Description
Fremstilling og farmasøytisk bruk av carboxyalkyldipeptider såvel som derivater og analoger derav, er blitt beskrevet i Europa-patentsøknad 12 401. Forbindelsene beskrevet i angitte Europa-patentsøknad er anvendbare som omdannende enzyminhibitorer og som antihypertensive midler.
Foreliggende oppfinnelse angår en fremgangsmåte for fremstilling av carboxyalkyldipeptider av den type som er beskrevet i den ovenfor angitte Europa-patentsøknad. Fremgangsmåten ifølge oppfinnelsen er således rettet mot fremstilling av carboxyalkyldipeptider av formel:
hvori R er aryl; substituert aryl hvori substituenten er lavere alkyl, lavere alkoxy eller halogen; en hetero-cyklisk gruppe som eventuelt er substituert med halogen, dihalogen, lavere alkyl, hydroxy, lavere alkoxy, acylamino, halogen-lavere alkyl; hvori aryl betyr fenyl, nafthyl eller bifenyl, og den heterocykliske gruppe betyr indolyl, thienyl, imidazolyl, furyl, benzimidazolyl, pyridyl, kinolinyl, isokinolinyl og benzothieny1, R^ er lavere alkyl med 1 til 4 carbonatomer; -NH-X- er alanin; glycin; isoleucin; leucin; lysin; fenylalanin; valin;
Y er alanin; glycin; isoleucin; leucin; lysin; fenylalanin; prolin; valin; og
farmasøytisk akseptable salter derav.
Foretrukne forbindelser av formel I hvori:
R er aryl, som eventuelt er substituert med lavere alkyl, lavere alkoxy, halogen;
-NH-X- og Y er som ovenfor definert.
Mere foretrukne forbindelser av formel I er slike hvori: R er fenyl;
er ethyl; og
X og Y er som ovenfor definert.
De mest foretrukne er forbindelser av formel I hvori: R er fenyl;
R^ er ethyl;
-NH-X- er alanin; og
Y er prolin.
De foretrukne forbindelser av formel I innbefatter også farmasøytisk akseptable salter derav.
Lavere alkylgruppene innbefatter rettkjedede, for-grenede og umettede hydrocarbonradikaler med 1-6 carbonatomer slik som methyl, ethyl, propyl, isopropyl, butyl, isobutyl, t-butyl, pentyl, isopentyl, hexyl. Aralkyl- og heteroaralkylgruppene har fra 1 til 6 carbonatomer i alkyl-delen og innbefatter benzyl, fenethyl, 3,3-difenylpropyl, 3-indolylmethyl. Halogen betegner klor, brom, jod eller fluor. Hvor aryl fremkommer i radikalene, betegner dette fenyl, nafthyl eller bifenyl. Heteroaryl innbefatter indolyl, thienyl, imidazolyl, furyl, benzimidazolyl, pyridyl, kinolinyl, isokinolinyl og benzothienyl. Acylamino angir lavere alkanoylami.no og aroylaminogrupper som acetylamino og benzylamino.
Fremgangsmåten ifølge foreliggende oppfinnelse er kjennetegnet ved at en forbindelse av formel
omsettes med et dipeptid under dannelse av et dicarboxydipeptid; dicarboxydipeptidet hydrogeneres i et surt medium i et alkohol-løsningsmiddel under ledsagende forestring til et dicarboalkoxydipeptid som en blanding av diastereomerer, hvoretter den diastereomere blanding separeres under dannelse av forbindelsene av formel I.
Som vist i reaksjonsskjemaet, omsettes 3-benzoylacrylsyre (II) med L-alanin-L-prolin-hemihydrat (III) i nærvær av ethanol og natriumbicarbonat under dannelse av dicarboxydipeptid IV. Dipeptid IV behandles deretter med ethanol og vannfritt hydrogenklorid, og den resulterende blanding under-kastes hydrogenering i nærvær av Pd/C-katalysator under dannelse av diester V som en blanding av diastereomerer. Denne diastereomere blanding V kombineres deretter med hydrogenbromid-eddiksyre og vandig hydrobromsyre, etterfulgt av ionebyttekromatografi og middeltrykks-væskekromatografi (MPLC) under dannelse av monoester I som en blanding av diastereomerer.
