Description
Den foreliggende oppfinnelse angår en analogi-fremgangsmåte for fremstilling av en gruppe av oksimer og aminer som er muskarin agonister, hvilket gjør dem nyttige som farmasøytiske midler. Nærmere bestemt er forbindelsene azabicykliske og azacykliske oksimer og aminer.
Kognisjonssykdommer er genereltkarakterisert vedsymptomer på glemsomhet, forvirring, hukommelsestap, oppmerksomhetssvikt og/eller i noen tilfeller emosjonelle forstyrrelser. Disse symptomer kan oppstå som et resultat av den almindelige aldringsprosess og/eller fra organisk hjerne-sykdom, cerebrovaskulær sykdom, hodelesjon eller utviklingsmessige eller genetiske defekter.
Den generelle svekkelse av kognitive funksjoner som følger aldringsprosessen, er anerkjent. Det samme fenomen er observert og dokumentert hos mange lavere pattedyr, inklusive slike som rutinemessig anvendes i farmakologiske testprogrammer for screening og forutsigelse av egnethet for særlige legemidler hos høyere dyr, herunder mennesker.
Selv om kognisjonssykdommer ofte følger den generelle aldringsprosess, er presenilitet og primær degenerativ alderdomssløvsinn de mest almindelig aksepterte årsaker til mental svekkelse hos eldre. Det er beregnet at minst ti prosent av personer over 60 år før eller siden vil lide av stor mental svekkelse. Et meget større antall vil oppleve kognitiv nedbrytning tilstrekkelig alvorlig til å hemme deres aktiviteter.
Mange av symptomene på kognitive sykdommer, særlig svekket hukommelse, er forbundet med nedsatt
acetylcholinsyntese og svekkelse av cholinreceptive neuroner.
I hippokampen og den cerebrale cortex hos pasienter som lider av primær degenerativ dementia, kan f.eks nivået av enzymet cholinacetyltransferase (CAT) reduseres med opptil 90%. (Se Davies et al, The Lancet, 1976 (vol. 2):1403; Perry et al, J. Neurol. Sei., 34:247-265 (1977); og White et al, The Lancet, 1977 (vol. 1):668- 670).
Eftersom CAT katalyserer syntesen av acetylcholin fra dets prekursorer cholin og acetylcoenzym A, avspeiler tapet av CAT tapet av cholinerge eller acetylcholin-frigivende nerve-spisser i hippokampen og den cerebrale cortex. Det er over- veidende beviser for at cholinerge terminaler i hippokampen er kritisk viktige for hukommelsesdannelse.
Den cholinerge hypotese foreslår at legemidler som gjenoppretter acetylcholin-nivåer eller som efterligner virkningen av acetylcholin (dvs. er cholinmimetiske), er effektive ved korrigering av denne mangel på neurotransmitter-kjemikalier og tilveiebringer behandling av hukommel-sessvekkelsesymptomet på cerebral utilstrekkelighet. Overveiende biokjemisk, farmakologisk og elektrofysiologisk bevis understøtter den hypotese at mangler i det cholinerge system ligger til grunn for geriatrisk kognitiv dysfunksjon.

