Description
Foreliggende oppfinnelse angår en fremgangsmåte for fremstilling av et formlegeme som inneholder et virksomt stoff, en voks, et tyngdemiddel og eventuelt et sukker, sukkeralkohol, celluloseeter eller en polyetylenglykol for administrering til drøvtyggere.
Virkestoff-frigivende systemer er tilberedelser, som kontrollert frigir et eller flere (fysiologisk) virksomme bestanddeler etter en definert frigivelsesprofil over en bestemt tid.
De viktigste indikasjonsområdene for anvendelse av disse terapeutiske systemene i dyremedisin er: 1) terapi i snever forstand, det vil si kontroll av parasittinfeksjoner og/eller sykdommer, så vel som i videre forsdtand for 2) tilføringen av mangelstoffer (for eksempel sporelementer), 3) reguleringen av metabolske prosesser (for eksempel ytelse), 4) reguleringene av endokrine forløp (for eksempel fertili-tet).
Anvendelseområdet for anvendelse av disse systemer i dyreoppdrett er særlig forbedringen av veksten og forutnyttelsen hos drøvtyggere.
Fordeler ved de virkestoff-frigivende systemene i sammenligning med tradisjonelle prosedyrer er et optimalt virke-stof f speil under hele anvendelsestiden, en lav virkestoff-dosering, en arbeids- og derved endelig en kostnadsbesparelse for dyreholderen.
De mulig opptredende ulemper, som resistensutvikling henholdsvis tilvenning, virkestoffresistans og ventetid, vevsirritasjon og rester av fremmedlegemer etter avsluttet anvendelse kan og må gjennom en optimal galenisk utvikling, utelukkes eller i det minste bli betydelig minimalisert.
Virkestoff-frigivende systemer for oralt bruk av drøvtyggere, i det følgende kalt "Bolus", er i faglitteraturen utførlig beskrevet (for eksempel Formulation of Veterinary Dosage Forms, av Jack Blodinger, Marcel Dekker Inc. New York, 1983). Oppholdet av Bolusen i dyrets gastrotrakt blir bevirket gjennom geometriske foranstaltninger eller gjennom tettheten på systemet, i siste tilfelle snakker man om en "high density device" (HDD). Fra litteraturen er det kjent at HDD skal ha en tetthet på mer enn 2 g/cm<3>(J.Riner et al., Am. J. Vet. Res. 4J3 (1982 ), S. 2038-2038), hvorved Bolusen trass i drøvtyggernes naturlige gjenoppgulping ikke igjen kan gulpes opp.
De over beskrevne "high density device" er derfor utstyrt med et vektelement, for å oppnå den nødvendige totaltettheten. Dette tyngdeelementet blir foretrukket utført som en massiv metallblokk for eksempel EP-A.0-333.311, EP-A-0 149.510, EP-A-0.062.391 eller som sylinder for eksempel EP-A-0.164.241 og i Vet. Parasitology 12 (1983) 223-232. Disse i litteraturen beskrevne metallelementene har den ulempen at de ikke er biodegraderbare og at de etter den ønskede brukstiden forblir som fremmedlegemer i dyrenes mage. Uheldige følger av dette er blant annet interaksjoner med andre bolus-systemer for gjentatt bruk, irritasjon av slimhinner og problemer med slakting og partering av dyrene.
Et visst fremskritt for overvinning av disse ulempene byr anvendelsen av nedbrytbare vektelementer på, som er beskrevet i EP-A-0.332.094. Riktignok er heller ikke de der beskrevne systemene optimalt egnet. De er sammensatt av flere ganske forskjellige komponenter. Disse komponentene er blant annet
a) en blanding (matrise) av en nedbrytbar polymer og virkestoff, b) en blanding (matrise) av en nedbrytbar polymer
og små biter med stor tetthet, c) en nedbrytbar polymer og d) smådeler med stor tetthet.
Den kostbare oppbyggingen av systemet er en ulempe, og består vanligvis av en adskilt fremstilling for senere sammenføying av enkeltdelene. Den maksimale affiserbare virkestoffmengden blir sterkt begrenset gjennom det begrensede volumet til den nedbrytbare polymeren. Dessuten må man legge merke til at under oppbyggingen av de forskjellige polymerene eller under den mekaniske påvirkningen i fordøyelsessystemet lett kan føre til en separering av de enkelte delene i systemet. De beskrevne bolus-systemene fremviser derfro kun i begrenset grad de praktiske krav for fremstilling og terapeutisk sikkerhet.
