Europeisk patentpublikasjon 106.489 og norsk patentansøkning 83.3206 (patent 164.418) beskriver 6-fluor-1,4-dihydro-4-okso-1,8-naftyridin-3-karboksylsyrer og 6,8-difluor-l,4-dihydro-4-oksokinolin-3-karboksylsyrer som antibakterieile midler.
Foreliggende oppfinnelse angår en fremgangsmåte for fremstilling av de korresponderende 5,6,8-trifluorkinoliner som har verdifulle terapeutiske, særlig antibakterieile egenskaper.
Oppfinnelsen angår således fremstillingen av forbindelser med formel I
hvor Z er
R2 er etyl eller cyklopropyl,
n er 0 eller 1, og
R3 er hydrogen eller alkyl med 1-4 karbonatomer.

formel med et amin som tilsvarer gruppen Z, med strukturformelen
hvor samtlige symboler er som angitt for formel I, og hvor L er en utgående gruppe, fortrinnsvis fluor eller klor.
Forbindelsene med formel I kan således fremstilles ved å behandle en korresponderende forbindelse med strukturformel IV med det aktuelle cykliske amin Va eller Vb. Alkylaminsubsti-tuenten i forbindelse Va eller Vb kan herunder eventuelt beskyttes med en gruppe som gjør den mer eller mindre inert overfor reaksjonsbetingelsene. Følgende beskyttelsesgruppe kan f. eks. benyttes: karboksylsyregrupper, så som formyl, acetyl, trifluoracetyl; alkoksykarbonylgrupper, så som etoksykarbonyl, t-butoksykarbonyl, 3,3,3-trikloretoksykarbonyl, 3-j odetoksy-karbonyl; aryloksykarbonylgrupper, så som benzyloksykarbonyl, p-metoksybenzyloksykarbonyl, fenoksykarbonyl; silylgrupper, så som trimetylsilyl; og grupper så som trityl, tetrahydropyranyl, vinyloksykarbonyl, o-nitrofenylsulfenyl, difenylfosfinyl, p-toluensulfonyl og benzyl. Beskyttelsesgruppen kan eventuelt fjernes efter reaksjonen mellom forbindelse IV og Va eller Vb ved hjelp av kjente fremgangsmåter. Eksempelvis kan etoksy-karbonylgruppen fjernes ved sur eller basisk hydrolyse og tritylgruppen fjernes ved hydrogenolyse.
Reaksjonen mellom forbindelsen med strukturformel IV og en passende beskyttet forbindelse med formel Va eller Vb kan utføres med eller uten oppløsningsmiddel, fortrinnsvis ved høyere temperatur i tilstrekkelig tid til at reaksjonen i det vesentlige blir fullstendig. Reaksjonen utføres fortrinnsvis i nærvær av en syreakseptor, så som alkalimetall- eller jordalkali-meta11-karbonat eller -bikarbonat, et tertiært amin så som trietylamin, pyridin eller pikolin. Et overskudd av forbindelse V kan alternativt benyttes som syreakseptoren.
Hensiktsmessige oppløsningsmidler for reaksjonen utgjøres av ikke-reaktive oppløsningsmidler som acetonitril, tetrahydrofuran, etanol, kloroform, dimetylsulfoksyd, dimetylformamid, pyridin, pikolin, vann og lignende. Blandinger av oppløsnings-midler kan også benyttes.
En hensiktsmessig reaksjonstemperatur ligger i området fra 20° til 150°C, idet høyere temperaturer vanligvis fordrer kortere omsetningstid.
Fjerningen av beskyttelsesgruppen kan foretas enten før eller efter isolering av produktet I. Beskyttelsesgruppen kan alternativt bibeholdes.
Utgangsforbindelsene med strukturformel IV er enten kjente forbindelser eller forbindelser som kan fremstilles fra kjente utgangsmaterialer ved standardmetoder eller varianter av sådanne.
