Fo sfinylalkanoylprolinen.
De uitvinding heeft betrekking op nieuwe fosfi-nylalkanoylprolinen met de algemene formule 1, waarin Ri lager alkyl, fenyl of fenyl-lager alkyl, waterstof, fenyl-lager alkyl of een metaalion, R^ waterstof of lager alkyl, R^ waterstof, lager alkyl, 5 fenyl-lager alkyl of een metaalion en n 0 of 1 voorstellen.
In de formule 1 zijn de door de symbolen voorgestelde lagere alkylgroepen rechte of vertakte alifatische koolwaterstof groepen met tot 7 koolstof atomen, bijvoorbeeld methyl, ethyl, propyl, isopropyl, butyl, sec.butyl, t-butyl en dergelijke.
10 Dergelijke koolwaterstofgroepen met 1-4 koolstofatomen en in het bijzonder 1-2 koolstofatomen genieten de voorkeur. De fenyl-lagere alkylgroepen zijn aralkylresten van hetzelfde type, waarbij fenyl-methyl en fenylethyl, in het bijzonder fenylmethyl, de voorkeur genieten.
15 De metaalionen, voorgesteld door R£ en R^, zijn ëënwaardige metaalionen, bij voorkeur de alkalime taal ionen, in het bijzonder natrium, kalium en lithium.
Voorkeursverbindingen volgens de uitvinding zijn de verbindingen met de formule 1, waarin n 0 of 1, in het bij-20 zonder 0, is, Rj voorstelt fenyl of fenyl-lager alkyl, in het bijzonder fenyl-Cj-Cg-alkyl, R2 voorstelt waterstof of fenyl-lager alkyl, in het bijzonder waterstof of fenylmethyl, R^ voorstelt waterstof of lager alkyl, in het bijzonder waterstof of methyl, en R^ voorstelt waterstof of alkalimetaal, in het bijzonder waterstof, natrium, 25 kalium of lithium.
De verbindingen volgens de uitvinding kunnen worden bereid door proline, bij voorkeur in de vorm van een lagere alkyl- of fenyl-lagere alkyl-ester, waarin de estergroep gemakkelijk wordt verwijderd, bijvoorbeeld de t-butylester, fenylmethyl-30 79075 68 f' ’* « <5
Tk 2 ester of dergelijke, in reactie te brengen met een fosfinylazijn-zuur met de algemene formule 2, waarin n 0 is, in tegenwoordigheid van een condensatiemiddel, zoals 1.1’-carbonyldiïmidazool of dicyclo-hexylcarbodiïmide, en in een inert organisch oplosmiddel, zoals 5 acetonitril, dichloormethaan, ether, tetrahydrofuran, dioxan of dergelijke, of met een fosfolan met de formule 3, waarin η 1 is, in tegenwoordigheid van een base, zoals triethylamine, pyridine, N.N-dimethylamine of dergelijke, en in een inert organisch oplosmiddel, zoals die welke hierboven zijn genoemd.
10 Wanneer of R^ fenylmethyl is kan deze groep worden omgezet in waterstof door katalytische reductie, bijvoorbeeld met palladium op koolstof of palladium op bariumsulfaat volgens gebruikelijke methodes.
Wanneer R^ een gemakkelijk verwijderbare ester-15 groep, zoals t-butyl, is leidt de behandeling van de ester met tri-fluorazijnzuur en anisool tot het vrije zuur, waarin dus R^ waterstof is. Wanneer R2 lager alkyl is leidt de behandeling met een ha-logeensilan, zoals broomtrimethylsilan of joodtrimethylsilan, en vervolgens water tot het.vrije zuur.
20 De zuren vormen metaalzouten met ëénwaardige metalen, zoals alkalimetaalzouten door behandeling met een metaal-hydroxyde, bijvoorbeeld in waterige oplossing, volgens gebruikelijke methodes.
De proline-esters worden bereid onder toepas-25 sing van willekeurige bekende veresteringsmethodes onder gebruikmaking van een lagere alkanol of fenyl-lagere alkanol R^OH (in het bijzonder bij peptide-syntheses) als geïllustreerd in het Amerikaanse octrooischrift 4.046.889, J, Org.Chem. 28, 176 (1963); Pettit, Synthetic Peptides, Vol. 3 (Academie Press, 1975), pag. 1724; Bo-30 danszky c.s., Peptide Synthesis, 2nd ed. (Wiley & Sons, 1976), pag. 49-56; Greenstein c.s., Chemistry of the Amino Acids, Vol. 2 (Wiley & Sons, 1961), pag. 782 e.v.; J. Chromatog 44, 269 (1969) en de daarin genoemde bronnen. De voorkeur genieten de verbindingen, waarin het proline-gedeelte van het molecuul in de L-vorm is. * <* \ \ optreden. In dit geval genieten de isomere D-vorm de voorkeur.
De uitgangsmaterialen met de formule 2 kunnen onder toepassing van variërende methodes worden bereid.