I EP-A-0.243.111 blir det til slutt beskrevet blant annet bolus-systemer, som er oppbygget av formlegemer, som under virkestoffet inneholde et jerngranualt og grafitt så vel som et hylster av magnesiumlegering og et beskyttelseshylster av ikke-nedbrytbart kunststoff. Magnesium, jern og grafitt danner i magesaften et galvanisk element, som bevirker korrosjon av magnesiumhylsteret. På denne måten blir virkestoff-frigjøringen styrt.
Ulempen med dette systemet er den samtidige frigivingen av magnesiumioner. Magnesium er en fysiologisk virksom substans og blir i veterinærmedisinen brukt som legemiddel, for eksempel mot gress-tetani hos kalver.
Fremstillingen av de ovennevnte bolus-systemene er til sammenligning kostbare og kostnadsintensive. Pressingen av jerngranulatet til formen bevirker en betydelig avrivning på det anvendte pressverktøyet. Den derav resulterende korte levetiden fører til en forhøying av produksjonskostnadene. Anvendelsen av et ikke-nedbrytbart kunststoff er ikke uproblematisk, siden kunststoffet etter endt virketid for bolusen blir igjen som fremmedlegeme i dyret eller blir utskilt og belaster omgivelsene.
Et praktisk rettet bolus-system må oppfylle følgende krav:
1) enkel, sikker administrerbarhet,
2) fullstendig retensjon på virkestedet under hele tiden for virkestofffrigivning,
3) kontrollert virkestoff-frigivning med klart mål,
4) ingen uønskede bivirkninger og ingen vevsskader,
5) ingen gjenværende fremmedlegemer etter avslutningen av behandl ingen.
Boliene fremstilt ifølge EP 0.385.106 og NO 172728, som begge er fremstilt ved smeltefremgangsmåter, oppfyller de ovenfor nevnte mål. Imidlertid er de tiltak som er beskrevet i de nevnte patenter kostbare, slik at det er ønskelig med en mindre kostnadseffektiv fremgangsmåte for fremstilling av slike boli.
Kravene ovenfor blir overraskende oppfylt på en enklere og rimeligere måte av et bolus-system, bestående av et formlegeme fremstilt ved en fremgangsmåte for fremstilling av et formlegeme for oral administrering til drøvtyggere, hvilket formlegeme inneholder 0,01 til 75 vekt-# av minst et virkestoff, 3 til 75 vekt-# voks, 25 til 90 vekt-# pulverformig tyngdemiddel og 0 til 30 vekt-# av minst en fysiologisk fordøybar sukker, sukkeralkohol, vannløselig celluloseeter eller polyetylenglykol, kjennetegnet ved at man fremstiller en granulatsmelte av formlegemets bestanddeler i en egnet blander under utnyttelse av friksjonsvarme, hvor granulatsmelten etter avkjøling, uten ytterligere oppvarming, blir presset til et formlegeme.
En bolus-tilberedning for oral administrering til drøvtyg- gere, fremstilt ifølge foreliggende fremgangsmåte, inneholder
0,001 til 75 vekt-#, fortrinnsvis inntil 50 vekt-#, aller helst opptil 30 vekt-# av minst et virkestoff,
3 til 75 vekt-56, fortrinnsvis 5 til 60 vekt-#, aller helst 5 til 50 vekt-# voks, 25 til 90 vekt-#, fortrinnsvis 30 til 85 vekt-#, aller helst 40 til 80 vekt-# pulverformig tyngdemiddel og 0 til 30 vekt-#, fortrinnsvis 0 til 14 vekt-#, aller helst 0 til 10 vekt-# av minst et fysiologisk lettfordøyelig sukker, sukkeralkoholer, vannløselige celluloseetere eller polyetylenglykoler eller blanding av disse.
Dersom et virkestoff ikke eller kun i liten grad er vann-løselig (som for eksempel Fenbendazol), er det fordelaktig at andelen av sukker, sukkeralkohol, vannløselig celluloseeter eller polyetylenglykol ikke senkes under 1 vekt-#. Det er da foretrukket med andel fra 1,5 til 27 vekt-#, aller helst 2 til 25 vekt-5é.
Tilberedningen kan dessuten i tillegg inneholde opptil 8 vekt-#, fortrinnsvis opptil 6 vekt-# av et overflateaktivt stoff, opptil 10 vekt-#, fortrinnsvis opptil 6 vekt-# slipmiddel, et smøremiddel og/eller stoffer som påvirker tilberedningens egenskaper.
Med drøvtyggere forstår man spesielt storfe, får og geiter.
