Description
Fra tyske patentsøknader P 20 32 433, P 23 08 584,
P 23 08 584, P 24 45 791, P 25 37 765 og P 26 37 162 er væske-forstøvere tidligere kjent som kan brukes til fremstilling av inhalerbare aerosoler av medikamentholdige oppløsninger. I dis-se apparater overføres svingninger fra et piezoelektrisk svingesystem til en forstøvningsplate. Den nevnte plate fremstiller av den tilførte væske fine smådråper som med luft danner en aerosol.
Ved anvendelse av inhaleringspreparater for bronkospasmo-lyse er det vesentlig at en tilstrekkelig andel av aerosolens dråper er så små at dråpene med åndeluften kan transporteres til påvirkningsstedet i lungen. Hvis dråpene er for store, vil de slå seg ned allerede i munnen eller i de øvre åndedrettsveier. Den tilstrebte virkning vil da ikke eller bare delvis kunne opp-nås. En ulempe ved den kjente væskeforstøver er at den leverer en altfor stor andel av forholdsvis store dråper.
Hensikten med oppfinnelsen er å unngå denne ulempe og oppfinnelsen angår derfor inhaleringsapparater med en forstøvnings-innretning på basis av et piezoelektrisk svingesystem og hensikten ifølge oppfinnelsen er oppnådd ved at arbeidsflaten av for-støverplaten er overtrukket med et tynt sjikt av en elastomer.
Det har nemlig overraskende nok vist seg at belegging av forstøvningsplaten med en tynn elastomerfilm resulterer i en be-tydelig forbedring av fordelingsspektret i den fremstilte aerosol .
Elastomerens sjikttykkelse ligger mellom 50 og 500 ym og fortrinnsvis mellom 100 og 300 ym. Forstøvningsplaten består fortrinnsvis av et metall med hensiktsmessige fysikalske og kje-miske egenskaper, særlig rustfritt stål. Til beskiktning kan brukes de forskjelligste elastomerer, f.eks. i form av konsen-trerte polymerløsninger, hvor løsningsmidlet fjernes ved fordamp-ning eller elastomerer kan anbringes på flaten ved polymerisering av monome-rer eller prepolymerer. Ved bruk av polyuretaner som dannes på overflaten, er ingen forbehandling nødvendig unntatt rengjøring av platen. Ved påføring av andre elastomerer kan det være nød-vendicr med forbehandling av arbeidsflaten med en primer for å forbedre bindestyrken mellom elastomeren og metallet.
Det nedenstående eksempel omtaler påføringen av en elastomer på en forstøvningsplate av rustfritt stål (ifølge den tyske
publikasjon DE-OS 2 308 584).
Eksempel
Arbeidsflaten til forstøvningsplaten av et inhaleringsapparat ifølge DE-OS 2 308 584 ble renset med metylenklorid og et fuktighetsherdende førstetrinn av polyuretan (50%-ig løsning av diisocyanatfenylmetan og en flerverdig polyeteralkohol i vann-fri toluol innstilt på en viskositet på 23 poise) ble påført arbeidsflaten. Når platen er innstilt horisontalt, fordeler blandingen seg slik over arbeidsflaten at et jevnt sjikt dannes. Herdningen ble avsluttet etter 24 timer (ved ca. 18°C og med relativ luftfuktighet på 50%) . Det ble fremstilt en film med 200 yra sjikttykkelse, en bruddutvidelse på 340% og en strekk-fasthet på 0,35 kg/mm<2>.
Ved bruken av det på denne måte utstyrte inhaleringsapparat ble det med en dosis på 200 yg fenoterol ved hundeforsøk (acetylkolin-bronkospasmus) oppnådd en like god virkning som med en handelsvanlig drivgassdreven doseringsaerosol med samme dosis pr. slag.
Ved anvendelse av apparatet ifølge DE-OS 2 308 584 oppnåd-de man derimot i sammenligningsforsøket ingen virkning.
Claims (3)
1. Inhaleringsapparat med en forstøvningsplate som svingnin-gene fra et piezoelektrisk svingesystem overføres til, karakterisert ved at forstøvningsplatens arbeidsflate er overtrukket med et tynt sjikt av en elastomer.
2. Apparat ifølge krav 1, karakterisert ved at elastomersjiktet på arbeidsflaten har en tykkelse fra 50 til 500 ym og fortrinnsvis fra 100 til 300 ym.
3. Apparat ifølge krav 1 eller 2, karakterisert ved at elastomersjiktet består av et polyuretan.