(1983)). Gamle mennesker og ikke-humane primater med svekket kognisjon viser forbedret hukommelse når de behandles med f.eks. acetylcholinesteraseinhibitorer, så som physostigmin. Disse midler øker den tilgjengelige forsyning av synaptisk acetylcholin ved inhibering av dets hydrolyse.
Aminopyridiner så som 3,4-diaminopyridin forbedrer aldersrelaterte kognitive mangler ved å øke frigivelsen av acetylcholin fra presynaptiske nerveterminaler, og således øke synaptisk acetylcholin. (Se H.P. Davies et al, Exp. Aging Res., 9:211-214 (1983)).
Det har i noen tid vært kjent at det naturlige alkaloid, muskarin, har evnen til å virke relativt selektivt ved autonome effektorceller for å produsere de kvalitativt samme virkninger som acetylcholin. To beslektede alkaloider, pilocarpin og arecolin (metylesteren av 1,2,5,6-tetrahydro-l-metyl-3-pyridinkarboksylsyre), har prinsipielt det samme virkningssted som muskarin og acetylcholin og klassifiseres således å ha "muskarin"-virkning. Selv om disse naturlig forekommende alkaloider har stor betydning som farmakologisk verktøy, er nuværende klinisk anvendelse stort sett begrenset til anvendelsen av pilocarpin som miotisk middel.
Det er nylig demonstrert at arecolin er virksomt ved forbedring av noen av symptomene på kognitive sykdommer hos pasienter som klinisk har fått stilt diagnosen presenil primær degenerativ dementia. Signifikant forbedring ble observert i et forsøk med bildegjenkjennelse efter administrering av arecolin til pasienter i en dobbel-blindundersøkelse. (Se Christie et al, Brit. J. Psychiatry, 138:46-50 (1981)).
Visse 3- eller 4-ketoksimer av l-(lavere alkyl)-1,2,5,6-tetrahydropyridiner i hvilke oksygen er usubstituert, er beskrevet i US patentskrift nr. 3.004.979 med anvendelighet som parasympatomimetiske midler som virker på ikke-tverrstripet muskel.
I US 4.710.508 beskrives O-substituert 1-(1,2,3,6-tetrahydro-l-metyl-3-pyridinyl)-ketonoksimer og O-substituert 1-(1,2,3,6-tetrahydro-4-pyridinyl)-ketonoksimer, som er nyttige som analgetiske midler eller midler til behandling av symptomene på cerebral utilstrekkelighet som erkarakterisertved nedsatt sentral acetylcholin produksjon.
I Dissertation Abstracts Int. B 1984, 45(7), 2120; CA102:113440 m beskrives oksim-O-etere med den nedenfor angitte formel å ha anticholinerge egenskaper

Den foreliggende oppfinnelse angår en analogi-fremgangsmåte for fremstilling av forbindelser med følgende generelle formler I, III og IV og farmasøytisk akseptable

R er (a) hydrogen,
(b) en uforgrenet eller forgrenet hydrokarbonkjede med 1-8 karbonatomer, som er mettet, eller er umettet og eventuelt inneholder 1-4 dobbelt- og/eller trippelbindinger,
(c) fenyl
(d) cykloalkyl med 3-6 karbonatomer,

