Abstract
본 발명은 3,6-안하이드로-L-갈락토오스(3,6-anhydro-L-galactose, L-AHG)를 유효성분으로 함유하는 면역 억제용 조성물, 상기 유효성분을 포함하는 면역성 질환의 예방 또는 치료용 약학적 조성물, 건강 기능 식품 및 기능성 화장료 조성물에 관한 것이다. 본 발명의 L-AHG를 포함하는 면역 억제용 조성물, 약학적 조성물, 건강 기능 식품 및 기능성 화장료 조성물은 면역 억제제의 주요 표적인 CD4 T 세포의 활성화, 증식 및 분화를 선택적이고 효과적으로 억제하는 우수한 효과가 있으므로 제약 산업, 식품 산업 및 화장품 산업에 매우 유용한 발명이다.
Description
본 발명은 3,6-안하이드로-L-갈락토오스(3,6-anhydro-L-galactose, L-AHG)를 유효성분으로 함유하는 면역 억제용 조성물, 상기 유효성분을 포함하는 면역성 질환의 예방 또는 치료용 약학적 조성물, 건강 기능 식품 및 기능성 화장료 조성물에 관한 것이다.
면역계는 어떤 경우에 부적절하게 작용할 수도 있는데, 과도하게 반응하여 과민성 반응(hypersensitivity)을 일으키거나 혹은 자기/비자기의 구별에 실패하여 자기 분자에 대한 면역반응을 유도한 결과로서 자가면역질환(autoimmune diseases)을 일으키는 경우가 대표적인 예들이다 [Clark and Kupper, 2005]. 또한, 치료 목적으로 이식한 동종이식편(allografts)에 대해 수혜자의 면역계가 일으키는 면역거부반응(immunorejection)은 이식편의 유지 및 생존을 위한 관점에서는 부적절한 면역반응이라 할 수 있다.
인체에서 일어나는 알러지(allergy) 또는 제1형 과민성 반응은 주로 IgE(immunoglobulin E) 항체가 관여하는 반응으로 체내 비만세포(mast cells) 및 호염구(basophils)의 세포 표면에 장착된 IgE에 특이적인 항원(알레르기 항원, allergen)이 결합하면, 이들 세포로부터 과민 상태와 염증을 유발하는 매개물질들이 방출된다. 과민성 반응의 주요 증상은 재채기(sneezing), 천명(wheezing) 및 천식(asthma) 등이 있으며, 아토피성 피부염 또는 피부 발진(두드러기, hives) 등이 있다.
면역계가 자기와 비자기를 구별하는 기능을 잃고 숙주를 면역적으로 공격하는 자가면역(autoimmunity)은 여러 가지 만성 쇠약성 질환의 원인이 된다. 주요 질환으로 다발성 경화증(multiple sclerosis) 류마티스 관절염(rheumatoid arthritis), 전신성 홍반루푸스(systemic lupus erythematosus), 제1형 당뇨병(Type 1 diabetes), 건선(Psoriasis), 하시모토씨 갑상선염(Hashimoto's thyroiditis) 및 최근 젊은 연령대에서 문제되는 급성 염증성 장질환[inflammatory bowel disease, IBD; Crohn's disease/ulcerative disease] 등이 있다.
인체의 면역계가 외래성 세포들이나 조직과 접촉했을 때, 이러한 외래성 요소를 제거하기 위해 강력히 반응한다. 그러나, 일부 경우에는, 비록 환자의 면역계가 외래성 요소로 인식하고 이를 거부하는 면역반응을 개시하더라도, 타 공여자로부터 제공된 세포, 조직 또는 장기의 이식이 환자의 질병에 대한 유일한 치료 방법일 수도 있다. 예를 들어, 미국에서만 매년 100,000명 이상의 사람들이 신장이식을 통한 치료를 받아야 하는 것으로 추산된다. 그러나, 면역계는 자기로 인식되지 않으면, 어떤 이식된 세포, 조직, 혹은 장기라 하더라도 면역거부반응(immunorejection response)을 통해 공격하고 제거한다. 따라서, 면역거부반응은 이러한 생명을 살리는 이식치료법의 심각한 장애요소가 되고 있다. 이식에 있어서 부가적인 위험은, 이식된 면역세포들(예, 면역능을 복구하기 위한 골수이식)이 새로운 숙주를 비자기로 인식하고 이에 대항하여 반응한다는 것이다. 이식편 대 숙주병(Graft-versus-host disease, GVHD)이라고 불리는 이 반응은 치명적일 수 있다.
따라서, 인체 면역계가 일으키는 과민성 반응(hypersensitivity), 자가면역질환(autoimmune diseases), 동종이식편(allografts)에 대한 면역거부반응(immunorejection) 및 이식편 대 숙주병 등과 같은 부적절한 면역반응을 조절하기 위하여, 면역억제 약물(면역억제제, immunosuppressants)을 사용하고 있다. 현재 사용되고 있는 면역억제제는 코르티코스테로이드계(프레드니손), 항증식제(마이코페놀레이트 모페틸 (CellCept®), 칼시네우린 저해제 [calcineurin inhibitors, 타크로리무스(Envarsus XR® 또는 Protopic), 사이클로스포린(Gengraf®, Neoral® 또는 Sandimmune®)], mTOR 저해제[sirolimus(Rapamun®)], 면역세포타깃 다중클론항체와 단일클론항체 등이 있다. 현재 사용되고 있는 면역억제제의 주요 문제점은 신장 기능 부전과 같은 타 장기 손상을 일으키는 부작용도 동반하고 효능면에서 만족스럽지 못하다 (Ojo et al., 2003; Bloom and Reese, 2007). 따라서, 약물의 부작용을 최소화하기 위한 안전하고 타깃 특이적 면역억제제 개발이 요구된다.
최근까지 인체 면역계에 대한 이해 증진과 생명공학의 발전에 힘입어, 임상적으로 면역억제제의 투여가 요구되는 과민성반응 환자, 자가면역질환 환자, 줄기세포 혹은 골수이식 환자를 포함한 장기이식 환자들에게는 부적절한 면역반응을 조절하기 위해서 여러가지 유형의 면역억제제들이 환자의 상태와 증상에 따라 단독 혹은 병용요법으로 치료에 활용되고 있다.
인체 면역기능을 인위적으로 억제시키는 목적으로 환자에게 사용되는 전형적인 면역억제제들의 작용 표적은 T 세포에 집중되어 있으며, 실제로는 T 세포들 중에서도 주로 CD4 T(TH) 세포의 기능 억제가 대표적인 작용기전이다. 이는 효과 TH 세포 유래의 사이토카인들이, T 세포가 주도적인 역할을 담당하는 세포성 면역반응은 물론이고 B 세포 유래의 항체 분자가 주도적인 역할을 담당하는 체액성 면역반응에도 중요하게 관련되기 때문이다. 특히, cytotoxic CD4+ 가 중요 질병의 원인이 된다고 보고된 쇼그렌증후군(Xiaoping Hong., 2020), 베체트병(Behcet's disease, Tong et al., 2019), 전신성 경화증(systemic sclerosis, Takashi Maehara., 2020), 다발성 근염(polymyositis, Paulius Venalis 1, Ingrid E Lundberg., 2013), 그리고 피부근염(dermatomyositis, Paulius Venalis 1, Ingrid E Lundberg; Jay Patel et al., 2021)에서의 T cells의 활성화 저해는 중요한 치료책이 될 수 있다.
