Description
Fremgangsmåte for fremstilling av antibiotika av spiramycin-typen.
Nærværende oppfinnelse angår antibiotiske
stoffer og fremgangsmåter for fremstilling av
slike stoffer og vedrører mere spesielt fremskaf-felsen av nye terapeutisk aktive derivater av
antibiotika av spiramycin-typen, slik som det
defineres i det følgende.
Den ukvalifiserte betegnelse «antibiotika av
spiramycin-type», som brukt i den følgende be-skrivelse og etterfølgende påstander, angir en
av de følgende antibiotiske stoffer:
1. Spiramycinkompleks er et antibiotisk stoff som har følgende egenskaper: en molekylarvekt mellom (400)n og (450)n, hvor n er lik 2, en venstredreiende aktivitet i alkoholisk 2 % i etanol), og sammensetningen er tilnær-met som følger:
— og dets ultra-fiolette absorpsjonsspektrum i

alkoholisk oppløsning (E-^ ^ er ca. 346) viser
en topp ved 231 m/i.
2. De individuelle bestanddeler av spiramycinkompleks, betegnet som Spiramycinene I,
II og III, har følgende karakteristikker: Spiramycin I — er et basisk stoff oppløselig i kloroform, klorerte oppløsningsmidler, alkoholer, heksan, benzol, ketoner, etylacetat og amylacetat og har evne til å danne salter med syrer, hvilket basiske stoff inneholder element-
ene kullstoff, vannstoff, surstoff og kvelstoff i det vesentlige i følgende vektsforhold:
hvilket basiske stoff i etylalkoholisk oppløs-ning viser en maksimal absorpsjon ved 232 mu og har en molekylarvekt bestemt ved den ebullioskopiske metode på ca. 800, en nøytral ekvivalent på 463, en dissosiasjonskonstant pK b på 7,7, et smeltepunkt bestemt på Maqu-enneapparat på 134—137° C, har optiske rotasjoner [a] y i metanol (c = 1 på i etanol (c = 1 %) på 91° og i kloroform (c = 1 %) på -1-57°, og som i fast form viser karakteristisk absorpsjon i det infra-røde området av spektret ved følgende frekvenser uttrykt i resiproke centimetere: 3470, 2970,2940, 1735, 1455, 1378, 1317, 1275, 1237, 1160, 1122, 1090, 1052, 1015, 993, 905, 865, 840, 810, 782.
Spiramycin II — er et basisk stoff oppløselig i kloroform, klorerte oppløsningsmidler, alkoholer, heksan, benzol, ketoner, etylacetat og amylacetat og med evne til å danne salter med syrer, hvilket basiske stoff inneholder element-ene kullstoff, vannstoff, surstoff og kvelstoff i det vesentlige i de følgende vektsforhold:
hvilket basiske stoff viser, i etylalkoholoppløs-
ning en maksimal absorpsjon ved 232 mn, har en molekylarvekt bestemt ved den ebullioskopiske metode på ca. 800, en nøytral ekvivalent på 464, en dissosiasjonskonstant pK b på 7,6, et smeltepunkt bestemt på Maquenne-apparat på 130—133° C, har optiske rotasjoner [a] d i metanol (c = 1 %) på -H 86°, i etanol (c = 1 %) på 4- 80°, og i kloroform (c = 1 %) på -=-55°, og som i fast form oppviser karakteristisk absorpsjon i det infra-røde området av spektret ved følgende frekvenser uttrykt i resiproke centimetere: 3460, 2970, 2940, 1740, 1457, 1372, 1300, 1275, 1232, 1160, 1122, 1085, 1052, 1015, 993, 940, 905, 860, 840, 810, 782, 685.
