Analogifremgangsmåte til fremstilling av terapeutisk virksomme fenoksyalicykliske karboksylsyrer.
Description
Foreliggende oppfinnelse angår fremstilling av nye, anti-aterosklerosevirkende fenoksyalicykliske karboksylsyrer som kan brukes for å senke forhøyede nivåer av kolesterol eller lipider. Disse nye forbindelser har den generelle formel: hvor n og m hver er 4 eller 5, og A er hydrogen eller en gruppe med formelen:
hvor m har den ovenfor angitte betydning, samt salter derav med baser.
Aterosklerose er en sykdom som rammer voksne pasienter,
og det kjenner ingen tilfredsstillende helbredelsesmetode. Til tross for mange undersøkelser er årsaken til aterosklerose ikke kjent. Det er imidlertid vanlig anerkjent at en av de mest betydelige histopatologiske manifestasjoner på aterosklerose er en avsetning av lipider i blodet. Undersøkelser har derfor rettet seg mot en forstyrret metabolisme av lipider, og særlig har man konsentrert seg om et ekstraordinært høyt nivå av kolesterol i
blodet.
En rekke eksperimenter og kliniske fakta har vært angitt som klart indikerende at det er et forhold mellom aterosklerose og et forhøyet nivå av kolesterol eller lipid i blodet. Man har derfor sett det som særlig viktig å utvikle midler for å kunne senke et for høyt nivå av kolesterol eller lipid i blodet, for derved å kunne hindre aterosklerose.
Det har i tidens løp vært utviklet en rekke midler for senking av kolesterol eller lipidinnholdet i blodet, og en rekke forbindelser har vært prøvet klinisk, men ingen av dem har vist seg fullt ut å være tilfredsstillende. Noen av disse forbindelser er relativt effektive, men gir til dels meget skadelige bivirkninger, mens andre har liten effektivitet, slik at de krever å bli tilført i større doser.
En gruppe forbindelser som praktisk anvendes for det oven-nevnte formål inkluderer etyl-a-(p-klorfenoksy)isobutyrat. -Imidlertid er nevnte forbindelses effektivitet ikke særlig høy, og den har en tendens til å gi skadelige bivirkninger, såsom hyper-trofi i leveren. De"'nye forbindelsene med formel I er effektive som kolesterolsenkende midler, og er i alt vesentlig ikke-toksiske.

Når man samtidig får fremstilt en fenoksyalicyklisk monokarboksylsyre (Ia) (dvs. A = H), og en fenoksyalicyklisk di-
karboksylsyre (Ia) (dvs.
så kan disse skilles på
vanlig kjent måte, f.eks. ved omkrystallisasjon eller kromato-grafi.
Reaksjonen kan utføres i nærvær av et overskudd av kloroform og ketoforbindelsen (III) i nærvær eller fravær av et inert reaksjonsmedium. Eksempler på et reaksjonsmedium i denne forbindelse innbefatter dioksan, benzen, toluen etc.
Bisfenolderivater med formel (II) som anvendes som utgangs-forbindelse, kan fremstilles ved den fremgangsmåte som f.eks. er beskrevet i J.A.C.S., 61, 345 (1939). I dette tilfelle representerer bisfenolderivater med formel (II) forbindelser som 1,1-bis(4 *-hydroksyfenyl)-cyklopentan og 1,1-bis(4'-hydroksy-fenyl)cykloheksan. Ketoforbindelser med formel (III) kan angis som cyklopentanon og cykloheksanon.

