Analogifremgangsmåte for fremstilling av terapeutisk aktive 6[a-(amidino- og imidoylamino-alkanoylamino)-fenylacetylamino]-penicillan-syrer.
Description
Oppfinnelsen gjelder en fremgangsmåte for fremstilling av 6-[2-fenyl-2-(amidino- og imidoylamino-alkanoylamino)-acetamido]-penicillansyrer og deres farmasøytisk akseptable salter, som har en høy antibakteriell aktivitet, spesielt mot gram-negative mikroorganismer.
Penicillinforbindelser skiller seg fra hverandre ved variabelen Rx og har den generelle formel som angis nedenfor hvor acyIdelen av 6-acylaminopenicillansyren
oppnås fra en karboksylsyre eller et funksjonelt derivat av denne som f.eks. et acylhalogenid eller -anhydrid.
De farmakodynamiske egenskaper og den antibiotiske profil for et gitt penicillin bestemmes i.høy grad av naturen av R -gruppen. De mest brukte penicilliner er de hvor R -delen
x x
har betydningen benzyl , fenoksymetyl eller o-fenoksyetyl . Mens disse velkjente analoger er meget aktive mot gram-positive mikroorganismer, har de begrenset gram-negativ aktivitet. Derfor er medisiner som vil bekjempe en økning av gram-negative infeksjoner, f.eks. E. coli,Pseudomonas eller Klebsiella, av verdi for den medisinske profesjon.
Nyere anstrengelser for å forbedre aktiviteten hos penicillin-familien har resultert i syntese av mange nye midler, a-karboksybenzylpenicillin (US-patent 3.142.673), et bred-spektret antibiotikum, er rapportert å ha større effektivitet mot gram-negative bakterier inngitt parenteralt, men har begrenset aktivitet ved oral administrasjon. a-aminoarylmetyl-penicilliner og slektninger av dem (US-patenter 2.985.648, 3.140.282, 3.373.156, 3.308.023 og 3.342.677) er kjente, mén har et begrenset spektrum når det gjelder aktivitet mot visse gram-negative mikroorganismer. Både gram-negativ og gram-positiv aktivitet angis for 6-ureido-penicillansyrederivater i US-patenter 3.180.863, 3.120.512 og 3.118.877 og for a-ureido-penicilliher i US-patent 3.352.851. Aktivitet mot graiti-negative bakterier, spesielt av slekten Pseudomonas, angis for a-karbamyl-ureidopenicilliner (US-patent 3.483.118) og a-alkoksykarbonyl-ureidopenicilliner (US-patent 3.481.922). Nylig er a-guanylureido-penicilliner (US-patent 3.579.501) og a-imidoylureidopenicilliner blitt rapportert å være nyttige mot infeksjonssykdommer, spesielt de som er forårsaket av slekten Pseudomonas.

er i besittelse av høy antibakteriell aktivitet, spesielt mot gram-negative.mikroorganismer.
Som fagmannen lett vil forstå er a-karbonatomet på penicillinsidekjeden til hvilken amidino- eller imidoylamino-alkanoylaminodelen er festet, et asymmetrisk karbonatom som gir grunnlag for eksistensen av to optisk aktive isomerer, D- og L-diastereoisomerene, såvel som racematet, DL-formen. Overens-stemmende med tidligere funn angående aktiviteten åv slike penicilliner som har asymmetriske a-karbonatomer, er forbindelsene fremstilt ifølge oppfinnelsen med D-konfigurasjon mer aktive enn de med L-konfigurasjon og er de foretrukne forbindelsene ,. selv om fremstillingen av L- og DL-formené av disse forbindelsene omfattes av oppfinnelsen.
Fremgangsmåten ifølge oppfinnelsen karakteriseres ved at

hvor Z er som ovenfor definert og X er -0H eller -Cl i nærvær av et middel til å fjerne henholdsvis vann eller HC1,
og om ønsket dannes de farmasøytisk akseptable salter derav.
