Abstract
재구성한 혈장칼리크레인·키닌계를 이용한 피검물질의 활성측정법을 제공한다.
재구성한 혈장칼리크레인·키닌계에 있어서 피검물질의 존재하, 혈액응고제 XⅡ인자의 활성화를 초발반응으로 하는 일련의 효소반응을 개시시킨 후, 이 반응을 정지시키고 상기한 반응에서 생성된 생리활성물질을 정량한다.
본 발명의 활성측정법에 있어서는 복잡한 그 외 인자를 실질적으로 제거한 반응계에 의해서, 생성되는 생리활성물질의 생성에 대한 피검물질의 순수한 활성(촉진능 또는 저해능)을 간단신속하고 또한 정확하게 측정할 수 있다. 따라서 본 발명의 활성측정법은 혈장칼리크레인·키닌계, 혈액응고계, 선용계 등의 생체기능의 조정에 유용한 작용을 알고 있고, 또는 이와 같은 약제를 개발하는데 매우 유효한 수단이 된다.

제1도는 본 발명의 실시예1 및 실시예2에 있어서, 각종 인큐베이션시간에 있어서의 흡광도의 변화를 나타낸다.
제2도는 본 발명의 활성측정법을 사용하여 각종 피검약의 혈장칼리크레인생성에 대한 저해활성을 측정한 결과를 나타낸다.
제3도는 혈장칼리크레인·키닌계의 효소반응기구를 설명한 것이다.
Claims (7)
- 재구성한 혈장칼리크레인·키닌계에 있어서 피검물질의 존재하, 혈액응고제 XⅡ인자의 활성화를 초발반응으로 하는 일련의 효소반응을 개시시킨 후, 이 반응을 정지시키고 상기한 반응에서 생성된 생리활성물질을 정량하는 것을 특징으로 하는 피검물질의 활성측정법.
- 제1항에 있어서, 생리활성물질의 생성량이 포화에 달하기 전에 그 생리활성물질의 생성을 정지시키는 피검물질의 활성측정법.
- 제2항에 있어서, 생리활성물질이 활성형 혈액응고제 XⅡ인자인 피검물질의 활성측정법.
- 제2항에 있어서, 생리활성물질이 혈장칼리크레인인 피검물질의 활성측정법.
- 제2항에 있어서, 생리활성물질이 브랜디키닌인 피검물질의 활성측정법.
- 제1항 내지 제4항중 어느 한 항에 있어서, 혈액응고제 XⅡ인자, 혈장프레칼리크레인으로 재구성한 계인 피검물질의 활성측정법.
