포스폰산 유도체의 제조방법
[발명의 상세한 설명]
본 발명은 하기 일반식(I)의 화합물 및 이의 염(R 및/또는 R1이 히드록시인 경우)의 제조방법에 관한 것이다.
상기식에서, R과 R1은 각각 서로 독립적으로 C1-C4알킬, C1-C4알콕시 또는 히드록시이며 ; X는 수소, 할로겐, C1-C4알킬, C1-C4알콕시, 트리메틸실릴, 시아노, 메톡시카보닐 또는 -CH2-CH(NH2)-P-(O)(R)(R1)라디칼이고 ; Y는 수소, 할로겐, C1-C2할로알콕시 또는 C1-C2할로알킬이다.
본 발명은 적어도 하나의 활성성분으로서, 일반식(I)의 1-아미노-2-아릴에탄포스폰산 유도체 또는 1-아미노-2-아릴에탄 포스핀산 유도체 또는 1-아미노-2-아릴에탄포스핀 옥사이드 유도체 및 이의 염을 함유하는 살균제 및 유해한 미생물에 제어시키는 이러한 화합물의 용도에 관한 것이다.
일반식(I)은 R과 R1이 히드록시 또는 알콕시인 경우 에탄포스폰산 유도체를 나타내고, 두 치환체 R과 R1중 하나가 알킬인 경우 에탄포스핀산 유도체를 나타내며, 치환체 R과 R1모두가 알킬인 경우 엔탄포스핀 옥사이드 유도체를 나타낸다.
알콕시그룹의 잔기로서 또는 그 자체로서의 C1-C4알킬은 메틸, 에틸, 프로필, 이소프로필, n-부틸, 2급-부틸, 이소부틸 또는 3급-부틸이다.
할로겐은 불소, 염소, 브롬 또는 요오드이다. 할로알콕시그룹의 잔기로서 또는 그 자체로서의 C1-C2할로알킬은 단일할로겐화된 메틸 또는 에틸그룹이거나, 특이한 할로겐원자에 의해서나 상이한 할로겐원자들에 의해 과(per)할로겐화된 메틸 또는 에틸그룹이다. 전형적인 예로는 CHCl2, CHF2, CH2Cl, CCl3, CH2F, CHBr2, CH2CH2Cl, CHCl-CHCl, CF3, C2F5, CF2Cl, CF2-CF2Cl 이다.

일반식(I)의 포스폰산유도체는 비치환되거나 치환된 2-페닐아세트알데히드를 암모니아 및 디알킬 포스파이트와 반응시키고 임의로 무시간을 사용하여 가수분해시켜 수득할 수 있다 :

무기산은 바람직하게는 염산 또는 브롬화수소산과 같은 하이드로할산 ; 황산, 인산, 질산, 톨루엔설폰산 등일 것이다.
더 일반적인 방법이 문헌[참조 : Berlin 등, J. Org. Chem. 33, 3090(1968)]에 제시되는데, 여기서, 치환된 페닐 아세틸할라이드를 a) O,O,O-트리알킬포스파이트, b) O,O-트리알킬포스포니트 또는 c) O-트리알킬포스피니트와 반응시키고 중간체를 히드록실라민의 존재하에서 추가로 반응시켜 포스포닐, 포스피닐 또는 옥소포스피노-옥심을 수득하고 이것을 수소첨가 반응시켜 아민을 수득할 수 있다 :

일반식(I*)은 R과 R1이 R*및 R1 *인 일반식(I)의 화합물만을 포함한다. R*및/또는 R1 *이 C1-C4알콕시인 경우, 바람직하게는 무기산을 사용하여 상기 화합물을 가수분해시켜 유리 히드록실 그룹을 수득할 수 있다. 필요한 경우, 유리 OH 그룹은 염기를 사용하여 염(예 : 금속염, 암모늄염, 알킬암모늄염, 디알킬 암모늄염 등)으로 전환시킬 수 있다.
일반식(I)의 화합물은 a) 아미노-메틸포스포네이트, 아미노메틸포스피네이트 또는 아미노메틸포스핀옥사이드, 및 b) 알데히드(예 : 벤즈알데히드) 또는 케톤(예 : 페닐아세톤 또는 벤조페논)으로부터 수득한 활성화된 쉬프 염기를 적합하게 치환된 벤질 할라이드로 아르알킬화시키는 추가의 방법에 의해 간단히 높은 수율로 수득할 수 있다[참조 : R. W. Radcliffe 및 B. G. Christensen, Tetrahedron Letters, 4645 (1973)] :

일킬화(벤진화)시키는 동안의 반응온도는 통상 -100 내지 +10℃, 바람직하게는 -80 내지 -20℃ 이다. 가수소분해는 통상 -20 내지 +80℃에서 실시한다.