I praksis omsettes forbindelsen med formel II i form av trietylamin- (TEA) salt og hydrokloridet av syrekloridet. med formel III, hvor X er klor, i et reaksjonsinert, aprotisk løsningsmiddel i nærvær av en hydrogenhalogenidfjerner, som f.eks. et tertiært amin, ved -3'0 til 0°C. Vanligvis er det fordelaktig å anvende et overskudd syreklorid så stort som 100-200%. Under disse betingelser er det nødvendig å tilsette en.tilstrekkelig mengde tertiært amin, fortrinnsvis trietylamin, for å nøytralisere hydrogenkloridet fra syrekloridet såvel som hydrogenkloridet som utvikles ved reaksjonen mellom syrekloridet og a-aminogruppen i penicillinet med formel II.
Det reaksjonsinerte løsningsmidlet som omfatter væskefasen i nevnte reaksjonsblanding, bør være et som ikke reagerer i særlig grad med noen av reaktantene eller reaksjonsproduktet. De foretrukne løsningsmidlene bør være vannfrie, aprotiske, polare løsningsmidler som f.eks. dimetylformamid eller heksametylfosforamid.
Selv om den første kontakt mellom reaktantene utføres ved isbadtemperatur, for å redusere forekomsten av biprodukter, er det ofte ønskelig efter noen få minutters blanding å la reaksjonsblandingen få oppvarmes til romtemperatur, inntil reaksjonen er fullført eller nesten fullført. Det rene produktet blir så filtrert og tørret.

Penicillinet med formel II er kjent og beskrevet i US-patenter 2.985.648 og 3.342.677 og av Long et al, J. Chem. Soc. 1920 (1971), mens syrekloridhydrokloridene lett syntetiseres fra de tilsvarende syrene ved anvendelse av tionylklorid eller fosforpentaklorid. De tilsvarende syrer fremstilles ved syntesemetoder som er velkjente for fagmannen. Verd å legge merke til i denne forbindelse er metodene som
angis av Ried et al., Ann. 661, 76 .(196 3) , Ried et al, Chem. Ber. , 95, 728 (1962), McElvain et* al. , J. Am. Chem. Soc, 71, 40 (1949), Bauer et al., J. Org. Chem. 27, 4382 (1962) og Cardellini et al., Ann. Chim. (Rom), 58, 183, 1199 (1958). ;Når X i formel III er OH, bringes hensiktsmessig ekvimolare mengder av hydrokloridet av den aktuelle alkan-syre med formel III, p-nitrofenol og dicykloheksylkarbodiimid (DCC) i kontakt i et reaksjons-inert løsningsmiddel, som f.eks. de som er angitt ovenfor, ved omgivelsestemperatur. Efter 1 til 3 timers forløp behandles mellomproduktet, p-nitrofenylesteren, ;som er frembragt in situ, med en ekvimolar eller litt mindre . mengde av penicillinet med formel II, fortrinnsvis som.et basisk tertiært aminsalt. Reaksjonstiden kan variere fra 1 til 6 timer avhengig av temperatur, konsentrasjon og reaktivitet for de anvendte reagenser. ;Produktet isoleres efter filtrering av reaksjonsblandingen. ved tilsetning av reaksjonsfiltratet til et stort volum diétyl-eter. Produktet kan renses., om ønsket, ved å anvende trietylamin som nevnt i den tidligere diskuterte fremgangsmåté. ;Forbindelsene fremstilt ifølge- oppfinnelsen er i stand ;til å danne salter. Forbindelsene ;fremstilt ifølge oppfinnelsen omdannes til basiske salter ved innvirkning av en passende base på nevnte syre i et vandig eller ;ikke-vandig medium. Slike basiske reagenser som passende anvendes ved fremstillingén av nevnte salter kan variere i natur og skal ;omfatte slike baser som f.eks. organiske aminer, ammoniakk, alkali-metallhydroksyder, -karbonater, -bikarbonater, -hydrider og -alkoksyder, såvel som jordalkalimetallhydroksyder, -hydrider, -alkoksyder