Bijvoorbeeld wordt een Grignard-reagens Rj-MgBr 5 in reactie gebracht met een dialkylhalogeenfosfiet met de formule 4, waarin Hal halogeen voorstelt, bij voorkeur chbor of broom, onder vorming van een verbinding met de formule 5. De reactie van deze verbinding met een halogeenacylester met de formule 6 resulteert in de vorming van een verbinding met de formule 7, die vervolgens 10 met waterig zuur, bijvoorbeeld chloorwaterstofzuur, wordt gehydro-lyseerd teneinde deze om te zetten in het fosfinylazijnzuur met de formule 8. Deze laatste verbinding wordt vervolgens veresterd met methanol en acetylchloride of dergelijke en behandeld met benzyl-p-tolyltriazeen onder vorming van de fenylmethylester met de formule 15 9. Anderzijds kan de verbinding met de formule 7 worden behandeld met trimethylsilylbromide en benzyl-p-tolyltriazeen onder vorming van de verbinding met de formule 9. Deze laatste wordt vervolgens gehydrolyseerd met een base, bijvoorbeeld natriumhydroxyde, ter verkrijging van het uitgangsmateriaal met de formule 10.
20 Anderzijds wordt, wanneer Rj lager alkyl of fenyl voorstelt, een alkylfosfinezuur met de formule 11 met fosfor-pentachloride omgezet in het chloride met de formule 12 en vervolgens met een aralkanol, zoals benzylalkohol, in het aralkylderivaat met de formule 13. De behandeling van dit produkt met een lithium-25 dialkylamide, zoals lithiumdiïsopropylamide of een alkyllithium, zoals sec.-butyllithium, en kooldioxyde leidt tot het produkt met de formule 14.
Als verder alternatief kan, wanneer Rj lager alkyl of fenyl-lager alkyl voorstelt, de verbinding met de formule 30 14, waarin Rj methyl is, worden veresterd met diazomethaan of der gelijke onder vorming van een verbinding met de formule 15, welke verbinding vervolgens wordt behandeld met een 1ithiumdialkylamide, zoals lithiumdiïsopropylamide, en een alkylhalogenide of aralkyl-halogenide Rj-hal, waarin Rj lager alkyl of fenyl-lager alkyl is, 35 onder vorming van het produkt met de formule 16. De lagere alkyl-ester kan vervolgens worden omgezet onder toepassing van gebruike- 790 75 68 v- ·$ \ 4 lijke methodes, bijvoorbeeld met natriumhydroxyde ter verkrijging van het vrije zuur.
De uitgangsmaterialen met de formule 3 worden bereid volgens de algemene methodes, beschreven in Zh. Obsh. Kim.
5 37, 411 (1967) en 38, 288 (1968).
Verdere experimentele details zijn aangegeven in de onderstaande voorbeelden.
De verbindingen volgens de uitvinding zijn angiotensine-omzettend enzym-remmers en zijn bruikbaar als hypoten-10 sieve middelen, in het bijzonder voor de vermindering van met renine-angiotensine in verband staande hypertensie, bijvoorbeeld renovasculaire hypertensie en kwaardaardige hypertensie. Door toe-doening van een preparaat, bevattende een of meer angiotensine-omzettend enzym-remmers. volgens de uitvinding, aan een hypertensief 15 zoogdier wordt ingegrepen in de reactie-volgorde renine -* angioten-sinogeen ^ angiotensine I -* angiotensine II en wordt de hypertensie verminderd of verzacht.
Een enkelvoudige dosis of bij voorkeur 2-4 dagelijkse deeldoses, verschaft op basis van 30-300 mg per kg per 20 dag en in het bijzonder ongeveer 10-100 mg per kg per dag, is geschikt voor het bewerkstelligen van een vermindering in de verhoogde bloeddruk. De dierproeven, beschreven door Engel., Proc. Soc. Exp. Biol. Med. 143, 483 (1973), voorzien in een waardevolle leidraad.
Het preparaat wordt bij voorkeur subcutaan, 25 intramusculair, intraveneus of intraperitoneaal toegediend, maar kan eveneens oraal worden toegediend in een dosis van 10-1000 mg per kg per dag, bij voorkeur ongeveer 10-100 mg per kg per dag. De verbinding of verbindingen met de formule 1 kunnen worden gerecepteerd als tabletten, capsules of elixers voor orale toediening. Steriele 30 oplossingen of suspensies kunnen worden gebruikt voor parenterale toediening.
Ongeveer 10-500 mg van een of meer verbindingen met de formule 1 kunnen worden gemengd met een fysiologisch aanvaardbaar vehikel, dragermateriaal, excipiëns, bindmiddel, conserveer-35 middel, stabiliseermiddel, smaakstof, etc., onder vorming van een gebruikelijke eenheidsdoseringsvorm zoals wenselijk is in de farma- 790 7 5 68 tr 4 * 5 \ ceutische praktijk. De hoeveelheid actieve stof wordt zodanig gekozen, dat voorzien wordt in een dosering binnen het aangegeven traject.
De uitvinding wordt verder toegelicht maar 5 niet beperkt door de volgende voorbeelden.
Voorbeeld I
/hydroxy(3-fenylpropyl)fosfinyl/azijnzuur.
(a) Magnesiunr-metaal (4,86 g, 0,2 mol) wordt gesuspendeerd in diethylether (100ml) en druppelsgewijze behandeld 10 met een oplossing van 3-broompropylbenzeen (39,8 g, 0,2 mol) in diethylether (100 ml). De toevoeging wordt zodanig ingesteld, dat een kalme terugvloeiing van het reactiemengsel wordt bewerkstelligd.
