Description
Fremqanqsmåte til å fremstille lavere alkylestere av 6,7-di(lavere)alkosy-4-hydroksy-3-kinolinkarboksylsyre som er virksomt mot coccidiose.
Oppfinnelsen vedrører fremstilling av lavere alkyl-
estere av 6,7-di(lavere)alkoksy-4-hydroksy-3-kinolinkarboksylsyre som er virksomt mot coccidiose, og med formelen
hvor R og R-^ hver betegner et rett eller forgrenet lavere alkyl-
radikal inneholdende fra 2 til 4 karbonatomer og R ? betegner en lavere alkylgruppe.
Disse nye forbindelser utmerkes av høy grad kjemo-terapeutisk virkning ved behandling av parasittiske infeksjoner ved dyr, hvis de administreres i mengder som ligger like under toxiske doser. De har vist seg a gi overraskende gode resultater ved oral administrering for bekjempelse av coccidiose.
Coccidiose er en generelt forekommende og av økonomisk hensyn betydningsfull sykdom hos fjærfe, som forår-sakes av typer av coccidia omfattende Eimeria tenella, Eimeria acervulina og Eimeria necatrix. De nye forbindelsene ifølge oppfinnelsen er i høy grad effektive midler mot slik coccidiose.
Når de inkorporeres i fra ernæringssynspunkt riktig avpasset f6r i en mengde av størrelsesorden fra omkring 0,006 til omkring 0,1 vektprosent, og kyllinger som er infisert av coccidiose, fores hermed, hindres dødelighet og sykdom som vanligvis etterfølger coccidiose, og dessuten har de en heldig virkning på kyllingenes vekst og utseende.
Prepareringen av disse nye forbindelser i en form
som medfører bekvem administrering, kan lett skje ved innblanding av dem i dyrenes for, som angitt ovenfor. De kan, alt etter ønske, inkorporeres i form av tabletter, gelatinkapsler, suspensjoner eller lignende komposisjoner bestående av hensiktsmessige bærere og innen farmasien vanlige tilsetningsmidler.
Fremgangsmåten ifølge oppfinnelsen erkarakterisertved at man
a ) oppvarmer en Schiffsk base med formelen
hvor R, R-j^ og R2har den innledningsvis angitte betydning, under cykliseringsbetingelser, eller
b) cykliserer et anilinderivat med formelen
hvor R og R-^ har den ovenfor angitte betydning, og R^betegner en formylsubstituert alkoksykarbonylgruppe med formel -CH(CHO)COOR2 eller gruppen R20.C0.CH2- og R^ betegner hydrogen henholdsvis formyl i nærvær av en base.
Det under punkt a) anvendte utgangsmateriale kan frem-stilles ved reduksjon av tilsvarende l,2-dialkoksy-4-nitrobenzol, og omsetning av den dannede aminoforbindelse med en passende di-alkylalkoksymetylenmalonsyreester. Den dannede Schlffske base kan deretter ved hjelp av et varmeoverføringsmedium oppvarmes til den for cyklisering egnede temperatur.
Reduksjonen utføres fortrinnsvis i nærvær av en katalysator som palladium på kull, og et oppløsningsmiddel som etanol. Eksempler på hensiktsmessige malonsyreestere er dietyletoksymetylen- og dimetylmetoksymetylenmalonat og som hensikts-messig varmeoverførings- eller temperaturreguleringsmiddel må nevnes det preparat som går under betegnelsen "Dowtherm A"
(Dowtherm-A er en blanding av difenyl og difenyloksyd).
Fremstillingen ifølge oppfinnelsen av forbindelsene
er beskrevet nedenfor ved hjelp av noen utførelseseksempler. Eksempel 1.
Etyl- 6 t7- dietoksy- 4- hydroksy- 3- kinolinkarboksylat♦
l,2-dietoksy-4-nitrobenzol (30 g), 0,42 mol) inn-føres i en sterkvegget kolbe eller beholder med 365%-ig Pd på kull og 130 ml metanol. I løpet av 1 time absorberes ca. 10 liter hydrogen (teor. ca. 9,5) (38»5$-ig resp. jS , 2%- ±g). Katalysatoren filtreres fra og vaskes med en liten mengde etanol.