인용한 출발 화합물은 자체에 공지되어 있거나 공지된 방법으로 제조한다. 쉬프 염기를 제조하기 위해서는, 질소 또는 아르곤과 같은 불활성 기체의 기류하에 공정을 수행하는 것이 유리하다. 치환된 벤질 브로마이드는 적합하게 치환된 톨루엔을 아조이소부틸니트릴(촉매)의 존재하에서 CCl4중의 N-브로모 석신이미드로 브롬화시켜 용이하게 수득할 수 있다.
일반식(I)의 화합물은 β-페닐알라닌의 포스포닐, 포스피닐 및 옥소포스피노 동족체이다. 이들은 P-원자에 인접한 비대칭 탄소원자를 가지므로 천연의 L-α-아미노산에 상응하는 구조는 (R)-배위인 반면, D-α-아미노산에 상응하는 일반식(I)의 구조는 (S)-배위이다. 부분입체이성체 (R,S)-화합물 각각의 (R)-또는 (S)-배위는 분별결정이나 크로마토그래피(HPLC)에 의해 순수한 것을 수득한다 (R)-부분 입체이성체와 (S)-부분입체이성체는 상이한 살균성을 가지고 있다.
본 발명은 또한 신규 화합물을 제조하는 상기의 제조 방법에 관한 것이다. 본 발명은 또한 R과 R1이 동시에 히드록시 또는 에톡시라는 조건으로 치환체 X 및 Y중 적어도 하나가 수소가 아닌 일반식(I)의 신규 화합물에 관한 것이다.
R 및 R1이 C1-C4알콕시인 일반식(I)의 화합물은 유용한 진드기 구충제[특히 테트라니쿠스(Tetranychus) 및 앰블리요마(Amblyomma)중]일 뿐만 아니라, 특히 흡층에 대해 효과적인 살중체(아피사이드)이다.
바람직한 살균제는 R이 C1-C4알킬 또는 OH이고 ; R1은 OH이며 ; X는 수소, 할로겐, C1-C4알킬, C1-C4알콕시, Si(CH3)3, -CN, -COOCH3또는 CH2-CH(NH2)-P(O)(R)(R1)이고 ; Y는 수소, 할로겐, 할로메톡시, 할로에톡시 또는 할로메틸인 일반식(I)의 화합물 및 이의 염이다.
상기 살균제중에서 R=R1=OH이고; X는 수소, 불소, 염소 또는 브롬, C1-C4알킬, CH3O, Si(CH3)3, -CN, -COOCH3또는 CH2-CH(NH2)-P(O)(OH2) 이며 ; Y는 불소, 염소, 브롬, 할로메톡시 또는 할로메틸인 화합물이 바람직하다.
마지막으로 언급한 살균제중에서 치환체 X 및 Y중 적어도 하나가 할로겐인 화합물이 특히 바람직하며, 이들중에서도, 치환체 X 및 Y중 하나가 불소인 화합물이 가장 바람직하다.
또 다른 중요한 서브그룹의 살균 작용을 가진 화합물은 R과 R1이 각각 서로 독립적으로 C1-C4알킬, C1-C4알콕시 또는 히드록시이고 ; X는 할로겐, C1-C4알킬, C1-C4알콕시, Si(CH3)3,시아노 또는 CH2-CH(NH2)-P(O)(R)(R1)이며 ; Y는 수소, 할로겐, 할로메톡시 또는 할로메틸인 일반식(I)의 화합물 및 이의 염이다.
마지막으로 언급한 살균제중에서 R, R1및 Y가 상기된 바와같고, X는 불소, 염소, 메틸, 에틸, 이소프로필, 3급-부틸, 메톡시, 에톡시, 이소프로폭시 또는 시아노인 화합물이 특히 중요하다.
또한 R과 R1이 일반식(I)에서 정의한 바와같고 ; X는 할로겐, 메틸, 메톡시 또는 시아노이고 ; Y는 수소, 할로겐, OCHF2또는 CF3인 살균제화합물 및 이의 염이 중요하다.
또 다른 살균제의 중요한 그룹은 R과 R1이 상기에서 정의한 바와같고 ; X는 수소이며 ; Y는 디플루오로메톡시 또는 트리플루오로메틸인 일반식(I)의 화합물이다.
펩티딜아미노메탄포스폰산 및 펩티딜아미노포스핀산의 유도체가 항생물질에 대한 상승제로서 미합중국 특허 명세서 제4,016,148호에 기재되어 있다. 그러나, 거기에는 해충 제어와 관련된 사항은 포함되어 있지 않았으며 그러한 용도로는 제안조차 되지 않았다.