Forbindelser med formel IV hvor R2 er etyl, kan f.eks. fremstilles fra tetrafluorbenzen som er et kommersielt til-gjengelig materiale. Tetrafluorbenzen blir først nitrert og nitrogruppen redusert til en aminogruppe som, uten isolering, omsettes med dietyl-etoksymetylenmalonat til dietyl-2,3,4,5-tetrafluoranilinmetylenmalonat som ved oppvarmning cykliseres til 5,6,7,8-tetrafluor-1,4-dihydro-4-okso-3-kinolinkarboksylsyre-etylester. Etylering av esteren og eventuell hydrolyse gir det passende mellomprodukt IV. Denne reaksjonsfølge er ytterligere illustrert i eksemplene.
Som et alternativ, spesielt når R2 er cyklopropyl, kan forbindelser med formel IV fremstilles fra kommersielt til-gjengelig pentafluorbenzoesyre. Syren omdannes til benzoyl-eddiksyre-etylester via syrekloridet. Mellomproduktet (2) omsettes med eddiksyreanhydrid og trietyl-ortoformiat og derefter med cyklopropylamin for å gi (3) som cykliseres ved oppvarmning med base. Forbindelsene anvendt i henhold til oppfinnelsen som har strukturformel Va eller Vb, er kjente forbindelser og kan fremstilles som beskrevet i europeisk patentpublikasjon 106.489.
Forbindelsene fremstilt i henhold til oppfinnelsen oppviser antibakteriell virkning ved undersøkelse etter mikrotitrering-fortynningsmetoden beskrevet i Heifetz et al., Antimicr. Agents & Chemoth., 6, 124 (1974).
Ved å benytte den siterte metode ble følgende verdier for minimal inhiberingskonsentrasjon (MIC) i ;jg/ml oppnådd for representative forbindelser fremstilt i henhold til oppfinnelsen.
SAMMENLIGNINGSFORSØK
CFI-mus, som var barbert på ryggen 24 timer tidligere, ble dosert med de forskjellige mengder av legemidlet på passende måte. 1 time efter doseringen ble musene utsatt for lys i UVA-området (320-400 nm) i en 3 timers periode i et tre-kammer-bur. Dette lange bølgelengde-området (UVA) i UVR-spektret er kjent for å fremkalle de fleste av de fototoksiske og fotoallergiske reaksjoner hos mennesker. Doseringen/eksponeringen ble gjentatt daglig i 4 dager eller inntil en tydelig positiv reaksjon var fremkalt. Den eksponerte huden til musen ble iakttatt daglig, ca. 24 timer efter bestråling for eventuelle fototoksiske reaksjoner, så som rødhet og svelning. De iakttatte reaksjoner ble bedømt med hensyn til graden av alvorlighet. Definisjonene av fototoksiske reaksjoner og graderingsskala er:
Graderingsskala - definisjon
= ingen reaksjon
1+ = noe rødflekket på huden
2+ = moderat sammenhengende rødhet på huden 3+ = alvorlig rødhet på huden med eller uten svelning.
Alle 15 mus som ble gitt 1-cyklopropylderivatet som er generisk beskrevet i norsk ansøkning 83.3206 (3 grupper i parallell på hver 5 mus), viste en første reaksjon (1+) efter den tredje orale dose på 10 mg/kg. 5 mus som hver fikk 4 mg/kg doser av forbindelsen ifølge eksempel 3 i henhold til foreliggende oppfinnelse, viste imidlertid ingen fototoksiske reaksjoner, mens det tidligere 1-cyklopropylderivat ifølge norsk ansøkning 83.6302 var kraftig fototoksisk ved 18 mg/kg ved dag 3. 1-cyklopropylderivatet ifølge norsk ansøkning 83.3206 viser seg derfor å ha et større fototoksisk potensiale enn forbindelsen ifølge eksempel 3 i henhold til oppfinnelsen.
