제1도는 실시예1의 항-IgA-피넛 어글루티닌 효소 면역 분석을 사용하여 시험한 845혈청 표면의 패널(panel)로부터의 결과를 도시한 것이고,
제2도는 신규분석을 사용한 음성-CEA환자 시료 시험의 결과를 도시한 것이며,
제3도는 실시예2의 변성 항-IgA-피넛 어글루티닌 효소 면역 분석을 사용하여 정상환자, 비암환자 및 암환자의 180혈청 표본 시험의 결과를 도시한 것이다.
Claims (11)
- a) 유체 시료를 항-lgA 항체와 접촉시켜 항-IgA/lgA 복합체를 형성시키고 ; b) IgA1이 시료내에 존재할 경우 항-IgA/IgA 복합체를 IgA1에 대해 특이성을 지닌 표식렉틴과 접속시켜 항-IgA1/IgA1/렉틴 복합체를 형성시킨 다음 ; c) 유체 시료에서의 IgA1의 존재 또는 양의 척도로서 항-IgA/IgA1/렉틴 복합체에 대한 결합 또는비결합 표식의 존재 또는 양을 측정함을 특징으로 하여, 유체시료에서 0-결합 올리고 당류를 지닌 IgA1의 존재 또는 양을 측정하는 방법.
- a) 유체시료를 고체상에 결합된 항-IgA향체와 접촉시켜 항-IgA/IgA 고체상 복합체를 형성시키고, b) IgA1이 유체시료내에 존재할 경우 항-IgA/IgA 고체상 복합체를 IgA1에 대해 특이성을 지닌 표식레틴과 접촉시켜 항-IgA/IgA1/렉틴 고체 상 복합체를 형성시킨 다음 ; c) 유체 시료에서의 IgA1의 존재 또는 양의 척도로서 항-IgA/IgA1/렉틴 고체상 복합체에 대한 결합 또는 비결합 표식의 존재 또는 양을 측정함을 특징으로 하여, 유체시료에서 0-결합 올리고 당류를 지닌 IgA1의 존재 또는 양을 측정하는 방법.
- 제1항 또는 2항에 있어서, 표식을 효소, 방사성 동위원소 및 형광성 분자중에서 선택하는 방법.
- a) IgA 면역글로부린에 대해 특이적인 항체를 고체상에 흡수시키고 ; b) 유체시료를 항체-피복 고체상에 가하고 배양시켜 항-IgA/IgA 고체상 복합체를 형성 시키며 ; c) 복합체를 세척하여 미반응 시료를 제거하고 ; d) IgA1에 대해 특이성을 지닌 효소 표식렉틴을 가하고 배양시켜 항-IgA/IgA1/렉틴 고체상 복합체를 형성시키며 ; e) 항-IgA/IgA1/렉틴 고체상 복합체를 세척하고 ; f) 효소 기질을 가한 다음 배양시키며 ; 9) 효소 반응을 퀀치시키고 ; h) 시료내의 IgA1의 존재 또는 양의 척도로서 표식을 검출함을 특징으로 하여, 유체시료에서 0-결합올리고 당류를 지닌 IgA1의 존재 또는 양을 측정하는 방법.
- 제4항에 있어서, 효소표식을 알카리성 포스파타아제, β-갈락토올시다아제, 글루코스 옥시다아제 및 호스래디쉬 퍼옥시다아제 중에서 선택하는 방법.
- a) IgA 면역 글로부린에 대해 특이적인 항체를 고체상에 흡수시키고 ; b) 시료를 항체-피복 고체상에 가하여 항-IgA/IgA 고체상 복합체를 형성시키며 ; c) 복합체를 세척하여 미반응 시료를 제거하고 ; d) IgA1에 대해 특이성을 지닌 방사성 표식 렉틴을 가하며 ; e) 배양시켜 IgA1이 시료내에 존재할 경우 항-IgA/IgA1/렉틴 고체상 복합체를 형성시키고 ; f) 고체상을 세척하며 ; g) 시료내의 IgA1의 존재 또는 양의 척도로서 표식을 검출함을 특징으로 하여, 유체시료에서 변성된 0-결합 올리고 당류를 지닌 IgA1의 존재 또는 양을 측정하는 방법.
- 제 1,2,4 및 6항에 있어서, 유체 시료를 혈액, 혈청 및 혈장 중에서 선택하는 방법.
- 제 1,2.4 및 6항에 있어서, 렉틴을 피넛 어글루티닌(Peanut agglutinin) 또는 바우히니아 푸푸레아 알바(Bauhinia purpurea alba) 어글루티닌 중에서 선택하는 방법.
- 제 1,2,4 및 6항에 있어서, IgA1이 IgA1의 폴리펩티드 쇄에 대해 0-결합된 비차단되거나 노출된 DGaIBeta(1-3) DgalNac 올리고 당류 구조를 함유하는 방법.
- 항-인체/IgA 항체로 피복된 폴리스티렌 비두, IgA1에 대해 특이성을 지닌 표식렉틴, 표본회석 완충액 및 콜로스트럼 IgA 표준물질로 이루어진 유체시료에서 0-결할 올리고 당류를 지닌 IgA1을 시험하기 위한 키트(Kit).
- a) 유체시료를 항-IgA 항체와 접촉시켜 항-IgA/IgA 복합체를 형성시키고 ; b) IgA1이 시료내에 존재할 경우 항-IgA/IgA 복합체를 IgA1에 대해 특이성을 지 닌 표식렉틴과 접촉시켜 항-IgA/IgA1/렉틴 복합체를 형성시키며 ; c) 유체시료내의 IgA1의 존재 또는 양의 척도로서 항-IgA/IgA1/렉틴 복합체에 대해 결합 또는 비결합 표식의 존재 또는 양을 측정함을 특징으로 하여, 태생기암 항원 음성 생물학적 유체시료에서 0-결합 올리고 당류를 지닌 IgA1의 존재 또는 양을 측정하는 방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