Nadat de toevoeging is voltooid wordt het reactiemengsel gedurende de nacht bij kamertemperatuur geroerd. Het mengsel wordt afgefil-15 treerd in een stikstofatmosfeer en druppelsgewijze toegfevoegd aan een gekoelde (0°C) oplossing van.diethylchloorfosfiet (31,3 g, 0,02 mol) in diethylether (60 ml) op zodanige wijze, dat de inwendige temperatuur beneden 10°G wordt gehouden. Nadat de toevoeging is voltooid wordt het reactiemengsel gedurende 1 uur onder terugvloei-20 koeling verhit. Het mengsel wordt vervolgens afgekoeld, afgefiltreerd en in vacuo geconcentreerd. Het residu wordt in vacuo gedestilleerd, waarbij 19 g (3-fenylpropyl)fosfonzuur-diethylester worden verkregen; kookpunt = 90-92°C/0,05 mm.
(b) Een mengsel van methylbroomacetaat 25 (9,33 g, 0,06 mol) en (3-fenylpropyl)fosfonzuur-diethylester, (2,16 g, 0,009 mol) wordt op 340°C verhit totdat de destillatie yan ethylbromide begint. Een verdere hoeveelheid van de fosfonzuurester (8, 64g, 0,036 mol) wordt vervolgens druppelsgewijze over een periode van 10 minuten toegevoegd. Nadat de toevoeging is voltooid wordt 30 het reactiemengsel gedurende nog 45 minuten op 140°C verhit. Het reactiemengsel wordt vervolgens afgekoeld tot 100°G en in vacuo geconcentreerd ter verwijdering van de overmaat methylbroomacetaat en niet-gereageerd uitgangsmateriaal. Uit TLC (EtOAc) en NMR blijkt dat het produkt /ethoxy(3-fenylpropyl)fosfinyl/azijnzuur-methyl-35 ester is met een zuiverheid van meer dan 90 %; opbrengst 11,8 g.
Dit materiaal wordt zonder verdere zuivering in de volgende trap 790 75 68 % f l \ 6 gebruikt.
(c) Een mengsel van /ethóxy(3-fenylpropyl)-fosfinyl/azijnzuur-methylester (3 g, 0,0106 mol) in 6N. chloorwater-stofzuur (25 ml) wordt gedurende 6 uren onder terugvloeikoeling ver-5 hit en vervolgens wordt het reactiemengsel in vacuo geconcentreerd. De resulterende vaste stof wordt tweemaal herkristalliseerd uit ethylacetaat-benzeen, waarbij 1,5 g /hydroxy(3-fenylpropyl)fosfi-nyl/azijnzuur worden verkregen; smeltpunt = 118—119°C.
Voorbeeld II
10 /hydroxy(2-fenylethyl)fosfinyl/azijnzuur.
(a) Magnesium-metaal (4,86 g,0,2 mol) wordt gesuspendeerd in diethylether (100 ml) en druppelsgewijze behandeld met een oplossing van 2-broomethylbenzeen (37 g, 0,2 mol) in diethylether (100 ml). De toevoeging'wordt zodanig ingesteld, dat 15 een kalme terugvloeiing van het reactiemengsel wordt bewerkstelligd. Nadat de toevoeging is voltooid wordt: het reactiemengsel gedurende de nacht bij kamertemperatuur geroerd. Het mengsel wordt afgefiltreerd in een stikstofatmosfeer en druppelsgewijze toegevoegd aan een gekoelde (0°C) oplossing van diethylchloorfosfiet (31,3 g, 20 0,2 mol) in diethylether (60 ml) op zodanige wijze, dat de inwen dige temperatuur beneden 10°C wordt gehouden. Nadat de toevoeging is voltooid wordt het reactiemengsel gedurende 1 uur onder terugvloeikoeling verhit. Het mengsel wordt vervolgens afgekoeld, afgefiltreerd en in vacuo geconcentreerd. Het residu wordt in vacuo 25 gedestilleerd, waarbij 20 g (2-fenylethyl)fosfonzuur-diethylester worden verkregen; kookpunt = 90-92oC/0,05 mm.
(b) Een mengsel van methylbroomacetaat (9,33 g, 0,06 mol) en (2-fenylethyl)fosfonzuur-diethylester (2,54 g, 0,011 mol) wordt op 140°C verhit totdat de destillatie van ethylbromide 30 begint. Een verdere hoeveelheid uitgangsmateriaal (7,63 g, 0,034 mol) wordt vervolgens druppelsgewijze over een periode van 10 minuten toegevoegd. Nadat de toevoeging is voltooid wordt het reactiemengsel gedurende nog 1 uur op 140°C verhit. Het reactiemengsel wordt vervolgens afgekoeld tot 100°C en in vacuo geconcentreerd ter ver- 35 wijdering van de overmaat uitgangsmaterialen. Uit TLC (EtOAc) en NMR blijkt dat het produkt een zuiverheid van meer dan 90 % bezit; 790 75 68 % * * 7 opbrengst lig. Het produkt /ethoxy(2-fenylethyl)fosfinyl/azijnzuur-methylester wordt zonder verdere zuivering gebruikt.