Efter å ha fått to doser av 10 mg/kg av forbindelsen
ifølge eksempel 11 i norsk ansøkning 83.3206, viste alle de 5 undersøkte mus en moderat (2+) fototoksisk reaksjon. 5 mus som
fikk identiske doser av forbindelsen ifølge eksempel 1 i henhold til foreliggende oppfinnelse, viste ingen fotoreaksjoner. Bare liten reaktivitet (1+) ble iakttatt efter fire doser på 10 eller 18 mg/kg av forbindelsen ifølge eksempel ld i henhold til foreliggende oppfinnelse. 1-etylderivatet i henhold til eksempel 11 i norsk ansøkning 83.3206 har derfor et større fototoksisk potensiale enn forbindelsen ifølge eksempel ld i henhold til foreliggende forbindelse.
Forbindelsene fremstilt i henhold til oppfinnelsen kan danne farmasøytisk akseptable syreaddisjons- og/eller basesalter. Basesalter dannes med metaller eller aminer, så som alkali- og jordalkalimetaller eller organiske aminer. Eksempler på metallkationer er natrium, kalium, magnesium, kalsium og lignende. Eksempler på passende aminer er N,N'-dibenzyletylen-diamin, klorprokain, cholin, dietanolamin, etylendiamin, N-metylglukamin og prokain.
Farmasøytisk akseptable syreaddisjonssalter dannes med organiske og uorganiske syrer.
Eksempler på syrer egnet for saltdannelse er saltsyre, svovelsyre, fosforsyre, eddiksyre, sitronsyre, oksalsyre, malonsyre, salicylsyre, eplesyre, glukonsyre, fumarsyre, ravsyre, ascorbinsyre, maleinsyre, metansulfonsyre og lignende. Saltene fremstilles ved å bringe den frie baseform i kontakt med en tilstrekkelig mengde av den ønskede syre for å gi enten et mono- eller di- etc. salt på konvensjonell måte. Den frie baseform kan regenereres ved å behandle saltformen med en base. Eksempelvis kan fortynnede oppløsninger av vandige baser benyttes. Fortynnet vandig natriumhydroksyd-, kaliumkarbonat-, ammoniumhydroksyd- og natriumbikarbonatoppløsninger egner seg for dette formål. De frie baseformer skiller seg noe fra de respektive saltformer i visse fysikalske egenskaper, så som oppløselighet i polare oppløsningsmidler, men forøvrig er saltene i denne sammenheng likeverdige med de respektive frie baser. Bruk av baseoverskudd når R' er hydrogen gir de korresponderende basiske salter.
Forbindelsene fremstilt i henhold til oppfinnelsen kan forekomme både i usolvatiserte og solvatiserte former, innbefattet hydratformer. Generelt er de solvatiserte formene, innbefattet hydratformene og lignende, likeverdige med de usolvatiserte former.
Noen av forbindelsene kan forekomme i optisk aktive
former. Den rene D-isomer, rene L-isomer og blandinger av disse, innbefattet racemiske blandinger, kan fremstilles i henhold til oppfinnelsen. Ytterligere asymmetriske karbonatomer kan forekomme i en substituent, f.eks. i en alkylgruppe. Fremstillingen av slike isomerer og blandinger av disse skal også omfattes av oppfinnelsen.
Eksempel 1
7-[ 3-( aminometyl)- 1- pyrrolidinyl]- 1- etyl- 5, 6, 8- trifluor- 1, 4-dihydro- 4- okso- 3- kinolinkarboksylsyre ( A)
Til 1,0 g (3,5 mmol) 1-etyl-5,6,7,8-tetrafluor-1,4-dihydro-4-okso-3-kinolinkarboksylsyre i 20 ml acetonitril ble det tilsatt 1,05 g (6,9 mmol) 1,8-diazabicyklo[5.4.0]undek-7-en og 0,35 g (3,5 mmol) 3-pyrrolidinmetanamin og blandingen ble omrørt ved 45°C i 18 timer og deretter tilbakeløpsbehandlet i 1 time. Blandingen ble avkjølt, fortynnet med 50 ml dietyleter og filtrert. Faststoffet ble vasket med etanol og deretter med dietyleter for å gi 1,04 g (82 %) av tittelforbindelsen,
smp. >170°C (dekomponering).
