Abstract
La presente invención comprende nucleasas modificadas genéticamente que reconocen y escinden una secuencia de reconocimiento dentro del primer exón del gen de la región constante alfa del receptor de linfocitos T (TCR) humanos. Las meganucleasas modificadas genéticamente pueden presentar al menos una característica optimizada, tal como mayor (es decir, aumentada) especificidad o eficiencia de escisión, en comparación con la meganucleasa de primera generación TRC 1-2x87EE. La presente invención también comprende métodos para usar dichas nucleasas modificadas genéticamente para producir células modificadas genéticamente, y el uso de dichas células en una composición farmacéutica y en métodos para el tratamiento contra enfermedades, tales como cáncer.