I en 1 liters trehalset kolbe innføres den ovenfor nevnte alkoholiske oppløsning og det tilsettes 30 g (o,142 mol) dietyletoksymetylmalonat. Nitrogen bobles gjennom blandingen,
som oppvarmes på dampbad for avdestillering av etanolen. Deretter tilsettes 3°0 ml "Dowtherm A" og temperaturen økes hurtig til 250°C ved hjelp av en oppvarmningsmantel. Blandingen bibeholdes med tilbakeløp i 30-40 minutter, hvoretter oppløsningen får avkjøle". Krystallene som dannes frafiltreres og vaskes med heksan. Derved fåes 18,6 g (utbytte 6l%) av etyl-6,7-dietoksy-4-hydroksy-3-kinolinkarboksylat med smeltepunkt 275 - 280°C.
Eksempel 2.
Etyl- 6 >7- dibutoksy- 4' a- hydroksy- 3- kinolinkarboksy lat.
I en sterkvegget kolbe innføres 29,48 g (0,11 mol) l,2-dibutoksy-4-nitrobenzol, 200 ml etanol og 3 g Pel på kull. Opptakning av den teoretiske mengde hydrogen ved reduksjon av denne blanding i et Parr-apparat oppnås på 1 time (ca. 7»4liter). Reduksjonen gjentas med en like stor mengde av nitroforbindelsen. De reduserte materialer slås sammen og filtreres og katalysatoren vaskes med etanol. Filtratet innføres i en 1-liters trehalset kolbe sammen med 47»5g dietyletoksymetylen-malonat. Blandingen oppvarmes, damp og etanol fjernes ved destillering. 600 ml "Dowtherm A" tilsettes og blandingen oppvarmes hurtig til tilbake-løpstemperatur (250°C). Etter oppvarmning under tilbakeløp i 40 minutter avkjøles blandingen til værelsetemperatur og heksan tilsettes. Den krystallinske utfelling frafiltreres og vaskes med heksan. Derved fåes 29% utbytte av et råprodukt av etyl-6,7-dibutoksy-4-hydroksy-3-kinolinkarboksylat (22,76 g) med smeltepunkt 260 - 285°C.
Dette produkt kan omkrystalliseres fra dimetylformamid, eventuelt under tilsetning av trekull, og gir derved I95& etyl-6,7-dibutoksy-4-hydroksy-3-kinolinkarboksylat med smeltepunkt 279<->284°C.
Eksempel 3»
Etyl- 6 > 7- dipropoksy- 4- hydroksy- 3- kinolinkarboksylat.
I en sterkvegget kolbe innføres 38 g (0,16 mol) l,2-dipropoksy-4-nitrobenzol, 200 ml etanol og 3 g 5_Pr°sentig Pd på kull. Blandingen hydreres i et Parr-apparat med et hydrogen-forbruk på ca. 10,8 liter. Dette gjentas til totalt 172 g (0,72 mol) og nitroforbindelsen er redusert. Katalysatoren frafiltreres og de etanoliske filtrater slås sammen for neste be-handlingstrinn.
Den etanoliske oppløsning blandes med dietyletoksy-metylenmalonat (0,72 mol) og alkoholen fjernes ved destillering på dampbad. Oppvarmningen fortsettes i 3 timer. Deretter til settes "Dowtherm A" (750 )°6blandingen oppvarmes hurtig til tilbakeløp. Oppvarmningen fortsettes med tilbakeløp i 30 minutter og oppløsningen får deretter avkjøle. Den derved dannede krystallinske utfelling frafiltreres og vaskes med heksan. Utbyttet av den i eksemplets titel angitte forbindelse er 96 g (40% utbytte beregnet på nitroforbindelsen). Produktet kan om-krystallisere fra dimetylformamid med smeltepunkt 271-273°C
Eksempel 4»
Etyl- 617- diisopropoksy- 4- hydroksy- 3- kinolinkarboksylat.