펩티딜아미노알칸포스폰산 유도체가 제초제 및 식물 성장과 식물 발아에 대한 지연제로서 미합중국 특허 명세서 제4,431,438호에 제시되어 있다. 그러나, 미생물 발병에 대한 식물의 보호에 대해서는 언급되어 있지 않다. 또한, 일반적인 제초 또는 성장 억제 성질을 지닌 조성물은 경작 식물에 보호하는데 당연히 매우 적합하지 않다는 것이 부언되어야만 한다.
놀랍게도 일반식(I)의 아미노에탄포스폰산 유도체 및 이의 염은 제초성질을 가지지 않을 뿐만 아니라 진균과 세균의 발병으로부터 지속적으로 보호해주고 상기 식물의 성장을 촉진시키는데 특히 적합한 예상외로 강력한 살균작용을 가지고 있다는 것이 밝혀졌다.
치환체 R과 R1중 적어도 하나가 OH 그룹인 경우에만 일반식(I)의 염은 바람직한 화합물의 범주에 든다. 일반식(I)의 금속염 특히 알루미늄, 니켈, 망간 및 구리염과 저급 알킬암모늄 염은 이들의 살진균작용 때문에 특히 바람직하며, 이들은 토양 적용을 위해 그리고 종자 처리제로서 특히 적합하다.
일반식(I) 화합물의 중요한 용도는 유해한 식물병원성 진균을 제어하는 것이다. 따라서, 실제적인 목적으로 일반식(I)의 화합물은 바람직하지 않은 부작용으로 인해 경작식물에 악영향을 끼치지 않으면서 경작식물을 보호하는데에 매우 유용한 치료적, 예방적 및 전신적 활성을 나타낸다. 본 발명의 범주내에 드는 경작식물의 예로는 하기와 같다. 시리얼(밀, 보리, 호말, 귀리, 쌀) ; 비이트(사탕무우 및 포더 비이트 fodder beet), 다육과(drupes), 이과(pomes) 및 소프트 과일(사과, 배, 플럼, 복숭아, 아몬드, 체리, 딸기, 나무딸기 및 검은딸기), 콩과 식물(콩, 렌즈콩, 완두콩, 대두) ; 기름 식물(평지, 겨자, 양귀비, 올리브, 해바라기, 코코닛, 피마자유, 식물, 코코아콩, 땅콩) ; 오이과 식물(오이, 메로우, 참외류) ; 섬유식물(목화, 아마, 대마, 황마) ; 감귤류 과일(오렌지, 레몬, 그레이프프루트, 만다린) ; 야채(시금치, 상치, 아스파라거스, 양배추, 당근, 양파, 토마토, 감자, 파프리카) ; 또는 옥수수, 담배, 견과류, 커피, 사탕수수, 홍차, 동굴식물, 홈, 바나나, 아보카도와 같은 식물, 및 천연고무 식물 뿐만 아니라 장식용 식물, 유용한 식물 및 관련된 작물에서, 식물, 또는 식물의 부분(열매, 꽃, 잎, 줄기, 괴경, 뿌리)에서 생기는 미생물을 일반식(I)의 화합물을 사용하여 박멸시키거나 억제시킬 수 있는 한편, 동시에 나중에 성장하는 식물의 부분도 또한 그러한 미생물의 공격으로부터 보호시킬 수 있다.
일반식(I)의 화합물은 하기의 부류에 속하는 식물병원성 진균에 대해 특히 유효하다 : 아스코마이세테스(Ascomycetes) [예 : 헬민토스포리움(Helminthosporium) 및 푸사라움(Fusarium)종] 및 바시도마이세테스(Basidomycetes) [예 : 틸레티아 (Tilletia) 및 우스틸라고(Ustilago)]. 따라서, 일반식(I)의 화합물은 종자(열매, 괴경, 낟알)보호를 위한 종자 처리제로서 사용될 수 있으며, 진균감염에 대해서 뿐만 아니라 토양에서 생기는 식물병원성 진균에 대한 식물 절단(cutting)에 사용될 수 있다.
따라서, 본 발명은 식물병원성 미생물을 제어하고 이러한 미생물의 질병으로부터 식물을 보호하기 위해 식물을 예방적 처리하기 위한 일반식(I)의 화합물의 용도에 관한 것이다.