따라서, 휴지기 TH 세포의 활성화, 증식, 및 효과 TH 세포로의 분화를 적절히 차단할 수 있는 수단에 의해서는 면역억제효과가 잘 발휘될 수 있다. 면역반응의 유도와 조절에 관련된 TH 세포의 중요성은, 인간 면역결핍바이러스(human immunodeficiency virus, HIV)에 감염되어 TH 세포의 기능 장애가 발생한 환자들에게 면역결핍증이 나타나는 현상에 의해서도 잘 증거된다.
현재까지 개발되어 있는 면역억제제의 사용에 의해 나타나는 부작용, 잠재적 위험, 혹은 합병증을 개선하기 위해서는, 부적절한 면역반응만을 특이적으로 억제할 수 있는 새로운 면역억제제의 개발이 계속 요구되고 있다.

본 발명은 상기와 같은 종래 기술의 문제점을 해결하기 위하여 안출된 것으로서, 본 발명에서 해결하고자 하는 과제는 면역 억제제의 주요 표적인 CD4 T 세포의 활성화, 증식 및 분화를 효과적으로 억제할 수 있는 신규한 면역 억제용 조성물, 면역성 질환의 예방 또는 치료용 조성물, 건강 기능 식품 및 기능성 화장료 조성물을 제공하고자 하는 것이다.
상기와 같은 과제를 해결하기 위하여, 본 발명은 3,6-안하이드로-L-갈락토오스(3,6-anhydro-L-galactose, L-AHG)를 유효성분으로 함유하는 면역 억제용 조성물을 제공한다.
상기 조성물은 CD4 T 세포의 활성화, 증식 또는 분화를 억제하는 것이 바람직하다.
상기 조성물은 G1 기 또는 S 기의 세포 주기 진행을 차단하는 것이 바람직하다.
상기 조성물은 TH1 세포 또는 TH2 세포로의 분화를 억제하는 것이 바람직하다.
상기 조성물은 인터페론-감마(INF-γ), 인터류킨-2(IL-2), 인터류킨-4(IL-4) 또는 인터류킨-13(IL-13)의 생성을 억제하는 것이 바람직하다.
또한, 본 발명은 3,6-안하이드로-L-갈락토오스(3,6-anhydro-L-galactose, L-AHG)를 유효성분으로 함유하는 면역성 질환의 예방 또는 치료용 약학적 조성물을 제공한다.
상기 면역성 질환은 과민성 면역 질환, 자가 면역 질환, 면역 거부 반응, 이식편 대 숙주 질환(graft versus host disease), 쇼그렌 증후군(sjogren's syndrome) 및 베체트병(behcet's disease)으로 이루어지는 군으로부터 선택되는 것이 바람직하다.

상기 과민성 피부 질환은 일광성 피부염, 접촉성 피부염 및 아토피성 피부염으로 이루어지는 군으로부터 선택되는 것이 바람직하다.
본 발명의 L-AHG를 포함하는 면역 억제용 조성물, 약학적 조성물, 건강 기능 식품 및 기능성 화장료 조성물은 면역 억제제의 주요 표적인 CD4 T 세포의 활성화, 증식 및 분화를 선택적이고 효과적으로 억제하는 우수한 효과가 있으므로 제약 산업, 식품 산업 및 화장품 산업에 매우 유용한 발명이다.
도 1은 마우스 비장으로부터 분리한 G0 T 세포의 CD4(TH) 및 CD8(TC) 세포의 분포를 나타낸다.
도 2는 아가로오스의 효소적 가수분해로부터 수득되는 L-AGH, NA2(neoagarobiose), NA4(heoagarotetraose) 및 NA6(neoagarohexose)의 immobilized anti-CD3/anti-CD28로 활성화된 T 세포의 증식 효과를 [3H] thymidine incorporation으로 평가한 결과를 나타낸다.
도 3은 L-AGH의 CD4 T 세포에서 세포 주기 진행을 특이적으로 차단하는 결과를 나타낸 것이다. G0 T 세포의 활성화 후 20시간이 경과되었을 때 nocodazole을 처리하였다.
도 4는 L-AGH의 nocodazole 처리하지 않은 CD4 T 세포에서 세포 주기 진행을 차단하는 양상을 나타낸 것이다.

이하, 본 발명을 상세하게 설명한다.
본 발명에서는 비-해양성 담수 환경에서 분리한 한천 분해 세균 Cellvibrio sp. KY-GH-1 균주(KCTC 13629BP)로부터 유래하는 재조합 GH16B β-아가레이즈(GH16B β-agarase), GH50A β-아가레이즈(GH50A β-agarase) [Kwon et al, 2020] 및 GH117 α-네오아가로비오스 가수분해효소(GH117A α-NABH) [Jang et al, 2021]를 병용하여 처리하는 효소공정으로 아가로오스를 당화시킨 후, 최종 분해 산물로 생성된 단량체인 3,6-안하이드로-L-갈락토오스(3,6-anhydro-L-galactose, L-AHG)와 D-갈락토오스(D-galactose) 혼합물을 Sephadex G-10 컬럼을 이용한 크기-배제 크로마토그래피(size-exclusion chromatography)로 분획하여 L-AHG를 최종 정제하였다. 정제한 L-AHG의 면역억제 활성(immunosuppressive activity)을 규명하기 위하여, 마우스 휴지기 (G0) T 세포의 in vitro 활성화 및 세포주기 개시 유도 모델을 활용하여, 휴지기 TH 세포의 활성화 및 증식에 미치는 L-AHG의 영향을 조사하였다. 그 결과, 휴지기 TH 세포가 T 세포 수용체 복합체(T cell receptor complex)에 결합하는 immobilized anti-CD3와 T 세포 공동자극수용체(T cell costimulatory receptor, CD28)에 결합하는 immobilized anti-CD28의 동시 자극에 의해 활성화되어 G0 기를 탈출하고 G1 → S → G2/M 기의 순서로 세포주기를 진행할 때, L-AHG 존재는 활성화된 TH 세포의 G1 기 및 S 기의 진행을 저해함으로써 T 세포의 증식을 감소시키는 면역억제효능을 발휘할 수 있음을 확인하였다.
따라서, 본 발명은 3,6-안하이드로-L-갈락토오스(3,6-anhydro-L-galactose, L-AHG)를 유효성분으로 함유하는 면역 억제용 조성물을 제공한다.
상기 유효성분은 아가로오스의 산가수분해 또는 효소적 분해로부터 수득될 수 있다.
바람직한 구체예로서, 본 발명의 유효성분은 아가로오스를 산가수분해 반응시켜 수득할 수 있다.
다른 바람직한 구체예로서, 본 발명의 유효성분은 한천 분해 세균 Cellvibrio sp. KY-GH-1 균주(KCTC 13629BP)로부터 유래하는 재조합 GH16B β-아가레이즈, GH50A β-아가레이즈, 및 GH117 α-네오아가로비오스 가수분해효소를 병용하여 처리하는 효소공정으로 아가로오스를 당화시킨 후, 최종 분해 산물로 생성된 단량체인 3,6-안하이드로-L-갈락토오스(3,6-anhydro-L-galactose, L-AHG)와 D-갈락토오스(D-galactose) 혼합물을 Sephadex G-10 컬럼을 이용한 크기-배제 크로마토그래피(size-exclusion chromatography)로 분획하여 수득할 수 있다.
또 다른 바람직한 구체예로서, 본 발명의 유효성분은 아가로오스의 아가레이즈-3,6-안하이드로-L-갈락토시데이즈-아라비노오즈 아이소머레이즈 효소 복합체 산물로부터 수득할 수 있다.
상기 설명된 L-AHG의 몇 가지 수득 방법 이외에 본 발명의 유효성분은 상업적으로도 입수할 수 있다. 예를 들어, Canada, Ontario, North York 소재의 Toronto Research Chemicals로부터 구입할 수 있다.
