Abstract
Se proporcionan métodos y composiciones para la integración de secuencias de ADN donante en el genoma de una célula de Bacillus sp. sin la integración de un marcador seleccionable en el genoma. Los métodos emplean un constructo de ADN recombinante lineal que comprende un ADN donante flanqueado por brazos de homología largos (cada uno de al menos 1000 nucleótidos de longitud) en combinación con un constructo de ADN recombinante que codifica una endonucleasa Cas9 y un ARN guía, para la introducción de un sistema de ARN guía/endonucleasa Cas (RGEN) en una célula de Bacillus sp., y como tal proporciona un sistema altamente eficaz para la integración de secuencias de ADN donante en el genoma de la célula de Bacillus sp., sin la necesidad de integrar un marcador seleccionable en el genoma de la célula de Bacillus sp.