일반식(I)의 화합물은 통상 조성물의 형태로 적용시키며, 경작 지역 또는 처리하려는 식물에 추가의 화합물과 함께 동시에 또는 연속적으로 적용시킬 수 있다. 이러한 화합물은 비료 또는 미량 영양소 공여체이거나 식물성장에 영향을 미치는 기타의 제제일 수 있다. 이들은 또한 필요한 경우, 추가의 담체, 계면활성제 또는 제형분야에서 통상적으로 사용되는 적용촉진 보조제(adjuvant)와 함께 선택적인 제초제, 살진균제, 살균제, 살선충제, 살연체동물제(molluscicide) 또는 이들 몇가지 제제의 혼합물일 수 있다. 적합한 담체 및 보조제는 고체 또는 액체일 수 있으며, 제형기술내에서 통상 사용되는 물질, 예를들면, 천연 또는 재생된 광물질, 용매, 분산제, 습윤제, 점착화제(tackifier), 농밀화제, 결합제 또는 비료에 사용한다.
일반식(I)의 화합물은 변형시키지 않은 형태로 사용하거나 바람직하게는 제형분야에서 통상 사용되는 보조제와 함께 사용하며, 따라서 공지된 방법에 따라 유화가능한 농축물, 피복가능한 페이스트, 직접 분사 또는 희석가능한 용액, 희석된 유제, 습윤성 분제, 가용성 분제, 분진제, 과립제 및 또한 예를들어 중합체성 물질내로의 캡슐화제로 제형화한다. 조성물의 성질 때문에, 적용방법, 예를들면, 분사, 분무, 분진, 산포 또는 유출은 의도하는 목적과 우세한 상황에 따라 선택한다. 농작에서 유리한 적용율은 통상 1헥타르당 50g 내지 5kg의 활성 성분(a·i·), 바람직하게는 100g 내지 2kg a.i./ha, 가장 바람직하게는 100g 내지 600g a.i./ha 이다.
적당한 용매는 하기와 같다 : 방향족 탄화수소, 바람직하게는 탄소수 8 내지 12의 분획, 예를들면, 크실렌 혼합물 또는 치환된 나프탈렌, 디부틸 프탈레이트 또는 디옥틸 프탈레이트와 같은 프탈레이트 ; 시클로헥산 또는 파라핀과 같은 지방족 탄화수소 ; 에탄올, 에틸렌 글리콜 모노메틸 또는 모노에틸에테르와 같은 알콜 및 글리콜과 이들의 에테르 및 에스테르 ; 시클로헥사논과 같은 케톤 ; N-메틸-2-피롤리돈, 디메틸설폭사이드 또는 디메틸포름아미드와 같은 강한 극성 용매 ; 뿐만 아니라 에폭시화된 코코닛 기름 또는 대두유와 같은 에폭시화된 식물성 기름 ; 또는 물.
실제로 적용율을 감소시킬 수 있는 특히 유용한 적용-축진 보조제는 또한 천연(동물성 또는 식물성) 또는 합성 포스포리피드의 세팔린과 레시틴 시리즈이며, 예를들면 포스파티딜 에탄올아민, 포스파티딜 세린, 포스파티딜 글리세롤 또는 리조레시틴이다.
제형화하려는 일반식(I) 화합물의 성질에 따라 적합한 계면활성 화합물은 우수한 유화성, 분산성 및 습윤성을 가진 비이온성, 양이온 및/또는 음이온 계면활성제이다. 또한, "계면활성제"란 용어는 계면활성제의 혼합물로 이루어진 것으로도 해석될 수 있을 것이다.
제형의 분야에서 통상 사용되는 계면활성제는, 예를들면, 문헌[참조 : "McCutcheon's Detergents and Emulsifiers Annual", MC Publishing Corp. Ridgewood, New Jersey, 1981 ; Helmut Stache "Tensid-Taschenbuch"(Surfactant Handbook) Carl Hanser Verlag, Munich/Vienna, 1981 ; M. & J. Ash, Encyclopedia of Surfactants, Vol I-III, Chemical Publishing Co., New York, 1980-1981]에 기술되어 있다.
농약 조성물은 통상 0.1 내지 99%, 바람직하게는 0.1 내지 95%의 일반식(I) 화합물 ; 99.9 내지 1%, 바람직하게는 99.8 내지 5%의 고체 또는 액체 보조제 ; 및 0 내지 25%, 바람직하게는 0.1 내지 25%의 계면활성제를 함유한다.
시판용 생성물은 바람직하게는 농축물로 제형화되는 반면, 최종 사용자는 통상 희석한 제형을 사용한다.
하기 실시예는 본 발명을 상세히 설명해주며 본 발명을 제한하지 않는다. 부와 백분율(%)은 중량에 의거한 것이다.