상기 조성물은 CD4 T 세포의 활성화, 증식 또는 분화를 억제하는 것이 바람직하다.
상기 조성물은 G1 기 또는 S 기의 세포 주기 진행을 차단하는 것이 바람직하다.
상기 조성물은 TH1 세포 또는 TH2 세포로의 분화를 억제하는 것이 바람직하다.
상기 조성물은 인터페론-감마(INF-γ), 인터류킨-2(IL-2), 인터류킨-4(IL-4) 또는 인터류킨-13(IL-13)의 생성을 억제하는 것이 바람직하다.
또한, 본 발명은 3,6-안하이드로-L-갈락토오스(3,6-anhydro-L-galactose, L-AHG)를 유효성분으로 함유하는 면역성 질환의 예방 또는 치료용 약학적 조성물을 제공한다.
상기 면역성 질환은 과민성 면역 질환, 자가 면역 질환, 면역 거부 반응, 이식편 대 숙주 질환(graft versus host disease), 쇼그렌 증후군(sjogren's syndrome) 및 베체트병(behcet's disease)으로 이루어지는 군으로부터 선택되는 것이 바람직하다.
상기 과민성 면역 질환은 알러지(allergy), 재채기(sneezing), 천명(wheezing), 천식(adthema), 아토피성 피부염 및 두드러기(hives)로 이루어지는 군으로부터 선택되는 것이 바람직하다.
상기 자가 면역 질환은 다발성 경화증(multiple sclerosis), 류마티스 관절염(rheumatoid arthritis), 전신성 홍반루프스(systemic lupus erythematosus), 제1형 당뇨병(Type 1 diabetes), 건선(Psoriasis), 하시모토시 갑상선염(Hashimoto’s thyroiditis), 및 전신성 홍반루프스(systemic lupus erythematosus) 및 급성 염증성 장 질환(acute inflammatory bowel disease)으로 이루어지는 군으로부터 선택되는 것이 바람직하다.
본 발명의 유효 성분을 포함하는 연역 억제용 조성물 및 약학적 조성물은 각각의 사용 목적에 맞게 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁제, 에멀젼, 시럽, 에어로졸 등의 경구 제형, 멸균 주사용액의 주사제 등 다양한 형태로 제형화하여 사용할 수 있으며, 경구 투여하거나 정맥 내, 복강 내, 피하, 직장, 국소 투여 등을 포함한 다양한 경로를 통해 투여될 수 있다.
이러한 약학적 조성물에는 추가적으로 담체, 부형제 또는 희석제 등이 더 포함될 수 있으며, 포함될 수 있는 적합한 담체, 부형제 또는 희석제의 예로는 락토오스, 덱스트로오스, 수크로오스, 솔비톨, 만니톨, 자일리톨, 에리쓰리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로스, 메틸 셀룰로스, 비정질 셀룰로스, 폴리비닐 피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유 등을 들 수 있다. 또한, 본 발명의 약학적 조성물은 충전제, 항응집제, 윤활제, 습윤제, 향료, 유화제, 방부제 등을 추가로 더 포함할 수도 있다.
바람직한 구체예로서, 경구 투여를 위한 고형 제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형 제제는 상기 면역 억제용 조성물 및 약학적 조성물에 적어도 하나 이상의 부형제, 예를 들면, 전분, 탄산칼슘, 수크로오스, 락토오스, 젤라틴 등을 혼합하여 제형화한다. 또한, 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 등과 같은 윤활제가 사용될 수도 있다.
바람직한 구체예로서, 경구용 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 예시될 수 있으며, 흔히 사용되는 단순 희석제인 물, 액체 파라핀 이외에 여러 가지 부형제, 예를 들면, 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다.
바람직한 구체예로서, 비경구 투여를 위한 제제에는 멸균된 수용액제, 비수성용제, 현탁제, 유제, 동결건조제, 좌제 등을 예시할 수 있다. 비수성용제, 현탁제에는 프로필렌글리콜, 폴리에틸렌글리콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 포함될 수 있다. 주사제에는 용해제, 등장화제, 현탁화제, 유화제, 안정화제, 방부제 등과 같은 종래의 첨가제가 포함될 수 있다.
본 발명의 유효 성분은 약제학적으로 유효한 양으로 투여한다. 본 발명에서, "약제학적으로 유효한 양"은 의학적 치료에 적용 가능한 합리적인 수혜/위험 비율로 질환을 치료하기에 충분한 양을 의미하며, 유효 용량 수준은 환자의 질환의 종류, 중증도, 약물의 활성, 약물에 대한 민감도, 투여 시간, 투여 경로 및 배출 비율, 치료 기간, 동시 사용되는 약물을 포함한 요소 및 기타 의학 분야에 잘 알려진 요소에 따라 결정될 수 있다. 본 발명의 면역 억제용 조성물 및 약학적 조성물은 개별 치료제로 투여하거나 다른 치료제와 병용하여 투여될 수 있고, 종래의 치료제와 순차적으로 또는 동시에 투여될 수 있으며, 단일 또는 다중 투여될 수 있다. 상기한 요소들을 모두 고려하여 부작용 없이 최소한의 양으로 최대 효과를 얻을 수 있는 양을 투여하는 것이 중요하며, 이는 당업자에 의해 용이하게 결정될 수 있다.
바람직한 구체예로서, 본 발명의 면역 억제용 조성물 및 약학적 조성물에서, 유효성분의 유효량은 환자의 나이, 성별, 체중에 따라 달라질 수 있으며, 일반적으로는 체중 ㎏ 당 1mg 내지 1,000mg, 바람직하게는 5mg 내지 100mg, 보다 바람직하게는 10mg 내지 20mg을 매일 또는 격일 투여하거나 1일 1 내지 3회로 나누어 투여할 수 있다. 그러나, 투여 경로, 질병의 중증도, 성별, 체중, 연령 등에 따라서 증감될 수 있으므로 상기 투여량이 어떠한 방법으로도 본 발명의 범위를 한정하는 것은 아니다.
본 발명의 면역 억제용 조성물 및 약학적 조성물은 다양한 경로를 통하여 대상에 투여될 수 있다. 투여의 모든 방식은 예상될 수 있는데, 예를 들면, 경구, 직장 또는 정맥, 근육, 피하, 자궁내 경막 또는 뇌혈관 내(intracerebroventricular) 주사에 의해 투여될 수 있다.
본 발명에서 "투여"는 임의의 적절한 방법으로 환자에게 소정의 물질을 제공하는 것을 의미하며, 본 발명의 면역 억제용 조성물 및 약학적 조성물의 투여 경로는 목적 조직에 도달할 수 있는 한 일반적인 모든 경로를 통하여 경구 또는 비경구 투여될 수 있다. 또한, 본 발명의 면역 억제용 조성물 및 약학적 조성물은 유효성분을 표적 세포로 전달할 수 있는 임의의 장치를 이용해 투여될 수도 있다.
본 발명에서 "대상"은, 특별히 한정되는 것은 아니지만, 예를 들어, 인간, 원숭이, 소, 말, 양, 돼지, 닭, 칠면조, 메추라기, 고양이, 개, 마우스, 쥐, 토끼 또는 기니아 피그를 포함하고, 바람직하게는 포유류, 보다 바람직하게는 인간을 의미한다.
또한, 본 발명은 3,6-안하이드로-L-갈락토오스(3,6-anhydro-L-galactose, L-AHG)를 포함하는 면역성 질환의 예방 또는 개선용 건강 기능 식품을 제공한다.