Spiramycin III — er et basisk stoff opp-løslig i kloroform, klorerte oppløsningsmidler, alkoholer, heksan, benzol, ketoner, etylacetat og amylacetat og med evne til å danne salter med syrer, hvilket basiske stoff inneholder ele-mentene kullstoff, vannstoff, surstoff og kvelstoff i det vesentlige i følgende vektsforhold:
hvilket basiske stoff viser, i etylalkoholoppløs-ning en maksimal absorpsjon ved 232 mu, har en molekylarvekt bestemt ved den ebullioskopiske metode på ca. 900, en nøytral ekvivalent på 473, en dissosiasjonskonstant pK b ved 7,6, et smeltepunkt bestemt på Maquenne-apparat på 130—131° C, har optiske rotasjoner [ a] d i metanol (c = 1 %) på — 83°, i etanol (c = 1 %) på 4- 79° og i kloroform (c = 1 %) på -r 50°, og som i fast form viser karakteristisk absorpsjon i det infra-røde området av spektret ved følgende frekvenser uttrykt i resiproke centimetere: 3470, 2970, 2940, 1740, 1460, 1380, 1370, 1300, 1280, 1240, 1185, 1162, 1122, 1085, 1052, 1015, 995, 906, 865, 842, 810, 782, 695, 685. 3. Hydrert spiramycin eller spiramycin-komponent som definert i henholdsvis (1) eller (2) foran eller blanding av spiramycinkompo-nenter, inneholdende inntil fire vannstoffatomer i tillegg til dem som er tilstede i det ikke hydrerte materiale. Individuelle, hydrerte produkter av viktighet er: (a) Tetrahydrospiramycin, som har den empiriske formel C46-48H83-870]5-]7N2, smeltepunkt ved 120-125° C, optisk rotasjon [ a] " = -f- 71,5° (c = 1 %, metanol), ultrafiolett absorpsjonsmaksimum 820 m/f, E * o/*1 = 14 (etanol), x /o og som i fast form viser karakteristisk absorpsjon i det infra-røde området av spektret uttrykt i resiproke centimetere ved følgende frekvenser: 3470, 2930, 2800, 1730, 1628, 1462, 1453, 1410, 1372, 1318, 1289, 1270, 1232, 1182, 1160, 1120, 1075, 1050, 1015, 991, 971, 956, 903, 841, 808, 783. (b) Dihydrospiramycin I, som har den empiriske formel C46-48H 81-85016-17N2, smeltepunkt ved 128 - 132° C, optisk rotasjon[a] " = -r- 83° (c = 1 %, metanol), og som i fast form oppviser karakteristisk absorpsjon i det infra-røde området av spektret ved følgende frekvenser uttrykt i resiproke centimetere: 3470, 2930, 2800, 1720, 1660, 1462, 1453, 1410, 1378, 1320, 1272, 1240, 1184, 1160, 1122, 1075, 1060, 1050, 1015, 993, 965, 903, 868, 841, 808, 783.
(c) Tetrahydrospiramycin I, som har den empiriske formel C4(i-48<H>83-870lc-J7N2, smeltepunkt ved 132—135° C, optisk rotasjon [a] "
= H- 79° (c = 10, metanol), ultrafiolett ab-1 cm sorpsjonsmaksimum 820 m/f, E ^y<1>=14(etanol), og som i fast form oppviser karakteristisk absorpsjon i det infra-røde området av spektret ved følgende frekvenser uttrykt i resiproke centimetere: 3470, 2930, 2800, 1730, 1720, 1650, 1462, 1453, 1410, 1377, 1318, 1272, 1242, 1185, 1160, 1120, 1080, 1052, 1015, 991, 903, 841, 808, 783.
(d) Tetrahydrospiramycin II, som har den empiriske formel C46-48H83-82015-K)<N>2, smeltepunkt ved 125—128° C, optisk rotasjon [a] %'
= 63° (c = 1 %, metanol), ultrafiolett absorpsjonsmaksimum 820 m/(, E^rym = 14
(etanol), og som i fast form oppviser karakteristisk absorpsjon i det infra-røde området av spektret ved følgende frekvenser uttrykt i resiproke centimetere: 3470, 2930, 2800, 1730, 1720, 1462, 1455, 1410, 1370, 1330, 1318, 1289, 1273, 1238, 1182, 1160, 1122, 1080, 1065, 1050, 1015, 991, 965, 903, 865, 841, 808, 783.
(e) Tetrahydrospiramycin III, som har den empiriske formel C40-48H83-87O16-16N2, smeltepunkt ved 135—140° C, optisk rotasjon [ a] "
= -i- 71° (c = 1 %, metanol), ultrafiolett
1 cm absorpsjonsmaksimum 820m/f, Eo/ = 14
(etanol), og som i fast form oppviser karakteristisk absorpsjon i det infra-røde området av spektret ved følgende frekvenser uttrykt i resiproke centimetere: 3470,2930,2800,1740, 1730, 1640, 1462, 1453, 1410, 1375, 1315, 1290, 1270, 1230, 1182, 1160, 1120, 1075, 1050, 1018, 993, 965, 903, 860, 841, 808, 783.
Spiramycinkompleks kan oppnås ved en fremgangsmåte (beskrevet i patent nr. 89771, som består i aerobisk dyrking av Streptomyces ambofaciens NRRL 2420 eller en mutant av denne i et medium inneholdende assimilerbare kilder av kullstoff, kvelstoff og mineralsalter inntil vesentlig antibiotisk aktivitet er fremkalt av nevnte mikro-organisme i nevnte kulturmedium og utvinning av spiramycin fra nevnte kulturmedium. Separeringen av komplekset i dets tre komponenter kan utføres ved bruk av Craig's motstrøms-distribusjonsmetode som beskrevet i patent nr. 89771 eller i patent nr. 92962, eller som beskrevet i sistnevnte patent ved å utsette en oppløsning av spiramycinkompleks for kromatografi på aluminium eller for fraksjonert krystallisasjon fra et aromatisk kullvannstoff-oppløsningsmiddel, fortrinsvis benzol.
