Abstract
La presente divulgación proporciona un plásmido de ADN que comprende una secuencia que codifica una molécula de ARNm y una cola de poli(A) modificada, en el que la parte de la secuencia que codifica la cola de poli(A) modificada se caracteriza porque ésta consiste de al menos dos elementos A cada uno definido como una secuencia de nucleótidos que consiste en de 55 a 65 nucleótidos T y al menos un elemento S cada elemento S consiste en un nucleótido que no es un nucleótido T, o 2 a 10 nucleótidos, preferiblemente 6 nucleótidos, en el que cada uno de los dos nucleótidos terminales no es un nucleótido T, en el que el número de elementos A es uno más que el número de elementos S, y en el que dos elementos A cualesquiera están separados por un elemento S; exhibiendo dicho plásmido de ADN una recombinación reducida durante la amplificación en una célula huésped bacteriana en comparación con el mismo plásmido de ADN sin dicho al menos un elemento S.
