Claims (38)
1. Ein Verfahren zum Inhibieren von Long Terminal Repeat Transposable Elements (LTR- TEs) während der Vermehrung einer Pflanze, wobei das Verfahren die Vermehrung der Pflanze in Gegenwart eines Reverse-Transkriptase-Inhibitors umfasst.
2. Verfahren nach Anspruch 1, wobei die Pflanze durch Pflanzengewebekultur vermehrt wird, umfassend das Kultivieren eines anfanglichen Pflanzengewebes in einem Kulturmedium, das einen Reverse-Transkriptase-Inhibitor umfasst, um das kultivierte Pflanzengewebe zu erhalten, wobei das Inhibieren von LTR-TEs zu einer Verringerung der somaklonalen Variation führt kultiviertes Pflanzengewebe
3. Ein Verfahren zur Verringerung der somaklonalen Variation in kultiviertem Pflanzengewebe, wobei das Verfahren einen Pflanzengewebekultur-Vermehrungsschritt umfasst, der das Kultivieren eines anfänglichen Pflanzengewebes in einem Kulturmedium umfasst, das einen Reverse-Transkriptase-Inhibitor umfasst, um das kultivierte Pflanzengewebe zu erhalten.
4. Verfahren nach Anspruch 2 oder Anspruch 3, wobei der Schritt der Pflanzengewebekultur eine Mikrovermehrung ist.
5. Verfahren nach einem der Ansprüche 2 bis 4, wobei das anfängliche Pflanzengewebe ausgewählt ist aus einem Samen, einem Embryoid, einem Embryo, einem Organ, einem Explantat, Pollen oder einem Staubbeutel, der optional ausgewählt ist aus Material von Sprossen. Stängel, Knollen, Wurzeln, Blüten oder Blatter, eine Zelle, ein Protoplast, ein Sämling, ein Pflänzchen, Kallus, der gegebenenfalls ein Spross und/oder bewurzelter Kallus ist.
6. Verfahren nach einem der Ansprüche 2 bis 5, wobei das anfängliche Pflanzengewebe eine genetische Modifikation umfasst.
7. Verfahren nach Anspruch 6, wobei das Verfahren den Schritt des Einführens einer genetischen Modifikation in das anfängliche Pflanzengewebe vor dem Schritt der Pflanzengewebekultur umfasst.

Verfahren nach Anspruch 6 oder Anspruch 7, wobei die genetische Modifikation ausgewählt ist aus der Liste bestehend aus: (i) Einführen eines zum ursprünglichen Pflanzengewebe heterologen Gens; (il) Gen-Editierung eines nativen Gens im ursprünglichen Pflanzengewebe; (iii) Silencing eines nativen Gens im anfänglichen Pflanzengewebe, gegebenenfalls durch Einführung eines Silencing-Mittels.

Verfahren nach Anspruch 7 oder Anspruch 8, wobei das Verfahren umfasst: (i) Einbringen einer genetischen Veränderung in ursprüngliches Pflanzengewebe, (ii) optional einen Selektionsschritt zum Selektieren von anfänglichem Pflanzengewebe, das die gewünschte genetische Modifikation umfasst, (ii) einen Gewebekulturschritt, umfassend das Kultivieren des anfänglichen Pflanzengewebes, das die gewünschte genetische Modifikation umfasst, in einem 1
BL-5585 Kulturmedium, das einen Reverse-Transkriptase-Inhibitor umfasst, um ein kultiviertes LU503053 Pflanzengewebe zu erhalten, das die gewünschte genetische Modifikation umfasst.
10. Verfahren nach einem der Ansprüche 2 bis 9, wobei der nukleotidanaloge Reverse- Transkriptase-Inhibitor in dem Kulturmedium in einer Konzentration von 1-50 uM vorhanden ist.
11. Verfahren nach einem der Ansprüche 2 bis 10, wobei das Kulturmedium 1/2 MS-Medium ist, das 1 % Saccharose umfasst.
12. Verfahren nach einem der Ansprüche 2 bis 11, wobei der Gewebekulturschritt bei einer Temperatur von 19-23°C durchgeführt wird.
13. Verfahren nach einem der Ansprüche 2 bis 12, wobei der Schritt der Pflanzengewebekultur ein Kallus-Induktionsschritt ist und das Kulturmedium ein Kallus-Induktionsmedium ist und das kultivierte Pflanzengewebe Kallus ist und optional das anfängliche Pflanzengewebe ist ein Samen, der optional Reis ist.
Verfahren nach einem der Ansprüche 2 bis 13, wobei das Verfahren ferner einen Kallus- Induktionsschritt vor dem Pflanzengewebe-Kultivierungsschritt umfasst, umfassend das Kultivieren von Pflanzenmaterial in …—Kallus-Induktionsmedium, um anfängliches Pflanzengewebe bereitzustellen, das Kallus ist.
15. Verfahren nach Anspruch 14, wobei das Kallus-Induktionsmedium einen Reverse- Transkriptase-Inhibitor umfasst.
16. Verfahren nach einem der Ansprüche 13 bis 15, wobei das Kallus-Induktionsmedium N6D- Medium ist.
17. Verfahren nach einem der Ansprüche 13 bis 16, wobei der Reverse-Transkriptase- Inhibitor in dem Kallus-Induktionsmedium in einer Konzentration von 20-40 uM vorhanden ist.
Verfahren nach einem der Ansprüche 13 bis 17, wobei der Kallus-Induktionsschritt umfasst: (i) Kultivierung im Kallus-Induktionsmedium für 7 — 8 Wochen und/oder (ii) Inkubation des Kallus-Induktionsmediums bei 24 bis 28 °C und/oder (iii) Inkubation des Kallus-Induktionsmediums im Dunkeln.
19. Verfahren nach einem der Ansprüche 13 bis 18, wobei das Verfahren einen Spross- induzierenden Schritt umfasst, der das Kultivieren des Kallus in einem Sprossmedium umfasst. 2