A. o- diisopropoksybenzol.
En langsom strøm av hydrogen føres gjennom en opp-løsning av 220 g (2 mol) pyrokatekin i 900 ml alkohol i en 3 liters trehalset kolbe utstyrt med omrører, tilbakeløpskjøler, dryppe-
trakt og nitrogentilføreselsrør. Etter noen minutters omrøring tilsettes l60 g (4 mol) natriumhydroksyd og omrøringen fortsettes til praktisk talt all natriumhydroksyd er oppløst. Til den om-
rørte, tykke suspensjon settes 750 g (6 mol) 2-brompropan i en periode på 2 timer under bibehold av svakt tilbakeløp ved hjelp av et dampbad. Etter tilsetningen avbrytes nitrogentilførselen og oppvarmningen med tilbakeløp fortsettes i 18 timer. Blandingen avkjøles omhyggelig og natriumbromidet frafiltreres og vaskes med alkohol. Det sammenlagte filtrat og vaskevann inndampes i vakuum til så meget alkohol som mulig har gått bort. En ytterligere mengde natriumbromid som avsetter seg under konsentreringen av oppløs-ningen, fjernes. Totalt fåes en mengde på 390 g natriumbromid (959» av den beregnede mengde).
Det oljeaktige residuet oppløses i 500 ml eter og ekstraheres med fire 200 ml porsjoner 10%-ig natriumhydroksydopp-løsning og deretter med to 100 ml porsjoner vann.
Eteroppløsningen tørkes over magnesiumsulfat og konsentreres først ved atmosfæretrykk og deretter i vakuum, idet det fåes en ambrafarget olje som veier 182 g (47% flv det teoretiske for diisopropoksybenzol).
B. 1, 2- diisopropoksy- 4- nitrobenzol.
o-diisoproplsybenzol (140 g, 0,72 mol) tilsettes dråpevis til en omrørt oppløsning av 90 ml kons. salpetersyre i 90 ml vann i en periode på 1 time. Temperaturen bibeholdes ved 20 - 25°C ved intermitterende anvendelse av et isbad. Omrøringen fortsettes til den eksotermiske reaksjon helt har opphørt. Reaksjonsblandingen fortynnes med et like stort volum isvann og det mørkfargede, oljeaktige produkt ekstraheres med tre 25O ml porsjoner kloroform. De sammenslåtte klorformekstrakter vaskes med tre 100 ml porsjoner vann og tørkes over magnesiumsulfat. -Et like stort volum karbontetraklorid tilsettes og oppløsningen helles i en 60 x 600 ml kolonne med aluminiumoksyd (Merck 71707»ca« 1>5kg). Kolonnen elueres med en blanding av like deler kloroform og karbontetraklorid med en strømhastighet på 30 ml/minutt. Etter opp-samling av ca. 3 liter gul oppløsning har eluatet blitt omtrent fargeløst, mens en mørk brun sone blir tilbake ved toppen av kolonnen. Konsentrering av det gule eluat i vakuum gir 100 g (58%) av den i titelen angitte nitroforbindelse som en gul olje.
