiddelet), 3,10-2,95 (m, 2H), 2,15-2,00 (m, 2H), 1,71 (br t, J=113,7 Hz, 2H). En oppslemming av 7-benzyloksy-3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kromanon (8,24 g, 18,5 mmol) i etanol (400 ml) og tetrahydrofuran (600 ml) ble tilsatt natriumborhydrid (7,0 g, 185 mmol). Blandingen ble rørt natten over. Ytterligere natriumborhydrid (7,0 g) ble tilsatt og reaksjonen ble rørt i 3 dager. Vann ble tilsatt, og løsningsmiddelet ble fjernet ved redusert trykk ved 45°C. Faststoffene som ble dannet, ble samlet og vasket godt med vann og deretter med eter. En blanding av 3R<*>4S<*> 7-benzyloksy-3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kroman-4-ol (15,22 g, 33,86 mmol), N-tert-butoksykarbonyl-L-prolin (14,65 g, 68,06 mmol),4-dimetylaminopyridin (4,18 g, 34,21 mmol), og 1-etyl-3-(3-dimetyl-aminopropyl)-karbodiimidhydroklorid (13,10 g, 68,3 mmol) i metylenklorid ble forsiktig tilbakeløpskokt i 3 timer. Blandingen ble inndampet ved redusert trykk, og resten ble fordelt mellom etylacetat og vann. Fasene ble adskilt, og det organiske sjiktet ble vasket med vann og saltvann; deretter ble den tørket over kalsiumsulfat og inndampet til et gult skum. Isopropyleter (200 ml) ble tilsatt, og blandingen ble opp-varmet kort til kokepunktet. Denne blandingen fikk vende tilbake til romtemperatur og rørt natten over. Bemerk at isopropyleterfiltratet inneholdt det diastereomere N-tert-butoksykarbonyl-L-prolinesterprodukt som kunne brukes til å fremstille det tilsvarende (-) tittelproduktet.
Produktet fra reaksjonen ovenfor (0,262 g, 0,405 mmol) ble satt til 0°C oppslemming av litiumaluminiumhydrid (0,020 g, 0,527 mmol) i tetrahydrofuran (10 ml). Blandingen fikk oppvarmes til romtemperatur og ble rørt i 10 min. Reaksjonen ble avbrutt med natriumsulfat decahydrat, tørket med natriumsulfat og filtrert gjennom celitt. En blanding av (+) (3R, 4S)-3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kroman-4,7 idol (2,11 g, 5,87 mmol) og vinsyre (0,885 g, 5,90 mmol, Mallinckrodt, høyreroterende med levo konfigurasjon) i etanol (20 ml under oppvarming) ble inndampet til et amorft lysegult fast stoff som ble tørket i vakuum natten over og gav 2,995 g (100%) av (+) (3 R,4S)-3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kroman-4,7-diol-tartrat-etanolat-hydrat som hadde smeltepunktområde 85-95°C; [oc]d = +55,6°C c=1,140 (metanol);<1> HNMR DMSOd6 6 7,53 (dd, J=5,6, 8,7 Hz, 2H), 7,15 (t, J=8,9 Hz, 2H), 7,08 (d, J=8,4 Hz, 1H), 6,81 (brs, 7H), 6,40 (dd, J=2,2, 8,3 Hz, 1H), 6,22 (d, J=2,2 Hz, 1H), 4,85 (s, 1H), 4,45 (d, J=-8,4 Hz, 1H), 4,21-4,13 (m, 3H), 3,44 (q, J=7,0 Hz, 2H) 3,34-3,08 (m, 5H), 2,19 (br, q, J=12,9 Hz, 2H), 1,72 (brt, J=11,8 Hz, 2H), 1,06 (t, J=7,0 Hz, 3H) <13>C NMR DMSOd6 6 174,02, 162,62, 159,41, 158,56, 154,24, 145,10, 131,80, 126,87, 126,77, 114,72, 114,45, 108,80, 101,94, 72,11, 68,55, 61,50, 61,08, 60,35, 56,08, 46,57 46,09, 36,39, 36,24, 18,57. Resultatene viser at forbindelsen ifølge foreliggende oppfinnelse er ti ganger mer aktiv når det gjelder reversering av katalepsi induset av haloperidol enn den forbindelsen som den blir sammenlignet med.
Claims (9)
1. Forbindelse, karakterisert ved at den er 3R<*>4S<*>3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kroman-4,7-diol, dens optiske isomer og farmasøytiske akseptable salter.
2. Forbindelse ifølge krav 1, karakterisert ved a t den er racemisk 3R4S 3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kroman-4,7-diol.
3. Forbindelse ifølge krav 1,karakterisert ved at den er (+) 3R4S 3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kroman-4,7-diol.
4. Forbindelse ifølge krav 1,karakterisert ved at den er (-)4R3S 3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kroman-4,7-diol.
5. Forbindelse ifølge krav 1,karakterisert ved at den er (+) (3R.4S)-3-[4-(4-fluorfenyl)-4-hydroksy-piperidin-1-yl]-kroman-4,7-diol-tartrat-etanolat-hydrat.