[실시예 1]
하기 구조식의 O,O-디이소프로필-1-아미노-2-(4-플루오로페닐)에틸포스네이트의 제조
a) O,O-디이소프로필-1-N-벤질리덴아미노-2-(4-플루오로페닐)에틸포스네이트(중간체)의 제조 750ml의 테트라하이드로푸란(THF)중의 디이소프로필아민 113.4g(0.8몰)에 500ml의 n-부틸리튬을 교반 냉각시키면서 가한다. 혼합물을 -75℃로 냉각시킨 다음 600ml THF중의 O,O-디이소프로필벤질리덴아민 메탄포스포네이트 226.6g(0,8몰) 용액에 1시간에 걸쳐 적가한다. 1시간 동안 교반한 후, 150ml THF중의 4-플루오로벤질 브로마이드 151.2%(0.8몰) 용액을 적가하고 1시간 동안 계속하여 교반한다. 반응혼합물을 실온에서 하루밤 정치시킨 후에 용매를 제거하고 잔류물을 1리터의 디클로로메탄에 용해시킨다. 각각 200ml의 물로 용액을 3회 세척하고 Na2SO4로 건조시킨다. 용매를 증발제거시켜 286.3g(이론치의 91.4%)의 중간체를 수득하고, 분자 증류시킨 후 순수한 생성물 261.2g(이론치의 83.4%)를 수득한다.
비점 : 160℃/0.1밀리바.
b) 최종생성물의 제조
200ml 이소프로판올중의 1a)에서 수득한 중간체 19.6g(0.05몰) 용액에 2g의 Pd/C(5% Pd)를 가하고 혼합물을 실온(20 내지 25℃)에서 수소첨가 반응시킨다. 62% 수소가 흡수된 후에 2g의 Pd/C를 다시 가하고, 90% 수소가 흡수된 후에 2g의 Pd/C를 더 가한다. 19시간 후에 수소 흡수가 완결된다. 반응혼합물을 여과하고 여액은 회전 증발시켜 농축한다. 잔류물(약 15g)을 봄브 튜브에서 증류시켜 순수한 최종 생성물을 무색의 기름 형태로 12.6g(이론치의 83.1%) 수득한다.
비점 : 170℃/0.08밀리바.
[실시예 2]
1-아미노-2-(4-플루오로페닐)에탄포스폰산(화합물 2.6)의 제조
227.5g(0.75)몰의 O,O-디이소프로필-1-아미노-2-(4-플루오로페닐)에틸포스포네이트와 750ml의 20% 염산의 혼합물을 교반하면서 5시간 동안 환류시키고 실온에서 5시간 더 교반한다. 500ml의 물을 가한 후에 침전물을 여과하고 250ml의 물과 500ml의 메탄올로 세척한다. 표제화합물 103.3g을 수득한다. 여액은 농축하고 잔류물은 메탄올/프로필렌 옥사이드로 재결정시켜 최종생성물 43.3g을 더 수득한다. 총 수득량은 146.6g(이론치의 89.2%)이고 ; 융점은 266 내지 270℃(분해)이다.
[실시예 3]
1-아미노-2-(4-플루오로페닐)에탄포스폰산의 광학이성체의 제조
A. O,O-디에틸-1-아미노-2-(4-플루오로페닐)에틸포스포네이트의 디벤조일 타타레이트의 제조
a) 750ml의 메탄올 및 750ml의 에탄올중의 O,O-디에틸-1-아미노-2-(4-플루오로페닐)에틸포스포네이트 68.82g(0.125몰) 용액에 47.04g의 디벤조일타타르산 (L) (-)xH2O를 가한다. 2시간 동안 교반한 후에 농밀한 백색 현탁액을 여과하고 잔류물을 건조시킨다 .에탄올로 2회 재결정하여 31.5g(이론치의 39.8%)의 염 "염((I)"을 수득하고, 융점은 179℃(분해)이다 ; [α]=-66.4±0.5°(c=메탄올중에서 2.077%).
b) 여액을 혼합하여 증발시키고 잔류물은 1N NaOH에서 교반한다. 용액은 염화나트륨으로 포화시키고 각각 400ml의 CH2Cl2로 3회 추출한다. 유기층은 Na2SO4로 건조하고 증발농축한다. 잔류하는 갈색오일(37.5g)을 400ml의 메탄올과 400ml의 에탄올중에 용해시키고 이 용액에 25.6g의 디벤조일 타타르산 (D) (+)xH2O를 가한다. 2시간 동안 교반한 후에 여과하여 63g의 염을 분리하고 1200ml의 메탄올로 재결정시켜 7.2g의 조 "염(II)"을 수득한다. 여액을 농축하고 잔류물을 500ml의 에탄올로 재결정시켜 17g의 순수한 "염(II)"을 수득한다 ; [α]=+67.3°±0.5°(c=메탄올중에서 1.998%).
