Description
Foreliggende oppfinnelse vedrører et kosmetisk preparat for
forhindring av hudens aldring. Preparatet er kjennetegnet ved at det som aktiv bestanddel inneholder en ekstrakt, tilstede i en konsentrasjon uttrykt som polyfenoler på fra 0,001 til 0,01 vekt%, av luftdelene av Cichorium intybus L, idet ekstrakten
er utvunnet ved hjelp av et polart løsningsmiddel fra luftdelen av Cichorium intibys L.
Ekstrakten av Cichorium intybus L (forbindelser) inneholder forskjellige polyfenoliske derivater som f.eks. kaffeylvinsyre.
Ekstrakten av Cichorium intybus L kan fremstilles ved ekstrak-sjon av de tørkede luftdeler ved hjelp av polare løsnings-midler som kokende vann, metanol, etanol og propylenglykol.
Bladene av denne planten er kjent for deres choleretiske, diuretiske og svakt avførende egenskaper som er grunnen til at infusjoner av ekstraktene har vært anvendt som et blodrensende middel innen folkemedisinen.
Grunnlaget for den foreliggende oppfinnelse er erkjennelsen om at en ekstrakt av luftdelene av Cichorium intybus L har en antiradikal aktivitet, særlig som et resultat av kompleksdan-nende jern, og at kosmetiske preparater inneholdende en slik ekstrakt kan forhindre de skadelige virkninger av de frie radikaler som delvis er ansvarlig for hudens aldring.
De frie radikalene frembringes under reaksjonene med celle-metabolisme, særlig som et resultat av ett-elektronreduksjonen av oksygen under reaksjonene med mitokondrien respirasjon.
De frembringes også i huden ved fotokjemiske reaksjoner under eksponering for sollys. Det foreligger et kraftig beskyt-telsessystem mot slike radikaler i en intakt celle. Peroksydanionet (02.-) omdannes ved hjelp av peroksyddismutaser til hydrogenperoksyd (H202) som i sin tur metaboliseres til vann (H20) ved katalase. Peroksyddismutase + katalase paret er et avgiftingssystem hvis effektivitet nedsettes under aldring. Under disse betingelser reagerer 02.- og H202 for å utvikle en kaskade av reaksjoner som katalyseres av jern og som er kjent som HABER-WEISS-reaksjonen, og som særlig leder til dannelse av hydroksyl (0H- )-radikalet. Dette radikal fremviser en meget høy reaktivitet og påvirker gjensidig biologiske struk-turer, særlig membranfosfolipidene, proteinene og nuklein-syrene, som det skader.
Det er kjent at hverken peroksydanionet eller hydrogenperoksyd kan indusere peroksydering av lipider eller et sukker som f.eks. deoksyribose direkte.
Denne reaksjon krever faktisk dannelse av mer reaktive radikaler som hydroksylradikalet OH*, som katalyseres ved hjelp av jern.
Når slike radikaler angriper fettsyrene i membranfosfolipidene sprer det seg en desorganisering gjennom cellemembranene som inhiberer deres funksjoner og særlig med hensyn til epidermis, forstyrrer deres rolle som barriere som sikrer adekvat kutan hydratisering. På samme måte, når disse radikaler gjensidig påvirker proteinene i huden som f.eks. elastin og kollagen, medfører de brudd og kryssbindingsegenskaper ved kutan aldring.
Som en følge av nærværet av ekstrakten av luftdelene av Cichorium intybus L som inhiberer disse frie radikaler, særlig ved å chelatere jern, er det kosmetiske preparat i samsvar med oppfinnelsen i stand til å forhindre aldring av huden.
Det kosmetiske preparat ifølge oppfinnelsen muliggjør forhindring av aldring av huden når minst en ektrakt av luftdelene av Cichorium intybus L påføres huden i en passende kosmetisk base.
Ekstrakten kan fremstilles på følgende måte.
Eksempel på fremstilling av ekstrakten
Luftdelene av Cichorium intybus L tørkes og findeles (uten faktisk å males til et pulver). De ekstraheres med fem ganger sin egen vekt av kokende vann (macerasjon i 30 minutter). Ekstrakten filtreres, og deretter ekstraheres resten med propylenglykol (fem ganger den initiale vekt av plantedelen). Propylenglykolekstrakten filtreres, resten tørkes ved hjelp av en presse og ekstraktene samles.
Vann/propylenglykolekstrakten (50:50) inneholder 1 ± 0,2 mg/ml polyfenoler som inkluderer kaffeylvinsyre (målt ved hjelp av ARNOW-reagenset).