Hydrerte spiramyciner kan oppnås, som beskrevet i patent nr. 92386 ved katalytisk hyd-rering av spiramycinkompleks eller komponenter av dette ved bruk av som katalysator, f. eks. et edelt metall som palladium og platina.
Spiramycin-antibiotika har, som kjent ytterst brukbare terapeutiske egenskaper, og de hydrerte spiramyciner har antibakterielle egenskaper langt overlegne dem for ikke-hydrerte produkter.
Det er en hensikt med nærværende oppfinnelse å fremskaffe nye derivater av antibiotika av spiramycin-typen som er i besiddelse av samme art og grad av terapeutisk aktivitet som de lignende stoffer, fra hvilke de er avledet, men også innehar den betydelige fordel å være praktisk talt uten smak.
Forbindelsene etter nærværende oppfinnelse er esterne dannet av antibiotika av spiramycin-typen med mono- eller di-karboksylsyrer av de alifatiske eller mono- eller bi-cykliske, aromatiske serier.
Antibiotikaene av spiramycin-typen er i besiddelse av fire esterifiserbare grupper. En eller flere av disse grupper kan esterifiseres i overensstemmelse med mengdene av brukte reaksjonsdeltakere.
De nye estere oppnås, ifølge nærværende oppfinnelse, ved acylering av et antibiotikum av spiramycin-typen, fortrinsvis ved hjelp av det egnede syrehalogenid eller -anhydrid. Reaksjonen utføres ved romtemperatur, hvilket forenkler fremgangsmåten, eller ved en forhøyet temperatur, hvilket øker farten av reaksjonen betraktelig. Det er imidlertid ingen fordel i å arbeide ved en temperatur over 120° C, da under slike betingelser utgangsantibiotikumet delvis kan spaltes, hvilket fører til tap. Reaksjonen kan utføres enten med eller uten et organisk oppløsningsmiddel og utføres fortrinsvis i nærvær av en basisk substans, som når oppløsningsmiddel er tilstede, kan foreligge i oppløsning eller suspensjon. Enten kan det brukes de teoretiske mengder av reaksjonskom-ponentene eller et svakt overskudd av en av dem. Blant de basiske stoffer , som kan brukes skal det nevnes f. eks. alkalimetallkarbonater og bikarbonater, tertiære aminer og hetero-cykliske, tertiære baser som pyridin eller alkyl-piperidiner.
Oppfinnelsen illustreres av de følgende eksempler, som beskriver fremstillingen og karakteristikkene for individuelle estere av særlig verdi. De angitte smeltepunkter er bestemt ved Koflerapparat.
Eksempel I.
Vannfritt natriumbikarbonat (15 g) og etyl-klorkarbonat (4,5 g) tilsettes til en oppløsning av spiramycinkompleks (9,5 g) i alkoholfri, vandig aceton (60 cm<3>). Etter tre timers om-røring filtreres det faste fra og, etter fordampning av aceton, oppnås det et produkt (11 g) som, etter omkrystallisering fra en blanding av toluol og cyklohcksan, har følgende karakteristikker: sm.p. 160—165° C; [ a]" = ~ 95,5°
(c = 1, metanol), og analysen viser at det er monoetoksyformylspiramycin med den empiriske formel: C51H86018N2.
Eksempel II.
Vannfritt natriumkarbonat (15 g) og ben-zoylklorid (5,6 g) tilsettes til en oppløsning av spiramycinkompleks (9,5 g) i vannfri aceton (60 cm<3>). Etter tre timers omrøring, filtreres det faste fra, og etter fordampning av aceton, blir det tilbake et produkt (12 g), sm.p. 90—
95° C; [a] "
lysen viser at det er tetrabenzoylspiramycin med den empiriske formel: C76H9802oN2.
Eksempel III.
Vannfritt trietylamin (2,2 g) fulgt av stea-roylklorid (6,1 g) tilsettes til en oppløsning av spiramycinkompleks (9,5 g) i toluol (60 cm<3>). Etter henstand i to timer filtreres det faste som har dannet seg fra, og fordampning av toluolet etterlater distearoylspiramycin (15 g), sm.p. 50° C.
Eksempel IV.
Ved å gå frem som i eksempel III, men begynne med trietylamin (1,1 g) og stearoyl-klorid (3 g) oppnås det monostearoylspiramycin (12 g), sm.p. 75—80° C; [ a] <2>J
1, metanol), med den empiriske formel:

Eksempel V.
