Description
Foreliggende oppfinnelse angår en fremgangsmåte ved fremstilling av visse forbindelser som er viktige utgangsmaterialer ved fremstilling av naturlige prostaglandiner og analoger derav.
Nærmere bestemt angår oppfinnelsen en fremgangsmåte ved fremstilling av 2-(6'-carbalkoxyhexyl)-4(R)-hydroxy-cyclopentan-1,3-dioner.
Prostaglandinene som er cykliske, oxygenerte C__ fettsyrer basert på prostansyreskjelettet, og analoger derav er blitt viet stor oppmerksomhet på grunn av de vidt utstrakte fysiologiske res-ponser de utviser i det cardiovaskulære system, respirasjonssystemet, det endokrine system, det hemostatiske system, nervesystemet, for-plantningssystemet, nyre- og mage-tarm-systernet i dyr, innbefattet mennesket.
Tilførselen av naturlige prostaglandiner er for tiden meget begrenset, og fremgangsmåter for fremstilling av disse innbefatter lange og trivielle trinn som bare gir små mengder av forbindelsene. En ytterligere ulempe ved de hittil kjente metoder ved fremstilling av prostaglandiner og analoger derav er de hyppige oppsplittings-trinn som må utføres for å oppnå de ønskede'optisk aktive forbindelser.
Et mål ved foreliggende oppfinnelse er å tilveiebringe en fremgangsmåte ved fremstilling av viktige utgangsmaterialer for syntesen av prostaglandiner og analoger derav, hvilke utgangsmaterialer er karakterisert ved den ønskede optiske aktivitet.
Foreliggende oppfinnelse angår således en fremgangsmåte ved fremstilling av optisk aktive 2-(6'-carbalkoxyhexyl)-4(R)-hydroxy-cyclopentan-1 , 3-dioner av generell formel;
hvori R er en lavere alkylgruppe med 1-3 carbonatomer, og frem-gangsmåten er særpreget ved at et 2-(6'-carbalkoxyhexyl)-cyclopentan-1,3,4-trion eller et 2-(6<1->carbalkoxyhexyl)-3-methoxy-cyclopent-2-en-l,4-dion underkastes fermentativ virkning av Dipodascus uninucleatus, hvorefter den ønskede forbindelse utvinnes fra reaksjonsblandingen.
Den anvendte mikroorganisme Dipodascus uninucleatus er deponert i American Type Culture Collection, 12031 Parklawn Avenue, Rockwille, MD 20852, USA og har fått deponeringsnummer 14626. Mikroorganismen er beskrevet i "The American Type Culture Collection", Catalogue of Strains, 12. utgave, side 209.
US patentskrift nr. 3432393 (Beilet) beskriver en mikrobiologisk fremgangsmåte ved fremstilling av optisk aktive l&-hydroxy-2-(substituert)-cycloalkan-3-oner av formelen
hvori n er 1 eller 2, og R' er en alifatisk kjede inneholdende 4-18 carbonatomer eller et radikal av strukturen
Både den alifatiske kjede og radikalet betegnet ved R' kan bære funksjonelle grupper slik som hydrogen, carbonyl, carboxyl, carbalkoxyi, hydroxyl, alkoxy, alkyl, aryl, aralkyl eller cyclo-alkyl- og begge kan ennvidere være mettet eller umettet.
Det er viktig å merke seg at i formel I er hydroxylgruppen ved orientert i 3-konfigurasjon, mens methyl og R'-gruppene ved C ? hver kan være i enten a- eller 3-konfigurasjon.