Den følgende forsøksprotokoll muliggjør påvisning av aktivi-teten av ekstrakten/den aktive bestanddel anvendt ved oppfinnelsen.
Inhibering av Fenton-reaksjonen
Fenton-reaksjonen er det tredje trinn i HABER-WEISS-reaksjonen.
Denne reaksjon gjomføres in vitro ved å omsette H202 (10"<4>M)
+ Fe<2+> (10"<5>M) i nærvær av hypoxantin og xantinoksydase, som utvikler peroksydanionet og deoksyribose (5 x 10"<3>M), som er substratet ved peroksydasjonsreaksjonen.
Når reaksjonen gjennomføres i nærvær av EDTA (etylendiamin-tetraeddiksyre) blir radikalet utviklet som 0H- i samsvar med den ovenfor beskrevne reaksjon. I motsetning til dette utvikles, i fravær av EDTA, en annen fremdeles ukjent type radikal (WINTERBOURN, CC. Free Radical Biology & Medicine, 3, 33 - 39, 1987).
I fravær eller nærvær av EDTA omdanner reaksjonen deoksyribose til oksydasjonsprodukter som kan måles ved hjelp av tiobar-bitursyre i samsvar med metoden beskrevet av GUTTERIDGE, J.M.C. (Biochem. J., 224, 761 - 767, 1984).
De aktive bestanddeler av ekstrakten av Cichorium intybus L inhiberer oksydasjonen av deoksyribose ved tilsetning til reaksjonsblandingen under de to sett av betingelser:
Inhibering av dannelse av kutant malonaldehyd
Et av tegnene på cellulær aldring som impliserer frie radikaler i aldringsprosessen er akkumulering av lipofuskiner i vevene. Disse pigmenter er Schiffske baser som resulterer fra reaksjonen av primære aminer med det malonaldehyd (MDA) som produseres ved radikal destruksjon av lipider og visse sukkerarter.
Den beskyttende virkning av ekstrakten mot denne prosess kan påvises ved en test hvor ekstrakten, når den er fortynnet i et kosmetisk hjelpestoff og påføres huden på et forsøksdyr, inhiberer dannelse av MDA som normalt induseres ved bestråling i det kutane vev. Et typisk forsøk er vist i det følgende: Hårløse mus ble sensitivert ved bading i fem minutter i en oppløsning av Bengalrose (10"<5>M) og deretter tørket i mørke. Den fortynnede ekstrakt eller hjelpestoffet ble påført over-flaten av den dorsale hud på dyrene ti minutter før de ble bestrålt. Bestålingen ble gjennomført i 3 0 munutter ved hjelp av en 75 watt KRYPTON glødelampe anbragt i en avstand på 25 cm fra dyrene. Kontrollmus ble holdt i mørke. Dyrene ble avlivet og prøver av deres epidemis ble tatt for analyse av MDA ved hjelp av den tiobarbitursyrereaksjon som er nevnt i det foregående. Resultatene er angitt i det følgende og viser den beskyttende virkning av ekstrakten mot kutan skade som skyldes peroksydasjon utløst av en fotokjemisk reaksjon.
Det kosmetiske preparat kan ytterligere om ønsket inneholde hvilke som helst andre substanser som er kjent å ha gunstige egenskaper med hensyn til skjønnhetspleie, som kollagen, elastin, hyaluronsyre, lipidforbindelser inneholdende umettede fettsyrer, liposomer, fosforlipider, fuktemidler, vitaminekst-rakter, parfymer, konserveringsmidler, fargestoffer. De kan også inneholde solfiltere.
Kosmetiske preparater kan gjøres tilgjengelige i alle former anvendt innen skjønnhetspleien, som kremer eller geler i krukker eller tuber, emulsjoner, lotioner i glass eller plastflasker, og om ønsket i glassdispensere eller endog ampuller eller spraypreparater.
For hvert preparat må man anvende passende hjelpestoffer. Disse hjelpestoffer må ha alle de egenskaper som vanligvis kreves, og eksempler som kan nevnes er glyserolstearat, propylenglykol, lanolin, glyserol, cetylalkohol, polyoler, vegetabilske, animalske og mineraloljer, fosfolipider som liposomer, voksarter, fuktemidler, bindemidler, stabilise-ringsmidler og emulgeringsmidler som vanlig anvendt.
De forskjellige kosmetiske former som nevnes i det foregående fremstilles ved hjelp av metoder som vanlig anvendt på området.
I det følgende gis eksempler på kosmetiske preparater (vekt-deler) fremstilt ved å anvende en ekstrakt av Cichorium intybus L inneholdende 1 mg/ml polyfenoler:
