Description
Foreliggende oppfinnelse vedrører et monoklonalt antistoff som gjenkjenner cx-2—>3-bindinger, og en hybridomcellelinje som produserer antistoffet.
Hittil har anti-sera blitt fremstilt ved å absorbere serumet fra et immunisert dyr på forskjellige sorter antigener. Anti-sera inneholder imidlertid et stort antall anti-stoffmolekyler som stammer fra forskjellige B-celler (poly-klonale), og forårsaker ofte kryssreaksjoner med annet antistoff. Det er derfor vanskelig å oppnå anti-sera med ut-merket spesifisitet.
Under slike omstendigheter utviklet Kohler i 1975 et hybridom som produserer anti-sau rødt blodlegeme-antistoff og som fremstilles ved å fusjonere miltceller utvunnet fra et immunisert dyr og mus-myelomceller for frembringelse av hybridomer. En klon som er et hybridom som stammer fra en enkel celle, kan isoleres fra slike hybridomceller (såkalt "kloning") på grunn av dens store proliferasjonskraft. Alle antistoffene fremstilt ved hjelp av et slikt klonet hybridom er identiske, har enhetlig spesifisitet til et antigen og gjenkjenner således det samme sted i et spesifikt antigen. I tillegg kan hybridomene lagres i nedfryst tilstand, f.eks. i flytende nitrogen. Stabil tilgang på de individu-elle antistoffer kan derfor sikres. Ved hjelp av cellefusjons-teknikk er det således blitt mulig å oppnå et monoklonalt antistoff som er svært spesifikt til et bestemt antigen.
Antistoffer er proteiner som kan gjenkjenne et molekyl eller stoff henvist til som "antigen" som hører naturlig til antistoffet, og kan bindes til dette. Det monoklonale antistoff betyr et antistoff som har et sted som spesifikt gjenkjenner et bestemt antigen, med andre ord gjenkjenner det bare en antigen determinant. I dag er forskjellige tek-nikker for fremstilling av monoklonale antistoffer og for frembringelse av hybridomer som er i stand til å produsere de monoklonale antistoffene, godt kjent innen teknikken.
I denne henseende kan det vises til en nyere publikasjon "Monoclonal Hybridoma Antibodies: Techniques and Applica-tions", redigert av John G. Hurrell, 1983.
Glycolipidet i pattedyrceller tilhører kategorien av såkalt spingoglycolipid og omfatter (a) en lipidstruktur henvist til som ceramid bestående av en langkjedet aminoalkohol kalt spingosin hvortil en fettsyre er syre/amido-bundet og (b) forskjellige kombinasjoner av sukkere. valgt fra gruppen bestående av glucose, galactose, N-acetylglucosamin, N-ace-tylgalactosamin, fucose og sialinsyre som er bundet til strukturen gjennom glycosidbindinger. Blant disse kalles glycolipidene som bærer sialinsyrerest for gangliosider.
De fleste av disse forbindelsene finnes vanligvis i den ytre cellemembran, og ut fra senere undersøkelser er det blitt antatt at gangliosider spiller en viktig rolle i funk-sjonene som en mottakelse av og respons på gjenkjennelse og informasjon, reseptorfunksjoner, differensiering, prolifera-sjon, malign endring eller oppførsel hos celler.
Blant disse gangliosidene er GD^-gangliosidet kjent
som det differensierte antigen i nerveceller og videre identifisert som det kreft-relaterte antigen i humane melanomceller.
Ettersom det monoklonale antistoff har høy spesifisitet til et bestemt antigen og således kan påvise dette med stor følsomhet som beskrevet ovenfor, ville det, dersom det ble erholdt et monoklonalt antistoff som er spesifikt til GD^-gangliosidene, forventes å finne anvendelse for et slikt monoklonalt antistoff til diagnose av kreft (som kreft-markør) in vitro, samt til belysningen av sukkerkjedens rolle i cellefunksjoner.