og -karbonater. Representative for slike baser er ammoniakk, primære aminer som f.eks. n-propylamin, n-butylamin, anilin, cykloheksylamin, benzylamin, p-toluidin, etylamin, oktylamin, sekundære aminer som f.eks. dicykloheksylamin og tertiære aminer som f.eks. dietylanilin, N-metylpyrrolidin, N-metylmorfolin og 1,5-diazabicyklo-(4,3 ,0)-5-nonen, natrium-hydroksyd, kaliumhydroksyd, ammoniumhydroksyd, natriumetoksyd, kaliummetoksyd, magnesiumhydroksyd, kalsiumhydrid og barium-hydroksyd. ;Som fagmannen lett vil forstå er forbindelsene fremstilt ifølge oppfinnelsen tilstrekkelig basiske på grunn av amidino- og imidoylaminodelene til å danne syreaddisjonssalter. ;Farmasøytisk akseptable syreaddisjonssalters fremstilling ligger også innenfor oppfinnelsens område. ;Ved utnyttelsen av den kjemoterapeutiske aktiviteten for de forbindelser fremstilt ifølge oppfinnelsen som danner basiske salter, må det naturligvis anvendes farmasøytisk akseptable salter. De nevnte farmasøytisk akseptable saltene omfatter fortrinnsvis natrium-, aluminium-, kalium-, kalsium-, magnesium-, ammonium- og substituerte ammoniumsalter, f.eks. procain, dibenzylamin, N>N,bis(dehydroabietyl)etylendiamin, 1-efenamin, N-etylpiperidin, N-berizyl-j3-fenetylamin, N,N '-dibenzyl-etylendiamin, trietylamin, så vel som salter med andre aminer som har vært brukt til å danne salter med benzylpenicillin. ;De nye penicillinene som er beskrevet her oppviser in vitro aktivitet mot en lang rekke mikroorganismer, omfattende både ;gram-pqsitive og gram-negative bakterier. Deres ;aktivitet kan lett demonstreres ved in vitro-tester mot forskjellige organismer i et hjerne-hjerte infusjonsmedium ved den vanlige dobbelt-seriefortynningsteknikken. In vitro-aktviteten for de her beskrevne forbindelser gjør dem nyttige for lokal anvendelse i form av salver, kremer og lignende. ;Disse nye penicilliner er også effektive antibakterielle ;midler in vivo i dyr, omfattende mennesket, ikke bare via parenteral administrasjon, men også ved oral administrasjon. ;Den dosering som er mest egnet for den enkelte pasient, ;vil variere med alder, vekt og responsen for den spesielle pasienten såvel som naturen og graden av symptomene, naturen av den bakterieinfeksjon som behandles og de farmakodynamiske egen-skapene for det spesielle midlet som skal administreres. Det vil ofte bli funnet at når forbindelsen administreres oralt, er det nødvendig med større mengder aktivt stoff for å gi det samme nivå,, som frembringes av en liten mengde administrert parenteralt. ;Idet det tas fullt hensyn til de foregående faktorer ansees ;det at en effektiv daglig dose av forbindelsene fremstilt ifølge oppfinnelsen for mennesker for effektivt å lindre symptomene på sykdommen er.ca. 10-100 mg/kg/dag, med et foretrukket område på ;ca. 50-75 mg/kg/dag i enkle eller oppdelte doser, og en parenteral dose på 25-100 mg/kg/dag, med et foretrukket område på ca. 20-75 mg/ kg/dag. Disse verdier er bare orienterende, og det kan naturligvis være individuelle tilfeller hvor høyere eller lavere doser kan brukes. ;Som nevnt tidligere er penicilliner fremstilt ifølge oppfinnelsen bredspektrede antibiotika som ulik mange penicillin-analoger er meget antagonistiske mot gram-negative mikroorganismer, spesielt mot E. coli, Pseudomonas og Klebsiella. Videre later de til å være mer resistente enn de