Abstract
La invención se relaciona con un sistema de expresión para la producción de uno o más polipéptidos objetivo/polipéptido objetivo, que comprende una célula huésped, en cuyo genoma la secuencia de AND que codifica la dehidrogenasa de glicerina-3-fosfato es inactivada o parcialmente o completamente suprimida y la cual es transformada por un elemento extracromosomal que comprende una secuencia de AND que codifica el polipéptido objetivo y la dehidrogenasa de glicerina-3-fosfato, en donde no solo el genoma de la célula huésped sino también el elemento extracromosomal no portan un gen de resistencia al antibiótico, así como una secuencia de ADN que codifica un polipéptido con actividad de dehidrogenasa de glicerina-3-fosfato caracterizada porque la secuencia de ADN es seleccionada de a) secuencias de ADN que comprenden una secuencia de nucleótido de conformidad con SEQ ID NO: 1, b) secuencias de ADN que comprende una secuencia de nucleótido representada por los nucleótidos 1338 a 2375 de SEQ ID NO: 1, c) secuencias de ADN que son codificadas por el plásmido pTP01 con el mapa de plásmido de conformidad con la Figura 4 y depositadas bajo el número de deposición DSM 18890, d) secuencias de ADN que codifican la secuencia de proteína de conformidad con SEQ ID NO: 2, e) secuencias de ADN que hibridizan bajo condiciones estrictas con una de las secuencias de ADN de conformidad con a), b), c) o d), f) secuencias de ADN que están relacionadas con las secuencias de nucleótido/ADN de conformidad con a), b), c), d) o e) debido a la degeneración del código genético, y g) filamentos complementarios a las secuencias de conformidad con a) a f), y la proteína codificada por lo mismo.
