Abstract

Den foreliggende oppfinnelse er relatert til en fremgangsmåte av filamentøs fagfremvisning for polypeptidene av interesse fremvist på fagpartikkelen er kotranslasjonell translokert over den cytoplasmiske membranen av gramnegative bakterier basert på signalgjenkjermings partikkelvelen. Derme fremgangsmåten er spesielt egnet for polypeptider, som er kjent til å være vanskelig å fremvise på fag, og for proteiner av cDNA bibliotek og andre kombinasjonsmessige bibliotek, spesielt når derivert fiia veldig raskt foldende, stabile proteinskaffblder. Oppfirmelsen er videre relatert til fag eller fagmidvektorer nyttige i fremgangsmåten omfattende en genkonstruksjon kodende for et fusjonspolypeptid omfattende polypeptidet som skal bli fremvist på fagpartikkelen og en N-terminal signalsekvens som fremmer kotranslasjonell translokasjon.