For terapeutiske virkestoffer dreier det seg i sæedeles om midler for kontroll av parasittinfeksjoner og/eller sykdommer, for tilføring av mangelstoffer, for regulering av metabolske prosesser og for regulering av endokrine forløp.
De følgende nevnte, terapeutiske virksomme stoffgruppene og forbindelsene kan eksempelvis bli satt inn i bolus-tilberedningen fremstilt ifølge foreliggende fremgangsmåte: Glucokortikoider for fødselsinduksjon, for eksempel
dexametason, betametason, flumetason og deres estere og
derivater,
gestagen for brunstsynkronisering, brunst- og løpetids-undertrykking,
^-adrenergika f°r behandling og profylakse for respira-sjon s sykdommer, for forhindring av abort og fødsel, for veksthjelp og stoffskifteforstyrrelser, som for eksempel clenbuterol,4-(2-tert.-butylamino-l-hydroksyetyl )-2-cyano-6-f luor-f enylkarbaminsyre-etylester-hydroklor id ,a[ [ [3-( 1-benzimidazolyl)-l,1-dimetylpropyl]-amino]-metyl]-2-fluor-4-hydroksy-benzyl alkohol-metansulfanat monohydrat, (cimaterol), l-(4-amino-3-cyanofenyl)-2-iso-propylamino-etanol,

Foretrukket er virkestoffer fra gruppen benzimidazol-, benztiazol-, derivater eller pro-benzimidazolen, i sær-deleshet forbindelser med formel I, II eller III

For forbedring av veksten og forutnyttelsen for drøvtyggere kan for eksempel de etterfølgende nevnte stoffgruppene og forbindelsene bli tilsatt bolustilberedningen ifølge foreliggende oppfinnelse.
Salinomycin, flavofosfoliol, monensin, dehydrometylmonensin, narsin, desoksynarsin, lacalocid A. alborixin, lysocellin, nigericin, dehydroksymetylnigericin, dianemycin, ionomycin, norbitomycin og andre polyeterantiobiotika, avoparcin, spiramycin, tylosin, virginiamycin, bacitracin A, carbadox, nitrovin, olaquindox, amprolium, arpinacid, dinitolmid, halofuginon, lasalocid A, metikloroindol, nicarbazin, decoquinate, triazinderivat og salter av nevnte forbindelse! som salinomycin-Na, monensin-Na, Tylosin-fosfat, Zn-bacitracin A, lasalocid A-Na.
Særlig foretrukket er fosfo-glykolipid, i særdeleshe* flavofosfolinol, salinomycin så vel som deres salter fortrinnsvis natriumsalter.
Under- voks i betydningen for anvendelse i forbindelse mei foreliggende oppfinnelse, forstår man et fysiologisk ufarlij naturlig eller kunstig fremstilt stoff som fremviser følgendi egenskaper: ved romtemperatur fast til hard og sprø, stor til finkrystallinsk, gjennomskinnende til opak, likevel ikki glassaktig; smelter over 40°C uten tilsetninger, likevel forholdsvis lav-viskøs og ikke-trekkbar til tråer rett over smeltepunktet, sterk temperaturavhengig konsistens og løselighet.
De skiller seg hovedsakelig fra andre syntetiske eller naturlige produkter (for eksempel harpikser, plastiske masser) disse smelter som regel i området fra omtrent 50 til 90°C. Egnet er for eksempel vokser som kan candelilla-, carnauba-vokser og fossile vokser som montanvoks og så videre, mineralvokser som cerasin, ozokerit (jordvoks), petrolatum, parafin- og mikrovokser; kjemisk endrede vokser som for eksempel de av Hoechst- og BASF-fremstilte gjennom oksydasjon av rohmontanvoks og hydrerte jojoba-oljer eller rizinus-oljer (for eksempel cutin HR) så vel som syntetiske vokser.
Det pulverformige tyngdemiddelet blir valgt under hensyntagen til sin spesifikke tetthet, sin sammensetning og sin mengde, slik at bolus-systemet trass i den naturlige oppgulpingen forblir i magen. Avhengig av bolussystemets størrelse og dets overflate skal dets spesifikke tetthet etter sammensetningen med de tetthetsgivende delene i alminnelighet være større enn 3,0 g/cm<3>, fortrinnsvis større enn 4,0 g/cm<3>.
Oppover er tettheten intet innskrenkende kriterium.
