Description
•k * α-Halogeenmethylaminozuurderivaten.
De uitvinding heeft betrekking op nieuwe farmaceutisch waardevolle a-halogeenaminozuurderivaten, die inhibitoren zijn van aromatisch aminozuurdecarboxylase.
De aminozuren tryptofaan, 5-hydroxytryptofaan, 5 3,4-dihydroxyfenylalanine (DOPA), tyrosine en fenylalanine worden door een aromatisch aminozuurdecarboxylase metabolisch omgezet in respectievelijk tryptamine, 5-hydroxytryptamine, 3,4-dihydroxyfenethylamine of dopamine, tyramine en fenethylamine. Men neemt aan, dat het aromatisch aminozuurdecarboxylase een niet specifiek enzyme is, in het bijzonder voor zover 10 het om perifere katalase gaat. Er zijn echter feiten, die er op wijzen, dat er in de hersenen specifieke decarboxyleringsenzymen bestaan voor DOPA en 5-hydroxytryptofaan elk.
Het is bekend, dat de hierboven opgesomde aromatische aminen bij verschillende pathofysiologische processen betrokken 15 zijn. Zo werd bijvoorbeeld gevonden, dat tryptamine, het decarboxylerings-produkt van tryptofaan, enzymatisch gemethyleerd wordt tot monomethyl-tryptamine, dat op zijn beurt in menselijke rode bloedlichaampjes, -plasma en -plaatjes enzymatisch tot dimethyltryptamine (DMT^ wordt gemethyleerd. Het methylerende enzyme is in vele zoogdierspecies aanwezig en het is aan-20 getoond, dat het in hersenweefsels van verschillende species, waaronder de mens, wordt geproduceerd. DMT, dat sterke hallucinogene of psychomime-tische eigenschappen heeft, kan een rol spelen bij de etiologie van schizofrenie en andere psychotische afwijkingen. Zodoende kan elk DMT-vorming blokkerend middel een waardevol antipsychoticum zijn. Blokkering van de 25 decarboxylering van tryptofaan leidt tot lagere tryptaminespiegels, waardoor het substraat voor de DMT-vorming wordt weggenomen. Zodoende kan een inhibitor van aromatisch aminozuurdecarboxylase, die omzetting van tryptofaan in tryptamine blokkeert, een waardevol antipsychoticum zijn.
Zowel 5-hydroxytryptamine (5-HT), het decarboxy-30 leringsprodukt van 5-hydroxytryptofaan, als 3,4-dihydroxyfenethylamine BAD ORIGtyl§L0 2 4 17 2 « (dopamine), het decarboxyleringsproduct van DOPA, zijn bij perifere en centrale fysiologische processen betrokken en middelen, die de spiegels van deze aminen doeltreffend regelen, zijn farmacologisch waardevolle middelen geworden. Het is gebleken, dat de centrale of hersenspiegel van 5-HT en norepinefrine, dat metabolisch door hydroxylering van dopamine en wordt gevormd, bij patiënt^ met manische afwijkingen hoger zijn dan bij personen zonder dergelijke afwijkingen. Ook is het aangetoond, dat monoamine centraal spiegel verlagende middelen, bijvoorbeeld 5-HT en in het bijzonder norepinefrine, bij toediening aan menselijke personen anti-manische eigenschappen hebben, terwijl monoamine spiegel verhogende geneesmiddelen bij hiertoe ontvankelijke personen manie kunnen opwekken. Zodoende kunnen middelen, die de vorming van 5-HT en dopamine blokkeren, bijvoorbeeld door inhibitie van het enzyme aromatisch aminozuurdecarboxy-lase, dat 5-hydroxytryptofaan en DOPA respectievelijk omzet in 5-HT en dopamine, van waarde zijn als antipsychotica of hoofdtranquilizers bij de behandeling van manische afwijkingen.
Ook werd aangetoond, dat middelen, die geschikt zijn voor de inhibitie van decarboxylering van DOPA tot dopamine, bij gelijktijdige toediening met exogeen DOPA of L-DOPA, geschikt zijn voor de behandeling van de ziekte van Parkinson. Men neemt aan, dat de ziekte van Parkinson tenminste ten dele aan verlaagde dopaminespiegels kan worden toegeschreven, aangezien exogene toediening van DOPA of L-DOPA een bekend # middel voor de behandeling van de ziekte van Parkinson is. Aangezien echter exogeen toegediend DOPA perifeer enzymatisch gemakkelijk in dopamine wordt omgezet, meet men grotere hoeveelheden toedienen teneinde de centraal-absorptie te vergroten. DOPA penetreert gemakkelijk in de hersen-bloedbarrière, terwijl dopamine dit niet doet. Toediening van DOPA of L-DOPA in combinatie met een perifeer actieve inhibitor van het enzyme, dat DOPA in dopamine omzet, vermindert de hoeveelheid L-DOPA, die ter verkrijging van een voor centraalabsorptie voldoende bloedspiegel moet worden toegediend. Men bereikt ook andere voordelen door gezamenlijke toediening van een aromatisch aminozuurdecarboxylase-inhibitor met L-DOPA. Door de perifere vorming van dopamine te voorkomen, kan men aan dopamine toegeschreven neveneffecten als cardiale arrhythmie, misselijkheid en overgeven voorkomen.

Onderzoekingen wijzen uit, dat 5-hydroxytryptamine (5-HT) spiegels bij patiënten met depressieve syndromen lager zijn dan bij personen zonder dergelijke syndromen. Ook is toediening van exogeen L-5-hydroxytryptofaan (L-5-HTP) voor de behandeling van bepaalde depressie-5 ve patiënten doeltreffend. Aangezien L-5-HTP echter evenals DOPA perifeer gemakkelijk tot 5-HT wordt gemetaboliseerd, moet men grote hoeveelheden L-5-HTP toedienen teneinde de verhoogde centraalspiegels van het aminozuur te bereiken. Het is aangetoond, dat door toediening van een inhibitor van aromatisch aminozuurdecarboxylase, dat de perifere vorming van 5-HT uit 10 5-HTP katalyseert, de hoeveelheid exogeen 5-HTP, die ter vorming van ver hoogde centraalspiegels nodig is, aanzienlijk wordt verminderd. Met andere woorden inhibitoren van aromatisch aminozuurdecarboxylase zijn bij combinatie met exogeen 5-HTP waardevol voor de behandeling van depressie gebleken.