(c) Een mengsel van /ethoxy(2-fenylethyl)fosfinyl/-azijnzuur-methylester (3 g, 0,011 mol) in 6N. chloorwaterstofzuur 5 (25 ml) wordt gedurende 6 uren onder terugvloeikoeling verhit en vervolgens in vacuo geconcentreerd. De resulterende vaste stof wordt tweemaal herkristalliseerd uit ethylacetaat-benzeen, waarbij 2,3 g /hydroxy(2-fenylethyl)fosfinyl/azijnzuur worden verkregen; smeltpunt = 120-121°C.
10 Voorbeeld III
Dimethylfosfinezuuf-fenylmethylester.
(a) Een suspensie van tetramethylbifosfine-bisulfide (25 g, 0,134 mol) in tetrachloorkoolstof (150 ml) wordt onder roeren onder terugvloeikoeling verhit en 30 %'s waterstof- 15 peroxyde (46 ml, 0,4 mol) wordt druppelsgewijze toegevoegd (40 minuten). Na de toevoeging wordt de verhitting onder terugvloakoeling gedurende 5 uren voortgezet. Na de afkoeling wordt de waterige laag verwijderd, millipore afgefiltreerd en in vacuo geconcentreerd. Het residu wordt opgelost in onder terugvloakoeling kokende 20 benzeen (1 liter); het in het mengsel achtergebleven water wordt verwijderd door azeotrppische destillatie. Na de filtratie van de hete oplossing en afkoeling worden 19 g (75 %) van het produkt, dimethylfosfinezuur, verkregen; smeltpunt = 82-84°C.
(b) De toevoeging in porties van dimethyl- 25 fosfinezuur (22,6 g, 0,024 mol) aan fosforpentachloride (50 g, 0,24 mol) resulteert in de vorming van een vloeibaar mengsel, welke vorming gepaard gaat met een heftige exotherme reactie. Na de toevoeging wordt het mengsel gedurende 1 uur op 115°C verhit alvorens te worden onderworpen aan een destillatie in vacuo, waarbij 24,3 g 30 (90 %) van een laag smeltende vaste stof, dimethylfosfinylchloride, worden verkregen; kookpunt = 82°C bij 0,5 mm druk.
(c) Aan een gekoelde (0°C) oplossing van dimethylf osf inylchloride (3,4 g, 0,03 mol) in dichloormethaan (50 ml) wordt druppelsgewijze (over een periode van 20 minuten) onder roe- 35 ren een oplossing van benzylalkohol (3,2 g, 0,03 mol) en triethyl-amine (3,4 g, 0,03 mol) in dichloormethaan 00 ml) toegevoegd. Na 790 75 68 i * i v 8 roeren bij 0°C gedurende I uur en bij kamertemperatuur gedurende de nacht wordt het mengsel afgefiltreerd en het filtraat in vacuo geconcentreerd. Het residu (5 g, 90 %) wordt half vast bij kamertemperatuur. Het wordt herkristalliseerd. uit pentaan (450 ml), 5 waarbij dimethylfosfinezuur-fenylmethylester wordt verkregen in een hoeveelheid van 3,6 g (65 %); smeltpunt = 41-45°C. Een tweede herkristallisatie van.0,5 g uit pentaan (75 ml) geeft 0,26 g pro-dukt; smeltpunt = 46-47°C.
Voorbeeld IV
10 (hydroxymethylfosfinyl)azijnzuur.
(a) Een oplossing van 0,0272 mol lithiumdi-isopropylamide in tetrahydrófuran wordt bereid door druppelsgewijze toevoeging van N-butyllithium (12,3 ml van een 2,22 N. hexaan-oplossing, 0,0272 mol) aan diïsopropylamine (5,5 g, 0,0544 mol) in 15 een gekoelde (0°C) oplossing van hexaan (70 ml). Het oplosmiddel wordt in vacuo verwijderd en vervangen door tetrahydrófuran (80 ml). De oplossing wordt afgekoeld tot -76°C en een opbssing van dimethyl-fosfinezuur-fenylmethylester (2,5 g, 0,0136 mol) in tetrahydrófuran (50 ml) wordt toegevoegd over een periode van 3-4 minuten. Na roe-20 ren gedurende 20 minuten wordt droog kooldioxyde in het mengsel geleid gedurende 30 minuten, het koelbad wordt verwijderd en de oplossing bij kamertemperatuur wordt verdund met ether (150 ml). De oplossing wordt geëxtraheerd met water (2 x 60 ml). De waterige fase (pH ongeveer 10) wordt gewassen met ether (25 ml) en aange-25 zuurd tot een pH van 1 met chloorwaterstofzuur. De zure oplossing wordt geëxtraheerd met dichloormethaan (9 x 100 ml). Na wassen met keukenzoutoplossing en drogen boven magnesiumsulfaat wordt de di-chloormethaanoplossing in vacuo geconcentreerd, waarbij een olie (2,4 g) wordt verkregen. De olie wordt opgelost in.dichloormethaan 30 (100 ml). De oplossing wordt geëxtraheerd met verzadigde natrium- hydrogeencarbonaatoplossing (3 x 25 ml). Nadat de alkalische oplossing is gewassen met dichloormethaan (4 x 50 ml) wordt deze aangezuurd tot een pH van 1 met chloorwaterstofzuur. De zure oplossing wordt geëxtraheerd met dichloormethaan (10 x 57 ml). Na wassen met 35 keukenzoutoplossing en drogen boven magnesiumsulfaat wordt het oplosmiddel in vacuo verwijderd, waarbij 2,2 g (84 %) van het produkt 790 7 5 68 % t 9 /methyl (fenylmethoxy)fosfinyl/azijnzuur, worden verkregen. TLC, silicagel, C^C^/MeOH/HOAc (8:1:1) geeft één enkele vlek;
Rf = 0,70.