Eksempelvis er oppført tyngdemidler som har en tilstrekkelig høy tetthet:
Ikke-toksiske stoffer blir foretrukket tilsatt, slik som bariumsulfat eller jernpulver, fortrinnsvis jernpulver.
Av stoffene med høy tetthet blir partikkelstørrelsen mindre enn 1 mm, eventuelt også mindre enn 0,1 mm foretrukket. Således har det i foreliggende oppfinnelse anvendte jern-pulveret partikler hvor 975°; < 0,15 mm, 80% < 0,1 mm.
For vannløselige virkestoffer kan andelen av sukker, sukkeralkohol, vannløselig celluloseeter eller polyetylenglykol bli redusert eller disse hjelpestoffene kan helt bortfalle.
Egnede sukkere ifølge foreliggende oppfinnelse er eksempelvis vannløselige farmasøytisk akseptable monosaccharider og disaccharider. Lactose er foretrukket, men også glycose, fructose, xylose, galactose, sucrose, maltose, saccharose og beslektede forbindelser som mannit, xylit og sorbit er egnet.
Egnede vannløselige celluloseetere er ®tylose-marken, ®tylose MB (metylcellulose) og ®tylose H (hydroksyetylcellulose), hvor metylcellulose er foretrukket.
Polyetylenglykoler (PEG) i foreliggende oppfinnelses mening er for eksempel PEG 400, 600, 1500, 2000, 3000, 4000, 6000 og 10000. De faste typene er foretrukket.
Av aktuelle overflateaktive stoffer blir fysiologisk fordøybare ikke-ioniske tensider med en HLB-verdi mellom 10 og 20 foretrukket, spesielt polyoksyetensorbitanesterene som monolaurat (®tween 20), monopalmitat (®tween 40) og mono-stearat (®tween 60). ®Tween 20 er foretrukket.
Som smøremiddel og formslippmiddel blir vanligvis talk, stearat eller andre metallsalter slik som fumarat eller palmitat tilsatt, fortrinnsvis magnesiumstearat.
Som hjelpestoffer som kan ha inflytelse på de mekaniske egenskapene, særlig avrivning og oppløsning, blir vanligvis finfordelt silisiumdioksyd (®aerosil) tilsatt.
Formuleringer for oral administrering til drøvtyggere, fremstilt ifølge foreliggende fremgangsmåte, består av et ovenfor definert middel, som inneholder en utregnet mengde av virkestoffet for en definert frigivelse i løpet av en fastsatt tid og eventuelt en hylse som er oppløselig eller degraderbar i fordøyelsessystemet.
Bolusen henholdsvis de enkelte tablettene har til hver dyreart en diameter som ligger mellom 1 og 4cm. Totallengden av bolusen skal ikke overskride 12 cm, hvor nedre grense er en enkel tablett. Bolusen kan bestå av et stykke (figur 1) eller bestå av et ønsket antall av enkeltstykker (for eksempel tabletter) (figur 2 og 3).
Totalvolumet kan derved ligge mellom 0,5 og 200m<3>.
Gjennom forstørrelsen av overflaten, det vil gjennom overgang fra et stykke (figur 1) til tablettformen (figur 2, 3) kan frigivningsraten ved samme sammensetning av matrisen bli forhøyet alt etter som antall, størrelse og tablettformen blir tilpasset kravene. Tapet av enkelttabletter under behandlingstiden, for eksempel gjennom oppgulping, blir forhindret ved at tablettene blir forbundet med hverandre. Denne sammenbindingen kan være fleksibel, for eksempel ved et bånd, en tråd eller en kjede, elastisk eller stiv for eksempel ved en stav (figur 2, 3). Forbindingsmaterialet kan eksempelvis være laget av fysiologisk ufarlig kunststoff (for eksempel silikon) eller av et fysiologisk ufarlig materiale, som blir ndbrutt i tarmen.
En tablett-bolus (figur 2, 3) unngår dessuten faren for at matrisen gjennom ytre påpvirkning kan bli knust henholdsvis beskadiget. Slik blir en rettmessig frigivelse under hele administreringstiden ivaretatt.
Høyden på tabletten kan variere avhengig av den ønskede frigivningsprofilen av virkestoffene. Likeledes er forskjellige tablettformer, for eksempel biplan, bikonvex mulige. I en bolus kan også tabletter med forskjellig høyde og form bli kombinert med hverandre. Likeledes er det mulig å kombinere tabletter som inneholder forskjellige konsen-trasjoner av virkestoff eller forskjellige virkestoffer med hverandre. For forenkling av bruken kan de med hverandre forbundne tablettene være sammenklebet ved hjelp av et vannoppløselig klebestoff eller være omgitt av en i fordøy-elsestrakten oppløselig eller degraderbar hylse. Materialet i klebestoffet henhodlsvis i hylsen kan for eksempel bestå av cellulose og cellulosederivater, gelatin og andre egnede polymerer og kopolymerer. Tablettene kan også være omgitt av et nett.
