Abstract
Un producto y proceso para extender la capacidad replicativa de las células somáticas metazoas usando enmiendas genéticas dirigidas para anular la inhibición de la progresión del ciclo celular durante la senescencia replicativa y derivar líneas celulares clonales para aplicaciones escalables y producción industrial de biomasa de células metazoas. Se crea una mutación de inserción o deleción utilizando ARN guía dirigidos al primer exón de la transcripción que codifica cada proteína usando CRISPR/Cas9. Las enmiendas dirigidas dan como resultado la inactivación de las proteínas p15 y p16, lo que aumenta la capacidad proliferativa de las poblaciones de células modificadas en relación con sus poblaciones parentales no alteradas. La combinación de estas enmiendas con la actividad de la telomerasa auxiliar de una construcción genética que dirige la expresión de un homólogo de la proteína de la telomerasa a partir de un gen TERT, aumenta la capacidad replicativa de las poblaciones de células modificadas de forma indefinida. Una aplicación es fabricar músculo esquelético para consumo dietético usando células de la especie aviar Gallus gallus; y otro es de la especie de ganado Bos taurus.
