Claims (13)

  1. 숙주 세포에 거의 침투할 수 없으며, 감염 세포내에서 및 숙주의 감염 세포로 부터 비감염 세포로 거의 확산될 수 없고, 숙주의 비감염 세포뿐만 아니라 감염 세포를 다량 죽일 수 있는 독소를 생성하지 못하도록 야생주 시겔라의 게놈을 형질 변환시키는 단계를 특징으로 하는 야생주 시겔라에 대한 왁찐을 제조하는데 사용할 수 있는 시겔라의 변이주를 생성하기 위한 장침투성 시겔라 야생주의 변이 방법.
  2. 제1항에 있어서, 시겔라의 야생주가 숙주의 조직뿐만 아니라, 세포에 침투하는데 필요한 단백질을 코오딩한 제1유전자와, 시겔라의 야생주가 숙주의 감염 세포와 비감염 세포 사이 및 감염 세포내에서 확산되는데 필요한 단백질을 코오딩한 제2유전자를 완전히 또는 부분적으로 제거하거나 영구적으로 불활성화 시킬수 있도록 시겔라 야생주의 게놈을 변이시키는 방법.
  3. 제2항에 있어서, 시겔라가 s. 플렉스너이고 제1유전자는 s. 플렉스너에 의한 에어로박틴의 사용 또는 생성을 코오드하는 방법.
  4. 제3항에 있어서, 제2유전자가 세포간-세포내 확산을 위해 코오드하는 방법.
  5. 제2항에 있어서, 시겔라는s.이질1 이고,s.이질1 에 의해 시가-독소의 사용 또는 생성을 코오드하는 제3유전자가 부분적으로 또는 완전히 제거되거나 영구적으로 불활성화시킬 수 있도록 게놈을 변이시키는 방법.
  6. 제5항에 있어서,s.이질1 의 제1유전자가s.이질1 에 의한 엔테로켈린의 사용 또는 생성을 코오드하는 방법.
  7. 제6항에 있어서, 제2유전자가 세포간-세포내 확산을 위해 코오드하는 방법.
  8. 제6항에 있어서, 제1유전자가s.이질1 의 엔테로켈린의 오페론의 ent F, Fep E, Fep C 및 Fep D 소단위 유전자를 포함한 방법.
  9. 제5항 내지 제8항중 어느 한 항에 있어서, 제1, 제2 및 제3유전자가 변이된 방법.
  10. 제2항 내지 제9항중 어느 한 항에 있어서, 하나 이상의 유전자가 하나 이상의 시험관내 변이 유전자를 이용한 대립형질 교환에 의해 불활성화된 방법으로서 특히 상기 변이 유전자는 중요 부위가 누락되었으며 마커 유전자가 그 변이 유전자로 삽입된 방법.
  11. 제1항 내지 제10항중 어느 한 항의 방법에 의해 변이되거나 그것의 후세대인 시겔라.
  12. 제11항의 변이된 시겔라로 부터 제조된 왁찐.
