제1도는 전체 돼지 성장호르몬 유전자에 해당하는 28종류의 합성 올리고 뉴클리오티드의 염기순서를 5′ 말단에서 3′말단순서로 나타낸 것이며,
제2도는 제1도의 올리고 뉴클리오티드의 접합전략을 도시한 것이며,
제3도는 합성 올리고 뉴클리오티드를 대장균 운반 pUC18에로의 클로닝 과정을 도시한 것이며,
제4도는 확인한 돼지 성장호르몬의 염기 서열 및 아미노산 서열을 나타낸 것이며,
제5도는 효모내에서 돼지 성장호르몬의 대량 생산을 위하여 대장균 운반체 pPGAP 및 효모발현 운반체 pCl/l에의 클로닝 과정을 도시한 것이다.
Claims (2)
- 돼지 성장호르몬의 아미노산 배열순서에 따라 효모에서 선택적으로 사용되는 코돈 및 Sac I, Xba I 및 Sal I의 인지부위가 존재하는 올리고 뉴클리오티드를 합성한 후 이를 접합전략에 따라 접합시켜 합성올리고 뉴클리오티드의 카복시쪽 Xba I/Sal I 절편을 Xba I/Sal I으로 처리한 대장균 운반체(pUC18)에 클로닝한 다음 이를 Sac I/Xba I으로 처리하여 합성 올리고 뉴클리오티디의 아미노쪽 Sac I/Xba I 절편을 클로닝시켜 아미노 말단의 일부가 없는 돼지 성장호르몬 유전자를 제조한 다음 이 유전자를 아미노말단 합성유전자와 함께 프로모터와 터니미네이터가 있는 대장균 운반체에 클로닝하여 포로모터-전체 돼지 성장 호르몬 유전자-터미네이터의 카세트를 제조한 다음 이를 효모발현 운반체에 재클로닝시켜 최종 효모발현 운반체를 만들어 이를 효모내에서 발현시킴을 특징으로 하는 돼지 성장호르몬의 제조방법.
- 제1항에 있어서, 아미노 말단 합성 유전자는 Nco I 인지부위 및 Sal I 인지부위가 존재하는임을 특징으로 하는 돼지 성장호르몬의 제조방법.※ 참고사항 : 최초출원 내용에 의하여 공개하는 것임.
