Abstract

본 발명은 미생물로부터 생산된 인지질 가수분해 효소를 이용하여 인지질증의 폴라스말로젠(Plasmalogen)을 고순도로 종축, 정제하는 방법에 관한 것으로, 더욱 상세히는 인지질증에 함유되어 있는 플라스말로젠을 분리하기 위해 세라시아종(Serratia)MKL으로부터 생산된 포스포리파제(Phosphlipse)Al을 이용하여 인지질증에 함유된 플라스말로젠 이외의 인지질을 가수분해하여 제거함에 의해 고순도의 플라스말로젠을 생산하는 방법에 관한 것이다.

효소를 이용한 고순도 인지질의 생산방법
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제 1도는 소심방에서 추출ㆍ정제하여 반응기질로 이용한 콜린 인지질의 HPLC 크로마토그램이다.
제 2도는 소심장에서 추출한 콜린 인지질증 플라스말로젠 함량을 조사한 HPLC 크로마토그램으로, 반응기질로 이용된 콜린 인지질을 10분간 염산가스로 처리후의 HPLC크로마토그램이다.

Claims (7)

  1. 동물에 존재하는 인지질을 전처리하여 분리된 콜린 인지질과 에탄올아민 인지질에 포스포리파제 Al을 반응시켜 다아실형 인지질을 가수분해하여 제거하고 플라스말로젠만 정제 농축시킴을 특징으로 하는 고순도 플라스말로젠의 제조방법.
  2. 제 1항에 있어서, 동물에 존재하는 인지질의 전처리는 클로로포름과 메탄을 혼합 용액으로 추출하고 균질 화한후, 아세톤등을 첨가하여 중성지질을 제거하고, 생성된 조인지질을 실리카겔이 들어있는 칼럼에 통과시켜 콜린 인지질 또는 에탄올아민 인지질을 용출시킨 후 이를 콜린 플라스말로젠과 에탄올아민 플라스말로젠의 분리에 이용함을 특징으로 하는 고순도 플라스말로젠의 제조방법.
  3. 제 2항에 있어서, 분리된 콜린 인지질과 에탄올아민 인지질을 포스포리파제 Al과 가수분해 반응시, 5~100mM의 칼슘이온 또는 칼슘염이 함유된 수용액 상에서 반응시키거나 물과 유기용매의 혼합용액으로 반응시킴을 특징으로 하는 고순도 플라스말로젠의 제조방법.
  4. 제 3항에 있어서, 유기용매는 에틸 아세테이트, 부틸 아세테이트, 이소프로필 에테르, 플로로포름, 부틸 에테르, 시클로핵산, 핵산, 헵탄, 이소옥탄, 에틸알코올, 데칸, 도데칸에서 선택된 1종 이상의 유기 용매임을 특징으로 하는 고순도 플라스말로젠의 제조방법.
  5. 제 1항에 있어서, 포스포리파제 Al은 토앙으로부터 스크리닝한 세라시아 속(Serratia spp)MKl 균주를 배양 배지에 접종하여 28~32℃에서 13~15시간 동안 흔들어 주면서 플라스크 배양시킨 후, 5,600 x g에서 18~22분간 원심분리한 후 미생물을 제거하여 얻어진 포스포리파제 Al임을 특징으로 하는 고순도 플라스말로젠의 제조방법.
  6. 제 5항에 있어서, 균주의 배양 배지는 NaHPO4ㆍ12H2O 14~16g, KH2PO42.5~3.5g, NaCl 0.4~0.6g, MgSO4ㆍ7H2O 0.2~0.3g, CaCl20.01~0.02g을 물 1L에 녹인 배지에 자일로스(xylose) 4~6g/L와 황산암모늄 0.9~1.1g을 첨가한 배지임을 특징으로 하는 고순도 플라스말로젠의 제조방법.
  7. 제 1항 내지 제 6항중 어느 한 항의 방법에 따라 제조된 고순도 플라스말로젠.