Følgelig er det et formål ved foreliggende oppfinnelse
å tilveiebringe et hybridom som er i stand til å produsere monoklonale antistoffer som er spesifikke for en bestemt antigen determinant både i GD^- og GM^-gangliosidet, som er identifisert som antigenene i humane melanomceller som er forbundet med kreft.
Det er videre et formål ved foreliggende oppfinnelse
å tilveiebringe et monoklonalt antistoff som utviser spesi-
fisitet til den bestemte antigene determinant både på GD^-og GM^-gangliosidet.
Oppfinnerne av foreliggende oppfinnelse har utført forskjellige undersøkelser for å eliminere de tidligere nevnte ulemper, og har funnet at i cellefusjonsteknikken anvendes det rensede GD3~gangliosid som et antigen for å frembringe et hybridom som er i stand til produsere et ønsket monoklonalt antistoff, og på denne måte har foreliggende oppfinnelse blitt fullført.
Ifølge et aspekt av foreliggende oppfinnelse er det tilveiebragt en hybridomcellelinje som er kjennetegnet ved at den har ATCC deponeringsnummer HB 9475 og produserer et monoklonalt antistoff som er et immunoglobulinmolekyl av IgM-type, binder både GD^-gangliosid og GM^-gangliosid immunologisk og gjenkjenner epitopen N-acetylneuraminsyre som har en a-2—>3-binding.
Ifølge et annet aspekt av foreliggende oppfinnelse er det også tilveiebragt et monoklonalt antistoff som er kjennetegnet ved at det er et immunoglobulinmolekyl av IgM-type, er fremstilt ved hjelp av en hybridomcellelinje med ATCC deponeringsnummer HB 9475, binder både GD^-gangliosid og GM^-gangliosid immunologisk og gjenkjenner epitopen N-acetylneuraminsyre som har en a-2->3-binding.
Figur 1 viser spesifisiteten av det monoklonale antistoff ifølge foreliggende oppfinnelse til antigener GD^ og GM^ bestemt i henhold til enzymmerket antistoffteknikk,
figurene 2 og 3 viser antigenspesifisitet av forskjellige slag av gangliosider mot det monoklonale antistoff ifølge foreliggende oppfinnelse, og
figur 4 viser et diagram som brukes til å klassifisere det monoklonale antistoff ifølge foreliggende oppfinnelse.
Foreliggende oppfinnelse vil nedenunder bli forklart nærmere.
Hybridomet ifølge foreliggende oppfinnelse kan fremstilles ved å fusjonere 'B-celler (B-lymfocytter) erholdt fra et dyr som er immunisert med et antigen i henhold til fremgangsmåten til Kohler & Millstein (se Nature 1975). "Antigenet" som her er brukt, er f.eks. gangliosider som inneholder N-acetylneuraminsyrerest med alfa-2—>3-binding, fortrinnsvis gangliosidglycolipider som inneholder N-acetyl-neuarminsyrerest med alfa-2—>3-binding. Bestemte eksempler på dette er:
Det monoklonale antistoff fremstilt ved hjelp av hybridomet ifølge foreliggende oppfinnelse gjenkjenner N-acetylneuraminsyre med alfa-2—>3 som epitopen eller den antigene determinant, og reagerer spesifikt med denne. Det monoklonale antistoff fremstilt ved hjelp av hybridomet viser derfor særlig høy reaktivitet med sialinsyre-holdige glycolipider som bærer N-acetylneuraminylrest med alfa-2—>3-binding, særlig GD^- og GM^-gangliosider. I tillegg utviser det monoklonale antistoff likt en slik spesifisitet til hvilke som helst av disse sialinsyre-holdige glycolipider uavhengig av typene av opphavsdyr.
"Dyr" som skal immuniseres med antigenet, det tidligere nevnte sialinsyre-holdige glycolipid, kan være enhver type dyr. Bestemte eksempler på slike er kanin, menneske, mus og rotte, fortrinnsvis mus og helst Balb/c-mus.