1 mol av et bisfenolderivat (II) oppløses eller suspen-deres i et inert reaksjonsmedium og bringes i kontakt med minst 1 mol av et alkalisk middel for dannelse av et alkalisalt, hvoretter minst 1 mol av en a-halogenert alicyklisk syre (IV) tilsettes den resulterende reaksjonsblanding, for derved å få utført reaksjonskondensasjonen. Når reaksjonen er fullstendig, kan reaksjonsblandingen ytterligere underkastes vanlige rensemetoder, hvorved man får fremstilt den ønskede fenoksyalicykliske karboksylsyre (I)• Eksempler på inerte reaksjonsmedia som kan brukes i denne fremgangsmåte er benzen, toluen, metanol, etanol, eter, dioksan, dimetylsulfoksyd, N,N-dimetylformamid etc. Eksempler på alkaliske midler som kan brukes er kaliumhydroksyd, natriumhydroksyd, alkalimetallalkoholater, alkalimetallkarbonater, metallisk natrium, natriumhydrid og organiske tertiære aminer, slik som trimetylamin, trietylamin og pyridin. Reaksjonen krever en temperatur på 20-120°C. For å kunne oppnå som hovedproduktet enten en fenoksyalicyklisk monokarboksylsyre (I) (dvs. A = H) eller en fenoksy
alicyklisk dikarboksylsyre (I)
må reaksjonsbetingelsene, slik som mengdeforholdet mellom reaktantene, reaksjonstemperaturen og reaksjonstiden, reguleres nøyaktig. Hvis det alkaliske middel og/eller nevnte a-halogen-alicykliske syre (IV) brukes i ekvimolar mengde med hensyn til bisfenolderivatet (II), så vil man i alt vesentlig oppnå en fenoksyalicyklisk monokarboksylsyre (I) (dvs. A = H). Hvis man på den annen side bruker to eller flere mol både av det alkaliske middel og nevnte a-halogen-alicykliske syre (IV), pr. mol av det angitte bisfenolderivatet (II), så vil man i alt vesentlig oppnå en fenoksyali
cyklisk dikarboksylsyre (I) For oppnåelse av det ønskede produkt, dvs. enten en fenoksyalicyklisk monokarboksylsyre (I) (dvs. A = H) eller en
fenoksyalicyklisk dikarboksylsyre (I)
må reaksjonsbetingelsene, slik som mengdeforholdet mellom reak-
tantene, reaksjonstemperaturen og reaksjonstiden, reguleres nøyaktig.
I foreliggende oppfinnelse kan den fenoksyalicykliske karboksylsyre omdannes til et salt ved en behandling med et alkali. Saltet dannes med den angitte karboksyl- og/eller fenol-iske hydroksylgruppe. Et alkalimetallsalt kan oppnås ved å
bringe den angitte fenoksyalicykliske karboksylsyre (I) i kontakt med natriumhydroksyd, kaliumhydroksyd, natriumkarbonat, kalium-karbonat, natriumbikarbonat eller ammoniakk, etc, eller med et alkoholat av et alkalimetall, slik som natriummetylat i et organisk oppløsningsmiddel, fortrinnsvis en lavere alkanol, som metanol eller etanol, eller med hydroksyd, karbonat eller bikarbonat av et alkalimetall i et organisk oppløsningsmiddel, fortrinnsvis i aceton eller metanol, hvis nødvendig, i nærvær av en mindre mengde vann. Det således fremstilte alkalimetallsalt kan omdannes til et alkalijordmetallsalt ved en behandling med et salt av et alkali-jordmetall, slik som kalsiumklorid.
De fenoksyalicykliske karboksylsyrer eller deres salter
(I) kan brukes i farmasøytiske preparater med eller uten til-blanding av minst ett farmasøytisk bærestoff, f.eks. laktose, potetstivelse, maisstivelse, cellulosederivater, gelatin, mais-olje, bommullsfrøolje etc, i form av kapsler, tabletter, pulvere eller lignende.
De kolesterolsenkende midler kan f.eks. tilføres oralt. Vanligvis vil en oral mengde være fra 0,01-10 g pr. døgn pr. voksen pasient, fortrinnsvis fra 0,05-3 g pr. døgn pr. voksen pasient.
Den kolesterolsenkende aktivitet for forbindelsene ble prøvet på følgende måte: Hannmus som veide fra 15-18 g, fra Nihon Animals Co., ble holdt på en normal kommersiell diett. De ble så oppdelt i eksperimentelle grupper hvor det var 6 dyr eller flere i hver gruppe, og de ble intravenøst injisert med 500 mg pr. kg kroppsvekt av "Triton WR 1339" (oksyetylert tert-oktylfenolformalde-hydpolymer). Prøveforbindelsene ble tilført oralt i en dose på
50 mg pr. kg kroppsvekt umiddelbart etter "Triton"-injeksjonen.