Illustrative eksempler på lavere alkoksy med 1-4 karbonatomer er metoksy, etoksy og n-propoksy.
Illustrative eksempler på en mettet uforgrenet eller forgrenet hydrokarbonkjede med 1-8 karbonatomer omfatter n-oktyl, n-heptyl, samt alle de illustrative eksempler på uforgrenet eller forgrenet lavere alkyl med 1-6 karbonatomer angitt ovenfor.
Illustrative eksempler på en umettet uforgrenet eller forgrenet hydrokarbonkjede med 1-3 umettetheter, som er dobbel-eller trippelbindinger, er etenyl, 2,4-pentadienyl, 1,4-pentadienyl, 2,4-pentadiynyl, 1,4-pentadiynyl, (E & Z)-2-penten-4-ynyl, 2-pentyn-4-enyl, 2-propenyl, 3-butenyl, 1-metyl-2-propenyl, 2-pentenyl, 2-etyl-3-butenyl, 4-heksenyl, etynyl, 2-propynyl, l-metyl-2-propynyl, l-etyl-2-propynyl, 1-metyl-3-butynyl,3-butynyl eller4-pentynyl. Illustrative eksempler på cykloalkyl med 3-6 karbonatomer er cyklopropyl, cyklobutyl og cykloheksyl.
Farmasøytisk akseptable syreaddisjonssalter av forbindelsene med formel I, III og IV er f.eks. saltsyre-, svovelsyre-, fosforsyre-, eddiksyre-, benzoesyre-, sitronsyre-, malonsyre-, salicylsyre-, eplesyre-, fumarsyre-, oksalsyre-, ravsyre-, vinsyre-, melkesyre-, glukonsyre-, ascorbinsyre-, maleinsyre-, asparaginsyre-, benzensulfonsyre-, metan- og etansulfonsyre-, hydroksymetan- og hydroksyetansulfonsyre-saltet. Foretrukne forbindelser som fremstilles ved fremgangsmåten ifølge oppfinnelsen, er forbindelser med formel I og III, hvor forbindelsene med formel I er de mest foretrukne. De mest foretrukne forbindelser som fremstilles ved fremgangsmåten ifølge oppfinnelsen, er forbindelser med formel I hvor n er 1, dvs. forbindelser med den følgende formel V
Generelt i formel V har R de samme betydninger som angitt ovenfor for formel I, III og IV. Forbindelser med formel V, hvor R er forskjellig fra hydrogen eller NH2foretrekkes.
Forbindelser med formel I, hvor n er 2, kan angis som vist nedenfor i formel VI
hvor R har de ovenfor angitte betydninger.
I forbindelsene med formel III og IV er =NOR-gruppen bundet til enten 2-, 3- eller 4-stillingen (formel III) eller 3- eller 4-stillingen (formel IV) i ringen.
Utover de hittil ukjente forbindelser med formel I-VIII ovenfor, fremstilt ved fremgangsmåten ifølge oppfinnelsen, tilveiebringes hittil ukjente farmasøytiske preparater.
De farmasøytiske preparater er nyttige som analgetiske midler og omfatter en analgetisk effektiv mengde av en forbindelse definert ovenfor i kombinasjon med en farmasøytisk akseptabel bærer.
De farmasøytiske preparater er også nyttige ved behandling av symptomer på senil kognitiv nedbrytning og omfatter en cholinerg virksom mengde av en forbindelse definert ovenfor i kombinasjon med en farmasøytisk akseptabel bærer.
Forbindelsene som fremstilles ved fremgangsmåten ifølge oppfinnelsen, kan opptre i begge de to Z- eller E- isomere former. Den foreliggende oppfinnelse omfatter fremstilling av begge former av forbindelsene samt blandinger av Z- og E-former. Formlene IX og X illustrerer Z- og E-formene av forbindelsene med formel I. I de forbindelser, hvor det er en dobbeltbinding i karbonkjeden, er videre begge Z- og E-former av olefinene omfattet av den foreliggende oppfinnelse.
Forbindelsene med formel I, III og IV fremstilles ved at man omsetter et keton med de etterfølgende formler XI, XII og XIII med et amin med formel NH20R<*>HC1, hvor R og ~R3 har de i formel I, II, III og IV angitte betydninger, som vist nedenfor:
I hver av de ovenfor viste reaksjoner anvendes typisk metanol som oppløsningsmiddel, og omsetningen utføres ved romtemperatur.
Saltene fremstilles ved å bringe den frie baseform av forbindelsene som fremstilles ved fremgangsmåten ifølge oppfinnelsen, i kontakt med en tilstrekkelig mengde av den ønskede syre for å danne saltet på konvensjonell måte. De frie baseformer kan regenereres, om ønsket, ved å behandle saltformen med en base. F.eks. kan fortynnede vandige oppløs-ninger av baser så som natriumhydroksyd, kaliumkårbonat, ammoniakk og natriumbikarbonat, anvendes til dette formål.
De frie baseformer av forbindelsene som fremstilles ved fremgangsmåten ifølge oppfinnelsen, avviker noe fra deres respektive saltformer med hensyn til slike fysiske egenskaper som smeltepunkt og oppløslighet i polare oppløsningsmidler, men ellers er saltene ekvivalente med deres respektive frie syre- eller baseformer, hva angår formål ifølge oppfinnelsen.
Utgangsmaterialene med formel XI, XII og XIII er kjente eller kan fremstilles ved i teknikken kjente metoder. Se f.eks. J. Saunders, et al, J. Chem. Soc, Chem. Comm. 1988, 1618 og R.J. Snow og L.J. Street, Tet. Lett. 1989, 30, 5795. Aminene med formel NH20R er også kommersielt tilgjengelige eller kan fremstilles ved i teknikken kjente metoder.

1- azabicyklo[ 2, 2, 11hept- 3- on- oksim, hvdroklorid l-azabicyklo[2,2,l]hept-3-on-oksim ble fremstilt ved fremgangsmåten beskrevet i J. Chem. Soc, Chem. Comm. 1988, 1618.
1-azabicyklo[2,2,1]hept-3-on-oksim (2 g, 18 mmol) og hydroksylamin,hydroklorid (1,25 g, 18 mmol) ble oppløst i 10 ml MeOH og ble omrørt ved romtemperatur i 18 timer. Reaksjonsblandingen ble konsentrert i vakuum. Det krystallinske residuum ble omkrystallisert fra etanol for å oppnå 2,04 g (70%) av tittelproduktet, smp. 211°C.

l-azabicyklo[2,2,l]hept-3-on (2 g, 18 mmol) ble oppløst i 25 ml metanol. Etoksyamin,hydroklorid (1,76 g, 18 mmol) ble tilsatt, og reaksjonsblandingen ble omrørt ved romtemperatur i 18 timer. Reaksjonsblandingen ble konsentrert i vakuum for å
oppnå et gult faststoff. Residuet ble oppløst i vann, gjort basisk med mettet vandig kaliumkårbonat og ble ekstrahert med