상기 면역성 질환은 과민성 면역 질환, 자가 면역 질환, 면역 거부 반응, 이식편 대 숙주 질환(graft versus host disease), 쇼그렌 증후군(sjogren's syndrome) 및 베체트병(behcet's disease)으로 이루어지는 군으로부터 선택되는 것이 바람직하다.

본 발명의 건강 기능 식품은 면역성 질환의 예방 또는 개선에 효과적인 식품 및 음료 등에 다양하게 이용될 수 있다. 본 발명의 유효성분을 포함하는 식품으로는, 예를 들어, 각종 식품류, 음료, 껌, 차, 비타민 복합제, 건강보조 식품류 등이 있고, 분말, 과립, 정제, 캡슐 또는 음료인 형태로 사용할 수 있다.
본 발명의 유효성분은 일반적으로 전체 식품 중량의 0.001 내지 10중량%로 가할 수 있으며, 건강음료 조성물은 100 ml를 기준으로 0.001 내지 10g, 바람직하게는 0.01 내지 1g의 비율로 가할 수 있다.
본 발명의 건강 기능 식품은 지시된 비율로 필수 성분으로서 상기 화합물을 함유하는 것 외에 식품학적으로 허용 가능한 식품보조 첨가제, 예컨대, 천연 탄수화물 및 다양한 향미제 등을 추가 성분으로서 함유할 수 있다.
상기 천연 탄수화물의 예로는 포도당, 과당 등의 단당류, 말토오스, 수크로오스 등의 이당류 및 덱스트린, 시클로덱스트린 등의 다당류와 같은 통상적인 당 및 자일리톨, 소르비톨, 에리쓰리톨 등의 당알코올이 있다.
상기 향미제로는 타우마틴, 레바우디오시드 A 또는 글리시르히진과 같은 스테비아 등의 천연 향미제 및 사카린, 아스파르탐 등의 합성 향미제를 사용할 수 있다. 상기 천연 탄수화물의 비율은 본 발명의 건강 기능 식품 100 ml 당 일반적으로 약 1 내지 20g, 바람직하게는 약 5 내지 12g을 사용한다.
상기 외에 본 발명의 건강 기능 식품은 여러 가지 영양제, 비타민, 광물, 합성 풍미제 및 천연 풍미제 등의 풍미제, 착색제 및 중진제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산음료에 사용되는 탄산화제 등을 함유할 수 있다.
그 밖에 본 발명의 건강 기능 식품은 천연 과일 주스 및 과일 주스 음료 및 야채 음료 등의 제조를 위한 과육을 함유할 수도 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 본 발명의 유효성분 100중량부 당 10 내지 약 50중량부의 범위에서 선택되는 것이 일반적이다.
또한, 본 발명은 3,6-안하이드로-L-갈락토오스(3,6-anhydro-L-galactose, L-AHG)를 유효성분으로 함유하는 과민성 피부 질환의 예방 또는 개선용 기능성 화장료 조성물을 제공한다.
상기 과민성 피부 질환은 일광성 피부염, 접촉성 피부염 및 아토피성 피부염으로 이루어지는 군으로부터 선택되는 것이 바람직하다.
본 발명의 화장료 조성물의 유효성분은 조성물 총 중량에 대해 0.001 내지 1중량%로 포함될 수 있으며, 바람직하게는 0.01 내지 0.1중량%, 더욱 바람직하게는 0.05 내지 0.5중량%이다. 이러한 함량 범위에서 적절한 제형 안정성을 확보할 수 있으며, 목적하는 항산화 효과를 기대할 수 있다.
본 발명의 상기 조성물은 본 발명의 유효성분 외에도 본 발명의 유효 활성을 저해하지 않는 범위에서 항산화, 피부 노화 방지 및 피부 재생 촉진의 효과를 위한 공지의 천연물 추출물이나 기타 성분을 추가로 포함시켜 이용할 수 있다.
또한, 상기 조성물은 화장료 조성물에 통상적으로 첨가되는 성분들, 예를 들면, 지방 물질, 유기 용매, 용해제, 농축제, 겔화제, 연화제, 항산화제, 현탁화제, 안정화제, 발포제(foaming agent), 방향제, 계면활성제, 물, 이온형 또는 비이온형 유화제, 충전제, 금속이온 봉쇄제 및 킬레이트화제, 보존제, 비타민, 차단제, 습윤화제, 필수 오일, 염료, 안료, 친수성 또는 친유성 활성제와 같은 화장품학 또는 피부과학 분야에서 통상적으로 사용되는 보조제나 담체를 포함하여 특정 제형으로 성형될 수 있다.
본 발명의 화장료 조성물은 당업계에서 통상적으로 제조되는 어떠한 제형으로도 제조될 수 있으며, 예를 들면, 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 팩, 비누, 계면활성제-함유 클린징, 오일 및 스프레이 등으로 제형화될 수 있으나, 이에 한정되는 것은 아니다. 보다 구체적으로는, 유연 화장수, 영양 화장수, 영양 크림, 마사지 크림, 에센스, 아이 크림, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션, 클렌징 크림, 클렌징 폼, 클렌징 워터, 팩, 스프레이, 파우더, 팩트, 립글로즈, 립스틱, 섀도우, 샴푸 및 린스 등의 제형으로 제조될 수 있다.
본 발명의 제형이 페이스트, 크림 또는 겔인 경우에는 담체 성분으로서 동물성유, 식물성유, 왁스, 파라핀, 전분, 트라칸트, 셀룰로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
본 발명의 제형이 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토스, 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 이용될 수 있고, 특히, 스프레이인 경우에는 추가적으로 클로로플루오로히드로카본, 프로판/부탄 또는 디메틸 에테르와 같은 추진체를 포함할 수 있다.
본 발명의 제형이 용액 또는 유탁액인 경우에는 담체 성분으로서 용매, 용해화제 또는 유탁화제가 이용되고, 예컨대, 물, 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코올, 벤질 벤조에이트, 프로필렌 글리콜, 1,3-부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 있다.
본 발명의 제형이 현탁액인 경우에는 담체 성분으로서 물, 에탄올 또는 프로필렌 글리콜과 같은 액상의 희석제, 에톡실화이소스테아릴 알코올, 폴리옥시에틸렌 소르비톨 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀룰로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가 또는 트라칸트 등이 이용될 수 있다.
본 발명의 제형이 계면-활성제 함유 클린징인 경우에는 담체 성분으로서 지방족 알코올 설페이트, 지방족 알코올 에테르설페이트, 설포숙신산 모노에스테르, 이세티오네이트, 이미다졸리늄 유도체, 메틸타우레이트, 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인, 지방족 알코올, 지방산 글리세리드, 지방산 디에탄올아미드, 식물성 유, 라놀린유도체 또는 에톡실화 글리세롤 지방산 에스테르 등이 이용될 수 있다.
또한, 본 발명은 L-AHG를 포함하는 T 세포의 활성화, 증식 또는 분화 억제용 시약 조성물을 제공한다.
상기 T 세포는 CD4 T 세포인 것이 바람직하다.
상기 활성화는 휴지기 T 세포의 활성화를 의미한다.
상기 증식은 T 세포의 세포주기 진행에 의한 개체수의 증가를 의미한다.
상기 분화는 TH1 세포 및 TH2 세포로의 분화를 의미한다.
상기 조성물은 생체 외 조건, 바람직하게는 시험관 내(in vitro) 조건에서 T 세포에 적용함으로서 T 세포의 활성화, 증식 및 분화를 억제시킬 수 있다.
또한, 본 발명은 생체 외(ex vivo) 조건 하에서, T 세포에 본 발명의 L-AHG를 처리하는 것을 특징으로 하는 T 세포, 바람직하게는 CD4 T 세포의 활성화, 증식 또는 분화 억제 방법을 제공한다.
