Abstract
Se proporcionan un anticuerpo dirigido a B7H4 o una variante del mismo, y un anticuerpo biespecífico que contiene el anticuerpo dirigido a B7H4, en donde el anticuerpo dirigido a B7H4 comprende una región variable de cadena ligera y una región variable de cadena pesada. La variante tiene una mutación de aminoácidos en la región variable de la cadena ligera o la región variable de la cadena pesada del anticuerpo y puede mantener la función del anticuerpo. El anticuerpo dirigido a B7H4 tiene la actividad de unirse a B7H4 humano y B7H4 de macaco cynomolgus, y no presenta reacción cruzada con otros miembros de la familia B7. El anticuerpo dirigido a B7H4 muestra una buena actividad antitumoral, actividad de activación de células T y actividad de internalización en experimentos in vivo e in vitro, y es más adecuado para usarse como un fármaco ADC. Además, se proporciona un anticuerpo biespecífico dirigido a B7H4 y CD3, que tiene una vida media más larga, conserva una buena estabilidad e hidrofilia, y reduce la toxicidad al tiempo que garantiza la eficacia.