For å forenkle sammenligningen med foreliggende fremgangsmåte-produkter kan den ovenfor angitte formel I også angis som
Hvis carbonatomet som bærer oxo-funksjonen angis som det
første carbonatom i ringen, er det klart at hydroxylgruppen da er på det tredje carbonatom i ringen. Således er betegnelsen l(3-hydroxy-2-(substituert)-cyclopentan-3-on ekvivalent med 2-substituert-3(3-hydroxy-cyclopentanon. Spesifikke eksempler på forbindelser av formel I som finnes i Beilet er: l(3-hydroxy-23-methyl-2a-(3'-oxo-6'-carbomethoxyhexylhcyclopentan-3-on [eller ekvivalent: 2a-(3'-oxo-6'-carbomethoxyhexyl)-20-methyl-33-hydroxy-cyclopentanon, eksempel 1 i Beilet],
13-hydroxy-2a-methyl-23-(3'-oxo-6<1->carbomethoxyhexyl)-cyclopentan-3-on teller 2a-methyl-2&-(3'-oxo-6<1->carbomethoxyhexyl)-33-hydroxy-cyclopentanon, eksempel 2 i Beilet], l&-hydroxy-2&-methyl-2a-(3' -oxo-butyl)-cyclopentan-3-on [eller 2a-(3'-oxo-butyl)-2|3-methyl-3&-hydroxy-cyclopentanon, eksempel 5 i Beilet] 1- hydroxy-2-methyl-2- (3 ' -klor-2 ' -butenyl>-cyclopentan-3-on [eller 2- methyl-2-(3 '-klor-2 1-butenyl>-cyclopentan-3-on; eksempel 6 i Beilet]
I de foregående formler betegner en stiplet linje en covalent binding rettet under referansecarbonatomets plan, dvs. a-binding, mens den kraftig opptrukne linje betegner en covalent binding over carbonatornets plan, dvs. B-binding. Alle eksempler i Beilet unntatt forbindelse V er forbindelser hvori hydroxylgruppen er B-orientert. I forbindelse V er bindingsretningen uspesifisert.
I forbindelse II og IV er methylgruppen i B-konfigurasjon, mens R' er er i a-konfigurasjon. I forbindelse III er det motsatte tilfelle.
Forbindelsene I - V er nyttige ved fremstilling av 19-nor-
steroider.
Fremstilling av forbindelsene av formel I skjer ved mikrobiologisk reduksjon av visse 2-substituerte 1,3-dioner av strukturen: hvori n og R' har de tidligere angitte betydninger. For sammen-lignings skyld kan VI representeres ved sin ekvivalente struktur:
x betegner den carbonylfunksjon som gjennomgår reduksjon. Reduk-sjonen av VI til I forløper ved anvendelse av: gjær, bakterier og klassene Fungi imperfecti og Actinomycetales, særlig Rhizopus, Saccharomyces og Pseudomonas, og spesielt Rhizopus nigricans, Rhizopus arrhizus, Micor spinosus, Curvularia falcata, Gongronella urceolifera, Aspergilus niger og Streptomyces platensis.
DQS 2038926 (Wang) beskriver en forbedring i Bellet-prosessen. Denne forbedring omfatter en sterospesifikk reduksjon av VI til I, hvori methylgruppen i VI er B-orientert, og R'-gruppen er a-orientert. Wang oppdaget at den sterospesifikke reduksjon erholdes ved anvendelse av mikroorganismer av slekten Schizosaccharomyces: S. japonicus, S. octosporus, S. pombe og S. versatilis. Wang beskriver fremstilling av følgende spesifikke forbindelser.
13-hydroxy-2a-(2'-carbomethoxyethyl)-2B-methyl-3-oxo-cyclopentan, lB-hydroxy-2a-(3'-oxo-6'-carbomethoxyhexyl)-2B-methyl-3-oxo-cyclo-pentan,
lB-hydroxy-2<x- (3 ' -oxo-6 ' -carbomethoxyhexyl) -2B-ethyl-3-oxo-cyclo-pentan,
13-hydroxy-2a-(3<1->oxo-butyl)-2B-methyl-3-oxo-cyclopentan, 13-hydroxy-2a-(3'-oxo-butyl)-23-ethyl-3-oxo-cyclopentan, 13~hydroxy-2a-(3<1->oxo-pentyl)-23-methyl-3-oxo-cyclopentan, 13-hydroxy-2a-(3',7'-dioxo-octanyl)-23-methyl-3-oxo-cyclopentan, og 13-hydroxy-2a-(3',7'-dioxo-nonanyl)-23-methyl-3-oxo-cyclopentan.
I alle forbindelser er hydroxylgruppen 3-orientert.
Foreliggende oppfinnelse angår en mikrobiologisk fremgangsmåte ved fremstilling av 2-(6<1->carbalkoxyhexyl)-4a-hydroxy-cyclo-pentan-1,3-dioner av formelen:
hvori R er en lavere alkylgruppe med 1-3 carbonatomer. Det er vesentlig å merke seg at C hydroxylgruppen er i a-konfigurasjon, og at C2 substituenten er i cyclopentanringens plan. Fremstillingen av VII krever som substrater enten et 2-(6'-carbalkoxy)-cyclopentan-1,3,4-trion av generell formel eller et 2-(6'-carbalkoxy)-4-(alkoxy)-cyclopent-2-en-l,4-dion av generell formel
hvor R har den tidligere angitte betydning.