En gruppe mus ble injisert "Triton" og fikk så bare fortynnings-midlet (uten prøveforbindelsen) og tjente som en kontroll på "Triton"-injeksjonen, mens en annen gruppe fikk ingen behandling og tjente som en normalkontroll. 24 timer etter "Triton"-injek-sjonen ble musene obdusert for analyse av serumkolesterol.
Den kolesterolsenkende aktivitet kan uttrykkes på følgende måte:
hvor parentesene betyr serumkolesterolnivået. Således betyr f.eks. [Tritoninjisert kontroll] en midlere verdi for serumkolesterolnivået (mg/dl) i den kontrollgruppe som bare ble injisert med "Triton".
Et eksempel på de resultater som ble oppnådd i denne prøve er vist i tabell 1. I tabell 1 er forbindelsene betegnet ved hjelp av nummerne fra eksemplene.
For å illustrere den fordelaktige terapeutiske virkning til forbindelsene som fremstilles ifølge foreliggende oppfinnelse, ble det foretatt sammenligningsforsøk med forbindelser fra norsk patent nr. 127.298 med hensyn til kolesterolsenkende aktivitet. Forsøkene ble utført under anvendelse av den ovenfor beskrevne metode, og følgende resultater ble oppnådd:
Forbindelse (a) er en monokarboksylsyre med den høyeste aktivitet og forbindelsen (b) er en dikarboksylsyre med den høyeste aktivitet, blant de forbindelser som omfattes av norsk patent nr. 127.298.
Forbindelsene nr. 4 og 5 som fremstilles ifølge foreliggende oppfinnelse og som er monokarboksylsyrer, har en kolesterolsenkende aktivitet som er bedre enn den samme aktivitet til alle forbindelsene i det norske patent (kfr. tabell 1 ovenfor). Også forbindelsene 1-3 (dikarboksylsyrer) og 8 (natriumsalt av en dikarboksylsyre), som fremstilles ifølge foreliggende oppfinnelse, har bedre kolesterolsenkende aktivitet enn dikarboksylsyren (b) i norsk patent nr. 127.298.
Selv om forbindelsene 1-3 og 8 har en virkning som ligger under virkningen til monokarboksylsyren (a) i det norske patent, er de førstnevnte forbindelser langt bedre kolesterolsenkende midler i betraktning av deres lavere toksisitet.
Med hensyn til levervekt-økende virkning, hvilket er
en av toksisitetene til kolesterolsenkende midler, ble forbindelse nr. 1, som fremstilles ifølge foreliggende oppfinnelse, sammenlignet med forbindelse (a) fra nevnte norske patent.
Hver forbindelse ble administrert oralt til 10 Wistar-hannrotter med en vekt på 130-160 g som en gruppe i en dose på 30 mg/kg i en uke. Levervekten ble målt og en økning i prosent ble beregnet idet man sammenlignet levervekten etter administra-sjon med den hos kontrolldyr. Følgende resultater ble oppnådd:
Levervektøkningen for forbindelse (a) i det norske patent var omkring 4 0%, mens den for forbindelse nr. 1 i foreliggende oppfinnelse viste en økning på kun 4%. Det er derfor .klart at forbindelsene som fremstilles ifølge foreliggende oppfinnelse er langt bedre kolesterolsenkende midler enn forbindelsene i norsk patent 127.298.
Følgende eksempler illustrerer fremgangsmåten ifølge oppfinnelsen.