(b) Een mengsel van /methyl(fenylmethoxy)-5 fosfinyl/azijnzuur (0,5 g, 0,022 mol) en 5 % palladium op koolstof (25 mg) in methanol (50 ml) wordt heftig geroerd onder een waterstof druk van 1 atmosfeer totdat 39 ml waterstof zijn verbruikt. Het mengsel wordt afgefiltreerd door diatomeeénaarde en in vacuo geconcentreerd. Het residu (0,37 g) wordt vast na samenwrijven met 10 pentaan; smeltpunt = 44-48°C. Het wordt herkristalliseerd uit ethyl-acetaat (15 ml), waarbij 0,17 g (57 %) (hydroxymethylfosfinyl)-azijnzuur worden verkregen; smeltpunt = 87-88°C.

15 (a) Een oplossing van /methyl(fenylmethoxy)- fosfinyl/azijnzuur in ethylacetaat-ether wordt behandeld met een overmaat diazomethaan in ether en gedurende 3 uren geroerd. De overmaat diazomethaan wordt ontleed door toevoeging van azijnzuur.
De oplossing wordt gewassen met natriumhydrogeencarbonaat en keuken-20 zoutoplossing, gedroogd boven magnesiumsulfaat en ingedampt, waarbij /methyl(fenylmethoxy)fósfinyl/azijnzuur-methylester wordt verkregen als een heldere olie in een opbrengst van 87 % (TLC: Rf = 0,18 (ethylacetaat)).
(b) Een oplossing van /methyl)fenylmethoxy)-25 fósfinyl/azijnzuur-methylester (1,0 g, 0,004 mol) in 20 ml tetrahydro-furan wordt over een periode van 50 minuten toegevoegd aan een oplossing van lithiumdiïsopropylamide (0,008 mol) in tetrahydrofuran, dat op een temperatuur van -78°C wordt gehouden in een argon-atmos-feer. Na de toevoeging wordt het roeren bij -78°C gedurende 20 minu-30 ten voortgezet. Fenylmethylbromide (0,684 g, 0,004 mol) in tetrahydrofuran (5 ml) wordt vervolgens toegevoegd en het mengsel wordt gedurende 2 uren bij -78°C en gedurende 1 uur bij 0°C geroerd. Het mengsel wordt vervolgens geneutraliseerd tot een pH van 5 met azijnzuur en uitgegoten in ether. De etherlaag wordt gewassen met water, 35 5 %'s kaliumhydrogeensulfaat en keukenzoutoplossing en gedroogd bo ven magnesiumsulfaat. De indamping in vacuo geeft een residu, dat 790 75 68 * ♦ \ \ * 10 wordt gechromatografeerd oyer silicagel .met dichlaormethaan-ethyl-acetaat. Het produkt /(2-fenylethyl)(fenylmethoxy)fosfinyl/azijn-zuur-methylester (0,66 g, 50 %) is blijkens TLC en NMR identiek aan het in voorbeeld VI bereide produkt.

(a) Een oplossing van (1-/hydroxy-(2-fenyl-ethyl)-fosfinyl/azijnzuur (9,1 g, 0,04 mol) en acetylchloride (1 ml) in methanol (100 ml) wordt gedurende de nacht onder terugvloeikoeling 10 verhit. Het reaetiemengsel wordt vervolgens in vacuo geconcentreerd, waarbij 9,73 g produkt, /hydroxy-(2-fenylethyl)fosfinyl/-azijnzuur-methylester, worden verkregen.
(b) Een afgekoelde (0°C) oplossing van 3-ben-zyl-l-p-tolyltriazeen (8,91 g, 0,04 mol) in diethylether (350 ml) 15 wordt in één keer behandeld met een oplossing van /hydroxy(2-fenyl-ethyl)-fosfinyl/azijnzuur-methylester (9,63 g, 0,04 mol) in ethyl-acetaat (15 ml). Het reaetiemengsel. wordt vervolgens gedurende 4 uren bij kamertemperatuur geroerd. De etheroplossing wordt daarna geëxtraheerd met 10 %'s chloorwaterstofzuur en keukenzoutoplos-20 sing, gedroogd boven magnesiumsulfaat en in vacuo geconcentreerd.
Het residu wordt gechromatografeerd op silicagel /silicAR CC-7/ (500 ml) onder elutie met mengsels van hexaan-ethylacetaat, waarbij 6 g produkt, /(2-fenylethyl)(fenylmethoxy)fosfinyl/azijnzuur-methylester, worden verkregen (TLC: silicagel; hexaan-ethylacetaat 25 (1:1); Rf = 0,15; zichtbaarmaking met UV).