상기 생체 외 조건은 T 세포가 생체로부터 물리적으로 완전히 분리되어 존재하는 상태를 의미하며, 시험관 내(in vitro) 조건인 것이 바람직하다.
상기 T 세포는 개체의 혈액으로 분리된 세포이거나, 이를 계대 배양한 세포이거나, 세포주일 수도 있다.
상기 처리는, 예를 들어, 세포 배양 접시 내의 T 세포를 포함하는 배지에 L-AHG를 첨가함으로써 수행될 수 있고, 처리 농도는 1.0 내지 1,000 μg/mL, 바람직하게는 12.5 내지 400 μg/mL인 것이 바람직하다.
상기 분화 억제는 TH1 및 TH2 세포로의 분화 억제일 수 있다. TH1 및 TH2 세포 분화 억제에 의하여 인터페론-감마(INF-γ), 인터류킨-2(IL-2), 인터류킨-4(IL-4) 또는 인터류킨-13(IL-13)의 분비량이 감소될 수 있다.
이하에서는, 구체적인 실시예를 통하여 본 발명을 더욱 상세하게 설명한다. 하기 실시예는 본 발명의 바람직한 일 구체예를 기재한 것이며, 하기 실시예에 기재된 사항에 의하여 본 발명의 권리범위가 한정되어 해석되는 것은 아니다.
[실시예]
1. 재료 및 방법
1.1. 동물
5~6주령 C57BL/6J 수컷 마우스는 오리엔트 사(경기도 성남시 중원구 갈마치로 322, 한국)에서 구입하여 경북대학교 실험동물자원 관리센터(대구시 중구 국채보상로 680, 한국)에서 특정 병원균이 없는 조건에서 유지하면서 실험에 사용하였다.
1.2. 화학물질, 항체, 시약 및 배지
마우스의 재조합 IL-2(rIL-2)는 Cambridge Bioscience (Cambridge, UK)에서 구입했다. 토끼 유래 anti-마우스 IgG는 Jackson ImmunoResearch (West Grove, PA, USA)에서, anti-CD3는 BD Pharmingen (Sandiego, CA, USA)에서 구입했다. IL-2 활성을 중화시키기 위한 염소 유래 anti-마우스 IL-2 항체는 R&D 시스템 (Minneapolis, MN, USA)에서 구입하였고, 렛트 유래 항-마우스 IL-2R 중화 항체는 Abcam (Cambridge, UK)에서 구입하였다. ECL 웨스턴 블로팅 키트는 Thermo Fisher Scientific (Rockford, IL, USA)에서 구입했다. 웨스턴 분석을 위한 Immobilon-P 막은 EMD Millipore Corporation (Temecula, CA, USA)에서 구입하였다. 미세소관 탈중합 약제인 노코다졸(nocodazole) [Han et al, 2013] 및 기타 모든 전기영동용 시약은 Sigma Chemical (St. Louis, MO, USA)에서 구입하였다.
아가로오스 가수분해산물인 네오아가로테트라오스 (NA4) 및 네오아가로헥사오스 (NA6)를 확보하기 위해, 열처리로 물에 용해시킨 9.0% 아가로오스를 GH16B β-아가레이즈 (1.6 μg/mL)로 최적의 반응 조건 (1 mM MnCl2 및 10 mM TCEP, 50 mM Tris-HCl, pH 7.0, 50°C) 에서 자석교반기로 계속 저으면서 14시간 동안 가수분해하여 NA4와 NA6를 생성시켰다. 그 생성 NA4/NA6 혼합물을 Sephadex G-15 column을 이용한 크기-배제 크로마토그래피로 분획하여 정제하였다 [Lee et al, 2022, in preparation]. 아가로오스로부터 네오아가로비오스 (NA2)를 확보하기 위해, 9% 아가로오스를 GH16B β-아가레이즈로 50℃에서 처리하여 NA4/NA6를 포함한 네오아가로올리고당 (neo-agarooligosaccharides, NAOSs)으로 1차 가수분해시키고, 이어서 온도를 35℃로 낮춘 다음 GH50A β-아가레이즈 (20 μg/mL)를 첨가하여 14시간 동안 반응시킴으로써 NA2를 생성시켰다. 반응 생성물 중의 NA2를 Sephadex G-15 column을 이용한 크기-배제 크로마토그래피로 분획하여 정제하였다. 또한, 아가로오스의 구성 단량체인 L-AHG를 확보하기 위해서는, 먼저 9% 아가로오스를 GH16B β-아가레이즈로 50℃에서 처리하여 NA4와 NA6를 포함한 NAOSs로 1차 가수분해시키고, 이어서 온도를 35℃로 낮춘 다음 GH50A β-아가레이즈 (20 μg/mL)와 GH117A α-NABH (60 μg/mL)를 동시에 첨가하여 14시간 동안 반응시킴으로써 최종 생성물로서 단량체인 L-AHG와 D-칼락토오스의 혼합물을 얻었다. 이 혼합물 중의 L-AHG를 최종 정제하기 위해서 Sephadex G-10 컬럼을 이용한 크기-배제 크로마토그래피로 분획하였다.
T 림프구 활성화 및 배양에 사용된 배지는 10% FBS (HyClone, Logan Utah, USA), 20 mM HEPES, 50 μM β-mercaptoethanol, 100 μg/mL gentamycin을 함유한 RPMI 1640으로, 완전배지로 명명하였다.
1.3. 휴지기 (G0) T 세포의 분리, 활성화 및 [

H]thymidine 삽입 측정
4~5 개월된 C57BL/6J 수컷 마우스 유래의 비장으로부터 휴지기 (G0) T 림프구의 분리는 T 세포 정제 컬럼 키트 (R&D 시스템, Minneapolis, MN, 미국)를 사용하여서 비장세포들로부터 음성선택 (negative selection)의 방법으로 분리하였다. 이때 G0 T 세포의 활성화를 위해서는 전술한 Kim 등의 방법 [Kim et al. 1992]에 변경하여 수행하였으며, 이때 배양 플라스틱 용기의 표면에 anti-CD3 (2.0 μg/mL) 및 anti-CD28 (2.0 μg/mL)를 원활히 코팅하기 위해, rabbit anti-hamster IgG 항체 (20 μg/mL)로 전처리하였다.
Immobilized anti-CD3/anti-CD28에 의해 활성화된 T 세포의 증식을 평가하기 위해, anti-CD3 (2.0 μg/mL)와 anti-CD28 (2.0 μg/mL)로 코팅된 96-웰 플레이트의 각 웰에 1 × 105개의 G0 T 세포와 각 농도별 L-AHG를 200 μL RPMI 1640 완전배지와 함께 넣고 배양함으로써 활성화를 시작하였으며, 활성화 후 29시간이 경과하였을 때, 각 웰에 0.25 μCi의 [3H] thymidine을 넣고 12 시간 동안 추가로 더 배양하면서 증식하는 T 세포의 DNA 내로 삽입되는 [3H]thymidine의 양을 Liquid scintillation counter로 정량하였다.
Immobilized anti-CD3/anti-CD28에 의해 활성화된 T 세포의 세포주기를 분석하거나 배양 상등액 속의 사이토카인 농도를 분석하기 위해, anti-CD3 (2.0 μg/mL)와 anti-CD28 (2.0 μg/mL)로 코팅된 35-mm 플레이트의 각 웰에 3 × 106개의 G0 T 세포와 각 농도별 L-AHG를 2 ml의 RPMI 1640 완전배지와 함께 넣고 배양함으로써 활성화를 시작하였으며, 활성화 후 30시간이 경과하였을 때, 배양을 중단하고 원심분리에 의해 세포와 배양상등액을 회수하였다.