B.
a) (+) O,O-디에틸-1-아미노-2-(4-플루오로페닐)에틸포스포네이트의 제조
적정으로부터 유리시키기 위해 25.34g의 염(I)을 실온에서 100ml의 1N NaOH중에서 2시간 동안 교반한다. 투명한 용액을 염화나트륨으로 포화시킨 다음 200ml의 CH2Cl2를 가하고 생성된 현탁액을 진공하에 여과시킨다. 잔류물을 각각 200ml의 CH2Cl2로 2회 세척한다. 여액을 혼합하여 유기층을 분리하고 Na2SO4로 건조시킨 다음 여과하고 증발 농축시켜 연노란색 오일로서 목적하는 최종 생성물 9.1g(이론치의 82.7%)을 수득한다 ; [α]=+10.6°±0.4°(c=메탄올중에서 2.5%).
1H-NMR(CDCl3) : 1.3(NH2, CH3)(t, 8H) ; 2.3-3.5(PCH-CH2)(m, 3H) ; 4.17(OCH2)(qu, 4H) ; 7.1(m)(4H, 페닐).
b) (+) 1-아미노-2-(4-플루오로페닐)에틸포스폰산의 제조
a)에서 수득한 (+) 포스포네이트 5.51g(0.02몰)을 20% 염산 40ml에서 4시간 동안 환류시킨다. 이어서, 용액을 농축하고 잔류물을 메탄올/프로필렌 옥사이드로 재결정시켜 목적하는 최종생성물 3.7g(이론치의 84.5%)을 수득한다. 융점은 259 내지 263℃(분해)이다 ; [α]=+37.5°±0.4°(c=1N NaOH 중에서 2.636%).

B. a) 방법에 따라 1N NaOH 용액을 사용하여 "염(II)"로부터 수득한다. 수율 : 이론치의 95.9% ; 담황색 오일 ; [α]=-10.3°±0.5°(c=메탄올중에서 2.036%).
b) (-) 1-아미노-2-(4-플루오로페닐)에탄포스폰산의 제조
B. b) 방법에 따라 20% 염산중에서 상기의 (-) 포스포네이트를 가수분해시켜 수득한다. 수율 : 이론치의 77.6% ; 융점 : 261 내지 263℃(분해) ; [α]=-36.9°±0.5°(c=1N NaOH 중에서 2.081%).
1H-NMR(D2O/NaOD) : 2.5-3.4(PCH-CH2)(m, 3H) ; 4.85(OH, NH2)(s) ; 6.8-7.4(페닐, 4H)(m).
구체적으로 달리 언급하지 않는한 부분입체 이성체의 혼합물로서 수득되는 하기의 일반식(I)의 화합물은 또한 이 방법으로 또는 상기한 다른 방법으로 제조할 수 있다.
1H-NMR 값은 바리안(Varian) EMK-360 스펙트로미터를 사용하여 60MHz에서 CDCl3중에서 (CH3)4Si를 표준물질로 하여 측정한다.
31P-NMR 값은 브루커(Bruker) WP 80 스펙트로미터를 사용하여 32.28MHz에서 85% H3PO4를 외부 표준물질로 하여 측정한다.

일반식(I)의 활성성분(화합물)에 대한 제형실시예(여기에서, 백분율은 중량에 의거한 것이다)
상기 액제는 미소적(microdrops) 형태의 사용에 적합하다.
활성성분을 메틸렌 클로라이드에 용해시키고 용액을 담체상에 분무한 다음 이어서 진공하에 용매를 증발제거한다.
사용시 만드는 분진제는 담체를 활성 성분과 긴밀하게 혼합하여 수득한다.
활성성분은 보조제와 완전히 혼합하고 혼합물은 적당한 분쇄기에서 완전히 분쇄하여 습윤분제를 수득하고 이것을 물로 희석하여 원하는 농도의 현탁액을 만들 수 있으며 이것은 특히 종자 처리에 적합하다.

생물학적 실시예 :
[실시예 B1]
콩에서 보트리티스 시네레아(Botrytis cinerea)에 대한 작용 및 잔류 보호 작용
약 10cm 크기의 콩식물에 시험 화합물의 습윤분제로부터 제조된 분무 혼합물(0.02%)을 사용하여 분무한다. 48시간후에, 처리한 식물을 진균 코니디아 현탁액으로 감염시킨다. 감염시킨 식물을 상대습도 95 내지 100% 및 21℃에서 3일 동안 배양시킨 다음 진균 발병에 대해 평가한다. 보트리티스 발병은 비처리한 감염된 콩식물에서는 100% 였다. 일반식(I) 화합물중의 하나로 처리한 후에는 발병은 20% 미만이었다. 예를 들어, 화합물 1.5, 2.6, 2.11, 3.4, 2.15, 2.37, 5.13 및 기타의 화합물로 처리한 후에는 발명(0 내지 5%)은 관찰되지 않았다.