fleste penicilliner mot øde-leggelse av penicillinase, et enzym som produseres av visse bakterier som nedbryter penicillin til en inaktiv penicillansyre. ;Det antimikrobielle spekter for et antall forbindelser fremstilt ifølge oppfinnelsen er gjengitt i de følgende tabeller. ;Tabell I sammenligner in vitro-profi len for 6-[D^-fenyl^-famidinopropionamido) acetamido] penicillansyre (PAP) med ampicillin (AMP) og carbenicillin (CAR). Testforbindelsen administreres til den infiserte mus ved et flerdoseringsregime hvor den første dose gis 0,5 time efter inokulering og gjentas fire og fireogtyve timer senere. ;De nye produktene fremstilt ifølge oppfinnelsen er av verdi som antibakterielle midler og er bemerkelsesverdig effektive ved behandling av infeksjoner forårsaket av følsomme' gram-negative og gram-posftive bakterier hos fjærfe og dyr, inkludert mennesket. For slike formål kan det rene materialet eller blandinger av dem med andre antibiotika anvendes. De kan administreres alene eller kombinert med en farmasøytisk bærer på basis av den valgte administrasjonsmåten og standard farmasøytisk praksis. Eksempelvis kan de administreres oralt i form av tabletter inneholdende slike bindemidlér som stivelse, melkesukker, visse ...typer leire osv., eller i kapsler al ene eller sammen med de samme eller, ekvivalente bindemidlér. De kan også administreres oralt i form av eliksirer eller orale suspensjoner som kan inneholde smaks- eller farve-stoff eir, eller kan injiseres parenteralt, dvs. intramuskulært eller subkutant. For parenteral administrasjon brukes forbindelsene best i form av en steril vandig oppløsning, som kan være enten vandig som f.eks. vann, isotonisk saltløsning, isbtonisk dekstrose, Ringer's oppløsning, eller ikke-vandig som f.eks. fete oljer av vegetabilsk opprinnelse (bomullsfrø-, peanøtt-, mais- sesamolje) og andre ikke-vandige hjelpemidler som ikke vil påvirke den terapeutiske effektiviteten av preparatet og er ikke-toksisk i det volum eller forhold det anvendes i (glyserin, propylenglykol, sorbitol). I tillegg kan det lages preparater for fremstilling av oppløsninger før administrering. Slike preparater kan inneholde flytende fortynningsmidler, f.eks. propylenglykol, dietylkarbonat, glyserol, sorbitol, osv, og buffermidler, såvel som lokalanestetiske midler og uorganiske salter for å gi ønskede farmakologiske egenskaper. .......... De, følgende eksempler angis for å illustrere oppfinnelsen. ;Eksempel 1 *
6-[ D- 2- fenyl- 2-( 3- amidinopropionamido) acetamido] penicillansyre
(Z = -CO-CH2-CH2-QNH^-NH2)
Til en suspensjon av 1,35 g (3 mmol) D-a-aminobenzylpenicillin-trietylaminsalt i 15 ml tørr dimetylformamid holdt under. : nitrogen tilsettes 0,63 ml (4,5 mmol) trietylamin og blandingen avkjøles i et isbad til 0°C. 3-amidinopropionylkloridhydroklorid (770 mg, 4,5 mmol) tilsettes og blandingen omrøres i 15 minutter ved 0°C og ytterligere 237 mg (1,5 mmol) av syrekloridhydro-kloridet og 0,21 ml (1,5 mmol) trietylamin tilsettes og reaksjonsblandingen omrøres i en time ved romtemperatur. De faste stoffene frafiltreres og filtratet tilsettes dråpevis til 200 ml dietyleter under omrøring. Det rå, tørkede produktet, 1,2 g, suspenderes i 200 ml metylenklorid og behandles med 1 ml trietylamin. Efter omrøring i 30 minutter filtreres det rensede produktet og tørkes under redusert trykk, 640 mg (47,4% utbytte).