동일한 배양조건에서, G0 T 세포의 활성화에 의해 개시되는 첫번째 세포주기진행이 nocodazole 처리에 의해 전중기 (prometaphase)에 정지되도록 하기 위해, T 림프구를 20시간 동안 활성화시킨 후 nocodazole을 0.2 μg/mL의 농도로 첨가하였으며, 이어서 추가로 10시간 동안 더 배양하였다. 배양 종료 후 배양물을 원심분리하여 세포와 배양 상등액을 회수하였다.
1.4. 형광표지 항-CD4/항-CD8 항체를 이용한 T 세포 표면 염색 및 DAPI를 이용한 세포 내 DNA 염색
마우스 비장세포들로부터 T 세포 정제 컬럼 키트 (R & D)를 사용하여 분리한 휴지기 (G0) T 세포와 G0 T 세포를 immobilized anti-CD3/anti-CD28로써 주어진 시간 동안에 활성화시킨 T 세포를 대상으로 anti-CD3, anti-CD4, anti-CD8, 및 DAPI 염색을 수행하였다. 약 5 × 105 세포를 100 μL 1X PBS/1% FBS 용액에 현탁 한 후 anti-CD3, anti-CD4 및 anti-CD8를 0.5 μg 씩 첨가하고 4℃에서 15분간 반응시키고 1X PBS/1% FBS 용액으로 2회 세척하였다. 이어서 세포를 1% paraformaldehyde용액으로 4℃에서 15분간 고정하였으며, 고정한 세포를 1X PBS 용액으로 2회 세척한 후 10 μM DAPI로 15분간 염색하였다. 염색된 세포를 유세포 분석기 (Attune NxT flow cytometer, Thermo Fisher Scientific, Waltham, MA, USA)로 측정하여 CD3 T 세포, CD4 T 세포, CD8 T 세포의 비율과 세포내 DNA에 결합한 DAPI 양에 따른 청색 형광 증가에 근거한 세포주기분포를 분석하였다.
1.5. 효소 결합 면역흡착 분석 (ELISA)
T 세포의 배양상등액을 회수한 후, ELISA 키트 (Mouse Uncoated Elisa Kit, Invitrogen)를 이용하여 제조사의 치침에 따라, IFN-γ, IL-2, IL-4 및 IL-13의 농도를 분석하였다. 요약하면, 먼저 포획항체 (capture antibody)가 희석된 용액을 96-웰 플레이트의 각 웰에 100 μl씩 넣고 4℃에서 16시간 처리한 후, 세척용액으로 3회 세척하였다. 각 웰에 200 μl의 블로킹 용액을 넣고 25℃에서 1시간 동안 블로킹하고 세척용액으로 1회 세척하였다. 이어서 표준시료 및 배양상등액을 적절히 희석한 용액을 각 웰에 100 μl씩 넣고 25℃에서 2시간 동안 반응시켰다. 세척용액으로 3회 세척한 다음, 2차 항체 (detection antibody, 검출항체) 용액 100 μl를 각 웰에 넣고 25ºC에서 1시간동안 반응시켰다. 세척용액으로 3회 세척한 후, 각 웰에 100 μl의 아비딘-HRP (Avidin-HRP) 용액을 넣고 25℃에서 30분간 반응시켰다. 세척용액으로 5회 세척한 다음, 각 웰에 100 μl의 기질용액을 넣고 빛을 차단한 상태로 25℃에서 15분간 반응시켰다. 각 웰에 100 μl 반응정지액을 첨가하여 반응을 종료시키고, 플레이트 리더 (Varioskan Lux Multimode Microplate Reader, Thermo Fisher Scientific)로 450 nm 및 570 nm 파장에서 흡광도를 측정하였다. 이때 450 nm 파장에서 측정된 흡광도에서 570 nm 파장에서 측정한 흡광도를 뺀 값을 표준곡선과 비교 환산하여 배양상등액에 존재하는 사이토카인의 농도를 산출하였다. 표준곡선 작성을 위해, 연속 희석된 재조합 IFN-γ, IL-2, IL-4 및 IL-13을 표준시료로 사용하였다.
2. 결과
2.1. 고정화 anti-CD3/anti-CD28에 의해 자극된 G0 T 세포의 [

H] 티미딘 삽입에 미치는 아가로스의 효소적 가수분해물의 영향
마우스 비장으로부터 휴지기 (G0) 림프구의 분리는 퍼콜 농도구배 원심분리 (Percoll gradient centrifugation) 방법 [Proust et al, 1987]으로 수행하였다. G0 림프구로부터 G0 T 림프구의 분리는 T 세포 정제 컬럼 키트 (R&D Systems, Minneapolis, MN, 미국)를 사용하여, 음성선택 (negative selection)의 방법으로 분리하였다 [Kim et al, 1992]. T 세포 정제 컬럼으로 분리된 G0 T 세포는 형광 표지된 anti-CD3, anti-CD4, 및 anti-CD8 항체들을 이용하여 표면 염색한 결과, 96% 세포가 CD3+인 T 세포로 확인되었다 (도 1). 또한, 이들 CD3+ T 세포 중에서 50.6% 가 CD4+ T (TH) 세포로, 그리고 43.8% 가 CD8+ T (TC) 세포로 확인되었다.
분리한 휴지기 T 세포를 96-웰 플레이트에서 immobilized anti-CD3/CD28로 자극하여 활성화시켰다. 이때 아가로오스를 GH16B β-아가레이즈로 가수분해시켜 확보한 NA4 및 NA6, 아가로오스를 GH16B β-아가레이즈로 1차 가수분해시킨 후 GH50B β-아가레이즈를 추가적으로 작용시켜 확보한 NA2, 그리고 아가로오스를 GH16B β-아가레이즈로 1차 가수분해시킨 후 GH50B β-아가레이즈 및 GH117A α-NABH를 추가적으로 동시에 작용시켜 확보한 L-AHG등을 12.5~400 μg/mL 농도로 각각 첨가하여, 휴지기 T 세포의 활성화에 따른 세포주기진행 및 증식에 미치는 이들 아가로오스 분해산물들 (L-AHG, NA2, NA4, NA6)의 영향을 조사하였다.
먼저, 96-웰 플레이트를 사용하여 휴지기 T 세포 (1 × 105 G0 T cells/well)를 여러 농도(0, 12.5, 25, 50, 100, 200, 및 400 μg/mL)의 L-AHG, NA2, NA4, 혹은 NA6의 존재 하에서 immobilized anti-CD3/anti-CD28로써 자극하여 활성화시켰다. 활성화 후 29시간이 경과하였을 때, 각 웰당 0.25 μCi의 [3H]thymidine을 넣고 12 시간 동안 추가로 더 배양하였으며, 이 추가 배양시간 동안에 일어나는 T 세포 증식에 따른 DNA에 삽입된 [3H]thymidine의 양을 Liquid scintillation counter로 정량함으로써 각 웰에서 일어난 T 세포의 증식 정도를 측정하였다.
그 결과로서, 도 2에서 보는 바와 같이, 12.5 μg/ml L-AHG는 활성화된 T 세포의 증식에 별다른 영향을 끼치지 못하여 [3H]thymidine 삽입의 수준이 L-AHG 무첨가 대조구의 경우와 거의 동일하였다. 이에 반해, 25~400 μg/ml L-AHG의 존재는 농도 의존적으로 [3H]thymidine 삽입을 저해하였다. 특히, 100 μg/mL L-AHG 존재 시에는 [3H]thymidine 삽입이 L-AHG 무첨가 대조구의 약 22% 수준으로 감소되었고, 200~400 μg/mL 농도에서는 [3H]thymidine 삽입이 거의 측정되지 않는 수준으로 현저하게 감소되는 것으로 나타났다. 그러나, 동일한 조건에서 NA2, NA4 및 NA6 등과 같은 아가로오스 분해산물들의 경우는 25~400 μg/mL 농도에서 활성화된 T 세포의 [3H]thymidine 삽입에 대해 크게 영향을 끼치지 않는 것으로 나타났다.