15 · Middelen, die perifere omzetting van 5-HTP in 5-HT
blokkeren, kunnen waardevol zijn voor de behandeling van andere aandoeningen, die reageren op verhoogde 5-HTP spiegels ten gevolge van exogene toediening van 5-HTP. Het is gebleken, dat men exogeen L-5-HTP kan gebruiken voor de behandeling van myocIonuswerking. Onderzoekingen hebben ook ont-20 huid, dat toediening van exogeen 5-HTP geschikt is voor de behandeling van insomnie. Zodoende kan gelijktijdige toediening van 5-HTP en een aromatisch aminozuurdecarboxylase-inhibitor voor de behandeling van deze aandoeningen gunstig zijn.
Blokkering van de perifere vorming van 5-hydroxy-25 tryptamine kan tot andere gunstige effecten leiden, aangezien het bekend is, dat 5-HT bijvoorbeeld is betrokken bij de etiologie van rheumatoide arthritis en het carcinoïdesyndroom door verhoging van de collageenspiegel. Ook wordt vermeld, dat 5-HT het primaire autocoïde is, dat verantwoordelijk is voor anafylactoïde reactiesobij menselijke personen alsmede voor 30 bronchoconstrictie bij astmatische patiënten en middelen, die de vorming van 5-HT antagoneren of inhibiteren, zijn dan ook voor de behandeling van deze aandoeningen van waarde. Ook is het bekend, dat 5-HT bloedplaat-jesaggregatie veroorzaakt en als causale factor betrokken is bij het postgastrectoraie-dumpingssyndrom en migraine hoofdpijn. Methylsergide, een 35 5-hydroxytryptamine-antagonist, is voor behandeling van postgastrectomie- BAD omcgiptg 2 4 17 4 dumpingssyndroom doeltreffend gebleken. *
Men heeft geopperd', dat fenethylamine, het decarboxy-leringsproduct van fenylalanine, als endogene verbinding bijdraagt tot schizofreniesymptomen en migraine hoofdpijnen uitlokt. Ook werd geopperd, dat endogeen tyramine, het decarboxyleringsproduct van tyrosine, bijdraagt tot toevallen.
Het is zodoende duidelijk, dat middelen, die men kan gebruiken voor het regelen van de spiegels van aromatische aminozuren en aminen in vele farmacologische situaties worden toegepast. De verbindingen van de uitvinding zijn inhibitoren van aromatisch aminozuurdecarboxylase, dat tryptofaan, 5-hydroxytryptofaan, 3,4-dihydroxyfenylalanine, tyrosine en fenylalanine omzet in respectievelijke aminen en hebben derhalve farmacologische waarde.
De verbindingen van de uitvinding hebben de algemene formule 1, waarin Y Fcn2~' F2CH~· C1CH2~ of C12CH- voorstelt, ^ waterstof, alkylcarbonyl met een rechte of vertakte alkylrest met 1-4 koolstofatomen, alkoxycarbonyl met een rechte of vertakte alkylrest met 1-4 koolstofatomen of een groep met de formule 1 3. waarin R22 waterstof, een rechte of vertakte alkylrest met 1-4 koolstofatomen, benzyl of p-hydroxybenzyl is, voorstelt, R2 hydroxyl, een rechte of vertakte altoxygroep met 1-8 koolstofatomen, -NR_R , waarin R_ en R elk waterstof of een rechte of vertakte alkylgroep % / o / 8 met 1-4 koolstofatomen zijn, of een groep met de formule 14» waarin R^ waterstof, een rechte of vertakte alkylgroep met 1-4 koolstofatomen, benzyl of*p-hydroxybenzyl is, voorstelt en elk der symbolen R^» Rj» Rg» R’4 en Rg de in onderstaande tabel A gedefinieerde betekenis heeft, waarbij Rwaterstof, een rechte of vertakte alkylgroep met 1-8 koolstofatomen, alkylcarbonyl roet een rechte of vertakte alkylrest met 1-6 koolstofatomen, benzoyl of fenylalkyleencarbonyl met een rechte of vertakte alkyleenrest met 1-6 koolstofatomen is.

Onder de uitvinding -allen ook farmaceutisch aanvaardbare zouten en individuele optische isomeren van de verbindingen met de algemene formule 1.
10 De verbindingen met de algemene formule 1 hebben grote farmacologische waarden, omdat deze verbindingen inhibito-ren zijn van aromatisch aminozuurdecarboxylase en als tussenproduk-ten bij de bereiding van waardevolle farmacologische middelen kunnen worden gebruikt.
15 De hier beschreven verbindingen met de algemene formule 10 en hun bereidingswijze vormt ook deel van de onderhavige uitvinding. De verbindingen met de formule 10 zijn waardevolle chemische tussenprodukten voor de bereiding van verbindingen met de formule 1 , waarin Y voorstelt.
20 Bij de definitie van de formule 1 wordt onder alkylcarbonyl een groep verstaan met de formule 15 met een rechte of vertakte alkylrest met 1-6 koolstofatomen.
De bij de definitie van de formule 1 gebruikte uitdrukking benzoyl heeft betrekking op een groep met de formule 25 16.
De in verband met de definitie van de algemene formule 1 gebruikte uitdrukking fenylalkyleencarbonyl heeft betrekking op een groep met de formule 17 met een rechte of vertakte alkyleenrest met 1-6 koolstofatomen, bijvoorbeeld methyleen, 30 ethyleen, isopropyleen of butyleen.
Goede voorbeelden van rechte of vertakte alkoxy-groepen met 1-8 koolstofatomen zijn methoxy, ethoxy, isopropoxy, n-butoxy, t-butoxy, n-pentyloxy, t-pentoxy, n-hexyloxy en n-octyl-oxy.
35 Goede voorbeelden van rechte of vertakte alkyl- groepen met 1-6 koolstofatomen zijn methyl, ethyl, n-propyl, iso- EJAD ORI0W 2 4 17 7 m propyl, n-butyl, t-butyl en n-pentyl.
Goede voorbeelden van farmaceutisch aanvaardbare zouten van de verbindingen van de uitvinding zijn niet-toxische zuuradditie-zouten, gevormd met anorganische zuren als zoutzuur, broomwaterstofzuur, 5 zwavelzuur en fosforzuur en organische zuren, als methaansulfonzuur, salicylzuur, maleïnezuur, malonzuur, wijnsteenzuur, citroenzuur en ascorbinezuur en niet-toxische zouten, gevormd met anorganische of organische basen, als die, afgeleid van alkalimetalen, bijvoorbeeld natrium, kalium en lithium, aardalkalimetalen, bijvoorbeeld calcium en magnesium, ^ lichte metalen van groep UIA, bijvoorbeeld aluminium, of van organische aminen, als primaire, secundaire of tertiaire aminen, bijvoorbeeld cyclohexylamine, ethylamine, pyridine, methylaminoethanol, ethanolamine en piperazine. Men bereidt de zouten op de gebruikelijke wijze.
Voorkeursverbindingen van de uitvinding zijn die met 1 5 · de algemene formule 1, waarin Rj waterstof of alkylcarbonyl met een rechte of vertakte alkylrest met 1-4 koolstofatomen voorstelt, waarbij de meeste voorkeur uitgaat naar verbindingen, waarin Rj waterstof is. Een andere voorkeursuitvoeringsvorm van de uitvinding zijn de verbindingen met de formule 1, waarin R2 hydroxyl of een rechte of vertakte alkoxygroep met nr\ , , . # 1-8 koolstofatomen voorstelt. Verbindingen, waarin R^ hydroxyl is, verdienen de meeste voorkeur. Ook gaat de voorkeur uit naar de verbindingen met de formule 1, waarin R^, R^, R^, R'^ en Rg elk waterstof of ORjq, waarin Rjq waterstof is, voorstellen. Ook verbindingen, waarin R OH is en een der symbolen R^, R^, R’^ of Rg OH, OCH^ of CH^ is en verbindingen, ^ waarin R^ OH is en een der symbolen R^ of R'^ OH, OCH^ of CH^ is, vormen voorkeursuitvoeringsvormen van de onderhavige uitvinding. Verbindingen met de formule 1, waarin Y FCH^- of F2CH- is, verdienen ook de voorkeur.

De verbindingen met de algemene formule ] zijn irreversibele inhibitoren van het enzyme, dat metabolisch de omzetting katalyseert van tryptofaan, 5-hydroxytryptofaan, 3,4-dihydrofenyl-alanine, tyrosine en fenylalanine in respectievelijk tryptamine, 5-hydroxytryptamine, 3,4-dihydroxyfenylethylamine, tyramine en fen-ethylamine. De verbindingen van de uitvinding inhibiteren irreversibel doeltreffend zowel centraal als perifeer de werking van niet specifiek aromatisch aminozuurdecarboxylase en de werking van 3,4-dihydroxyfenylalanine (DOPA decarboxy- 25 lase). Als boven reeds is aangegeven kan men de verbindingen met de algemene formule 1 bijzonder goed gebruiken als men ze samen met exogeen L-DOPA toedient, in welk geval men afzonderlijke preparaten van een verbinding met de algemene formule 1 en van L-DOPA kan toedienen, of beide actieve bestanddelen tot één enkel gecombineerd farmaceutisch preparaat kan verwerken. Bij elk van beide toedieningswijzen varieert de hoeveelheid verbinding met de algemene formule 1 vergeleken bij de toegediende hoeveelheid L-DOPA van 1:1 tot 1:10. Een gecombineerd preparaat kan bestaan uit een binnengedeelte, dat L-DOPA bevat en een buitengedeelte, dat een verbinding met de algemene formule 1 bevat, waarbij elk actief bestanddeel op geschikte wijze is verwerkt. Men kan een bijzonder geschikt gecombineerd preparaat bereiden door L-DOPA, eventueel met geschikte dragers, tot een kern te persen, deze kern van een tegen maagsap bestande bekledingslaag te voorzien en over de beklede kern een buitenlaag aan te brengen, die een op een geschikte wijze verwerkte verbinding met de algemene formule 1 bevat. Bij gebruikmaking van een dergelijke combinatie wordt de decarboxylase-inhibitor, dat wil zeggen een verbinding met de algemene formule 1, bij voorkeur 30 tot 60 minuten voor het L-DOPA afgeweven. De bekledingslaag kan worden gevormd door gebruikmaking van een niet-waterige oplossing van glyceriden of een in water onoplosbaar polymeer als ethylcellulose of celluloseacetaatftalaat. Men kan ook preparaten ίgebruiken, waarin het L-DOPA enterisC5s bekleed door gebruikmaking van mengsels van schellakken en schellakderivaten en celluloseacetaatftalaten.
Onder worden nog goede voorbeelden van geschikte farmaceutische preparaten beschreven.
De verbindingen met de algemene formule 1 zijn behalve waardevolle farmacologische middelen ook tussenproducten voor het bereiden van cefalosporinen met antibiotische werking. Zo kan men de verbindingen met de algemene formule 1, waarin R2 hydroxyl is, gebruiken'voor de bereiding van cefalosporinederivaten met de algemene formule 2. In deze formule 2 hebben Y, R^, R^, R^, Rg, R'4 en Rg dezelfde betekenis als in de algemene formule 1, stelt M waterstof of een negatieve lading voor en stelt X waterstof of acetoxy voor.
De verbindingen met de algemene formule 2 en hun BAD cffedbOlAL 4 17 14 k ê farmaceutisch aanvaardbare zouten en individuele optische isomeren zijn nieuwe verbindingen, die men als antibiotica kan gebruiken en op dezelfde wijze kan toedienen als vele bekende cefalosporinederivaten, bijvoorbeeld cefalexine, cefalothine en cefaloglycine. De verbindingen met de algemene 5 formule 2 en hun farmaceutisch aanvaardbare zouten en isomeren kunnen alleen of in de vorm van farmaceutische preparaten hetzij oraal, hetzij parenteraal, hetzij plaatselijk worden toegediend aan warmbloedige dieren, dat wil zeggen vogels en zoogdieren, bijvoorbeekd katten, honden, runderen, schapen, paarden en mensen. Oraal kan men de verbindingen toedienen in de 10 vorm van tabletten, capsules of pillen of in de vorm van elixirs of sus pensies. Parenteraal kan men de verbindingen het beste gebruiken in de vorm van een steriele oplossing in water, die andere opgeloste stoffen kan bevatten, bijvoorbeeld een ter verkrijging van een isotone oplossing voldoende hoeveelheid zout oi glucose. Ter plaatselijke toediening kan men 15 de verbindingen met de algemene formule 2 of hun zouten en isomeren in crèmes en zalven verwerken.
Goede voorbeelden van bacteriën, waartegen de verbindingen met de algemene formule 2 en hun farmaceutisch aanvaardbare zouten en afzonderlijke optische isomeren werkzaam zijn, zijn Staphylococcus 20 aureus, Salmonella schotmuehlero, Klebsiella pneumoniae, Diplococcus 'pneumoniae en Streptococcus pyogenes.
Goede voorbeelden van farmaceutisch aanvaardbare niet-toxische anorganische zuuradditiezouten van de verbindingen met de algemene * formule 2, zijn het hydrochloride, hydrobromide, sulfaat, sulfamaat, en 25 fosfaat en organische zuuradditiezouten zijn bijvoorbeeld het maleaat, acetaat, citraat, oxalaat, succinaat, benzoaat, tartraat, fumaraat, malaat en ascorbaat. De zouten kunnen op de gebruikelijke wijzen worden gevormd.

Men kan de verbindingen met de algemene formule 2, waarin waterstof is, bereiden door koppeling Vein 7-aminocefalosporaan-zuur of een derivaat daarvan met de formule 3, waarin X en M dezelfde betekenis hebben als in formule 2, met een zuur met de formule 4 of een functioneel derivaat daarvan als het zuurchloride of een zuuranhydride en in aanwezigheid van een dehydrateermiddel als dicyclohexylcarbodiimide, als men het vrije zuur gebruikt, in welke formule 4 R^, R^, R^, R'^ en Rg dezelfde betekenis hebben als in formule 2 en de aminogroep is beschermd met een geschikte blokkerende groep, bijvoorbeeld t-butoxycarbonyl, gevolgd door zure hydrolyse ter verwijdering van de beschermende groep vein de aminogroep.
Men voert de koppelingsreactie in het algemeen uit in een oplosmiddel, bijvoorbeeld ethylacetaat, dioxan, chloroform of tetrahydrofuran, in aanwezigheid van een base, bijvoorbeeld alkalibicarbo-naar. De reactie temperatuur kan van -10 tot 100° C en de reactietijd lean een half uur tot 10 uur verschillen. De cefalosporineproducten worden op de gebruikelijke wijzen gewonnen. Men bereidt de verbindingen met de algemene formule 4 op de hierboven beschreven wijzen, terwijl de verbin- * dingen met de formule 3 in de handel verkrijgbaar zijn of op algemeen bekende wijzen kunnen worden bereid. t Men kan de verbindingen met de algemene formule 2, waarin R^ geen waterstof is, uit de overeenkomstige derivaten, waarin R^ waterstof is bereiden volgens de algemene werkwijzen, die hieronder zijn beschreven voor verbindingen met de algemene formule 1, waarin Rj geen waterstof is.
Men kan de verbindingen met de algemene formule 1, waarin R^ waterstof is, R^ hydroxyl is, en R^ en R4 beide OR^0 zijn, waarbij rjq waterstof is, of R^ en R<. beide OR10 zijn, waarbij Rj0 waterstof is, of R^ en Rj. samen -O-CH^-O- zijn, of waarin R^, R^, R^» R'4 en Rg elk de in tabel A gedefinieerde betekenissen hebben, behalve, dat R^q methyl is, bereiden door een geschikt beschermd fenylpropionaat met de formule 5 met een sterke base te behandelen onder vorming van een carbaanion, BAD O0|Q|Oa2 4 17 dat men behandelt met een geschikt halogeenmethylalkyleermiddel, gevolgd « 16 door zure hydrolyse. In de algemene formule S stelt R een rechte of vertakte alkoxygroep met 1-8 koolstofatomen voor, R^ waterstof, fenyl, een rechte of vertakte alkylgroep met 1-8 koolstofatomen, methoxy of 5 ethoxy voor, Rc fenyl of een rechte of vertakte alkylgroep roet 1-8 kool- stofatomen voor of vormen R^ en Rq samen een alkyleengroep met 5-7 koolstof atomen, dat wil zeggen -C^-iC^^-C^-, waarin m een geheel getal van 3-5 is. Goede voorbeelden van rechte of vertakte alkylgroepen met 1-8 koolstofatomen, die door R^ en Rc kunnen worden voorgesteld, zijn 10 methyl, ethyl, n-propyl, isopropyl, n-butyl, t-butyl, n-hexyl, n-octyl en neopentyl. Lithiumacetylide, dilithiumacetylide, natriumhydride en lithiumdiisopropylamide worden bij voorkeur als base gebruikt.
Geschikte halogeenmethylalkyleermiddelen voor boven-35 staande reactie zijn bijvoorbeeld chloorfluormethaan, broomfluormethaan, bad orkS&O 2 4 17 18 ê fluorjoodmethaan, chloordifluormethaan, broomdifluormethaan, difluor-joodmethaan, broomchloormethaan, dichloormethaan, chloorjoodmethaan, broomdichloormethaan en dichloorjoodmethaan. De halogeenmethylalkyleermiddelen zijn in de techniek bekend.
5 Men kan de alkyleringsreactie uitvoeren in een apro- tisch oplosmiddel, bijvoorbeeld benzeen, tolueen, ether, tetrahydro-furan, dimethylsulfoxyde, of hexamethylfosfortriamide. De reactietempe-ratuur kan van -120 tot 65°C variëren, waarbij men bij voorkeur 40°C gebruikt. De reactietijd kan van een half tot 24 uur variëren.
10 Zure hydrolyse ter verwijdering van eventueel niet gereageerd hebbende uitgangsstof eb beschermende groepen kan in één trap of trapsgewijze geschieden. Bij een ééntrapshydrolyse varieert de gebruikte zuurconcentratie natuurlijk met de duur van de hydrolysetrap en de gebruikte temperatuur. Zo kan men bijvoorbeeld een ééntrapshydrolyse uit-15 voeren door behandeling met geconcentreerd zoutzuur bij 25-120°C gedurende 1-4 dagen. Trapsgewijze hydrolyse kan geschieden met verdund zuur bij 25°C gedurende een half tot 6 uur ter verwijdering van niet gereageerd hebbende uitgangsstof, herhaaldelijke behandeling met verdund zuur ter verwijdering van eventuele beschermende groepen van de aminegroep, 20 gevolgd door behandeling met geconcentreerd zuur bij 25-125°C gedurende 1-3 dagen ter verwijdering van eventuele ester of ethergroepen. Trapsgewijze hydrolyse verdient de voorkeur.
De verbindingen met de algemene formule 1, waarin Y FCH^- voorstelt, hydroxyl is, Rj waterstof is, R^ en R^ beide ORjq 25 voorstellen, waarbij R waterstof is, of R^ en R,. beide ORjq voorstellen, waarbij R waterstof is, of R^ en R^ beide -0-CH2-0- voorstellen, of waarin elk van de symbolen R^, R^, R^, R’^ en R^ een der andere in tabel A gedefi-nieerde betekenissen heeft, behalve Rjq is methyl of waterstof, kunnen allen worden bereid volgens de werkwijze, die in bijgaand reac-30 tieschema is weergegeven.
In dit reactieschema stelt Xa chloor of broom voor, hebben Rj j, RJ2> Rj3» R’i2 en R14 deze^e betekenis als in tabel B en hebben R^, R^, R^, R'^ en R^ dezelfde betekenis als in tabel A, behalve Rjq is methyl of waterstof.
35 Volgens bijgaand reactieschema voegt men een benzyl- halogenide met de formule 7 zeer langzaam toe aan magnesiumkrullen in BAD ORIGir&P 0 2 4 17 19 * een geschikte ether als oplosmiddel, bijvoorbeeld tetrahydrofuran, diethyl-ether of een mengsel daarvan en laat de reactie 30 minuten tot 24 uur voortgaan bij een temperatuur van -20°C tot 70°C, bij voorkeur van 25°C tot het kookpunt van het oplosmiddel. Bij het begin van de reactie voegt 5 men een spoor methyljodide toe. Ook, indien hetzij rjj» hetzij Rjj methoxy is, start men de reactie in tetrahydrofuran.
Bij het aldus gevormde Grignardreagens, dat wil zeggen een verbinding met de formule 8, voegt men fluorazijnzuurnitril in een verhouding, die van 0,5 tot 3 kan variëren toe in een aprotisch oplosmid-10 del, bijvoorbeeld tetrahydrofuran, diethylether, dioxan, benzeen, dime-thoxyethaan of dimethoxymethaan of een mengsel daarvan. De reactietempe-ratuur van varieert van -20°C tot -70°C, bij voorkeur van -20°C tot -25°C en de reactietijd van 10 minuten tot 12 uur, bij voorkeur van 10 minuten tot 1 uur. Men hydrolyseert het aldus gevormde ketiminezout, 15 dat wil zeggen een verbinding met de formule 9, met zuur tot het keton.
Zure hydrolyse kan worden verkregen door het ketiminezout uit te gieten in water en geconcentreerd zoutzuur onder handhaving van een sterk zuur medium. Men wint het keton met de formule 10 op de gebruikelijke wijzen, bijvoorbeeld door extractie met petroleumether of pentaan of hexaan en 20 daarna met een ether als diethylether.
Men zet het keton om in het aminonitril met de formule 11 onder de omstandigheden van een reactie van Strecker door behandeling met 1 tot 10 equivalenten natriumcyanide en 1 tot 10 equivalenten ammonium-zout, bijvoorbeeld ammoniumchloride in hetzij een basisch medium onder 25 gebruikmaking van ammoniumhydroxyde in water (1 molair tot geconcentreerd) in een lagere alkohol, als methanol of ethanol, hetzij een neutraal medium onder gebruikmaking van water en een lagere alkohol.
Hydrolyse van het aminonitril tot het aminozuur met de formule 12 kan op verschillende wijzen geschieden. Zo kan men de hydrolyse 30 bijvoorbeeld uitvoeren door behandeling met broomwaterstof gedurende een half tot 24 uur bij 25 tot 100°C. Als men echter broomwaterstof gebruikt is Rjq in de formule 12 waterstof. Hydrolyse kan ook met zwavelzuur op algemeen bekende wijze geschieden. Ook kan men hydrolyse bewerkstelligen door behandeling met een lagere alkohol, bijvoorbeeld methanol, 35 die met watervrije chloorwaterstof is verzadigd, gedurende 1 tot 24 uur, bij voorkeur 10 uur, bij 0 tot 50°C, bij voorkeur 25°C, onder ver- BAO origii?aC Π 2 4 1 7 9 20 krijging van het overeenkomstige amino amide, dat men hydrolyseert door behandeling met waterig zoutzuur of 50 % water bevattend zwavelzuur gedurende 2 tot 6 uur bij 60 tot 100°C, bij voorkeur 95°C, gevolgd door neutralisatie met bariumhydroxyde als men zwavelzuur heeft gebruikt.
De meest nabijliggende stand der techniek voor wat betreft de bereiding van de ketonen met de formule 10 is de bekende werkwijze voor het bereiden van fluormethyl m-tolylketon, E.D. Bergmann et al., J.Chem.Soc. 1961. 3452. De in bijgaand reactieschema beschreven werkwijze onderscheidt zich duidelijk van de stand der techniek en biedt ook ten ^ opzichte \an de stand der techniek bepaalde voordelen. Bij boven beschreven werkwijze zijn de Grignardverbindingen ten eerste benzylverbindingen, moet men ten tweede de temperatuur van het reactiemengsel tijdens de Grignardadditie op een temperatuur van -20°C of daar beneden houden, terwijl het bij de onderhavige werkwijze gebruikte fluorazijnzuurnitril-^ reagens veel minder toxisch is dan de monofluorazijnzuurderivaten, die men volgens de stand der techniek ter bereiding van fluormethylbenzylke-ton gébruikt.
Men kan de verbindingen met de algemene formule 1, waarin Rj waterstof en R2 hydroxyl is en R^, R^, R1^ of R^ ORjq voorstelt, ^ waarbij Rjq waterstof is, bereiden door het overeenkomstige derivaat, « waarin R^, R^, R'^ of R^ ORjq voorstelt, waarbij Rjq methyl is, 4-24 uur bij 25-125°C te behandelen met broomwaterstof in water of azijnzuur.
Men kan verbindingen met de algemene formule 1, waarin 25 Rj waterstof en R2 hydroxyl is en R^, R^, R'^ en/of R^ OR^ voorstelt, waarbij Rjq een rechte of vertakte alkylgroep met 1-8 koolstofatomen is, bereiden door de overeenkomstige verbinding, waarin Rjq waterstof is, 1-24 uur bij 25-85°C in een lagere alkohol, bijvoorbeeld methanol of ethanol, of een koolwaterstof als benzeen of tolueen, in aanwezigheid van een or-30 ganische base, als triethylamine of pyridine, of in een aprotisch oplosmiddel, als dimethylformamide, dimethylaceetamide, of dimethylsulfoxyde, in aanwezigheid van natriumhydride te alkyleren met een alkylhalogenide met de formule R2JY2* waar^n R21 een rechte vertakte alkylgroep met 1-8 koolstofatomen en Y2 halogeen, bijvoorbeeld broom of jodium, voor-35 stelt, gevolgd door hydrolyse met een waterige base, met dien verstande, • dat men voor de alkyleringsreactie de α-aminogroep en eventueel de carboxyl- groep van de hydroxylverbinding, waarvan men uitgaat, geschikt zijn be- BAD ORIGI^aQ 0 2 4 17 21 e schermd, bijvoorbeeld met t-butoxycarbonyl of benzyloxycarbonyl of benzyl, welke bescherming vervolgens weer wordt verwijderd door hydrogenolyse of door behandeling met zuur, bijvoorbeeld met trifluorazijnzuur. De bij deze werkwijze gebruikte alkylhalogeniden zijn algemeen bekend of kunnen op 5 algemeen bekende wijzen worden bereid.
Men kan de verbindingen met de algemene formule 1, waarin R.2 hydroxyl, en Rj waterstof is en Rg, R^, Rr^ en/of Rg ORjq voorstelt, waarbij R alkylcarbonyl met een rechte of vertakte alkylrest met 1-6 koolstofatomen, benzoyl of fenylalkyleencarbonyl met een rechte of ver-10 takte alkyleenrest met 1-6 kooIstofatomen is, bereiden door het overeenkomstige derivaat, waarin Rjq waterstof is, 1-24 uur bij 25-100°C in aanwezigheid van een organische base als pyridine, chinoline of triethyl-amine, die als oplosmiddel dient, te behandelen met een zuuranhydride met de formule 18 of een zuurhalogenide met de formule 19, waarin Hal 15 chloor of broom is en R^2 een rechte of vertakte alkylgroep met 1-6 kool-stofatomen, fenyl of fenylalkyleen met een rechte of vertakte alkylrest met 1-6 kooIs tofatomen voorstelt, met dien verstande, dat v66r de reactie de α-aminogroep en eventueel de carboxylgroep van de hydroxylverbinding, waarvan men uitgaat, geschikt zijn beschermd, bijvoorbeeld met t-butoxy-20 carbonyl of benzyloxycarbonyl of benzyl, die men vervolgens verwijdert door behandeling met zuur, bijvoorbeeld met trifluorazijnzuur en hydrogenolyse. De bij deze reactie gebruikte zuuranhydriden en zuurhalogeniden zijn algemeen bekend of kunnen op algemeen bekende wijzen uit de geëigende zuren worden bereid.
25 Men kan de verbindingen met de algemene formule 1, waarin R2 een rechte of vertakte alkoxygroep met 1-8 koolstofatomen is, bereiden doot'-'het overeenkomstige derivaat, waarin R2 hydroxyl is, met thionylchlo-ride te behandelen onder vorming van het zuurchloride, dat met 4-12 uur bij 25°C laat reageren met een alkohol met de formule R2gOH, waarin R22 30 een rechte of vertakte alkylgroep voorstelt met 1-8 koolstofatomen als methyl, ethyl, n-propyl, isopropyl, n-butyl, hexyl of octyl.
Men kan de verbindingen met de algemene formule 1, waarin R2 -NR^Rg voorstelt, waarin R^ en Rg elk waterstof of een rechte of vertakte alkylgroep met 1-4 koolstofatomen zijn, bereiden door een 35 acyleringsreactie van een zuurhalogenide, bijvoorbeeld een zuurchloride, van de overeenkomstige verbinding, waarin R2 hydroxyl is en Rj dezelfde betekenis heeft als in de formule 1, met dien verstande, dat een eventuele badorigiï^iQ 0 2 4 17 22 0 vrije aminogroep geschikt is beschermd, bijvoorbeeld met carbobenzyloxy of t-butoxycarbonyl en als R^, R^» R^ en/of R'^ OR^g voorstelt en rjq waterstof is, een dergelijke groep is beschermd als de overeenkomstige alkyl-carbonyloxygroep, met een overmaat geëigend amine, dat men kan voorstellen 5 als HNR^Rg. Men voert de reactie 1-4 uur bij 25° C uit in methyleenchloride, chloroform, dimethylformamide, ethers als tetrahydrofuran of dioxan, of benzeen. Geschikte aminen zijn bijvoorbeeld ammoniak, of een verbinding, die een potentiële ammoniakbron is, bijvoorbeeld hexamethyleentetramine, primaire aminen, bijvoorbeeld methylamine, ethylamine of n-propylamine of 10 secundaire aminen als dimethylamine, diethylamine of di-n-butylamine. Na de acyleringsreactie verwijdert men de beschermende groep van de aminogroep door behandeling met zuur, bijvoorbeeld broomwaterstof in dioxan of hydro-genolyse en verwijdert de bescherming van de hydroxylgroep, indien van toepassing, door basische of zure hydrolyse.
15 Men kan de verbindingen met de algemene formule 1, waarin R^ een groep voorstelt met de formule 14, bereiden door het overeenkomstige derivaat, waarin R^ hydroxyl of een functioneel derivaat daarvan, bijvoorbeeld een 'zuuranhydride is en Rj dezelfde betekenis heeft als in de formule 1, met dien verstande, dat een eventuele vrije amino-<20 groep geschikt is beschermd, bijvoorbeeld als benzyloxycarbonyl of t-butoxy carbonyl, 1-24 uur bij 0-50° C in een ether, bijvoorbeeld tetrahydrofuran of dioxan, te laten reageren met een verbinding met de formule 20, waarin .Rg dezelfde betekenis heeft als in de formule 1 en R24 een lagere alkyl-groep voorstelt, bijvoorbeeld methyl of ethyl, gevolgd door zure hydrolyse 25 ter verwijdering van de beschermende groep, met dien verstemde, dat, wanneer men het vrije zuur gebruikt, waarvan de aminogroep is beschermd, de reactie uitvoert onder gebruikmaking van een derhydrateermiddel, bijvoorbeeld di-cyclohexylcarbodiimide.
Men kan de verbindingen met de algemene formule 1, 30 waarin R^ alkylcarbonyl met een rechte of vertakte alkylrest met 1-4 kool stof atomen voorstelt, bereiden door het overeenkomstige derivaat, waarin Rj waterstof en R^ hydroxyl is, een half tot 6 uur bij 0-25° C in water, in aanwezigheid van een base als natriumhydroxyde of natriumboraat, te behandelen met een zuurhalogenide met de formule 21 , waarin hal een halo-35 geenatoom, bijvoorbeeld chloor of broom is en R2g een rechte of vertakte BAD ORIGINAL 8 0 0 2 4 1 7 23 # alkylgroep met 1-4 koolstofatomen voorstelt. Men kan deze verbindingen ook bereiden uit het esterderivaat, dat wil zeggen uit een verbinding met de algemene formule lf waarin R^ waterstof en een alkoxygroep met 1-8 koolstofatomen is, door een dergelijk derivaat een half tot 24 uur bij 0-25° C, in water, methyleenchloride, chloroform of dimethylaceetamide in aanwezigheid van een base als natriumhydroxyde, kaliumhydroxyde of overmaat triethylamine te behandelen met het zuurhalogenide met de formule 21 als boven beschreven.
Men kan de verbindingen met de algemene formule 1, waarin Rj alkoxycarbonyl met een rechte of vertakte alkoxyrest met 1-4 koolstofatomen is, bereiden door het overeenkomstige derivaat, waarin Rj waterstof en R^ hydroxyl is, een half tot 6 uur bij 0-25° C in water in aanwezigheid van een base als natriumhydroxyde of natriumboraat te behandelen met een alkylhalögeenformiaat met de formule 22, waarin hal een halogeenatoom als chloor of broom is en een rechte of vertakte alkylgroep met 1-4 koolstofatomen voorstelt.
Men kan de verbindingen met de algemene formule 1, waarin Rj een groep voorstelt met de formule 23, waarin R^ waterstof, een rechte of vertakte alkylgroep met 1-4 koolstofatomen, benzyl of p-hy-droxybenzyl is, bereiden door het overeenkomstige derivaat, waarin Rj ^waterstof is en Rj een rechte of vertakte alkoxygroep met 1-8 koolstof-atomen voorstelt, 1-12 uur bij 0-35° C te behandelen met een zuur met de formule 24 of een anhydride daarvan, waarin de aminogroep geschikt is »beschermd, bijvoorbeeld als benzoxycarbonyl of t-butoxycarbonylgroep en Rjy bovengenoemde betekenis heeft, in een ether, bijvoorbeeld tetrahydro-furan of dioxan, methyleenchloride of chloroform en in aanwezigheid van een dehydrateermiddel, als men het vrije zuur gebruikt, gevolgd door zure en basische hydrolyse ter verwijdering van de beschermende groepen. Eventueel kan men de basische hydrolyse weglaten, waardoor men het esterpep-tide derivaat verkrijgt.
De individuele optische isomeren van verbindingen met de formule 1, waarin waterstof en R2 hydroxyl is, kunnen worden afgescheiden onder gebruikmaking van een (+) of (-> binaftylfosforzuur-zout volgens de methode van R. Viterbo et al., Tetrahedron Letters 48, 4617 (1971). Men kan ook andere splitsmiddelen gebruiken als (+) camfer- BAD ORIGI&Ö. 0 2 4 17 24 10-sulfonzuur. De Individuele optische isomeren van de verbindingen met de formule 1» waarin R^ en R2 iets anders dan waterstof of hydroxyl zijn, kunnen als beschreven voor het racemische mengsel worden verkregen, maar dan uitgaande van het gesplitste aminozuur.
5 Men kan de verbindingen met de algemene formule 5 bereiden door één equivalent glycinaat met de formule 6, waarin Rft, R^ en Rc dezelfde betekenis hebben als in de algemene formule 5 te behandelen met één equivalent van een sterke base als een alkyllithium, bijvoorbeeld butyllithium of fènyllithium, lithium-di-alkylamide, bijvoorbeeld lithium-10 diisopropylamide, of lithiumamide, t-kaliumbutylaat, natriumamide, een metaalhydride als natriumhydride, een tertiair amine als triethylamine, lithiumacetylide of dilithiumacetylide, gevolgd door behandeling met een alkyleermiddel met de formule 7, waarin R^, R12' R13' R'l2 en R14 ^eze^^e betekenis hebben als in de formule 5 en X halogeen, bijvoorbeeld chloor d 15 of broom is. Men kan de alkylering uitvoeren in een aprotisch oplosmiddel, bijvoorbeeld benzeen, ethers, tetrahydrofuran, diraethylsulfoxyde of hexa-methylfosfortriamide. De reactietijd varieert van een half tot 24 uur en de temperatuur van -120 tot 25° C.
Men kan de verbindingen met de algemene formule 6 20 . bereiden door een geëigend alkylglycinaat te behandelen met een carbonyl- 4verbinding onder vorming van een base van Schiff, op algemeen bekende wijze, met name (a) als R^ waterstof is, door behandeling van de geëigende aminozuurester met benzaldehyde of een recht of vertakt alkanal met 1-9 «koolstofatomen, bijvoorbeeld propanal-1, butanal-1, 2,2-dimethylpropanal-25 1 of 2,2-diethylbutanal-l, (b) als ^ fenyl is, door behandeling van de geëigende aminozuurester met benzofenon of een fenyl of alkylketon met een rechte of vertakte alkylrest met 1-8 koolstofatomen, bijvoorbeeld fenylmethylketon, fenylethylketon, fenylisopropylketon, fenyl-n-butylketon of fenyl-t-butylketon en (c) als Rj^ een rechte of vertakte alkylgroep met 30 1-8 koolstofatomen is, behandeling van de geëigende aminozuurester met een fenylalkylketon als boven omschreven, met een dialkylketon, waarin elke alkylrest recht of vertakt is en 1-8 koolstofatomen bevat, bijvoorbeeld dimethylisopropylketon, di-n-butylketon of methyl-t-butylketon. De carbo-nylverbindingen zijn algemeen bekend of kunnen op algemeen bekende wijzen 35 worden bereid.
BAD ORIGINA& 0 0 2 4 1 7 25 *
Als in de verbindingen met de formule 6 methoxy of ethoxy is, laat men een geëigend alkylglycinaat reageren met een benzo-ylhalogenide, bijvoorbeeld het chloride of een alkaanzuurhalogenide, bijvoorbeeld het chloride, waarbij het alkaanzuur recht of vertakt is en 1-9 koolstofatomen bevat, bijvoorbeeld acetylchloride, propionylchloride, butyrylchloride, t-butyrylchloride, 2,2-diethylbutyrylchloride of valeryl-chloride, bij 0° C in een ether, methyleenchloride, dimethylformamide, dimethylaceetamide of chloorbenzeen in aanwezigheid van een organische base als triethylamine of pyridine, waarna men het reactiemengsel gedurende 1 uur tot 25° C laat opwarmen. Men combineert het verkregen amidederivaat, bij 25° C, opgelost in een chloorkoolwaterstof als methyleenchloride, chloorbenzeen of chloroform, roet een alkyleermiddel als methylfluorsulfo-naat, dimethylsulfaat, methyljodide, methyl-p-tolueensulfonaat of trime-thyloxoniumhexafluorfosfaat, -als R^ methoxy is, of triethyloxoniumtetra-fluorboraat, als ethoxy is, en verwarmt het verkregen reactiemengsel 12-20 uur aan een terugvloeikoeler. Daarna koelt men het reactiemengsel tot 25° C af, voegt een organische base toe, bijvoorbeeld triethylamine of pyridine, waarna men de oplossing met pekel extraheert en het product wint.
Als en Rc in de verbindingen met de formule 6 ..samen een alkyleengroep met 5-7 koolstof atomen vormen, kan men genoemde aminozuuresterderivaten verkrijgen door behandeling van de aminozuurester met een cyclisch alkanon, te weten cyclopentanon, cyclohexanon of cyclo-♦heptanon, onder vorming van een base van Schiff op algemeen bekende wijze.
Men kan de alkylglycinaten verkrijgen door glycine bij 25° C gedurende 12-36 uur te behandelen met een alcohol met de formule R H, waarin R dezelfde betekenis heeft als in de formule 6, die met a a HCl gas is verzadigd, of men betrekt de verbindingen uit de handel,
De verbindingen met de algemene formule 7 zijn algemeen bekend of kunnen worden bereid uit de overeenkomstige geëigend gesubstitueerde benzoëzuur- en benzaldehyde-derivaten, die algemeen bekend zijn. Zo kan men bijvoorbeeld de benzylhalogeniden met de formule 7 uit het overeenkomstige benzaldehyde bereiden door reductie met natriumborium-hydride, lithiumaluminiumhydride, of door katalytische reductie van de overeenkomstige benzoëzuurester met lithiumaluminiumhydride of boran of BAD ORI(Sr(k0 2 4 17 9 26 reductie van het overeenkomstige benzoëzuurderivaat met lithiumhydride en behandeling van het aldus gevormde benzylalcoholderivaat met bijvoorbeeld thionylchloride» fosforoxychloride, fosfortrichloride, fosfortribromide of fosforpentachloride.
5 Bepaalde verbindingen met de algemene formule 5 kunnen rechtstreeks uit het geëigende aminozuur worden verkregen door omzetting van dit zuur in een geëigende ester» gevolgd door bescherming van de α-aminogroep volgens de algemeen hierboven beschreven werkwijzen voor het beschermen van de aminogroep van het alkylglycinaat. Zo kan men 10 op een dergelijke wijze bijvoorbeeld het esterderivaat van tyrosine of fenylalanine met beschermde aminogroep verkrijgen. Men kan deze werkwijze niet gebruiken voor het bereiden van verbindingen met de formule 5» waarvan de aromatische ring een hydroxylgroep in de meta-stand bevat.
Het volgende voorbeeld I ligt het gebruik toe van 15 een verbinding met de algemene formule 1» waarin Rj hydroxyl is, als chemisch tussenproduct bij de bereiding van een cefalosporine met de formule 2.

1- fluor-3-(3,4-dimethoxyfenyl)-2-propanon.
Onder een stikstofatmosfeer voegt men 12,20 g 3,4-di-methoxybenzylchloride in 80 ml tetrahydrofuran (THF) langzaam (binnen 5 1 uur) toe aan 3,0 g magnesiumkrullen in 50 ml THF, waarbij de kolf is
gedompeld in een waterbad van kamertemperatuur. Men zet het roeren 1 uur voort, waarna men het Grignardreagens scheidt van de overmaat magnesium en overbrengt in een tweede kolf (onder stikstof) en tot -20°C afkoelt. Men voegt 4,0 g CH^FCN in 25 ml THF met zodanige snelheid van-10 uit een druppeltrechter toe, dat de inwendige temperatuur tussen -25 en -20°C gehandhaafd blijft. Nadat men alles heeft toegevoegd zet men het roeren nog 1 uur bij -20°C voort en giet daarna de oplossing uit op een mengsel van 200 g ijs, 240 ml water en 120 ml geconcentreerd zoutzuur en extraheert met 2 x 100 ml petroleumether en 4 x 100 ml diethylether. 15 Na drogen boven natriumsulfaat en afdamping van het etherextract verkrijgt men 4,53 g of 33 % zuiver keton met kookpunt 100°C/0,35 mm Hg. Bij een suspensiejvan 31,5 g of 0,149 mol l-fluor-3-20 (3,4-dimethoxyfenyl)-2-propanon en 9,57 g of 0,179 mol ammoniumchloride in 170 ml 28 % ammoniak in water voegt men 8,77 g of 0,179 mol natriumcyanide. Men roert het reactiemengsel 20 uur onder stikstof bij kamertemperatuur. De vaste stof, die zich afscheidt, wordt afgefiltreerd op gesinterd glas en daarna gewassen met 50 ml 28 % ammoniak in water.
25 De vaste stof wordt opgelost in 1,3 liter ether en de organische fase wordt gewassen met water en daarna met pekel en boven magnesiumsulfaat gedroogd. Bij concentratie van het oplosmiddel kristalliseert er 21,8 g 2-amino-2-fluormethy1-3-(3,4-dimethoxyfenyl)propionzuur uit met smeltpunt 76°C.

2-fluormethyl-2-amino-3-(3,4-dihydroxyfenyl)propionzuur.
Men verhit een oplossing van 20,7 g of 0,086 mol 2-amino-2-fluormethyl-3—(3,4-di-methoxyfenyl)propionzuur in 300 ml 47 % broomwaterstofzuur in water 32 uur onder stikstof op 100°C. Con-35 centratie van het oplosmiddel onder verlaagde druk levert een overblijf- badobigi^i00 24 u 0 29 sel op, dat men in 300 ml isopropanol oplost. Men laat de oplossing 12 uur bij 4°C rijpen. Het ammoniumbromide, dat zich afscheidt, wordt afgefiltreerd en gewassen met 3 x 40 ml isopropanol. Men neutraliseert het filtraat met 16 g triethylamine in isopropanol tot de pH van de 5 oplossing een waarde van 4,5-5 bereikt. Het precipitaat, dat zich vormt, wordt verzameld en intensief met chloroform gewassen. Het overblijfsel van 19,4 g wordt opgelost in 500 ml water en behandeld met actieve kool. Bij concentratie kristalliseert er 13 g a-fluormethyl-DOPA uit. Uit de moederloog verkrijgt men een tweede lading van 1,2 g 2-fluormethyl-2-10 amino-3-(3,4-dihydroxyfenyl)propionzuur met smeltpunt 230°C.

Als men bij de werkwijze van voorbeeld V een passende hoeveelheid p-methoxybenzylbromide in plaats van 3,4-dimethoxybenzyl-^ chloride gebruikt, verkrijgt men l-fluor-3-(p-methoxyfenyl)-2-propanon dat, wanneer men het bij de werkwijze van voorbeeld VI in een passende hoeveelheid gebruikt in plaats van l-fluor-3-(3,4-dimethoxyfenyl)-2-propanon, de verbinding 2-amino-2-fluormethyl-3-(p-methoxyfenyl)-propionzuurnitril oplevert.

Men verzadigt een oplossing van 1,0 g 2-amino-2-fluormethyl-3-(p-methoxyfenyl)propionzuurnitrilhydrochloride in 40 ml watervrije methanol onder ijskoeling met droge HC1. Men laat het mengsel 25 een nacht bij kamertemperatuur staan. Afdamping van het oplosmiddel levert het overeenkomstige amidehydrochloride met kwantitatieve opbrengst op. 35 Men verhit 920 g vrij amide en 7 ml 50 vol.% zwavelzuur BAD OR!Gir$l0 0 2 4 17 30 # in water 2 uur op 95°C. Men verdunt het mengsel met water tot een volume van 50 ml en neutraliseert met een bariumhydroxyde-oplossing (waarna het totale volume 150 ml bedraagt).