(c) Een oplossing van /(2-fenylethyl)-(fenyl-methoxy)-fosfinyl/azijnzuur-methylester (5,62 g, 0,017 mol) en IN. natriumhydroxyde (17,1 ml, 0,017 mol) in methanol (30 ml) wordt gedurende de nacht bij kamertemperatuur geroerd. Het reaetiemengsel 30 wordt in vacuo geconcentreerd. Het residu wordt opgelost in water en gewassen met ether. De waterige laag wordt vervolgens aangezuurd met kaliumhydrogeensulfaatoplossing en verscheidene malen geëxtraheerd met ethylacetaat. De gecombineerde ethylacetaat-extracten worden gedroogd boven magnesiumsulfaat en in vacuo gecon-35 centr’eerd, waarbij 5,6 g produkt, /(2-fenylethyl)-(fenylmethoxy)- fosfinyl/azijnzuur, worden verkregen (elektroforese: 2000 V; 20 mi- 7907 5 68 V * Λ 11 nut en, 0,1 M. NH^HCO^ + 4,5 cm; ëén enkele vlek, zichtbaar gemaakt met carboxyl-reagena).
(d) Carbonyldiïmidazool (2,86 g, 0,018 mol) wordt opgelost in acetonitril (200 ml), afgekoeld in een ijsbad, 5 en behandeld met /(2-fenylethyl)-(fenylmethoxy)-fosfinyl/azijnzuur (5,6 g, 0,018 mol) in acetonitril (15 ml). Het mengsel wordt gedurende 1 uur bij 0°C geroerd. Het mengsel wordt vervolgens in één keer behandeld met een oplossing van L-proline—fenylmethylester /bereid volgens de methode, beschreven in J.Org.Chem. 28, 174 10 (1963)/ (3,6 g, 0,018 mol) in acetonitril (5 ml). Het reactiemeng- sel wordt gedurende 1 uur bij 0°C geroerd, waarna men het gedurende de nacht bij kamertemperatuur laat staan. Het reactiemengsel wordt in vacuo geconcentreerd. Het residu wordt opgelost in ethylacetaat en gewassen met 5 %’s kaliumhydrogeensulfaat en 5 %!s natriumhydro-15 geencarbonaat. De ethylacetaat-laag wordt gedroogd boven natriumr-sulfaat en in vacuo geconcentreerd, waarbij 8 g ruw produkt worden verkregen. Dit produkt wordt gechromatografeerd over silicagel /silicAR CC-7/ (250 ml) onder elutie met mengsels van C^Clg/
EtOAc, waarbij 7,11 g produkt, l-//(2-fenylethyl)-(fenylmethoxy)-20 fosfinyl/acetyl/-L-proline-fenylmethylester, worden verkregen (TLC: silicagel; ethylacetaat; Rf = 0,2; zichtbaarmaking met UV).
(e) Een mengsel van l-//(2-fenylethyl)-(fenyl-methoxy)-fosfinyl/-acetyl/-L-proline-fenylmethylester (7,11 g, 0,014 mol) en 10 %fs palladium op koolstof (800 mg) in absolute 25 ethanol (400 ml) wordt heftig geroerd onder een waterstofdruk van 1 atmosfeer totdat 630 ml waterstof zijn vebruikt. Het reactiemengsel wordt igefiltreerd door Celite (diatomeeënaarde) en in vacuo geconcentreerd. (a) Carbonyldiïmidazool (1,3 g, 0,0079 mol) 35 wordt toegevoegd aan een.afgekoelde oplossing (0°C) van /methyl- (fenylmethoxy)fosfinyl/azijnzuur (1,8 g, 0,0079 mol) in acetonitril 790 7 5 68 * ’*» \ ♦ ' 12 (50 ml) en het mengsel wordt gedurende 1 uur geroerd. Het wordt vervolgens behandeld met een oplossing van L-proline-benzylester (1,61 g, 0,0079 mol) in acetonitril (25 ml). Het reactiemengsel wordt gedurende 1 uur bij 0°C geroerd, waarna men het gedurende de 5 nacht bij kamertemperatuur laat staan. Het reactiemengsel wordt in vacuo geconcentreerd. 15 (b) Een mengsel van l-//methyl(fenylmethoxy)- fosfinyl/-acetyl/-L-proline-fenylmethylester (2,3 g, 0,0052 mol) en 5 %’s palladium op koolstof (100 mg) in absolute methanol (125 ml) wordt heftig geroerd onder een waterstofdruk van. 1 atmosfeer totdat 190 ml waterstof zijn verbruikt. Het reactiemengsel wordt afgefil-20 treerd door diatomeeënaarde en in vacuo geconcentreerd. (a) Methanol (12,8 g, 0,4 mol) en triethyl-amine (40,4 g, 0,4 mol) worden opgelost in 750 ml. ether en afgekoeld 25 in een ijsbad. Fenyldichloorfosfine (35,8 g, 0,2 mol) wordt druppelsgewijze toegevoegd en het reactiemengsel wordt gedurende de nacht geroerd. De resulterende suspensie wordt afgefiltreerd en het filtraat geconcentreerd tot een olie, die in vacuo wordt gedestilleerd, waarbij het produkt, fenylfosfonzuur-dimethylester, 30 wordt verkregen; kookpunt = 100-105°C/10 mm; opbrengst 20 g (59 %).