Ytterligere behandling av I med acetonitril og maleinsyre tilveiebringer et urent maleatsalt som deretter kan om-krystalliseres under dannelse av N-(1(S)-ethoxycarbonyl-3-fenylpropyl)-L-alanin-L-prolinmaleatsalt.
Fremgangsmåten ifølge oppfinnelsen tilveiebringer således en enkel, effektiv og økonomisk måte for å syntetisere forbindelser av formel I ettersom utgangsmaterialene og hovedreaktantene enten lett kan fremstilles eller er kommer-sielt tilgjengelige. I en foretrukken utførelsesform kan diesteren V fremstilles for lagring og transport og kan deretter omdannes til enten monoestersaltet eller en forbindelse av formel I.
Oppfinnelsen illustreres nærmere i de etterfølgende eksempler.
Eksempel 1
Ethyl- N-( l- carboethoxy- 3- fenylpropyl)- L- alanyl- L- prolinat
Til en løsning av 1,00 g (5,12 mmol) L-alanyl-L-prolin-hemihydrat og 1,04 g (5,91 mmol) 3-benzoylacrylsyre i 15 ml absolutt ethanol ble tilsatt 0,497 g (5,91 mmol) natriumbicarbonat, og blandingen ble omrørt i 48 timer. Etter til-setning av 15 ml ethanol ble blandingen avkjølt (isbad), og vannfritt hydrogenklorid ble ført inn i løsningen til denne var mettet. 5 ml ethanol ble tilsatt, og blandingen ble underkastet hydrogenering i nærvær av 0,10 g 10%-ig palladium-på-carbon i 48 timer ved 2,8 kg/cm 2. Etter fjerning av kata-lysatoren ved filtrering og fordampning ble det oljeaktige residuum tatt opp i CH2Ci0, og vannfri ammoniakk,ble ført gjennom løsningen inntil denne var basisk. Etter fordampning ble residuet oppslemmet med EtOAc og filtrert. Etter fordampning ble residuet underkastet kolonnekromatografi på silicagel med EtOAc som elueringsmiddel, under dannelse av 1.1 g(2,8 mmol) av diesteren i et utbytte på 54% som en blanding (1:1) av diastereomerer. TLC (silicagel; EtAc)
Rf = 0,50, 0,55.
NMR (CDC13): 1,1-1,3 (9H, m); 1,8-2,2 (6H, m); 2,6-2,8 (2H, m); 2,9-3,1 (1H, bred); 3,1-3,3 (1H, m); 3,4-3,6 (3H, m);
4,1-4,3 (4H, m); 4,4-4,6 (1H, m); 7,1-7,3 (5H, m).
IR (CHC13): 2960, 1730, 1640, 1430 cm"<1>
MS: m/e 404 (M+).
Eksempel 2
N-( l- carboethoxy- 3- fenylpropyl)- L- alanyl- L- prolin eller
dets maleatsalt
0,508 g (1,26 mmol) av diesteren ifølge eksempel 1 ble kombinert med hydrogenbromid-mettet (32%) eddiksyre (10 ml)
og 2,5 ml konsentrert vandig hydrobromsyre, og den resulterende løsning ble kraftig omrørt i 48 timer. Løsningen ble konsentrert og deretter rekonsentrert flere ganger fra H00. Det oljeaktige residuum ble brakt på en kolonne av 10 g Dowex® 50W-X4. Etter en begynnelses-vannvask av harpiksen ble produktet eluert med 25:1 H20:pyr. Konsentrering ga en farve-løs olje som TLC (silicagel; 100:20:3:0,5
CHCl^zCH^OHrH^O:H0Ac) viste besto av monoester som en blanding (1:1) av diastereomerer, Rf = 0,65, 0,70, ledsaget av en liten mengde disyre, Rf = 0,2. Middeltrykks-væskekromato-graf i på silicagel (E. Merck Lobar-kolonne, størrelse A) i 100:20:3:0,5 CHCl3:CH30H:H20:H0Ac ga monoester (0,382 g;
1.02 mmol; 81%) som en blanding (1:1) av diastereomerer.