C. Etyl- 6, 7- diisopropoksy- 4- hydroksy- 3- kinolinkarboksyl at.
En 20 g porsjon (0,084 mol) 1,2-diisopropoksy-4-nitrobenzol hydreres over 2 g 10%-ig palladium-trekull-katalysator i 200 ml alkohol ved et initialtrykk på 2,8l kg/cm . Reduksjonen opphører etter et trykkfall på 1,62 kg/cm (87% av det beregnede) på 30 minutter. Katalysatoren filtreres fra, l8 g (0,084 mol) dietyletoksy-metylenmalonat tilsettes og oppløsningen oppvarmes under tilbakeløp i 2 timer. Det ambrafargede oljeaktige residuum som fåes etter avdestillering av alkoholen i vakuum, settes til 9OO ml "Dowtherm A" ved ca. 245°C Denne oppløsning kokes i 30 minutter og hensettes deretter til avkjøling natten over. Råproduktet frafiltreres og vaskes først med "Dowtherm" og deretter med benzol samt tørkes ved 80°C. Derved fåes 9,7 g (35$) av den i titelen angitte forbindelse som lysebrune krystaller, med smeltepunkt 220 - 228°C. Omkrystallisering fra dimetylformamid gir 6,4g hvite krystaller med smeltepunkt 233 - 234°C.
Eksempel 5»
Metyl- 6, 7- diisopropoksy- 4- hydroksy- 3- kinolinkarboksylat.
En 25 g prøve (0,11 mol) av l,2-diisopropoksy-4-nitrobenzol (eksempel 4B) hydreres over 4 g 5%-ig palladium-trekullkatalysator inneholdende ^ 0% vann i 200 ml alkohol ved et initialtrykk på 2,8l kg/cm . Reaksjonen opphører etter et trykk fall på 1,62 kg/cm<2>(87% av beregnet) på 30 minutter. Katalysatoren frafiltreres og til de sammenslåtte filtrater fra to slike reduksjoner settes 38 g (0,22 mol) dimetylmetoksy-metylenmalonat i 200 ml alkohol. Denne oppløsning oppvarmes under tilbakeløp ill/4 time og konsentreres deretter i vakuum til et viskøst residuum.
Dette residuet oppvarmes på dampbad og settes til 1400 ml "Dowtherm A", som forut oppvarmes til 245°C Oppløsningen kokes i 20 minutter og får deretter avkjøle. Råproduktet filtreres fra og vaskes først med "Dowtherm" og deretter med aceton som lufttørkes. Derved fåes 25 g (40%) av den i eksemplets titel
angitte forbindelse, med smeltepunkt 244_252°C.
Omkrystalliseringen av 52 g av dette produkt fra
550 ml dimetylformamid gir 43 g hvite krystaller, med smeltepunkt253-256°C.
Eksempel 6.
Etyl- 6, 7- diisopropoksy- 4- hydroksy- 3- kinolin- karboksylat.
A. 4, 5- diisopropoksy- 2- aminoacetofenon.
112 g (0,4 mol) 4»5-diisopropoksy-2-nitroacetofenon, 760 cm^ isopropanol og 47,5 g 5%-ig Pd/C, inneholdende 50% fuktig-het, has i et egnet hydreringsapparat, under 112,5 kg trykk hydrogen ved 40 - 50°C Reaksjonen er avsluttet i løpet av ca. 1/2 time. Katalysatoren filtreres fra, vaskes med en liten mengde isopropanol, og oppløsningen inndampes i vakuum. Den maksimale temperatur er 40 - 50°C. Det faste amin veier 95 gi me<* smeltepunkt 63 - 64°C.
B. 4>5- diisopropoksy- 2- formamidacetofenon.
34 cm^ 98%-ig maursyre settes langsomt under omrøring til 8l cm^ eddiksyreanhydrid. Reaksjonen er eksoterm og temperaturen øker til 50 - 60°C. Denne blanding omrøres kontinuerlig og oppvarmes ved 50 - 60°C i to timer, deretter avkjøles til 30°C. 50 g 4»5_diisopropoksy-2-aminoacetofenon tilsettes langsomt under
omrøring, således at temperaturen ikke overstiger 45°0« Omrøring og oppvarmning fortsettes ved 40 - 45°0 1 tre timer. Reaksjonsblandingen helles ut i 100 cm^ vann og ekstraheres med 100 cm^ eter. Eterlaget vaskes med vann og konsentreres til en olje. Oljen krystalliserer langsomt og etter flere dager oppslemmes
krystallene i heksan og filtreres. Utbyttet er 42 g, smeltepunkt 55 - 56°C.