따라서, 이러한 결과로써, 조사한 아가로오스의 효소적 가수분해산물들 중에서 L-AHG 만 활성화된 T 세포의 증식을 현저하게 억제하는 생리활성이 있음을 확인하였다.
최근까지 한천 분해산물들 (NA2, L-AHG, NAOS, AOS)의 다양한 생리적 활성이 보고되었다. 즉, 항산화 [Chen and Yan 2005; Xu et al. 2018], 프리바이오틱 [Hu et al. 2006; Yun et al. 2021], 항종양 [Enoki et al. 2012; Lee et al. 2017; Yun et al. 2021], 항염증 [Wang et al. 2017; Yun et al. 2013], 항당뇨 및 항비만 [Hong et al., 2017], 그리고 피부 보습 및 미백 [Kim et al., 2017b; Kobayashi et al. 1997; Yun et al. 2013] 등의 활성이다. 이외에도 마우스 모델에서 NA4의 경구 투여가, 장내 미생물총 (마이크로바이오타)의 조절을 통해, 격렬한 운동으로 인한 피로 [Zhang et al. 2017]와 간 손상 [Chen et al. 2019]에 대한 보호 효과가 있음이 보고되었다. 그러나, 활성화된 T 세포의 증식에 대한 L-AHG의 억제 효능은 현재까지 보고된 바가 없는 새로운 생리적 활성이다.
2.2. 고정화 anti-CD3/anti-CD28에 의해 자극된 G0 T 세포의 세포주기진행에 미치는 L-AHG의 영향
면역계를 구성하는 T 림프구들은 CD4 T (TH) 림프구와 CD8 T (TC) 림프구의 두 유형으로 크게 나눌 수 있다. [3H]thymidine 삽입 측정법으로 확인된 결과로서, L-AHG가 보여주는 활성화된 T 세포의 DNA 합성 저해를 통한 증식 억제 활성이 CD4 T (TH) 세포와 CD8 T (TC)세포에서 공통적으로 일어나는지, 혹은 두 유형의 T 세포들 중 어느 한 유형에만 국한되어 일어나는지를 조사하였다.
이를 위해, 휴지기 T 세포를 immobilized anti-CD3/anti-CD28로 자극하여 활성화시켜서 휴지기 탈출과 G1 기 진입 및 진행에 뒤이은 S 기의 개시 시점에 도달하였을 때, 즉 활성화 후 20 시간이 경과되었을 때, 미세소관 탈중합 약제인 nocodazole [노코다졸, 진핵세포의 세포주기진행을 유사분열기 (M phase)의 전중기 (prometaphse)에서 정지시키는 세포주기 정지제] [Han et al, 2013]을 첨가하여 10시간을 더 배양하였다. 이와 같은 독특한 실험 방법을 통해, 휴지기 T 세포가 활성화됨에 따라 개시하는 첫 번째 세포주기의 진행이 G1, S, 및 G2 기를 거친 후 nocodazole의 영향에 의한 미세소관 손상 때문에 M 기의 전중기에서 정지되도록 하였다. 이어서 이들 세포를 회수한 다음, anti-CD4 형광 항체 및 anti-CD8 형광 항체, 그리고 DNA 형광염색시약인 DAPI를 처리하여 염색하고 유세포분석기 (flow cytometer)로 분석하였으며, 그 결과로써 휴지기 CD4 T 세포와 휴지기 CD8 T 세포가 활성화됨에 따라 개시하게 되는 첫 번째 세포주기진행 (G1, S, 및 G2/M 기)의 양상이 L-AHG에 의해 어떤 영향을 받는지를 규명하고자 하였다.
도 3의 A와 B에서 보는 바와 같이, 염색된 세포를 유세포분석기 (flow cytometry)로 분석한 결과, 휴지기 T 세포를 immobilized anti-CD/anti-CD28로 자극하여 활성화시켰을 때, CD8 T 세포는 4.4%, 8.8%, 77.9%의 세포가 G1, S, G2/M 기에 각각 분포하는데 반해, CD4 T 세포는 9.9%, 35.4%, 49.9%의 세포가 G1, S, G2/M 기에 분포하는 것으로 나타났다. 즉, CD8 T 세포는 활성화 후 30시간이 경과되었을 때에 G2/M 기로 진입하지 못하고 G1 + S 기에 머물러 있는 세포의 비율이 13.2% 정도에 불과하였으나, CD4 T 세포는 G1 + S 기에 머물러 있는 세포의 비율이 45.3% 정도로 높았다. 이러한 결과는 휴지기 T 세포들 중에서 CD4 T 세포에 비해 CD8 T 세포가 immobilized anti-CD/anti-CD28에 의한 활성화 후에 세포주기를 비교적 더 신속하고 원활히 진행함을 보여준다.
이와 같이 확인된 휴지기 CD4 T 및 CD8 T 세포들의 활성화에 따른 첫 번째 세포주기진행은 25~100 μg/ml 농도의 L-AHG 첨가에 의해 서로 현저히 다르게 영향을 받는 것으로 나타났다. 즉, 휴지기 CD4 T 세포는 immobilized anti-CD3/anti-CD28에 의한 활성화 후에 G2/M 기로 진입하지 못하고 G1 및 S 기에 남아있는 세포의 비율이 L-AHG의 농도에 의존적으로 증가하였다. 특히, 100 μg/mL 농도의 L-AHG가 첨가되었을 때는 G1 와 S 기 세포의 비율이 각각 35.4%와 54.6% 였고, 동시에 G2/M 기 세포의 비율은 단지 8.8% 정도로 나타나, L-AHG에 의해 세포주기 진행이 현저하게 억제됨을 알 수 있었다. 이에 반해, 휴지기 CD8 T 세포는 활성화 후의 세포주기 진행은 25~50 μg/mL L-AHG 첨가에 의해서 거의 영향을 받지 않았으며, 100 μg/mL L-AHG 첨가에 의해서는 G1 기 세포의 비율 변화는 무시할 정도이면서 S 기 세포의 비율이 46.6%로 증가하였고 G2/M 세포의 비율은 41.8% 였다.
이러한 연구결과는, 휴지기 CD4 T 세포의 활성화에 따른 세포주기진행이 L-AHG (25~100 μg/mL)의 첨가에 의해 농도 의존적으로 주로 G1 및 S 기에서 저해됨을 나타내며, 특히 100 μg/ml L-AHG를 첨가 시에는 G2/M 기로 진입하지 못하고 G1 + S 기에 남아있는 세포의 비율이 전체의 약 90.0%임을 보여준다. 동일한 조건에서, 휴지기 CD8 T 세포의 활성화에 따른 세포주기진행은 25~50 μg/mL L-AHG 첨가에 의해서는 별다른 영향을 받지 않았으며, 100 μg/mL L-AHG를 첨가 시에도 G1 기의 진행은 영향을 받지 않았으나 S 기의 진행이 다소 저해됨을 보여준다. 이와 같이, 비록 휴지기 CD4 T 및 CD8 T 세포들의 활성화에 따른 세포주기진행이 L-AHG (25~100 μg/mL) 존재에 의해 서로 다른 양상으로 저해되었지만, 이들 세포에 있어서 L-AHG에 의한 아폽토시스성 세포사멸 (apoptotic sub-G1 세포의 비율에 근거해 볼 때)은 유도되지 않는 것으로 확인되었다.
