Abstract

Invenţia se referă la biotehnologie, în particular la un procedeu de obţinere a somaclonelor de varză.Procedeul include infectarea mecanică a plantelor în faza de două frunzuliţe cotiledonate cu virusul mozaicul conopidei, colectarea explantelor de ţesut mezofil din sectoarele învecinate zonelor clorotice, sterilizarea lor, inocularea explantelor pe mediul nutritiv Murashige şi Skoog, ce conţine agar 7 g/l, chinetină 0,01 mg/l şi suplimentar 6-benzilaminopurină 0,5 mg/l, acid naftilacetic 0,5 mg/l, cu incubarea ulterioară a lor la întuneric la temperatura de 25°C pentru iniţierea formării calusului morfogen, apoi cu o fotoperioadă de 16 ore la iluminare de 2000 lx şi temperatura de 27°C, transferarea calusurilor morfogene pe medii nutritive proaspete cu obţinerea somaclonelor şi înrădăcinarea ulterioară a lor pe mediul nutritiv Murashige şi Skoog, ce conţine suplimentar acid naftilacetic 0,5 mg/l cu adaptarea ulterioară la condiţii ex-vitro şi plantarea în sol a somaclonelor.Rezultatul invenţiei constă în sporirea variabilităţii somaclonale la varză şi în posibilitatea selecţiei somaclonelor care posedă caractere cantitative, nespecifice pentru soiul iniţial.