(b) Fenyldichloorfosfine (8,95 g, 0,05 mol) wordt opgelost in 50 ml ether, afgekoeld in een ijsbad in een stikstof atmosfeer . Fenylfosfonzuur-dimethylester (8,50 g, 0,05 mol) in 20 ml ether wordt druppelsgewijze toegevoegd en het mengsel 35 wordt gedurende 2 uren geroerd. De oplossing wordt in vacuo geconcentreerd en het residu gedestilleerd, waarbij 8,5 g (49 %) fenyl- 790 7 5 68 « 15 fosfonochloridigzuur-methylester worden verkregen; kookpunt = 66^/0,6 mm.
(c) Fenylfosfonochloridigzuur-methylester (8,5 g, 0,049 mol) wordt druppelsgewijze behandeld met acrylzuur 5 (3,4 ml, 0,049 mol) in een stikstofatmosfeer bij een zodanige toe- voegingssnelheid, dat de temperatuur niet boven 60°C stijgt. Nadat de exotherme reactie is verlopen wordt een gedeelte (2,6 g) van het resulterende heldere glasmateriaal onderworpen aan een kogel-buis ("Kugelrohr") destillatie bij 0,005 mm. Wanneer de badtempera-10 tuur een waarde van 160°C bereikt destilleert een vluchtige vloeistof. Een verdere verhitting tot 240°C geeft een hoofdfractie, 2.5-dioxo-2-fenyl-I.2-oxafosfolan, van in totaal 1,35 g (65 %) in de vorm van een glasachtige vaste stof.
(d) L-proline-1.1-dimethylethylester (1,18 g, 15 0,0069 mol) en triethylamine (0,95 ml, 0,0069 mol) wordt opgelost in 30 ml dichloormethaan en afgekoeld in een ijsbad. Het produkt van (c) (1,35 g, 0,0069 mol) in 10 ml dichloormethaan wordt druppelsgewijze toegevoegd en het mengsel wordt gedurende de nacht geroerd.
De resulterende oplossing wordt ingedampt tot een residu, dat wordt 20 opgenomen in water en wordt aangebracht op een 2,5 x 60 cm kolom van AG-50 ionuitwisselaarhars (H+ vorm). Elutie met water geeft een hoofdfractie, die wordt gelyofiliseerd tot een amorfe witte vaste stof, l-/3-(hydroxyfenylfosfinyl)-l-oxopropyl/-L-proline-I.1-dimethylethylester, totale opbrengst 1,40 g (55 %). TLC: Rf = 0,53, 25 silicagel, butanol-azijnzuur-water (3:1:1). Elektroforese: enkelvoudige vlek +3,0 cm, 2000 V, mA. 35, pH 6,5, 30 minuten.
(e) 1-/3-(hydroxyfenylfosfinyl)-1-oxopropyl/-L-proline-1.1-dimethylethylester (1,40 g, 0,0038 mol) wordt opgelost in 20 ml trifluorazijnzuur en 5 ml anisool en gedurende 1 uur 30 geroerd. Het zuur wordt in vacuo verwijderd en het residu wordt verscheidene malen samengewreven met ether-hexaan. Het residu wordt opgenomen in water en de oplossing wordt gewassen met ether, milli-pore afgefiltreerd en gelyofiliseerd. De resulterende schuim wordt gekristalliseerd uit ethylacetaat, waarbij 300 mg (25 %) 1-/3-35 (hydroxyfenylfosfinyl)-l-oxopropyl/-L-proline worden verkregen; smeltpunt = 125-128°C.

(a) Een oplossing van fenylfosfonigzuur-di-methylester (15,6 g, 0,09 mol) en methyljodide (3 ml) in benzeen 5 (75 ml) wordt gedurende 2 uren onder terugvloeikoeling verhit. Het mengsel wordt afgefiltreerd en het filtraat in vacuo geconcentreerd, waarbij een vloeibaar residu van 15,2 g (97 %) wordt verkregen.
Het het NMR-spectrum blijkt de aanwezigheid van twee afzonderlijke doublets, elk equivalent met eën methylgroep. Het produkt, methyl-10 fenylfosfinezuur-methylester, wordt zonder verdere zuivering bij de volgende reactie gebruikt.
(b) Trimethylsilylbromide (15 ml, 0,1 mol) wordt in porties toegevoegd aan methylfenylfosfinezuur-fenethylester (15,2 g, 0,1 mol) met een zodanige snelheid, dat de terugvloei-15 temperatuur wordt gehandhaafd. Na de toevoeging wordt het mengsel gedurende 1 uur bij kamertemperatuur geroerd. De vluchtige materialen worden in vacuo verwijderd. De toevoeging van water (10 ml) aan het vloeibare residu (20,6 g) resulteert in de afscheiding van een witte vaste stof. Het mengsel wordt gedurende de nacht bij ka-20 mertemperatuur geroerd en vervolgens afgefiltreerd, waarbij 14 g vaste stof worden verkregen. Deze wordt herkristalliseerd uit di-chloormethaan (80 ml), waarbij methylfenylfosfinezuur wordt verkregen; 9,9 g (72 %); smeltpunt = 133—135°C.