Eksempel 37
1- azabicvklo[ 2, 2. 21oktan- 3- on- O- metvloksim
En blanding av ketoamin,HCl (5,56 g, 0,03 mol), 3-kinuklidinon (2,51 g, 0,03 mol) og kaliumkårbonat (11,0 g, 0,08 mol) i metanol (250 ml) ble tilbakeløpskjølt i 2 timer. Efter fjernelse av metanol under redusert trykk, ble oppløsningen suspendert i vann og ble ekstrahert med etyleter (3x125 ml), tørret over MgS04, filtrert og konsentrert for å oppnå en blanding av syn- og antiisomerer. Oksalatsaltet ble dannet fra oljen, smp. 125-128°C.

( E)- og ( Z)- 1- azabicyklo[ 2, 2, 21oktan- 3- on- O-( 2- propynyl) oksim
3-kinuklidinon (1,5 g, 12 mmol) og 2-propynyl-hydroksyl-amin,hydroklorid (1,29 g, 12 mmol) ble oppløst i 20 ml metanol og omrørt ved romtemperatur i 16 timer. Reaksjonsblandingen ble inndampet i vakuum for å oppnå et mørkebrunt halvfast stoff. Det rå halvfaste stoff ble gjort basisk med vandig K2C03og ble ekstrahert med kloroform. Kloroformekstrakten ble inndampet for å oppnå det rå oksim som en blanding av E-og Z-isomerer. Separering av isomerene ved kromatografi (silikagel, diklormetan-aceton-metanol (150:10:5)), ga henholdsvis isomer A, 1,0 g (smp. 81-83°C) og isomer B, 0,42 g (smp. 100-101°C).

1-azabicyklo[2,2,l]heptan-3-on (2 g, 18 mmol) og O-allylhydroksylamin,hydroklorid,hydrat (1,97 g, 18 mmol) ble oppløst i 25 ml metanol og omrørt ved romtemperatur i 18 timer. En oppløsning av metoksyamin,hydroklorid (4,18 g, 0,05 mol) i metanol ble satt til en hurtig omrørt oppløsning av N-metyl-8-azabicyklo[3,2,1]okta-3-on (kommersielt tilgjengelig)
(6,96 g, 0,05 mol) i metanol under avkjøling i isbad. Reaksjonsblandingen ble oppvarmet til romtemperatur, omrørt ved romtemperatur i 16 timer og derefter ble oppløsningsmidlet fjernet. Det resulterende faste residuum ble fordelt mellom CH2C12og kons. vandig K2C03oppløsning. Den organiske fase

Fremstilling av denne forbindelse er vist i det følgende reaksjonsskjerna, og er beskrevet nedenfor. (a) Fremstilling av etyl- 1, 3. 5- cykloheptatrien- l- karboksvlat ( 2) . Etyldiazoacetat (43,4 g, 0,38 mol) ble satt langsomt (sprøytepumpe) i løpet av 7 timer til en blanding av benzen (594,32 g, 7,61 mol) og tetrakis(perfluorbenzoat)dirhodium(II) (0,84 g, 0,016 mol). Efter omrøring ved romtemperatur i 16 timer, ble oppløsningsmidlet fjernet, og det oljeaktige residuum ble destillert i vakuum for å oppnå 50,44 g (81%) etyl-1,3,5-cykloheptatrien-l-karboksylat, kp. 90-96°C (2,4-3,0 mmHg) , ^"H-NMR (CDC13) , U: 6,6 (m, 2H) , 6,2 (m, 2H) , 5,3 (m, 2H), 4,2 (q, 2H OCH2CH3), 2,5 (t, 1H), 1,3 (t, 3H CH2CH3). En blanding av etyl-1,3,5-cykloheptatrien-l-karboksylat
(12,72 g, 0,077 mol), metylamin (23,13 g, 0,74 mol) og NaOH (3,1 g, 0,078 mol) i MeOH (60 ml) ble behandlet i autoklav ved 125°C i 26 timer. Reaksjonsblandingen ble filtrert, og filtratet ble konsentrert for å oppnå en olje. Oljen ble oppløst i MeOH (400 ml) og mettet med HCl-gass, kons. svovelsyre (8 ml) ble tilsatt, og blandingen ble tilbakeløpskjølt i 12 timer. (c) Fremstilling av N- metyl- 2- acetyl- 8- azabicvklor3. 2, llokt-2- en ( 4 ).
Til en blanding av etyltrop-2-en-2-karboksylat (3,43 g, 0,019 mol) og Me3SiCl.(10,32 g, 0,095 mol) i THF (70 ml) avkjølt til -100°C, ble satt dråpevis MeLi (1,4 M, 17,07 ml, 0,0239 mol). Efter oppvarmning til romtemperatur ble reaksjonsblandingen omrørt i 16 timer. Overskudd av Me3SiCl og THF ble avdampet, og det resulterende residuum ble omrørt med EtOH (6 ml) efterfulgt av vann (6 ml). Blandingen ble derefter gjort sur (6 N HC1) og ble ekstrahert med eter. Det vandige lag ble gjort basisk (fast K2C03) og ble ekstrahert med kloroform, tørret (K2C03) og konsentrert, hvilket ga 3,14 g (81%) 2-acetyltrop-2-en som en olje: IR (ren) 1670 (C0CH3), "<*>"H-NMR (CDC13), U: 6,7 (bt, 1H) , 2,3 (s, NCH3) , 2,2 (s, C(_0)CH3).