Eksempel 2
6- fD- 2- fenvl- 2-( acetimidoylaminoacetamido) acetamido] penicillansyre
(Z= -C0-CH2-NH-Ct«i-CH3)
En suspensjon av 1,58 g (3,5 mmol) D-a-aminobenzyl-penicillintrietylaminsalt i 35 ml tørr dimetylformamid behandles med 0,5 ml (3,5 mmol) trietylamin og blandingen avkjøles til 0°C i et isbad under kraftig omrøring. Acetimidoylaminoacetylklorid-hydroklorid (595 mg, 3,5 mmol) tilsettes og reaksjonsblandingen om-røres kaldt i 30 minutter. Ytterligere 595 mg (3,5 mmol) av syrekloridet og 0,5 ml (3,5 mmol) trietylamin tilsettes og omrøringen fortsettes i 30 minutter ved 0°C og ved romtemperatur i 30 minutter. Blandingen filtreres og det klare filtratet helles i 400 ml dietyleter. Efter filtrering suspenderes råproduktet i 5 ml metylenklorid som er tilsatt 1 ml trietylamin. Efter omrøring ved romtemperatur i 5 timer filtreres produktet og tørkes i vakuum.
755 mg (48% utbytte).

Ved å anvende fremgangsmåten fra eksempel 2 ble oppnådd fra 1,35 g (3 mmol) D-a-aminobenzylpenicillin-trietylaminsalt, 1,1 g (7 mmol) formimidoylaminoacetylklorid-hydroklorid og 0,9 8 ml (7 mmol) trietylamin i 15 ml tørr dimetylformamid 913 mg (72% utbytte) av det ønskede produkt, 6-[D-2-fenyl-2-(formimidoylaminoacetamido)acetamidojpenicillan-syre (Z -C0-CH2-NH-^H)

6-[ D- 2- fenyl- 2-( propionimidoylaminoacetamido) acetamido] penicillari-syre (Z. = -CO-CH2-NH-C{Jty-C2H5
En blanding av 4,5 g (10 mmol) D-a-aminbbenzyl-penicillin-trietylaminsalt og 1 g (10 mmol) trietylamin i 80 ml dimetylformamid avkjølt til -30°C, behandles med 1,0 g (5,2 mmol) propionimidoyl-aminoacetylklorid-hydroklorid og
blandingen omrøres ved -20 til -30°C i 20 minutter, da ytterligere 1 g syreklorid tilsettes og omrøringen fortsettes i 20 minutter;
Ytterligere 1 g trietylamin og 1 g syreklorid tilsettes og fremgangsmåten gjentas igjen efter 20 minutters omrøring i kulde slik at ialt 4,0 g syreklorid bg 3 g amin er tilsatt. Reaksjonsblandingen oppvarmes til 20°G, filtreres og det gule filtratet tilsettes til 1 1 kloroform. Efter vasking med kloroform og tørring (i vakuum) oppløses råproduktet i 20 ml dimetylformamid som ér tilsatt 1 ml trietylamin. Faste stoffer filtreres fra og filtratet tilsettes- til 200 ml kloroform. Produktet filtreres og rensefremgångsmåtén;gjentas^igjen for å
gi 1,6 g av det ønskede produkt. Spormengder av løsningsmiddel fjernes ved utgnidning av produktet med dietyleter og efter-følgende tørring i vakuum, 1,5 g (33% utbytte).

Ved å anvende fremgangsmåten fra eksempél 4 utgående fra D-a-aminobenzylpenicillin-trietylaminsalt og de passende syre-klorider, syntetiseres følgende forbindelsen

En oppslemming av 1,7 g (3,8 mmol) 6-[D-2-fenyl-2-(3-amidinopropionamido)acetamido]penicillans<y>re i-20 ml vann behandles med "0/5'ml (3,8 mmol) triétylamin. Efter omrøring i 5 minutter frafiltreres en liten mengde faste stoffer og filtratet frysetørkes.