[실시예 B2]
호밀에서 푸시리움 니발레(Fusarium nivale)에 대한 작용(종자처리)
푸사리움 니발레로 자연적으로 감염된 테트라헬(Tetrahell) 변종의 호밀 종자를 활성 성분 600ppm 및 200ppm농도(종자의 중량기준)의 시험 살진균제로 회전 혼합기에서 처리한다. 감염되고 처리된 종자는 10월에 파종기를 사용하여 야외에서 3미터 길이의 소지구에 6줄로 파종한다. 시험 화합물 각각을 사용하여 주어진 농도에서 3종을 실시한다. 평가할때까지 시험식물은 통상의 들판 상태하에서 바람직하게는 겨울 몇달동안 눈 덮힌 것이 부서지지 않는 지역에서 경작한다. 식물독성을 평가하기 위해 가을의 발아와 봄의 새싹 및 개체의 밀립상태를 측정한다. 시험 화합물의 효과를 판정하기 위해서는 눈이 녹은 직후 봄에 푸사리움으로 발병한 식물의 백분율(%)로 평가한다.
일반식(I)의 시험 화합물에서는 식물독성이 거의 없거나 전혀 없었다. 화합물 1.5, 1.8, 2.1, 2.2, 2.6, 2.8, 2.34 및 기타의 주어진 농도 둘 모두에서 푸사리움 발병을 완전히 억제했다.

보리에서 헬민토스포리움 그라미네움(Helminthosporium gramineum)에 대한 작용(종자처리)
헬민토스포리움 그라미네움으로 자연적으로 감염된 "Cl" 변종의 겨울 보리의 종자를 활성성분 600ppm 및 200ppm(종자의 중량기준) 농도의 시험 살진균제로 회전혼합기에서 처리한다. 감염되고 처리된 보리는 10월에 파종기를 사용하여 야외에서 2미터 길이의 소지구에 3줄로 파종한다. 시험 화합물 각각을 사용하여 주어진 농도에서 생중으로 실시한다. 평가할 때까지 시험 식물은 통상의 들판상태하에서 경작한다. 식물독성을 평가하는데에 가을의 발아와 봄의 새싹 및 개체의 밀집 상태를 측정한다. 시험 화합물의 효과를 판정하기 위해서는 이삭 발아 시간에 헬민토스포리움으로 발병된 줄기의 백분율(%)로 평가한다.
일반식(I)의 시험 화합물에서는 식물독성이 거의 없거나 전혀 없었다. 주어진 두가지 농도 모두에서 화합물 1.28, 2.2, 2.6, 2.9, 2.30, 3.5 및 기타의 비처리된 대조식물에 비해 20% 미만으로 진균발병을 억제했다.
[실시예 B4]
보리에서 우스틸라고 누다(Ustilago nuda)에 대한 작용(종자처리)
우스틸라고 누다로 자연적으로 감염된 "RM1" 변종의 겨울보리 종자를 활성성분 600ppm 및 200ppm(종자의 중량 기준) 농도의 시험 살진균제로 회전혼합기에서 처리한다. 감염되고 처리된 보리는 10월에 파종기를 사용하여 야외에서 2미터 길이의 소지구에 3줄로 파종한다. 시험 화합물 각각을 사용하여 주어진 농도에서 3중으로 실시한다. 평가할 때까지 시험 식물은 통상의 들판상태하에서 경작한다. 시험 화합물의 효과를 판정하기 위해서는 꽃이 피어있는 동안 우스틸리고에 의해 발병된 이삭의 백분율(%)로 평가한다.
화합물 2.6, 2.10, 2.15, 2.19, 5.21 및 기타는 주어진 농도를 모두에서 20% 미만으로 진균발명을 감소시켰다.
[실시예 B5]
틸레티아 트리티시(Tilletia tritici)에 대한 작용(종자 처리)
틸레티아 트리티시의 흑수병 포자(종자 1kg당 건조포자물질 3g)로 인공적으로 감염시킨 프로부스(Probus) 변종의 겨울 밀 종자를 활성 성분 600ppm 및 200ppm(종자의 중량 기준) 농도의 시험 살진균제로 회전혼합기에서 처리한다. 감염되고 처리된 밀은 10월에 파종기를 사용하여 야외에서 2m 길이의 소지구에 3줄로 파종한다. 시험 화합물 각각을 사용하여 주어진 농도에서 3중으로 실시한다. 이삭이 익을때까지 시험식물은 통상의 들판상태하에서 경작한다. 식물독성은 평가하는데 가을의 발아와 봄의 새싹 및 개체의 밀집상태를 측정한다. 시험 화합물의 효과를 판정하기 위해서는 이삭이 익을때에 틸레티아 트리티시로 발병한 이삭의 백분율(%)로 평가한다.