B-cellene (B-lymfocyttene) som produserer det monoklonale antistoff ifølge foreliggende oppfinnelse utvinnes fortrinnsvis fra miltceller. På den annen side kan "myelomceller" som anvendes i foreliggende oppfinnelse, også være de som er utvunnet fra hvilke som helst dyr, slik som menneske, mus, rotte og kanin, fortrinnsvis de som er utvunnet fra mus og helst myelomceller (X63-Ag8.653) utvunnet fra Balb/c-mus. Disse myelomcellene utviser svært høy proliferasjonskraft, slik at hybridomet som er frembragt ved å fusjonere dem med B-cellene, kontinuerlig kan produsere det monoklonale antistoff ifølge oppfinnelsen.
Hybridomet ifølge foreliggende oppfinnelse kan fremstilles ved hjelp av fremgangsmåten forklart nærmere nedenunder .
Først av alt injiseres GD^-gangliosid i mus intraperi-tonealt, subkutant eller intravenøst, fortrinnsvis intra-venøst, for å immunisere dem. Ved denne immunisering kan det anvendes enten mangelfulle adjuvantia eller komplette adjuvantia som et adjuvans, dvs. et middel for immunologisk forsterkning, og bestemte eksempler på dette er oljer, emul-geringsmidler, drept tuberkelbacillus, drept salmonella og blanding derav, fortrinnsvis drept Salmonella minesota, for intraperitoneal eller subkutan injeksjon og for intravenøs injeksjon. Disse antigener og adjuvantia anvendes fortrinnsvis i form av en oppløsning med omtrentlig fysiologisk akseptabel sammensetning, slik som en oppløsning av fosfatbufret medisinsk salt.
Ved denne immunisering bør det legges merke til at det er vanskelig å sensibilisere et dyr som skal immuniseres, med egen-bestanddelene og det er derfor i prinsippet foretrukket å velge et dyr som fenogenetisk er fjernt fra det som det antigene stoff erholdes fra. Immuniseringen kan imidlertid også utføres in vitro for å unngå slike vanskelig-heter .
Så fusjoneres B-cellene og myelomcellene sammen. Som et middel for cellefusjon kan det anvendes polyethylenglycol og HVJ, fortrinnsvis polyethylenglycol 6000. I tillegg er det foretrukket å anvende HAT-medium som dyrkningsmedium for lett å isolere hybridomceller fra myelomceller som forblir ufusjonert.
Deretter underkastes de resulterende hybridomceller slik kloningsoperasjon som methylcelluloseteknikk, bløt-agaroseteknikk eller begrensende fortynningsteknikk for å isolere et ønsket enkelt klonet hybridom.
Antistofftiteren av det monoklonale antistoff-produ-serende hybridom frembragt på denne måte kan undersøkes på vanlig måte for å velge ut hybridom som utviser høy anti-stof f titer. Det således utvalgte hybridom lagres så.
Det monoklonale antistoff fremstilt ved hjelp av hybridomet ifølge foreliggende oppfinnelse kan anvendes til in vitro påvisning av glycolipidantigenet som naturlig forekommer i kreftvev hos menneske eller dyr. Det monoklonale antistoff kan derfor brukes f.eks. i ELISA- og RIA-analyser som på vanlig måte er tilpasset til påvisningen av kreft-assosi-erte gangliosider. D.et monoklonale antistoff ifølge foreliggende oppfinnelse kan dessuten anvendes i affinitetskromato-grafi for å rense antigener som er i stand til å binde seg til det monoklonale antistoff. Det monoklonale antistoff ifølge foreliggende oppfinnelse kan anvendes til å diagnos-tisere arter in vitro med antigenet, slik...som menneske og dyr, slik som mus. Det er forventet at det monoklonale antistoff ifølge foreliggende oppfinnelse kan anvendes til grunnleggende undersøkelse av nerveceller og kan anvendes til forskjellige kliniske formål.