oppnås ved katalytisk reduksjon (H2/RaNi) av forbindelse (4) og omdannes til N-metyl-8-azabicyklo[3,2,1]oktan-2-on, forbindelse (7) ved i teknikken kjente fremgangsmåter, se J. Org. Chem. 55, 5025 (1990). I forbindelse (6) står TBDMSO for prøvning-butyldimetylsilyloksy. Ketonet, forbindelse (7) omdannes til metyloksimen som generelt beskrevet ovenfor i eksempel 66.
Når N-metyl-2-acetyl-8-azabicyklo[3,2,1]oktan, forbindelse (5) erstattes med N-metyl-2-acetyl-9-azabicyklo[4,2,l]nonan, hvilket kan fremstilles fra 2-acetyl-9-azabicyklo[4,2,l]non-2-en, som er kommersielt tilgjengelig, oppnås på lignende måte N-metyl-8-azabicyklo[4,2,l]nonan-2-on-metyloksim.
Forbindelsene som fremstilles ved fremgangsmåten ifølge oppfinnelsen, er sentraltvirkende muskarinmidler og er således nyttige som analgetiske midler ved behandling av smerter hos pattedyr herunder mennesker, som sovemidler, og som midler ved behandling av symptomene på senil demens, Alzheimer's sykdom, Huntington's korea, tardiv dyskinesi, hyperkinesi, mania eller lignende tilstander av cerebral utilstrekkelighet karaterisert ved nedsatt cerebral acetylcholin- produksjon eller frigivelse.
Den biologiske aktivitet av forbindelsene som fremstilles ved fremgangsmåten ifølge oppfinnelsen, ble vurdert under anvendelse av en rekke tester. Aktiviteten av forbindelsene som sentral-muskarin-bindingssted-agonister og - antagonister ble målt. Ved RQNB screeningsassayet som er beskrevet nærmere av Mark Watson, et al, J. Pharmacol. and Exp. Ther., 237(2):411 (1986), ble cerebral cortex vev fra rotter behandlet med radiomerket kinuklidinylbenzilat, en kjent muskarin-bindingssted-antagonist. Den konsentrasjon av testforbindelse som er nødvendig for å inhibere 50% av bindingen av denne muskarinantagonist, ble derefter bestemt.
Ved RCMD screeningsassayet som er beskrevet nærmere av T.W. Vickeroy, et al, 229(3):747 (1984), ble cerebral cortex vev fra rotter behandlet på lignende måte med radiomerket cis-metyldioksolan, en kjent muskarin-bindingssted-agonist. Den konsentrasjon av testforbindelse som er nødvendig for å inhibere 50% av bindingen av denne muskarinagonist, ble derefter bestemt. Disse verdier er angitt som IC5Q-konsen-trasjoner i tabell 2 og viser at forbindelsene som fremstilles ved fremgangsmåten ifølge oppfinnelsen, har signifikant muskarin-aktivitet.
Ved terapeutisk anvendelse som midler til behandling av smerte eller behandling av cerebral utilstrekkelighet admini-streres forbindelsene ved en farmasøytisk metode til pasienten i dosenivåer fra 0,07 til 700 mg pr. dag. For et normalt voksent menneske på ca. 70 kg betyr dette en dose på 0,01 til100mg/kg legemsvekt pr. dag. De spesifikke doser som anvendes kan imidlertid varieres avhengig av pasientens behov, alvorlighetsgraden av den tilstand som behandles og aktiviteten av den forbindelse som anvendes. Bestemmelse av den optimale dose for en bestemt situasjon ligger innenfor fagmannens kompetanse.
Ved fremstilling av farmasøytiske preparater fra forbindelsene som fremstilles ved fremgangsmåten ifølge oppfinnelsen, kan inerte, farmasøytisk akseptable bærere være enten faste eller flytende. Fastformspreparater omfatter pulvere, tabletter, dispergerbare granulater, kapsler, oblatkapsler og suppositorier.