Omdannelsen av VIII eller IX til VII skjer under anvendelse av mikroorganismen Dipodascus uninucleatus.
Den etterfølgende tabell viser de vesentlige forskjeller mellom "Bellet-patentet", "Wang-patente.t" og foreliggende oppfinnelse:
Det er ennvidere kjent at ketofunksjonen i 4-stilling i de ovenfor angitte forbindelser kan reduseres ved hydrogenering over en palladium-carbonkatalysator (se Chemical Abstracts, 7_5 (1971) 4849 m). Imidlertid er den resulterende hydroxyforbindelse racem-isk. Ved anvendelse av den fermentative virkning av Dipodascus uninucleatus kan hydrogeneringen av oxo-funksjonen ved 4-stillingen utføres stereospesifikt under dannelse av 4(R) epimer konfigurasjon (under anvendelse av Cahn-Ingold-Prelog tegnsystemet). Denne konfigurasjon er nødvendig for omdannelse til de naturlige prostaglandiner. Et eksempel på slik omdannelse hvor det erholdte prostaglandin er PGE1 er vist skjematisk nedenfor.
I det ovenfor angitte reaksjonsskjema utføres omdannelsen fra VHI til VU i overensstemmelse med den foreliggende fremgangsmåte. VII kan deretter omdannes til X som følger: Til en avkjølt (-11° C) omrørt løsning på 102,4 mg (0,4 mmol) hydroxydionmethylester (VII) og 0,112 ml (ca. 0,84 mmol) triethylamin (destillert over kalsiumhydroxyd) i 8 ml tørr tetrahydrofuran (under N2)ble tilsatt 0,046 ml (0,4 mmol) benzoylklorid i løpet av 5 min. Den resulterende løsning ble omrørt ved —10° C i 35 min. 4 ml methanol ble tilsatt og løsningen fikk oppvarmes til romtemperatur og deretter heldt over i 60 ml vann. Denne løsning
ble ekstrahert fire ganger med ethylacetat (50 ml pr. ekstraksjon)
og det kombinerte ekstrakt ble vasket henholdsvis med 10 ml 0,1N HC1, 10 ml vann, 10 ml mettet natriumbicarbonat og 10 ml mettet natriumklorid, og tørket over magnesiumsulfat. X ble utvunnet ved fordampning av løsningsmidlet.
Omdannelsen fra X til XI ble utført som følger:
Til en avkjølt (-78° C) omrørt løsning av 50 mg (0,147 mol) X i 10 ml tørr tetrahydrofuran under N2~atmosfære ble natriumdihydro-bis-(2-methoxyethoxy)-aluminat (Na(A1H2(OCH2CH2OCH3)2)) 1 toluen (1,5M) tilsatt i fire 9,38 ml's alikvoter i løpet av 15 min. Den resulterende løsning ble omrørt ved -78° C i ytterligere 30 min. 2 ml iseddik ble deretter tilsatt, løsningen fikk oppvarmes til romtemperatur og ble heldt over i 40 ml vann. Den resulterende blan-ding ble ekstrahert med fire 30 ml's porsjoner ethylacetat. Ethyl-acetatlaget ble fraskilt, vasket henholdsvis med 10 ml mettet natriumbicarbonatløsning og 10 ml mettet natriumkloridløsning og tørket over MgSO^. Løsningsmidlet (ethylacetat) ble deretter fordampet og den resulterende olje ble løst i 10 ml eddiksyre-vann (75:25). Den resulterende løsning ble omrørt ved romtemperatur i 24 timer og eddiksyre-vannet ble fordampet under redusert trykk.
Den resulterende olje ble løst i 10 ml ethylacetat, vasket henholdsvis med mettet natriumbicarbonat og mettet natriumkloridløsninger, og tørket over magnesiumsulfat. XI ble utvunnet ved fordampning av løsningsmidlet.
Omdannelsen fra XI til XII kan utføres som beskrevet av Charles J. Sih et al., (J. Amer. Chem. Soc., 94: 3643 (1972)). Når følgelig det stereospesifike 2(6'-carbomethoxyhexyl)-4(R)-hydroxy-cyclopentan-1 , 3-dion er erholdt, kan omdannelsen til prostaglandin lett utføres under bibeholdelse av den ønskede stereospesifike konfigurasjon.