Foreliggende oppfinnelse vil bli forklart nærmere i eksemplene nedenunder.
Eksempel
(A) Fremstilling av antigen
(1) Ekstraksjon og rensing av GD3~ og GM3~gangliosider
Etter at binyre fra svin var homogenisert i kaldt aceton og tørket, ble det tørkede produkt ekstrahert med
kloroform/methanol på vanlig måte.
Den resulterende prøve ble behandlet med et alkali og så ble GD^-gangliosid renset under anvendelse av DEAE-Sepha-dex A-25 anionbytterharpiks og "Iatrobeads"-kolonne.
GM^-gangliosid ble fremstilt ved å sentrifugere hunde-serum behandlet med heparin, vaske det resulterende sediment tre ganger med fosfatbufret saltoppløsning (PBS(-)), lyofi-lisere det og så ekstrahere produktet med kloroform/methanol. Rensingen derav ble utført på samme måte som ovenfor.
Gangliosidene GD]_a» ^Dib°^ ^lb' Gal-Ser og Glc-Cer, og GTj^ og GM2 var alle kommersielt tilgjengelige og ble brukt uten ytterligere rensing.
Disse gangliosidglycolipidene ble oppløst i kloroform/ methanol (1:1 (volum/volum)) eller ethanol og lagret ved -20°C.
(2) Dyr
Seks 6 uker gamle Balb/c-mus av hunnkjønn ble brukt
til forsøkene etter oppaling under vanlige betingelser.
(3) Fremstilling av antigenoppløsning
Gangliosid GD^ som er ekstrahert fra binyre fra svin og så renset, og Salmonella minesota R 595 behandlet med eddik-syre (vektforhold = 1:4) og med ditehylether, ble blandet i PBS(-) slik at konsentrasjonen av GD^-gangliosid var 100 ug/ ml, og den resulterende oppløsning ble brukt som antigen-oppløsning .
(4) Dyrkningsmedium
Dyrkningsmedium: Nissui RPMI 1640 ble brukt som dyrkningsmedium. Til mediet ble det tilsatt kanamycinsulfat og bovint fosterserum (FBS) slik at sluttkonsentrasjonen derav var lik med henholdsvis 50 ug/ml og 10% før bruk.
HAT-medium: 0,038 8 g thymidin og 0,13 61 g hypoxanthin ble oppløst i 100 ml destillert vann under oppvarming, og den resulterende oppløsning (a) ble lagret ved 20°C som en standardoppløsning med en konsentrasjon som var 100 ganger høyere enn den ønskede. Likeledes ble 0,0176 g aminopterin oppløst i 100 ml destillert vann ved å tilsette en liten mengde IN natriumhydroxyd i vandig oppløsning, så ble denne fortynnet 10 ganger med RPMI 1640 dyrkningsmedium og den resulterende oppløsning (b) ble lagret ved -20°C som en standardoppløsning med en konsentrasjon som var 100 ganger høyere enn den ønskede, mens det ble skjermet mot lys. HAT-medium ble fremstilt ved å tilsette 1/100-dels volum
hver av de to oppløsningene til 10% FBS RPMI 1640 medium øyeblikkelig før bruk.
Videre ble HT-medium fremstilt ved ganske enkelt å tilsette 1/100-dels volum av standardoppløsningen (a) inne-holdende hypoxanthin og thymidin til det samme 10% FBS RPMI 1640 medium.
(5) Opphavsceller
Som opphavscellene for cellefusjon ble det brukt myelomceller (C63-Ag8-6.5.3-celler) som var blitt utvunnet fra Balb/c-mus. Disse cellene ble underkastet subdyrking i RPMI 1640 medium hvortil 10% FBS var tilsatt, mens genereringen av mutant ble inhibert ved å tilsette 6-thioguanin til mediet, slik at konsentrasjonen av dette var lik med 3 yg/ml.