På analog måte ble følgende forbindelser fremstillet:
(B) 1-etyl-5,6,8-trifluor-1,4-dihydro-7-(7-metyl-2,7-diazaspiro[4.4]nonan-2-yl)-4-okso-3-kinolinkarboksylsyre,
smp. 253-255°C; (C) 1-etyl-5,6,8-trifluor-1,4-dihydro-7-[3-[(metylamino) metyl]-1-pyrrolidinyl]-4-okso-3-kinolinkarboksylsyre,
smp. 225-227°C; og
(D) 1-etyl-7-[3-[(etylamino)metyl]-1-pyrrolidinyl]-5,6,8-trif luor-1 ,4-dihydro-4-okso-3-kinolinkarboksylsyre,
smp. 224-227°C.
Eksempel 2
1- cyklopropyl- 7-[ 3-[( etylamino) metyl]- 1- pyrrolidinyl]- 5, 6, 8-trifluor- 1, 4- dihydro- 4- okso- 3- kinolinkarboksylsyre ( A)
Til 1,0 g (3,32 mmol) 1-cyklopropyl-5,6,7,8-tetrafluor-1,4-dihydro-4-okso-3-kinolinkarboksylsyre i 9 ml acetonitril ble det tilsatt 0,5 g (en ekvivalent) 1,8-diazabicyklo[5.4.0]-undek-7-en og 0,42 g N-etyl-3-pyrrolidinmetanamin i ytterligere 3 ml acetonitril. Blandingen ble oppvarmet til 6 0°C i 4 timer og omrørt ved 25°C i 18 timer. Blandingen ble filtrert og faststoffet vasket med dietyleter for å gi 1,12 g tittelforbindelse,
smp. 24 7-248°C.
På analog måte ble følgende forbindelse fremstillet:
( B.) 7-[ 3-(aminometyl)-1-pyrrolidinyl]-1-cyklopropyl-5,6 ,8-trif luor-1,4-dihydro-4-okso-3-kinolinkarboksylsyre , sm.p. 180-240°C (d)
Eksempel 3
7-[ 3- amino- 1- pyrrolidinyl]- 1- cyklopropyl- 5, 6, 8- trifluor- 1,4-dihydro~ 4- okso- 3- kinolinkarboksylsyre
Til 2,0 g (6,64 mmol) 1-cyklopropyl-5,6,7,8-tetrafluro-1,4-dihydro-4-okso-3-kinolinkarboksylsyre i 10 ml acetonitril ble det tilsatt 1,5 g (8,3 mmol) 3-t-butoksykarbonylamino-
pyrrolidin og 1,01 g (6,64 mmol) 1,8-diazabicyklo[5.4.0]-
undek-7-en. Blandingen ble omrørt over natten og tilbakeløps-behandiet i 2 timer. Faststoffene ble frafiltrert og vasket med eter. De ble behandlet med 5 ml trifluoreddiksyre og omrørt ved 25°C i 1,5 timer. Trifluoreddiksyren ble fjernet og blandingen tatt opp i vann. Deretter ble pH justert til 7,0 og fast-
stoffene frafiltrert og tørket for å gi 2,0 g av tittelfor-
bindelsen, smp. 290-292°C.
Fremstilling av utgangsmaterialene
Eksempel A
2, 3, 4, 5- tetrafluor- 1- nitrobenzen
Til en liter konsentrert svovelsyre ble det ved 5°C tilsatt 10 ml 1,2,3,4-tetrafluorbenzen. Deretter ble en blanding av 100 ml 70 % salpetersyre, som på forhånd var blandet med 200 ml konsentrert svovelsyre, langsomt tilsatt ved 0°C. Reaksjonsblandingen ble omrørt i 1 time ved 0°C og deretter 1 time ved 25°C. Blandingen ble helt over is og ekstrahert med diklormetan som ble tørket og konsentrert for å gi 120 g av et tykt residuum som ved tynnskiktkromatografi ga én flekk. Dette produkt ble benyttet i det neste trinn uten rensing.
