Abstract

La presente invencion proporciona un metodo para separar una molecula de acido nucleico de un soporte solido al cual esta enlazada, en donde se incorpora un nucleotido no convencional en un sitio predeterminado en la molecula de acido nucleico, el metodo comprende desdoblar selectivamente la molecula de acido nucleico en el sitio del nucleotido no convencional. La molecula de acido nucleico puede ser una molecula quimerica que comprende un acido nucleico (secuencia de nucleotidos) y otro componente molecular de una naturaleza quimica diferente. Este aspecto de la invencion tambien proporciona una molecula quimerica (o constructo) que comprende una secuencia de enlace nucleotido que comprende un nucleotido no convencional selectivamente desdoblado en un sitio predeterminado, acoplado a un grupo funcional, que puede ser un grupo de enlace de afinidad o un grupo reportero. La invencion encuentra utilidad en un numero de aplicaciones, por ejemplo, procedimientos biologicos moleculares tales como sintesis de acido nucleico, amplificacion o formacion de secuencias, aislamiento de acido nucleico o en separacion basada en afinidad o procedimientos de ensayo.