(B) Fremstilling av hybridom
(1) Immuniseringsmetode
6 uker gamle Balb/c-mus av hunnkjønn ble immunisert
ved intravenøs injeksjon av den følgende immunogenoppløsning i henhold til den følgende immuniseringsplan (mengde inji-sert ble uttrykt i mengden av gangliosid): til å begynne med 5 yg, 10 ug på dag 4, 15 ug på dag 7 HL, 2 0 yg på dag 12 og 20 yg på dag. 79 etter start. På dag 3 etter slutt-immuniseringen ble musene avlivet for å skjære bort milten, og en suspensjon av individue-He miltceller ble fremstilt for etterfølgende bruk i cellefusjon.
Immunogenoppløsning: 100 \ iq GD^-gangliosid ekstrahert fra svinebinyre og renset, og 400 ug Salmonella minesota T 595 som et adjuvans, ble blandet il ml PBS(-) (fri for Ca<++> og-Mg<++>), og den resulterende oppløsning ble så passende fortynnet til bruk som immunogenoppløsning.
(2) Cellefusjon
Fusjon av miltcellene erholdt ovenfor og mus-myelomcellene ble utført ifølge fremgangsmåten til Kohler og Millstein. Nærmere bestemt ble 1 x 10 g miltlymfocytter fusjonert med 2 x 10 7 myelomceller i nærvær av 50% polyethylenglycol (PEG 6000) i et dyrkningsmedium.
(3) Utvelgelse og dyrking av hybridom
Etter cellefusjonen ble de resulterende celler dyrket
i HAT-medium (hypoxanthin, aminopterin og thymidin) ved 37°C i nærvær av 5% C02.
Det derved frembragte hybridom er blitt deponert ved American Type Culture Collection (ATCC) under deponeringsnummer ATCC HB 9475.
(C) Beregning av reaktiviteten til det monoklonale antistoff med gangliosider
96-brønners flatbunnet plate ble forbehandlet i ethanol før anvendelse i forsøkene. 50 yl av antigenoppløsningen (immunogenoppløsningen) som var blitt fortynnet med ethanol for å regulere konsentrasjonen av GD^-gangliosid til 20 yg/ml (optimal konsentrasjon), ble pipettert over i hver brønn i platen, så ble oppløsningsmidlet avdampet, 200 yl 1% ovalbumin PBS(-)-oppløsning ble innført i hver av brønnene og de fikk stå i 30 minutter ved værelsetemperatur. Platen ble rystet med oppsiden ned for å fjerne oppløsningen, så ble 50 yl av supernatanten fra hybridomkulturmediet som det primære antistoff, tilsatt til hver brønn og platen fikk stå i 1,5 timer ved værelsetemperatur. På samme måte ble det primære antistoff fjernet fra brønnene, så ble brønnene vasket 3 ganger ved å tilsette 150 yl PBS(-)-oppløsning og fikk stå i 30 minutter ved værelsetemperatur etter tilsetning av 100 yl i hver brønn av 1% ovalbumin-PBS(-)-oppløsning. Etter fjern-ingen av denne oppløsning ble 50 yl sekundært antistoff fortynnet med 1% ovalbumin-PBS(-)-oppløsning til dets opti-
male konsentrasjon tilsatt til hver brønn og platen ble hen-satt i 1,5 timer ved værelsetemperatur. Som i tilfellet med det primære antistoff ble brønnene vasket 3 ganger med PBS(-)-oppløsning og 100 yl av en reaksjonsoppløsning ble tilsatt til hver brønn for å forårsake reaksjon i mørket. Reak-sjonsoppløsningen ble fremstilt ved oppløsning i citrat/fos-fat-buffer (pH = 5), O-fenylendiamin og hydrogenperoxyd slik at konsentrasjonene var henholdsvis 0,4 mg/ml og 0,01%. Reaksjonen ble stanset ved tilsetning av 30 yl 8N svovelsyre-oppløsning til hver brønn og så ble produktet undersøkt ved kolorimetri ved 500 nm. Som det sekundære antistoff ble det brukt geit-anti-mus-IgG-, -IgM- og -IgA-antistoffer merket med pepperrotperoxydase (HRP).