Eksempel B
Dietyl- 2, 3, 4, 5- tetrafluoranilinmetylenmalonat
Til 48,5 g (0,25 mol) 2,3,4,5-tetrafluor-1-nitrobenzen i 500 ml 2-propanol ble det tilsatt 3,0 g Raney-nikkel og ledet inn hydrogengass under et trykk på 1,4 kg/cm 2. Etter 19 timer ble blandingen luftet og filtrert direkte over i en blanding av 58 ml dietyletoksymetylenmalonat og 500 ml toluen. Toluenen ble avdestillert i løpet av 3 timer og residuet behandlet med pentan. Faststoffet ble filtrert for å gi 6 2,5 g av tittelforbindelsen, smp. 115-116°C.
Eksempel C
5, 6, 7, 8- tetrafluor- 1, 4- dihydro- 4- okso- 3- kinolinkarboksylsyre-etylester
Til 700 ml kokende "dowtherm" ble det tilsatt 62,5 g (187 mmol) dietyl 2,3,4,5-tetrafluoranilinmetylenmalonat i tre porsjoner. Etter 30 minutter ble blandingen avkjølt og fortynnet med dietyleter og filtrert. Faststoffet ble vasket med dietyleter for å gi 40,0 g (74 %) av tittelforbindelsen,
smp. 282-283°C.
Eksempel D
1- etyl~ 5, 6, 7, 8- tetrafluor- 1, 4- dihydro- 4- okso- 3- kinolin-karboksy lsyre- etylester
Til 38,5 g (134 mmol) 5,6,7,8-tetrafluor-1,4-dihydro-4-okso-3-kinolinkarboksylsyre-etylester i 700 ml N,N-dimetylformamid ble det tilsatt 37,15 g (to ekvivalenter) kaliumkarbonat og 50 ml (fem ekvivalenter) etyljodid. Blandingen ble brakt til 50°C og kraftig omrørt over natten. Oppløsnings-midlene ble fjernet og residuet fordelt mellom vann og diklormetan. Diklormetanfasen ble tørket (magnesiumsulfat) og konsentrert. Residuet ble utgnidd med dietyleter for å gi 30,5 g av et materiale som ble omkrystallisert fra etanol. Filtrering førte til 23,5 g tittelforbindelse, smp. 217-220°C.
Eksempel E
1- etyl- 5, 6, 7, 8- tetrafluor- 1, 4- dihydro- 4- okso- 3- kinolin-karboksylsyre
Til 20,0 g (63 mmol) 1-etyl-5,6,7,8-tetrafluor-1,4-dihydro-4-okso-3-kinolinkarboksylsyre-etylester ble det tilsatt 200 ml eddiksyre og 150 ml 6N saltsyre i 70 ml vann. Blandingen ble holdt ved 100°C i 2 timer. Den ble deretter avkjølt, filtrert og vasket med dietyleter for å gi 17 g av tittelforbindelsen smp. 229-230°C.
Eksempel F
Pentafluorbenzoyleddiksyre- etylester
Til 20,0 g (0,094 mmol) pentafluorbenzosyre i 175 ml diklormetan ble det tilsatt 9,0 ml (1,1 ekvivalenter) oksalyl-klorid og 3 dråper N,N-dimetylformamid. Blandingen fikk stå
over natten og ble deretter konsentrert. Residuet ble oppløst i 100 ml tetrahydrofuran og benyttet uten rensing i det neste trinn.