(c) Een afgekoelde (0°C) oplossing van 3-ben-25 zyl-l-p-tolyltriazeen (12,5 g, 0,056 mol) in diethylether (150 ml) wordt druppelsgewijze over een periode van 45 minuten toegevoegd aan een geroerde suspensie van methylfenylfosfinezuur (8 g, 0,051 mol) in ethylacetaat (250 ml). Na roeren bij kamertemperatuur gedurende 3 uren wordt het mengsel gewassen met verzadigde natrium-30 hydrogeencarbonaatoplossing, 5 %’s kaliumhydrogeensulfaat en keuken- zoutoplossing. Na droging boven magnesiumsulfaat wordt de etherische oplossing in vacuo geconcentreerd, waarbij 14,3 g van een olieachtig residu worden verkregen. Dit wordt gechromatografeerd over silicagel onder elutie met mengsels van dichloormethaan-ethylacetaat, 35 waarbij 9,25 g (74 %) methylfenylfosfinezuur-fenylmethylester worden verkregen. • (d) Een oplossing van 0,033 mol lithiumdiiso-propylamide wordt bereid door druppelsgewijze toevoeging van N-bu-tyllithium (15 ml van een 2,22 N. hexaanoplossing, 0,033 mol) aan 5 diïsopropylamine (4,1 g, 0,067 mol) in een gekoelde (0°C) oplossing van hexaan (70 ml). Het oplosmiddel wordt in vacuo verwijderd en vervangen door tetrahydrofuran (85 ml). De oplossing wordt afgekoeld tot -76°C en een oplossing van methylfenylfosfinezuur-fenylmethyl-ester (4,1 g, 0,0166 mol, 1 equivalent) in tetrahydrofuran (50 ml) 10 wordt toegevoegd over een periode van 5 minuten. Na roeren gedurende 20 minuten wordt droog kooldioxyde gedurende 30 minuten in het mengsel geleid. Het koelbad wordt verwijderd en de oplossing wordt bij kamertemperatuur verdund met ether (100 ml). Het mengsel wordt geëxtraheerd met water (125 ml, in twee porties). De waterige fase 15 (pH 10) wordt gewassen met ether (25 ml) en aangezuurd tot een pH van 1 met chloorwaterstofzuur. Een olie scheidt zich af uit de zure oplossing. Deze wordt geëxtraheerd in dichloormethaan (2 x 100 ml), gewassen met keukenzoutoplossing, gedroogd (MgSO^), afgefiltreerd en het oplosmiddel wordt verwijderd, waarbij het produkt, /fenyl-20 (fenylmethoxy)fosfinyl/-azijnzuur in de vorm van een olie (3,85 g, 77,6 %) wordt verkregen. TLC: silicagel, dichloor-ethylacetaat-azijnzuur (8:1:1), één vlek, Rf = 0,85 (zichtbaarmaking met PMA plus verhitting).
(e) Carbonyldiiunidazool (2,28 g, 0,014 mol) 25 wordt toegevoegd aan een gekoelde oplossing (0°C) van /fenyl(fenyl-methoxy)fosfinyl/azijnzuur (4,2 g, 0,014 mol) in acetonitril (200 ml). Het mengsel wordt gedurende 1 uur geroerd en een oplossing van L-proline-fenylmethylester (2,85 g, 0,014 mol) in acetonitril (20 ml) wordt in één keer toegevoegd. Het mengsel wordt gedurende 30 nog 1 uur bij 0°C geroerd, waarna men het gedurende de nacht bij kamertemperatuur laat staan. Het oplosmiddel wordt in vacuo verwijderd en het residu wordt opgelost in ethylacetaat (600 ml) en gewassen met 5 %’s kaliumhydrogeensulfaat, verzadigd natriumhydro-geencarbonaat en keukenzoutoplossing. Na droging (MgSO^) wordt het 35 oplosmiddel in vacuo verwijderd, waarbij 6,7 g (theorie) ruw produkt worden verkregen. Een totale hoeveelheid van 8 g ruw produkt, 790 7 5 68 18

verkregen bij de boven beschreven procedure, wordt gechromatogra-feerd over silicagel (Baker φφ 5-3405, 60-200' mesh, 1200 ml) onder elutie met mengsels van CH2Cl2/EtOAc, waarbij 7 g produkt, l-//fenyl-(fenylmethoxy)fosfinyl/acetyl/-L-proline-fenylmethylester, in de 5 vorm van een olie (83 %) worden verkregen. TLC: silicagel, ethyl-acetaat, Rf = 0,16, zichtbaarmaking door PMA plus verhitting.
(f) Een mengsel van l-/fenyl(fenylmethoxy)fosfinyl/-acetyl/-L-proline-fenylmethylester (3 g, 0,006 mol) en 5 % palladium op koolstof (100 mg) in absolute methanol wordt heftig geroerd onder 10 een waterstofdruk van 1 atmosfeer totdat 226 ml waterstof zijn opgenomen. Het reactiemengsel wordt afgefiltreerd door diatomeeenaarde en in vacuo geconcentreerd. Het residu wordt opgelost in dubbel gedestilleerd water, millipore gefiltreerd ën gelyofiliseerd, waarbij 1,7 g (94 %) l-/(hydroxyfenylfosfinyl)acetyl/-L-proline als een 15 amorfe vaste stof worden verkregen; / α 7p = -65° (c = 11,5 in methanol).