NMR (D20): 1,30, 1,34 (3H, 2 x t, J = 7); 1,53, 1,58 (3H,
2 x d, J = 7); 1,8-2,0 (2H, m); 2,1-2,4 (4H, m); 2,7-3,0
(2H, m); 3,3-3,7 (2H, m); 3,8-4,2 (2H, m) ; 4,2-4,4 (3H, m); 7,3-7,5 (5H, m). IR (CHClj): 3500-2500, 2960, 1720, 1640, 1430 cm"<1>. Ms (FAB): m/e 377 (M<+> +1).
Separering av diastereomerene ble oppnådd ved motsatt-fase HPLC. Alternativt kan separeringen oppnås ved behandling av en løsning av 1:1-blandingen av isomerene i acetonitril-løsning med en acetonitrilløsning inneholdende en halvmolar ekvivalent av maleinsyre. Etter omrøring i 1 time ved rom-temperatur ble det faste materiale oppsamlet og skylt med acetonitril og lufttørket under dannelse av maleatsaltet,
som ved HPLC viste seg å være 96% rent. Det urene maleatsalt ble omkrystallisert fra acetonitril under dannelse av N-(l-(S)-ethoxycarbony1-3-fenylpropyl)-L-alanyl-L-grolinmaleatsalt, smp. 148-150°C, ved HPLC ca. 99% rent. [a]p<5>= -42° (CH3OH).
Eksempel 3
N-( l- carboxy- 3- oxo- 3- fenylpropyl)- L- alanyl- L- prolln
Til en løsning av 2,26 g (10,2 mmol) L-alanyl-L-prolin-hydroklorid og 1,97 g (11,2 mmol) 3-benzoylacrylsyre i 50 ml methanol ble tilsatt 1,71 g (20,3 mmol) natriumbicarbonat, og blandingen ble omrørt i 48 timer. Etter filtrering og fordampning ble residuet renset på Dowex® 50W-XY under anvendelse av 10:1 H20:CH30H og deretter 50:1 H-,:pyr som eluerings-midler. Frysetørking av E0:pyr-fraksjoner ga produktet som en blanding (1:1) av diastereomerer (2,68 g; 7,40 mmol; 73%).
MS: m/e 461 (bis-trimethylsilyl M® - C02TMS). TLC (silicagel; 3:1:1:1 EtOAc, BuOH, HOAc, H20), Rf = 0,35. NMR (D20): 1,08 (mindre), 1,18 (mindre), 1,26, 1,34 (3H, 4 xd, J=7)
(2 rotomerer); 1,5-2,2 (4H, m); 2,1-3,8 (4H, m); 3,9-4,3
(3H, m); 7,3-7,8 (5H, m) .
Disyreproduktet ifølge eksempel 3 kan erholdes anriket med diastereomer av (S,S,S)-konfigurasjon ved å utføre reaksjonen i et tofase-reaksjonsmedium bestående av vann og f.eks. ether eller diklormethan eller annet løsningsmiddel som ikke er blandbart med vann. Følgelig kan en løsning av en molar ekvivalent av 3-benzoylacrylsyre i det organiske løsnings-middel kombineres med en vandig løsning av L-alanyl-L-prolin inneholdende en molarekvivalent av en uorganisk base slik som kaliumbicarbonat. Reaksjonen kan utføres under forsiktig omrør-ing i nærvær av et kvartært ammoniumsalt slik som hexadecyl-
(r)
trimethylammoniumbromid eller Tritorr-^B. Spesielt fordelaktig med hensyn til å oppnå optimale utbytter av den angitte diastereomer er chirale ammoniumsalter slik som N,N-dimethyl-efedriniumbromid eller N-dodecyl-N-methylefedriniumbromid. Også anvendbare er kroneethere slik som 18-krone-6 og dibenzo-18-krone-6.