Til 26 g (to ekvivalenter) malonsyre-halv-etylester i
600 ml tetrahydrofuran ble det ved -10-0°C tilsatt 179 ml
(fire ekvivalenter) 2,2N n-butyllitium. Etter at dianion-dannelsen var fullført, ble blandingen avkjølt til -78°C og det ovenfor fremstilte syreklorid langsomt tilsatt. Etter endt
tilsetning ble blandingen brakt til -3 5°C og omrørt i 1 time. Den ble deretter helt over 94 ml 2N saltsyre og 200 g is.
Hele blandingen ble ekstrahert med diklormetan og oppløsningen deretter ekstrahert med mettet natriumbikarbonat. Diklormetanfasen ble tørket og konsentrert for å gi 14,3 g av tittelforbindelsen som ble isolert i form av en lysegul olje etter søylekromatografi (toluen:heksan:eter; 6:3:1).
Eksempel G
2-( pentafluorbenzoyl)- 3- cyklopropylaminoakrylsyre- etylester
Til 14,0 g (49,6 mmol) pentafluorbenzoyleddiksyre-etylester ble det tilsatt 12,1 g eddiksyreanhydrid og 10,9 g tri-etylortoformiat. Blandingen ble tilbakeløpsbehandlet ved 150°C i 2,2 timer. Den ble deretter avkjølt til 8 0°C og konsentrert i 1,5 timer. Blandingen ble deretter avkjølt til 45°C og omsatt med 2,83 g cyklopropylamin i 100 ml 2-propanol. Blandingen ble omrørt over natten og konsentrert. Residuet ble behandlet med pentan og faststoffet frafiltrert for å gi 11,36 g av tittelforbindelsen som et blekgult pulver, smp. 85-86°C.
Eksempel H
1- cyklopropyl- 5, 6, 7, 8- tetrafluor- 1, 4- dihydro- 4- okso- 3-kinolinkarboksylsyre
Til 11,43 g (32,75 mmol) 2-(pentafluorbenzoyl)-3-cyklopropylaminoakrylsyre-etylester i 250 ml dioksan ble det tilsatt 4,99 g 1,8-diazabicyklo[5.4.0]undek-7-en ved 85°C. Blandingen ble omrørt i 2 timer og ble deretter fordelt mellom vann og diklormetan. Diklormetanfasen ble vasket med 1N saltsyre, tørket og behandlet med aktivkull. Blandingen ble filtrert, konsentrert og renset ved søylekromatografi (kloroform:heksan: 2- propanol; 6:3:1) for å gi 2,7 5 g av et produkt som ble opp-løst i 40 ml eddiksyre og 8 ml 2N saltsyre. Etter 2 timer ved 100°C ble 20 ml vann tilsatt. Blandingen ble avkjølt og faststoffet frafiltrert for å gi 2,5 g av tittelforbindelsen,
mp. 179-180°C.
Claims (3)
1. Analogifremgangsmåte for fremstilling av terapeutisk aktive forbindelser med formel I
hvor Z er
R2 er etyl eller cyklopropyl,
n er 0 eller 1, og
R3 er hydrogen eller alkyl med 1-4 karbonatomer, karakterisert ved omsetning av en forbindelse med den generelle formel
med et amin som tilsvarer gruppen Z, med strukturformelen
hvor samtlige symboler er som angitt for formel I, og hvor L er en utgående gruppe, fortrinnsvis fluor eller klor.
2. Fremgangsmåte ifølge krav 1, for fremstilling av l-etyl-7-[3-[(etylamino)metyl]-1-pyrrolidinyl]-5,6,8-trifluor-1,4-dihydro-4-okso-3-kinolinkarboksylsyre, karakterisert ved anvendelse av tilsvarende utgangsmaterialer.
3. Fremgangsmåte ifølge krav 1, for fremstiling av 7-[3-amino-l-pyrrolidinyl]-l-cyklopropyl-5,6,8-trifluor-1,4-dihydro-4-okso-3-kinolinkarboksylsyre, karakterisert ved anvendelse av tilsvarende utgangsmaterialer.