Verfahren nach Anspruch 19, wobei entweder: (i) das kultivierte Pflanzengewebe ein Kallus ist, und das Verfahren ferner den Spross- induzierenden Schritt umfasst, und das Sprossmedium optional einen Reverse-Transkriptase- Inhibitor umfasst, oder (ii) das anfängliche Pflanzengewebe ein Kallus ist und der Pflanzengewebe-Kulturschritt ein Spross-induzierender Schritt ist und das Kulturmedium ein Sprossmedium ist.
21. Verfahren nach einem der Ansprüche 13 bis 20, wobei das Verfahren einen Bewurzelungsschritt umfasst, der das Kultivieren des Kallus in einem Bewurzelungsmedium umfasst.
22. Verfahren nach Anspruch 21, wobei entweder: () kultiviertes Pflanzengewebe ist ein Kallus, und das Verfahren umfasst ferner den Bewurzelungsschritt, und das Bewurzelungsmedium umfasst optional einen Reverse- Transkriptase-Inhibitor, oder (ii) das anfängliche Pflanzengewebe ein Kallus ist und der Pflanzengewebekulturschritt ein Bewurzelungsschritt ist und das Kulturmedium ein Bewurzelungsmedium ist.
23. Verfahren zur Herstellung einer Pflanze, wobei das Verfahren die Schritte umfasst: (a) Durchführen eines Verfahrens nach einem der Ansprüche 2 bis 22, und (b) Regenerieren einer Pflanze aus dem kultivierten Pflanzengewebe, optional durch Übertragen des kultivierten Pflanzengewebes auf Wachstumsmedien oder Erde zur Vermehrung.
24. Verfahren nach Anspruch 1, wobei die Pflanze aus einem gekeimten Samen vermehrt wird.
25. Verfahren nach Anspruch 1, wobei die Pflanze asexuell vermehrt wird
26. Verfahren nach Anspruch 25, wobei die Pflanze durch vegetative Vermehrung vermehrt wird
27. Verfahren nach Anspruch 25, wobei die Pflanze durch Pfropfen vermehrt wird
28. Verfahren nach einem der Ansprüche 25 bis 27, wobei das Hemmen von LTR-TEs die somaklonale Variation in der Pflanze verringert.
29. Verfahren nach Anspruch 24 oder Anspruch 25, wobei die Vermehrung in vitro in einem Vermehrungsmedium erfolgt, das den Reverse-Transkriptase-Inhibitor umfasst. 3

30. Verfahren nach einem der Ansprüche 24 bis 28, wobei die Vermehrung in einer Umgebung LU503053 erfolgt, die gegebenenfalls Erde ist, und der Reverse-Transkriptase-Inhibitor auf die Pflanze durch Wurzeltrankung oder durch Besprihen der Blatter aufgebracht wird.
31. Verfahren nach einem der Ansprûche 1 bis 30, wobei der Reverse-Transkriptase-Inhibitor ein nukleotidanaloger Reverse-Transkriptase-Inhibitor ist.
32. Verfahren nach Anspruch 31, wobei der nukleotidanaloge Reverse-Transkriptase-Inhibitor Tenofovir oder ein Analogon oder Prodrug davon ist.
33. Verfahren nach Anspruch 32, wobei der nukleotidanaloge Reverse-Transkriptase-Inhibitor Tenofovirdisoproxil oder ein Salz davon ist.
34. Verfahren nach einem der Ansprüche 1 bis 33, wobei die Pflanze oder das Pflanzengewebe Arabidopsis, Reis, Tabak, SüBkartoffel, Karotte, Medicago, Bambus oder Dattelpalme ist.
35. Gezüchtetes Pflanzengewebe oder vermehrte Pflanze, erhalten oder erhältlich durch das Verfahren nach einem der Ansprüche 1 bis 34.
36. Pflanzenvermehrungsmedium, Pflanzengewebekulturmedium, Kallusinduktionsmedium, Sprossmedium oder Bewurzelungsmedium, wobei das Medium einen Reverse-Transkriptase- Inhibitor umfasst und das optional zur Verwendung in einem Verfahren nach einem der Ansprüche angepasst ist 1 bis 34, oder das wie in einem Verfahren nach einem der Ansprüche 1 bis 34 definiert ist.
37. Verwendung eines Reverse-Transkriptase-Inhibitors in einem Verfahren zur Pflanzengewebekultur-Pflanzenvermehrung, das optional ein Verfahren nach einem der Ansprüche 1 bis 34 ist.
38. Verfahren zur Bewertung der LTR-TE-Transposition, umfassend den Vergleich der LTR- TE-Transpositionsrate zwischen kultiviertem Pflanzengewebe, das auf Medien kultiviert wurde, die einen Reverse-Transkriptase-Inhibitor umfassen, mit kultiviertem Pflanzengewebe, das auf Medien kultiviert wurde, die nicht umfassen: a Reverse-Transkriptase-Hemmer. 4