Eksempel 1
En blanding av 15 g 1,1-bis (4'-hydroksyfenyl)cykloheksan og 300 g cykloheksanon ble tilsatt 75 g kaliumhydroksyd. Deretter ble 40 g kloroform tilsatt blandingen under omrøring ved 20-30°C, hvoretter blandingen ble holdt på en temperatur i området 60-70°C i 3 timer for å få en fullstendig reaksjon. Deretter ble reaksjonsblandingen konsentrert til en rest. Etter at resten var oppløst i vann, ble oppløsningen behandlet med aktiv trekull og surgjort med fortynnet saltsyre, hvorved man fikk et oljeaktig stoff. Dette oljeaktige stoff ble ekstrahert med eter, og eteroppløsningen ble igjen ekstrahert med en vandig fortynnet Na2CO^-oppløsning. Det utskilte alkaliske vandige lag ble surgjort og ekstrahert med eter. Eterlaget ble tørket over vannfritt natriumsulfat og konsentrert til et råprodukt, og dette ble renset ved omkrystallisasjon fra toluen. Man oppnådde 12 g av den forønskede fenoksyalicykliske dikarboksylsyre, smeltepunkt 174-175°C.

Ved en fremgangsmåte som tilsvarer den som er beskrevet
i eksempel 1, ble de følgende forbindelser fremstilt slik det er angitt i tabell 2.
Eksempel 6
Til en blanding av 2 g 1,1-bis(4<1->hydroksyfenyl)cyklo-heksan og 20 ml tørr toluen, ble det tilsatt 1,2 g natrium-metoksyd. Deretter ble 3,1 g 1-bromcyklopentylkarboksylsyre oppløst i toluen gradvis og dråpevis tilsatt til blandingen under omrøring. Blandingen ble oppvarmet ved 60-80°C i 6 timer. Reaksjonsblandingen ble svakt surgjort med vandig HC1 og deretter vasket med vann. Deretter ble toluen avdestillert og resten ble renset ved omkrystallisering fra toluen. Den ønskede fenoksyalicykliske dikarboksylsyre ble oppnådd med et smeltepunkt på 188-190°C.

Eksempel 7
Til en blanding av 3 g 1,1-bis(4'-hydroksyfenyl)cyklo-heksan og 30 ml tørr toluen, ble det tilsatt en toluensuspen-sjon av natriumhydrid under avkjøling. Etter omrøring av blandingen i 1/2 time, ble det dråpevis tilsatt en blanding av 4,5 g 1-bromcykloheksylkarboksylsyre og 10 ml tørr toluen,
og blandingen ble oppvarmet under omrøring i 6 timer. Etter av-kjøling ble reaksjonsblandingen svakt surgjort med vandig HC1 og deretter vasket med vann. Toluen ble så avdestillert. Resten ble renset ved omkrystallisering, hvilket ga den ønskede syre, 3,1 g, smp. 172-173°C.

Eksempel 8
Den dikarboksylsyre som ble fremstilt i eksempel 1 ble behandlet med en 10% NaOH vandig oppløsning under svak oppvarm-ing, hvorved man fikk fargeløse plater som var svakt oppløselige i vann, semltepunkt over 260°C.

- Analogifremgangsmåte til fremstilling av terapeutisk virksomme fenoksyalicykliske karboksylsyrer med den generelle formel:hvor n og m hver er 4 eller 5, og A er hydrogen eller en gruppe med formelen: hvor m har den ovenfor angitte betydning, samt salter derav med baser, karakterisert ved at man (a) omsetter et mol av et bisfenolderivat med formelen: hvor n har den ovenfor angitte betydning, med minst et mol av et keton med formelen: hvor m har den ovenfor angitte betydning, og minst et mol kloroform i nærvær av minst 3 mol alkalimetallhydroksyd ved 15-150°C i 3-4 0 timer for dannelse av en fenoksyalicyklisk karboksylsyre med formelen: hvor n, m og A har den ovenfor angitte betydning, eller (b) omsetter et mol av et bisfenolderivat med formelen: hvor n har den ovenfor angitte betydning, oppløst eller sus-pendert i et inert reaksjonsmedium, med minst et mol av et alkalisk middel, og deretter omsetting av det resulterende salt ved 20-120°C med minst et mol av en a-halogenert alicyklisk karboksylsyre med formelen: hvor X er halogen og m har den ovenfor angitte betydning, og, om ønsket, omdannelse av den således oppnådde fenoksyalicykliske karboksylsyre til salter derav med baser.
