Claims (21)
- 인자 XIII의 서브유니트(Subunit)를 이황화물 교환반응에 적합한 담체 매트릭스에 가역적으로 결합시키고 이것을 환원제와 반응시켜 담체 매트릭스로부터 제거함을 특징으로 하여, 인자 XIII의 서브유니트를 친화성 크로마토그래피하여 정제하는 방법.
- 제 1항에 있어서, 인자 XIII의 서브유니트를 태반 균질물 또는 혈소판 균질물로부터 수득하는 방법.
- 제 1항에 있어서, 인자 XIII의 서브유니트를 혈장으로부터 수득하는 방법.
- 제 1항 내지 제 3항중 어느 한 항에 있어서, 인자 XIII이 용액중에 존재하는 방법.
- 제 1항 내지 제 4항중 어느 한 항에 있어서, 인자 XIII 또는 인자 XIII-함유 용액을 알킬화제로 전처리하는 방법.
- 제 5항에 있어서, 알킬화제가 요오아세트아미드 또는 요오드아세트산인 방법.
- 제 6항에 있어서, 요오도아세트산이 5 내지 250밀리몰/l, 바람직하게는 5 내지 50밀리몰/1의 농도로 존재하는 방법.
- 제5항 내지 제7항중 어느 한 항에 있어서, 알킬화반응을 Ca2+및 Mg2+의 부재하에 수행하는 방법.
- 제6항 및 제 7항중 어느 한 항에 있어서, 알킬화 반응을 킬레이트화 물질, 바람직하게는 0.001 내지 0.1몰/l, 특히 바람직하게는 0.002 내지 0.02몰/l 농도의 에틸렌디아민테트라아세트산(EDTA)의 존재하에 수행하는 방법.
- 제 9항에 있어서, 인자 XIII-함유 용액을 pH 6.5 내지 8.5에서 0.001 내지 0.04몰/l 농도의 EDTA의 존재하에 1 내지 360분 바람직하게·는 5 내지 60분 동안, 0.005 내지 0.25몰/1 농도의 요오도아세트산으로 처리하는 방법.
- 제 5항 내지 제10항중 어느 한 항에 있어서, 알킬화반응을 수행한 후, 바람직하게는 단백질-투석 또는 염침전시켜 알킬화 물질을 제거하는 방법.
- 제1항 또는 제3항 내지 제11항중 어느 한 항에 있어서, 혈장 인자 XIII 복합체를 0.05 내지 0.6몰/l, 바람직하게는 0.05 내지 0.2몰/l농도의 Ca2+또는 Mg2+으로 배양하는 방법.
- 제 1항 내지 제12항중 어느 한 항에 있어서, 인자 XIII-함유 용액을 머캅토그룹, 바람직하게는 티올-세파로즈(Sepharose) 4B를 함유하는 담체 매트릭스와 접촉시키는 방법.
- 제 1항 내지 제13항중 어느 한 항에 있어서, 담체 매트릭스에의 결합단계를, 바람직하게는 pH 6.5 내지 8.5에서 바람직하게는 0.01 내지 0.2몰/l 농도의 Ca2+또는 Mg2+의 존재하에서 수행하는 방법.
- 제13항 또는 제14항에 있어서, 담체 매트릭스를 디-2-피리딜 이황화물, 2,2′-디티오비스(피리딘 N-산화물), 5,5′-디티오비스(2-니트로벤조산), 이들 화합물의 유도체, 또는 아조디카복실산의 유도체와 반응시켜 미리 활성화시킨 방법.
- 제13항 내지 제15항중 어느 한 항에 있어서, 인자 XIII을 포함하는 담체 매트릭스를, 0.01 내지 0.2몰/l농도의 Ca2+또는 Mg2+을 함유하는 용액으로 세척하는 방법.
- 제13항 내지 제16항중 어느 한 항에 있어서, 담체 매트릭스에 결합되는 물질을 0.01 내지 0.2몰/l 농도의 Ca2+또는 Mg2+및 0.005 내지 0.05몰/l 농도의 환원제, 바람직하게는 디티오트레이톨, 머캅토에탄올, 에탄디티올, 글루타티온 또는 시스테인, 및 경우에 따라서는 0.1 내지 0.7g/ml 농도의 자당으로 용출시키는 방법.
- 제 7항 내지 제17항중 어느 한 항에 있어서, 인자 XIII이 천연단백질이거나, 유전학적으로 촉진시켜 제조된 인자 XIII 또는 이의 서브유니트이고/이거나, 인자 XIII활성을 나타내는 천연발생적이거나 합성 제조한 단백질일 수 있는 방법.
- 제 1항 내지 제18항중 어느 한 항에 있어서, 인자 XIII와 서브유니트는 활성을 거의 완전히 보유하는 방법.
- 제 1항 내지 제19항중 어느 한 항에서 청구한 바와같은 방법으로 정제된 인자 XIII, 및 알부민, 포도당 또는 자당과 같은 생리학적으로 허용되는 통상의 첨가제를 함유하는 치료 조성물.
- 인자 XIII 친화력에 의해 유발된 응혈장해를 치료하는데 사용하기 위한, 제20항에 청구한 바와같은 치료조성물의 용도.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