I de etterfølgende eksempler ble stereoisomerien av 4-hydroxylfunksjonen fastslått ved sirkulær dikroismeanalyse. R-epimeren utviste en negativ "cotton" effekt, ((e) x 10 3 -85°) ved X 281 my og en positiv "cotton" effekt '(•{©) x 10~<3> +87°) ved
X 262 my, hvilket er karakteristisk for denne type av system. Like-ledes ble stereoisomeri en korrigert med stereoisomeri en til prostaglandin E-, (PGE-j^) ved å omdanne 2 (6 '-carbomethoxyhexyl)-4 (R)-hydroxy-cyclopentan-1, 3-dion til PGE^ i samsvar med den ovenfor beskrevne prosedyre.
Produktet fremstillet ifølge de efterfølgende eksempler ble identifisert med hensyn til struktur ved bruk av ultrafiolett, infrarødt, og kjernemagnetisk resonansspektrum, og ved tynnskikts-kromatografi.
Eksempel 1
2 (6'-carbomethoxyhexyl)-4(R)-hydroxy-cyclopentan-1,3-dion (VII) ble fremstillet som følger:
A. Fermentering
Overflateveksten fra en 1 ukes gammel skråstivnet agar av Dipodascus uninucleatus ble suspendert i 5 ml saltvann (0,85 %) .
2 ml's porsjoner av denne suspensjon ble anvendt for å innpode 50 m av soyabønne-dextrosemediet i en 250 ml Erlenmeyer-kolbe (F-l trinn Kolbene ble inkubert ved 25° C på en rotasjonsrister (250 omdr./min 5 cm radius) i 24 timer, hvoretter 10 % av volumet ble overført til hver av fire 2 liter Erlenmeyer-kolber (trinn F-2), inneholdende
500 ml soyabønne-dextrosemedium. Etter 24 timers inkubering på en rotasjonsrister, ble 250 mg 2(6'-carbomethoxyhexyl)-cyclopentan-1,3,4-trion (I) (J. Katsube ogM.Matsui, J. Agr. Biol. Chem., 33: 1078, (1969)), løst i 2 ml dimethylformamid tilsatt til hver kolbe. Trinn F-2-kolbene ble deretter inkubert i ytterligere 24 timer unde de betingelser som ble anvendt ved inkubering av trinn F-l-kolbene.
B. Isolering
24 timer efter tilsetning av 2-carbomethoxyhexyl-cyclo-pentan-1,3,4-trion (I) ble cellene fjernet ved sentrifugering. Den overliggende vasske ble justert til pH 2,0 med 6N HC1, og ble der-efter kraftig ekstrahert med 1,5 liter ethylacetat tre ganger. Ethylacetatet ble tørket over Na2S04 og fordampet under dannelse av et oljeaktig residuum. Dette residuum ble oppløst i 0,5 ml benzen-ethylacetat (1:1) og tilsatt til en 32 x 2,5 cm kolonne av silici-umsyre-Celite (85:15). Kolonnen ble eluert med et gradientsystem bestående av 500 ml 50 %-ig ethylacetat-benzen i blandekammeret og 500 ml rent ethylacetat i reservoaret, og 7 ml fraksjoner ble opp-samlet. Fraksjonene 25 - 105 inneholdende det ønskede produkt ble samlet og konsentrert til tørrhet under dannelse av 1,2 g krystal-linsk residuum. Omkrystallisasjon fra ethylacetat-petroleumether ga 750 mg (75% utbytte) av 2-carbomethoxyhexyl-4(R)-hydroxy-cyclopentan-1,3-dion (VII), sm.p. 90 - 92°C, [>]<23> = +19,93° ( C± 1 ,02 CHClg),

Eksempel 2
2-(6'-carbomethoxyhexyl)-4(R)-hydroxy-cyclopentan-1,3-dion ble fremstilt som beskrevet i eksempel 1 med det unntak at 2-(6'-carbomethoxyhexyl)-3-methoxy-2-cyclopenten-l,4-dion ble anvendt som substrat for omdannelsen til det ønskede produkt.

- Fremgangsmåte ved fremstilling av optisk aktive 2-(6'-carbalkoxyhexyl)-4(R)-hydroxy-cyclopentan-1,3-dioner av generell formel:hvori R er en lavere alkylgruppe med 1-3 carbonatomer,karakterisert ved at et 2- (6'-carbalkoxyhexyl)-cyclo-pentan-1,3,4-trion eller et 2-(6<1->carbalkoxyhexyl)-3-methoxy-cyclo-pent-2-en-l,4-dion underkastes den fermentative virkning av Dipo-dascus uninuclea tus, hvorefter den ønskede forbindelse utvinnes fra reaksjonsblandingen.