De på ovennevnte fremgangsmåte fremstilte produktene kan bestå av opptil 30, fortrinnsvis opptil 20 og helst opptil 10 enheter. Matrisen blir i løpet av behandlingstiden fullstendig nedbrutt; etter avslutning av behandlingen blir ingen rester tilbake.
Det beskrevne virkestoff frigivelsessystemet er i prinsippet egnet for bruken av alle virkestoffer som i området vete- rinærmedisin og dyrenæring skal bearbeides over lengre tidsrom. Forskjellige fysikalske egenskaper hos virkestoffet, som for eksempel løselighetsforskjeller, kan bli påvirket gjennom egnede tilsatsstoffer som styrer virkestoff-frigivingen. Egnede tilsatsstoffer for regulering av fri-givingsprofilen i henhold til de terapeutiske krav er kjent for fagmannen.
Foreliggende oppfinnelse angår således også fremstillingen av de ovennevnte enheten og frembringelsen for profylakse og/eller behandling av drøvtyggere av sykdommer så vel som påvirkningen av veksten, stoffskiftet, kroppsvekten, vevssammensetningen og/eller forutnyttelsen.
De underfølgende eksemplene tjener som forklaring av oppfinnelsen, uten å være begrensende for den.
A. Alminnelig fremstillingseksempel av en smelte.
Voksen (for eksempel Hoechst voks E eller hydrert rizinus-olje, som cutina HR) blir smeltet i en beholder (temperatur ca 80 til 100°C).
Under omrøring blir virkestoffet, tyngdemiddelet (for eksempel jernpulver) og de øvrige hjelpestoffene tilsatt.
Den tregtflytende smeiten blir overført i egnede forvarmede former og langsomt avkjølt til romtemperatur.
B. Alminnelig fremstillingseksempel for en granulatsmelte
Formuleringens bestanddeler blir veid opp i en egnet blander (for eksempel fluidblander Fa. Henschel). Den oppståtte varmen fra friksjonen i blanderen fører til dannelse av en granulatsmelte. Etter avkjøling av granulatet til romtemperatur blir agglomeratet findelt ved passering gjennom en sil (for eksempel frewitt granulator, maskevidde:lmm ).
Det på denne måten standardiserte granulatet blir presset til de ønskede enhetene med en presse. Ved behov kan et smøre-middel henholdsvis formsliptmiddel bli tilsatt. De fremstilte tilberedningene i de følgende eksemplene har fortrinnsvis følgende sammensetning (angitte prosenter er vektprosent):
Virkestoff: opptil 30%
Voks: 5 - 50%
Jern: 40 - 80%
Lactose, metylcellulose eller polyetylenglykol: 0 - 25% Finfordelt silisiumoksid: opptil 2%
Polyoksyetylen-sorbitanmonolaurat: opptil 6%
Mg-stearat: opptil b%
På analogt vis ble salinomycin-Na inneholdende tilberedninger fremstilt.
Hvis voksandelen ligger under 20%, blir smeiten høyviskøs og fremstillingen kritisk; ved voksandler < 10% er ikke smelte-fremgangsmåten anvendbar.
Claims (2)
1.
Fremgangsmåte for fremstilling av et formlegeme for oral administrering til drøvtyggere, hvilket formlegeme inneholder 0. 01 til 75 vekt-# av minst et virkestoff, 3 til 75 vekt-# voks, 25 til 90 vekt-# pulverformig tyngdemiddel og 0 til 30 vekt-# av minst en fysiologisk fordøybar sukker, sukkeralkohol, vannløselig celluloseeter eller polyetylenglykol,karakterisert vedat man fremstiller en granulatsmelte av formlegemets bestanddeler i en egnet blander under utnyttelse av friksjonsvarme, hvor granulatsmelten etter avkjøling, uten ytterligere oppvarming, blir presset til et formlegeme.

Fremgangsmåte for fremstilling av formlegemene ifølge krav 1,karakterisert vedat man knuser den fremstilte granulatsmelten og presser til de ønskede formstykkene med en presse, og utstyrer de oppnådde formstykkene med en hylse som er oppløselig eller nedbrytbar i fordøyeIsestrakten.