De oppnådde resultater er plottet på figur 1 hvor den igjenfyllte ring representerer gangliosid GM^ og den åpne ring representerer gangliosid GD^.
Som det sees av resultatene plottet på figur 1, forårsaker gangliosidene GD^ og GM^ antigen/antistoff-reaksjoner med det monoklonale antistoff ifølge foreliggende oppfinnelse ved høy sensitivitet. Således bærer begge gangliosidene GD^ og GM^ den antigene determinant mot det monoklonale antistoff ifølge oppfinnelsen. I denne forbindelse utviser gangliosidet GD^ høyere reaktivitet med det monoklonale antistoff enn gangliosidet GM3. Det er derfor funnet at gangliosidet GD^ har en epitop eller antigen determinant med mye høyere spesifisitet til det monoklonale antistoff ifølge foreliggende oppfinnelse.
(2) TLC-immunofargingsteknikk
En tynnsjiktkromatografiplate av silikagel (TLC(poly-gram SIL G)) ble skåret opp i stykker av passende størrelse og ble flekket med en liten dråpe av oppløsningen av et glycolipid i kloroform/methanol (1:1 (volum/volum)). Avhengig av formål ble platen fremkalt med en fremkallingsoppløsning som f.eks. kloroform/methanol/vann (60/40/10, volum/volum), kloroform/methanol/0,5% CaC^-oppløsning (55/45/10, volum/ volum) eller kloroform/methanol/2,5N NH4OH-oppløsning (60/ 40/9, volum/volum), og så fikk platen stå i 1% ovalbumin/1% polyvinylpyrrolidon (K-30) PBS(-)-oppløsning ved 4°C over natten. Den ble så rystet i 2 timer ved værelsetemperatur under neddypping i oppløsningen av primært antistoff. Etter tilstrekkelig vasking med PBS(-), ble den dyppet i 1% ovalbumin/1% polyvinylpyrrolidon (K-30) PBS(-)-oppløsning ved værelsetemperatur i 30 minutter. Platen ble tatt ut av opp-løsningen, ble dyppet i 2 timer i det sekundære antistoff som var fortynnet til sin optimale konsentrasjon med 3% polyvinylpyrrolidon (K-30) PBS(-)-oppløsning under rysting, så vasket tilstrekkelig med PBS(-) og en reaksjonsoppløsning ble tilsatt. Reaksjonsoppløsningen ble fremstilt ved å oppløse 4 mg 4-klor-l-nafthol i 1 ml methanol, tilsette 50 mmol tris-(hydroxymethyl)-aminomethan, 2 00 mmol NaCl og 5 ml av en buffer (pH = 7,4), og så tilsette hydrogenperoxyd slik at konsentrasjonen derav var 0,01%. Reaksjonen ble stanset ved å vaske platen med vann og så ble den lufttørket. (3) Identifisering av epitoper som er spesifikke til det monoklonale antistoff ifølge foreliggende oppfinnelse Resultatene av kvalitative tester og immunoreaksjoner av forskjellige typer glycolipider erholdt ved hjelp av TLC-fargingsteknikk (orcinol^farging) (A) og TLC-immunofargingsteknikk (B), er vist i figur 2. Det ble funnet fra resultatene vist i figur 2 at gangliosidet GD^ i felt 5 har en antigen determinant som er spesifikk for det monoklonale antistoff ifølge foreliggende oppfinnelse.
Dessuten påvises CDH (Gal-Glc-Cer), som ble fremstilt ved å la Vibrio cholera sialidase virke på gangliosidet GD^
i felt 3, ikke når det ble underkastet TLC-immunofarging. Dette viser klart at Gal-Glc-Cer (CDH)-strukturen ikke er en antigen determinant for det monoklonale antistoff ifølge foreliggende oppfinnelse. I tillegg indikerer båndet som kommer til syne i felt 3 i TLC-immunofargingen, gangliosid GD^ som ikke påvirkes av den ovenfor nevnte sialidase. I figur 2 viser CMH i felt 1 Gal-Cer og CMH i felt 2 Glc-Cer,
og CTH i felt 4 viser Gal-Gal-Glc-Cer.