En fast bærer kan være én eller flere substanser som også kan virke som diluenter, smaksstoffer, oppløsningsmidler, smøremidler, suspensjonsmidler, bindemidler eller tablett desintegrasjonsmidler. Den kan også være et innkapslende materiale.
I pulvere er bæreren et findelt faststoff som er i blanding med den findelte aktive komponent. I tabletter blandes den aktive komponent med bæreren som har de nødvendige bindingsegenskaper i passende proporsjoner og forarbeides i den ønskede form og størrelse.
Ved fremstilling av suppositorier smeltes først en lavt-smeltende voks så som en blanding av fettsyreglycerider og kakaosmør, og den aktive ingrediens dispergeres deri f.eks. ved omrøring. Den smeltede homogene blanding helles derefter i former av passende størrelse og står til avkjøling og størkning.
Pulvere og tabletter inneholder fortrinnsvis mellom ca. 5% og ca. 70 vekt% av den aktive ingrediens. Egnede bærere er magnesiumkarbonat, magnesiumstearat, talkum, laktose, sukker, pektin, dextrin, stivelse, tragant, metylcellulose, natriumkarboksymetylcellulose, en lavt-smeltende voks, kakaosmør eller lignende.
Uttrykket "preparat" skal omfatte formuleringen av den aktive forbindelse med innkapslende materiale som en bærer, hvilket tilveiebringer en kapsel, hvor den aktive komponent (med eller uten andre bærere) er omgitt av en bærer, hvilken således er i assosiasjon med denne. På lignende måte omfattes oblatkapsler.
Tabletter, pulvere, oblatkapsler og kapsler kan anvendes som faste doseformer egnet for oral administrering.
Preparater i flytende form omfatter oppløsninger som er egnet for oral eller parenteral administrering, eller suspensjoner og emulsjoner som er egnet for oral administrering. Sterile vannoppløsninger av den aktive komponent eller sterile oppløsninger av den aktive komponent i oppløsningsmidler omfattende vann, etanol eller propylenglykol kan nevnes som eksempler på flytende preparater som er egnede for parenteral administrering.
Sterile oppløsninger kan fremstilles ved å oppløse den aktive komponent i det ønskede oppløsningsmiddelsystem, og derefter lede den resulterende oppløsning gjennom et membran-filter for å sterilisere den, eller alternativt, ved å oppløse den sterile forbindelse i et tidligere sterilisert oppløsningsmiddel under sterile betingelser.
Vandige oppløsninger for oral administrering kan fremstilles ved å oppløse den aktive forbindelse i vann og tilsette passende smaksstoffer, farvestoffer, stabilisatorer og fortykningsmidler efter ønske. Vandige suspensjoner for oral anvendelse kan lages ved å dispergere den findelte aktive komponent i vann sammen med et viskøst materiale så som naturlig eller syntetisk gummi, harpikser, metylcellulose, natriumkarboksymetylcellulose og andre suspensjonsmidler kjent i teknikken.
Det farmasøytiske preparat er fortrinnsvis i enhetsdose-form. I denne form oppdeles preparatet i enhetsdoser inne-holdende passende mengder av den aktive komponent. Enhetsdoseformen kan være et emballert preparat, hvor emballagen inneholder adskilte mengder av preparatet, f.eks. emballerte tabletter, kapsler og pulvere i hetteglas eller ampuller. Enhetsdoseformen kan også være en kapsel, oblatkapsel eller tablett i seg selv eller den kan være det passende antall av enhver av disse i emballert form.
