Abstract

La invención se refiere a una molécula de ácido nucleico que codifica al menos un polipéptido rep del AAV, en donde uno o más de los promotores p5, p19 y p40 de AAV se han modificado para reducir o eliminar la expresión de uno o más de los polipéptidos Rep, o la molécula de ácido nucleico no codifica polipéptidos Rep52 o Rep40 funcionales, o la molécula de ácido nucleico no codifica una secuencia inhibidora de adenovirus funcional. La invención también se refiere a un proceso para producir vectores AAV recombinantes mediante el uso de un sistema de 2 adenovirus, en donde todos los genes necesarios para la replicación y el empaquetamiento de AAV (es decir, una secuencia de rep del AAV de la invención, cap del AAV y el vector de transferencia de AAV que comprende un transgén) pueden estar codificados dentro de dos adenovirus.