På lignende måte fremstilles ammoniumsaltene og de farmasøytisk akseptable saltene oppnådd fra organiske aminer for det ovenfor beskrevne penicillin og de andre penicilliner fremstilt ifølge oppfinnelsen.
Eksempel 7 6-[ D- 2- Fenyl- 2-( 4- pyridoimidoylaminoacetamido) acetamido] penicillansyre
A. 4- Pyridoimidoylaminoacetyiklorid- dihydroklorid
En blanding av 21,4 g glycin og 97 g metyl-4-pyri-dylimidat i loo. ml amylalkohol oppvarmes ved tilbakeløpstempe-råtur i 3 timer. De faste stoffer frafiltreres mens reaksjonsblandingen er varm og vaskes deretter med amylalkohol (2 x loo ml) og dietyleter (4 x loo ml). Tørring natten over under nitrogen gir 51,4 g av det ønskede materiale, 4-pyridoimidoylaminoeddiksyre, sm.p. 223 - 226°C.
lO g 4-pyridoimidoylaminoeddiksyre i 5o ml dietyleter behandles med tilstrekkelig hydrogenkloridgass til å gi då-hydrokloridsaltet, 13,8 g, smeltepunkt 145 15<b°C .
Til 35o ml metylenklorid settes lo,l g 4-pyridoimidoylaminoeddiksyre-dihydroklorid, 9,2 g fosfor-pentaklorid og 3,5 ml dimetylformamid, og den resulterende reaksjonsblariding omrøres ved romtemperatur natten over.. Det gule, faste stoff frafiltreres, vaskes i rekkefølge med kloroform (1 x 3oo ml)
og hexan (1 x 3oo ml). Det ønskede 4-pyridoimidoylaminoace-tylklorid-dihydroklorid tørres under nitrogen, sm.p. 15o°C (spaltning) .
B . 6-[ D- 2- Fenyl- 2- ( 4- pyridoimidoylåminoacetamido) acetåfttido] penicillansyre
Til 4,o3 g D-a-aminobenzylpenicillin-trihydrat i 6o ml dimetylformamid og 2,72 ml trietylamin i en 150 ml kolbe utstyrt mod magnetisk rører, tørrerør og termometer for lave temperaturer,
og avkjølt til -3o°C,settas 1„ 33 g 4-pyridoimidoylamindaaetyl-klorid-dihydroklorid. Etter 3o. min. omrøring ved -3o - -4o°
C tilsettes ytterligere 1,36 ml trietylamin, fulgt etter 5 min.
av ytterligere 1,35 g av syrekloridet. Omrøring fortsettes i 3o min. ved -3o - -4o°C, og deretter tilsettes 5,44 ml trietylamin, fulgt etter 5 min. av 2,7 g av syrekloridet. Etter 45 min. omrøring ved den lave temp. tilsettes 4,o8 ml trietylamin fulgt etter lo min. av 1,3 g av syrekloridet. Omrøring fortsettes ved den lave temp., i 3o min., og deretter får reaksjonsblandingen oppvarmes i løpet av 45 min. til romtemp.
De uoppløselige bestanddeler frafiltreres og vaskes med 2o ml dimetylformamid. De faste bestanddeler.kastes, og vaskevæskene kombineres med filtratet og settes til 1 1 kloroform.
De faste stoffer som utfelles (9oo ml) filtreres og kastes, og filtratet fortynnes med 2 1 hexan. Det resulterende bunn-
fall, som består av en blanding av D-a-aminobenzylpenicillin og det ønskede produkt, filtreres og. tørres, 2,87 g. Råproduktet, settes til 8o ml metylenklorid, og til denne suspensjon settes under omrøring 1 ml trientylamin. Suspensjonen omrøres i 3o min. og filtreres deretter, 1,31 g. Rent produkt oppnås ved krystalisasjon ved romtemp. fra metanol og fra vann, 325
mg.
