Abstract

Los tumores atraen los vasos sanguineos para crecer, mediante un proceso llamado angiogenesis. La cantidad relativa de estimulantes e inhibidores es un factor de determinacion importante para la iniciacion de la angiogenesis. Las trombospondins-1 y -2 son glicoproteinas adhesivas que tienen la habilidad de inhibir la angiogenesis. Esta actividad inhibidora se ha dirigido a las repeticiones del tipo 1 de TSP-1 y TSP-2. La invencion incluye proteinas quimericas que contienen porciones anti-angiogenicas de TSP-1, TSP-2, endostatina, angiostatina, factor 4 de plaqueta o prolactina, enlazada a una porcion de la region N-terminal de la proteina de la matriz oligomerica del cartilago (COMP) que permite la formacion de pentameros. Aqui tambien se describen las moleculas de acido nucleico, vectores y celulas huesped para expresar y producir dichas proteinas quimericas. Otras modalidades de la invencion incluyen metodos para tratar humanos u otros mamiferos con proteinas anti-angiogenicas para reducir el tamaño del tumor o el indice de crecimiento. Ya que la region de repeticion del tipo 1 de TSP-1 y TSP-2 inhibe la infeccion por VIH, las proteinas quimericas que comprenden estas repeticiones tambien pueden utilizarse para este proposito, asi como para inhibir la angiogenesis.