C. Metyl- 6, 7- cliisopropoksy- 4- hydroksy- 3- kinolinkarboksylat.
34 g 4>5-diisopropoksy-2-formamidoacetofenon opp-løses i 150 cm^ dimetylkarbonat, og 14 g natriummetylat tilsettes i 2 omtrent like deler. Reaksjonen er eksoterm til omtrat 50 - 60°C. Etter omtrent 1/2 time blir reaksjonsblandingen tykk og det foregår ikke ytterligere tilstandsforandring. Blandingen hensettes natten over. Produktet filtreres, vaskes med metanol ved oppslemming og omfUtreres deretter. Kaken vaskes med ny metanol og produktet lufttørkes. Utbyttet er 20 g, 51% utbytte, smeltepunkt 235 - 240°C.
Det er undersøkt og sammenlignet følgende forbindelser med hensyn til virkning mot coccidiose. Herved inngås over f6ret på kyllinger som var infisert med Eimeria tenella, følgende forbindelser:
etylester av 6,7-dietoksy-4-hydroksy
3-kinolinkarbonsyre, kalt U-854 eksempel 1 etylester av 6,7-dibutoksy-4-hydroksy-
3-kinolinkarbonsyre, kalt U-926 eksempel 2 etylester av 6,7-dipropoksy-4-hydroksy-
3-kinolinkarbonsyre, kalt U-927 eksempel 3 metylester av 6,7-dietoksy~4-hydroksy-
3-kinolinkarbonsyre, kalt U-955 eksempel 4 etylester av 6,7-diisopropoksy-4-hydroksy-
3-kinolinkarbonsyre, kalt U-1041 eksempel 5 metylester av 6,7-diisopropoksy-4-hydroksy-
3-kinolinkarbonsyre, kalt U-IO63 eksempel 6 etylester av 6,7-dimetoksy-4~hydroksy-
3-kinolinkarbonsyre, kalt U-833 JACS 68 (I964) I264-6
For prøvingen ble det valgt grupper av ti dager gamle kyllinger (Cobbs White Rock), som var oppfødd under betingelser som sikret at dyrene var fri fra sykdom og podet ved hjelp av svelgsonde med oocyster av Eimeria tenella for å fremkalle infeksjon. En infeksjon som fremkalles eksperimentelt på denne måte ligner i det minste den som forekommer i praksis. Herav følger at hvis en komposisjon gir bra resultat på kyllinger som er infisert eksperimentelt kan man regne med bra resultater også i praksis. Forbindelsene ble blandet i hvert spesielt tilfelle med en bærer inneholdende kullhydrat, protein, mineraler og vitaminer, således at dyrene underglkk maksimal tilvekst. En komposisjon bestående av bærer og en forutbestemt mengde av den aktuelle forbindelse ble gitt til dyrene ad libitum.
Ved prøvene ble det anvendt positive kontroller.
Følgende resultater fremkom:
Av ovenstående tabell fremgår at produktene fremstillet ifølge oppfinnelsen har en betraktelig og ønskelig virkning mot coccidiose, mens kjente forbindelser savner denne egenskap.

- Fremgangsmåte til & fremstille lavere alkylestere av 6,7-di(lavere)alkoksy-4-hydroksy-3-kinolinkarboksylsyre som er virksomme mot coccidiose med formelenhvor R og R^hver betegner et rett eller forgrenet lavere alkyl- radikal med 2-4 karbonatomer, og Rg betegner en lavere alkylgruppe,karakterisert vedat man a) oppvarmer en Schiffsk base med formelen hvor R, R-j^ og Rg har den innledningsvis angitte betydning, vinder cykliseringsbetingelser, eller b) cykliserer et anilinderivat med formelenhvor R og Rjl har den ovenfor angitte betydning, og R^betegner en formylsubstituert alkoksykarbonylgruppe med formelen -CH(CH0)C00R2eller gruppen RgO.CO.CHg- og R^ betegner hydrogen, henholdsvis formyl, i nærvær av en base.