Det er således funnet at disse strukturene ikke er antigene determinanter for det monoklonale antistoff ifølge foreliggende oppfinnelse ettersom de ikke ble farget ved hjelp av TLC-immunofargingsteknikken.
Som det sees ovenfor, ble det funnet at den antigene determinant for det monoklonale antistoff ifølge oppfinnelsen er N-acetylneuraminsyre-rester.

På samme måte er resultatene oppnådd ved å farge forskjellige gangliosider med N-acetylneuraminsyre-rester ved hjelp av TLC-immunofarging, oppsummert i tabell I nedenunder.
(4) Identifisering av epitop eller antigen determinant for det monoklonale antistoff ifølge foreliggende oppfinnelse Som i figur 2, viser figur 3 resultatene av kvalitative tester og immunoreaksjoner av forskjellige typer glycolipider erholdt ved TLC-fargingsteknikk (A) og TLC-immunofargingsteknikk (B). I figur 3 viser feltene 2 og 3 resultatene av naturlig forekommende gangliosid, med andre ord N-acetylneuraminsyre med alfa-2—>3-binding. Disse feltene viser klart at N-acetylneuraminsyren med alfa-2—>3-binding har sterk spesifisitet til det monoklonale antistoff ifølge foreliggende oppfinnelse.
I tillegg viser feltene 4 og 5 resultatene av kjemisk syntetiserte N-acetylneuraminsyrederivater. Disse resultatene indikerer at det syntetiserte gangliosid ikke har spesifisitet til det monoklonale antistoff ifølge foreliggende oppfinnelse.
Når man tar i betraktning både de foreliggende observa-sjoner og de som ble oppnådd under punkt (3), kan det konklu-deres med at det monoklonale antistoff ifølge foreliggende oppfinnelse gjenkjenner alfa-2—>3-bindingen i N-acetylneuraminsyre som sin epitop eller antigene determinant.

(5) Kvalitativ immunodiffusjonsteknikk (Ouchterlony-teknikk)
Figur 4 viser resultatene oppnådd ved immunodiffusjonsteknikk. Som det sees fra figur 4, er det monoklonale antistoff fremstilt ved hjelp av hybridomet ifølge oppfinnelsen IgM. Ved denne fremgangsmåte ble antiseraene anti-MIgM, anti-MlgG^, anti-MIgG2a, anti-MIgG2b og anti-MIgG3 erholdt kommersielt.

(6) Det faktum at det monoklonale antistoff ifølge foreliggende oppfinnelse er IgM demonstreres også gjennom resultatene oppnådd ifølge enzym-merket antistoffteknikk, idet resultatene som ble oppnådd ved fremgangsmåten er oppsummert i tabell II nedenunder.
Claims (3)
1. Monoklonalt antistoff,
karakterisert ved at det er et immunoglobulinmolekyl av IgM-type, er fremstilt ved hjelp av en hybridomcellelinje med ATCC deponeringsnummer HB 9475, binder både GD3-gangliosid og GM3-gangliosid immunologisk og gjenkjenner epitopen N-acetylneuraminsyre som har en a-2-*3-binding.
2. Monoklonalt antistoff ifølge krav 1, karakterisert ved at gangliosidet er GD3-gangliosid.
3. Hybridomcellelinje,
karakterisert ved at den har ATCC deponeringsnummer HB 9475 og produserer et monoklonalt antistoff som er et immunoglobulinmolekyl av IgM-type, binder både GD3-gangliosid og GM3-gangliosid immunologisk og gjenkjenner epitopen N-acetylneuraminsyre som har en a-2-»3-binding.