Spiramycinkompleks (2 g) oppløses i eddik-anhydrid (40 cm<3>) og pyridin (0,1 cm<3>) og las henstå i 3 dager ved romtemperatur. Det helles derpå på is (200 g) og, når hydrolysen er full-
stendig, bringes oppløsningen til pH 7 med natriumhydroksyd (d = 1,33). Det oppnådde bunnfall filtreres fra, vaskes med vann (5 cm<3>)
og tørres i vakuum. Det oppnås således tri-acetylspiramycin (2,0 g), sm.p. 145—147° C,
som kan omkrystalliseres fra petroleumeter;
[a] 2D7 = 4- 99° (c = 1, metanol).
Eksempel VI.
En blanding av tetrahydrospiramycin (0,5 g) eddiksyreanhydrid (10 cm<3>) og pyridin (0,25
cm<3>) las henstå i 24 timer. Den helles derpå på
is (50 g) og bringes til pH 8 med natriumhydroksyd (d = 1,33, ca. 20 cm<3>), og det opp-
nådde hvite faste filtreres fra. Det oppnås så-
ledes triacetyltetrahydrospiramycin (0,5 g), som kan omkrystalliseres fra petroleumeter og som så smelter ved 120—124° C.
Eksempel VII.
Vannfritt trietylamin (0,5 g) fulgt av etyl-klorkarbonat (0,5 g) tilsettes til en oppløsning av tetrahydrospiramycin (3,1 g) i toluol (40
cm<3>). Etter henstand i 24 timer filtreres trietylamin-hydrokloridet fra, og toluolen fordampes.
Det oppnås monoetoksyformyltetrahydrospira-
mycin (3,2 g), som kan omkrystalliseres fra en blanding av like mengder av cykloheksan og petroleumeter og som derpå smelter ved 102
— 106° C [ a] ™ = -4 74° (c = 1, metanol).
Eksempel VIII.
Ved å gå frem som i eksempel VI, men gå
ut fra Spiramycin I (0,5 g) oppnås det triacetyl-spiramycin I (0,46 g), sm.p. 142—143° C.
Eksempel IX.
Ved å gå frem som i eksempel VI, men gå
ut fra Spiramycin II (0,5 g), oppnås det tri-acetylspiramycin II (0,45 g), sm.p. 140—141° C.
Eksempel X.
Ved å gå frem som i eksempel VI, men gå
ut fra Spiramycin III (0,5 g), oppnås det tri-acetylspiramycin III (0,38 g), sm.p. 144° C.
Eksempel XI.
Ved å gå frem som i eksempel VII, men
gå ut fra Spiramycin I (3,1 g), oppnås det monoetoksyformylspiramycin I, (3,2 g), sm.p.
155—160° C; [a] " = 92° (c=l, metanol.)
Eksempel XII.
Ved å gå frem som i eksempel VII, men gå
ut fra spiramycin II (3,1 g), oppnås det monoetoksyformylspiramycin II (3,2 g), sm.p. 145—
150° C; [ a] ™ = —86° (c = 1, metanol).
Eksempel XIII.
Ved å gå frem som i eksempel VII, men gå
ut fra Spiramycin III (3,1 g), oppnås det monoetoksyformylspiramycin III (3,2 g), sm.p. 130
—135° C; [a] " = — 73° (c = 1, metanol).
Eksempel XIV.
En oppløsning av spiramycinkompleks (9,25
g) og ravsyreanhydrid (1,00 g) i etylacetat (100
cm<3>) tillates å stå ved romtemperatur i 24
timer, etter hvilken periode anhydridet har reagert fullstendig. Etter fordampning av opp-løsningsmidlet oppnås monosuccinylspiramycin (10,1 g) i form av et krempulver, som er hurtig oppløselig i vann, sm.p. 150—155° C; [a] 19 =
4-99° (c = 1, metanol).
Eksempel XV.
Ved å gå frem som i eksempel XIV, men
bruke spiramycinkompleks (9,25 g) og ravsyre-
anhydrid (2,00 g), oppnås det disuccinylspira-
mycin (10,9 g), sm.p. 150—155° C; [ a] <19> —
4- 93° (c = 1, metanol).
Eksempel XVI.
Spiramycinkompleks (27,76 g) og malein-syreanhydrid (5,88 g) i benzol (300 cm<3>) tillates å stå ved romtemperatur i 48 timer. Det dannes et krystallinsk bunnfall av dimaleylspiramycin som tillates å tørre. Det oppnås et hvitt pulver (30 g), sm.p. 165—170° C; [ a] ^ = 79° (c =
1, metanol).

- Fremgangsmåte for fremstilling av antibio-tika av spiramycin-typen, som er praktisk talt uten smak, karakterisert ved esterifisering av et spiramycinkompleks, eller spiramycin I,II,III eller blandinger av disse eller av et hydrert spiramycinprodukt ved reaksjon med en mono-eller di-karboksylsyre av den alifatiske eller mono- eller bicykliske aromatiske serie, eller et syrehalogenid eller anhydrid herav, eller etyl-klorkarbonat.