주어진 농도들 모두에서 일반식(I)의 화합물 예를 들면, 1.5, 2.8, 1.28은 틸레티아 트리티시에 의한 발병에 대해 현저한 보호작용을 나타내며, 따라서 밀의 발아와 성장이 상당히 촉진 되었다.
Claims (11)
- 일반식(II)의 쉬프염기의 CH2그룹을 활성화시키고 ; 활성화된 염기를 적절하게 치환된 일반식(III)의 벤질 할라이드로 아르알킬화시킨 다음 ; 촉매의 존재하에서 일반식(IV)의 중간체를 가수소분해적으로 분리시키고 ; 단, R 및/또는 R1이 알콕시이고, 또한 히드록실그룹으로 전환될 경우, 승온에서 무기산을 사용하여 임의로 더 처리하는 단계를 포함함을 특징으로 하여 일반식(I)의 화합물 및 이의 염을 제조하는 방법.상기식에서, R과 R1이 각각 서로 독립적으로 C1-C4알킬, C1-C4알콕시 또는 히드록시이며 ; X는 수소, 할로겐, C1-C4알킬, C1-C4알콕시, 트리메틸실릴, 시아노 또는 CH2-CH(NH2)-P(O)(R)(R1)라디칼이고 ; Y는 수소, 할로겐, C1-C2할로알콕시 또는 C1-C2할로알킬이며 ; T1은 수소, 또는 지방족이나 방향족 라디칼이고 ; T2는 지방족 또는 방향족 라디칼이며 ; Hal은 할로겐 원자이고 ; 단, R과 R1이 동시에 히드록시 또는 에톡시일 경우, 치환체 X와 Y중에서 적어도 하나는 수소가 아니다.
- 제1항에 있어서, R과 R1이 동시에 히드록시 또는 에톡시인 경우 치환체 X와 Y중의 적어도 하나는 수소가 아닌 일반식(I)와 화합물을 제조하는 방법.
- 제1항에 있어서, R=R1=OH이고, X는 수소, 불소, 염소 또는 브롬, C1-C4알킬, CH2O, Si(CH3)3, -CN, -COOCH3또는 -CH2-CH(NH2)-P(O)(OH)2이며, Y는 불소, 염소, 브롬, 할로메톡시 또는 할로메틸인 일반식(I)의 화합물을 제조하는 방법.
- 제1항에 있어서, 치환체 X와 Y중에서 적어도 하나가 할로겐이거나 할로겐을 함유하는 일반식(I)의 화합물을 제조하는 방법.
- 제1항에 있어서, 치환체 X와 Y중에서 하나가 불소이거나 불소를 함유하는 일반식(I)의 화합물을 제조하는 방법.
- 제1항에 있어서, R과 R1이 각각 서로 독립적으로 C1-C4알킬, C1-C4알콕시 또는 히드록시이고, X는 할로겐, C1-C4알킬, C1-C4알콕시, Si(CH3)3, 시아노 또는 CH2-CH(NH2)-P(O)(R)(R1)이며, Y는 수소, 할로겐, 할로메톡시, 할로에톡시 또는 할로메틸인 일반식(I)의 화합물을 제조하는 방법.
- 제1항에 있어서, R, R1및 Y가 제6항에서 정의한 바와 같고, X는 불소, 염소, 메틸, 에틸, 이소프로필, 3급-부틸, 메톡시, 에톡시, 이소프로폭시 또는 시아노인 일반식(I)의 화합물을 제조하는 방법.
- 제1항에 있어서, R 및 R1이 제1항에서, 정의한 바와 같고, X는 할로겐, 메틸, 메톡시 또는 시아노이며, Y는 수소, 할로겐, 디플루오로메톡시 또는 트리플루오로메틸인 일반식(I)의 화합물을 제조하는 방법.
- 제1항에 있어서, R 및 R1이 제1항에서, 정의한 바와 같고, X는 수소이며, Y는 디플루오로메톡시 또는 트리플루오로메틸인 일반식(I)의 화합물을 제조하는 방법.
- 제1항에 있어서, 1-아미노-2-(4-플루오로페닐)에탄포스폰산을 제조하는 방법.
- 제1항에 있어서, 1-아미노-2-(2-클로로-4-플루오로페닐)에탄포스폰산을 제조하는 방법.