Også anvendbare for å øke mengden av den ovenfor angitte diastereomer er en fremgangsmåte ved hvilken en løsning av 3-benzoylacrylsyre i et organisk løsningsmiddel slik som f.eks. benzen, diklormethan eller acetonitril, kan tilsettes til L-alanyl-L-prolin-kaliumsalt, som fremstilt ved behandling av dipeptidet med en molarekvivalent vandig kaliumhydroxyd etterfulgt av frysetørking. Reaksjonen kan utføres under tilbake-løpskokning i nærvær av et kvartært ammoniumsalt slik som dodecyltrimethylammoniumbromid eller spesielt N-dodecyl-N-methylefedriniumbromid, eller en kroneether, f.eks. 18-krone-6.
I de ovenfor beskrevne prosesser kan ethyl-3-benzoyl-acrylat anvendes istedenfor 3-benzoylacrylsyre, hvilket fører til esteren N-(l-carboethoxy-3-oxo-3-fenylpropyl)-L-alanyl-L-prolin anriket med diastereomeren med (S,S,S)-konfigurasjonen.
I hvert tilfelle kan produktet isoleres ved konsentrering til tørrhet og rensing som ovenfor beskrevet på Dowex® 50W-X4 sur harpiks. Analyse av det diastereomere materiale kan utføres ved omvendt fase HPLC.
Alternativt kan residuet etter konsentrering taes opp i absolutt ethanol, mettes med vannfri hydrogenklorid og behandles videre som beskrevet i eksempel 1 under dannelse av ethyl-N-(l-carboethoxy-3-fenylpropyl)-L-alanyl-L-prolinat.
Claims (3)
1. Fremgangsmåte for fremstilling av carboxyldipep-tider av formel
hvor R er aryl; substituert aryl hvori substituenten er lavere alkyl, lavere alkoxy eller halogen; en hetero-cyklisk gruppe som eventuelt er substituert med halogen, dihalogen, lavere alkyl, hydroxy, lavere alkoxy, acylamino, halogen-lavere alkyl; hvori aryl betyr fenyl, nafthyl eller bifenyl, og den heterocykliske gruppe betyr indolyl, thienyl, imidazolyl, furyl, benzimidazolyl, pyridyl, kinolinyl, isokinolinyl og benzothienyl, R^ er lavere alkyl med 1 til 4 carbonatomer; -NH-X- er alanin; glycin; isoleucin; leucin; lysin; fenylalanin; valin;
Y er alanin; glycin; isoleucin; leucin; lysin; fenylalanin; prolin; valin; og
farmasøytisk akseptable salter derav, karakterisert ved at en forbindelse av formel
omsettes med et dipeptid under dannelse av et dicarboxydipeptid; dicarboxydipeptidet hydrogeneres i et surt medium i et alkohol-løsningsmiddel under ledsagende forestring til et dicarboalkoxydipeptid som en blanding av diastereomerer, hvoretter den diastereomere blanding separeres under dannelse av forbindelsene av formel I.
2. Fremgangsmåte ifølge krav 1, karakterisert ved at behandlingen av dicarboxydipeptidet utføres i nærvær av Pd/C-katalysator.
3. Fremgangsmåte ifølge krav 1, karakterisert ved at separeringen av den diastereomere blanding utføres ved ionebyttekromatografi og middeltrykk-væskekromatografi.