한편, 동일한 조건으로 휴지기 T 세포를 immobilized anti-CD3/anti-CD28로 자극하여 활성화시키고 nocodazole의 첨가 없이 30시간 동안 배양하였을 때는, 휴지기 CD4 T 및 CD8 T 세포의 활성화에 따른 세포주기진행에 미치는 L-AHG의 저해효과를 유세포분석으로 확인하는 것이 용이하지 않은 것으로 나타났다 (도 4의 A와 B). 이는 휴지기 CD4 T 세포 혹은 휴지기 CD8 T 세포가 활성화되어 개시하게 되는 세포주기진행이 일부 세포들은 L-AHG의 존재에 의해 저해되는 영향을 받지만, 나머지 세포들은 L-AHG에 의해 저해되는 영향을 받지 않고 첫 번째 세포주기진행을 완료하고 재차 G1, S, 및 G2 기 단계로 진행하게 되어, 결국에는 L-AHG에 의해 세포주기진행이 저해된 세포들과 L-AHG의 존재에 의해서도 세포주기진행이 일어나는 세포들이 함께 섞여서 존재하는 이유 때문으로 보인다.
현재의 연구결과를 종합하면, 활성화된 T 세포의 증식에 미치는 L-AHG의 억제 효능은 CD8 T 세포가 아니라 CD4 T 세포를 주요 표적으로 하며, 그 세포주기의 진행을 G1 기 및 S 기에서 저해하기 때문임을 시사한다.
2.3. 고정화 anti-CD3/anti-CD28에 의해 자극된 G0 T 세포의 사이토카인 생성에 미치는 L-AHG의 영향
CD4 T 세포는, CD8 T 세포와는 달리, 기능면에서 차별화되는 여러 가지 효과 CD4 T 세포 아형들로 분화되어 다양한 면역반응을 조율한다. 효과 CD4 T 세포 아형으로는 TH1, TH2, TH17, TFH 및 Treg 세포 등이 알려져 있다. 이들 각 아형들은 분비하는 사이토카인 조합의 특징을 기준으로 서로 구별할 수 있다.
TH1 세포는 IFN-γ를 생산하여 대식세포의 활성화를 촉진함으로써 대식세포 내에서 생존하거나 증식할 수 있는 미생물을 더 효율적으로 파괴할 수 있도록 한다. TH2 세포는 IL-4, IL-5, IL-13 와 같은 사이토카인들을 생산하여 호산구 (IL-5)와 비만세포/호염구 (IL-4)를 모집하고 활성화시키는데 관여하고, 또한 점막표면 (IL-13)의 장벽면역 (barrier immunity)을 향상시킨다. 주로 IgE를 매개로 방어반응을 촉진시킴으로써 기생충에 의한 감염을 통제하는데 기여한다. 특히, IL-4 및 IL-13은 IgE를 생산하기 위한 B 세포의 클래스 전환에도 필요하다. 이렇게 TH2 세포의 도움에 의해 B 세포가 생산하는 IgE는 알레르기와 천식과 같은 과민성 반응을 일으키는 원인이 되기도 한다. TH17 세포는 IL-17A, IL17F, IL-22를 분비하여, 국소 상피세포 및 기질세포들이 감염 부위에 호중구를 모집하는 케모카인을 생산하도록 유도한다. 이를 통해 세포 외 세균 및 진균과 같은 병원체를 제거하는 호중구의 반응을 증폭시켜 준다. 아울러 점막상피조직 및 피부의 상피세포를 활성화시켜 세균을 공격하는 항균펩티드를 생산하도록 한다. TFH 세포는 림프조직내에서 B 세포에 도움을 주는 효과 CD4 T 세포 아형이다. TFH 세포는 B 세포 여포로의 소통 (traffic to B cell follicles)과 항원에 대한 연관인식 (linked recognition)등의 기전을 통해, 미경험 B 세포와 상호작용 관계를 형성하고 배중심 반응을 촉진하여 B 세포의 항체생산 및 개별형 전환 (class switching)에 도움을 준다. TFH 세포는 B 세포의 증식과 항체생산 혈장세포로의 분화를 지원해주는 사이토카인 IL-21을 대량으로 생산한다. Treg 세포는 TGF-β와 IL-10의 생산을 통해 T 세포 활성과 내재면역세포 활성을 보통 억제함으로써 면역반응을 제한하고, 면역반응 동안에 자가면역이 전개되지 않도록 도운다.
효과 CD4 T 세포 아형들의 안정성에는 서로 간에 차이가 있다. 미경험 CD4 T 세포는 다양한 잠재력을 가진 반면에, TH1 및 TH2 세포는 비교적 안정한 바닥 상태 (ground state)를 보이며 다른 아형으로의 전환에 대한 내성이 높다. 한편, iTreg 세포와 TH17 세포는 덜 안정적이며 우세하게 영향을 끼치는 사이토카인들에 의해 다른 아형으로 전환될 수 있다. iTreg 세포는 IL-6 및 IL-1에 의해 반응하면 TH17 세포로 전환될 수 있고, IL-12에 의해 반응하면 TH1 세포가 될 수 있다. TH17 세포의 경우도 IL-12에 반응하면 TH1 세포로 전환될 수 있다. 이때 iTreg 세포가 TH17 세포로 전환되고 TH17 세포가 TH1 세포로 전환되는 것은 단방향성 (unidirectional)이거나 비가역적 (irreversible)인 것으로 이해되고 있다.
효과 CD4 T 세포 아형들의 차별화되는 면역 관련 기능에 근거해 볼 때, 과민성반응 환자, 자가면역질환 환자, 줄기세포 혹은 골수이식 환자를 포함한 장기이식 환자들에게 일어나는 부적절한 면역반응을 필요한 수준으로 조절하기 위해서는 효과 CD4 T 세포의 아형 중에서도 TH1 및 TH2를 표적으로 하는 특이적인 억제 능력을 발휘하는 면역억제제를 사용하는 것이 더 효과적일 것으로 기대된다.
L-AHG (25~100 μg/mL)는 활성화된 CD4 T 세포의 세포주기 진행을 주로 G1 기 및 S 기에서 농도의존적으로 저해함으로써 증식을 현저하게 억제하는 것으로 나타났다. 동일한 조건에서 immobilized anti-CD3/ant-CD28의 자극으로 휴지기 T 세포를 활성화시킨 후 30시간이 경과하였을 때, 배양 상등액을 회수하고, TH1 세포가 특징적으로 분비하는 것으로 알려진 TH1 기호 사이토카인들 (TH1 signature cytokines; IFN-γ, IL-2)과 TH2 세포가 특징적으로 분비하는 것으로 알려진 TH2 기호 사이토카인들 (TH2 signature cytokines; IL-4, IL-13)의 농도를 ELISA 방법으로 측정하였다.
그 결과, 활성화시킨 T 세포의 배양 상등액 속에 함유된, TH1 아형에 의해 분비되는 IL-2와 IFN-γ 뿐만 아니라 TH2 아형에 의해 분비되는 IL-4와 IL-13의 경우에도 L-AHG (25, 50, 및 100 μg/mL)의 첨가에 의해 농도 의존적으로 현저하게 감소하는 것으로 나타났다 (도 5의 A와 B). 흥미롭게도, TH1 기호 사이토카인들에 비해 TH2 기호 사이토카인들의 농도 감소가 더 현저하게 나타났다. 특히, 25 μg/mL 농도의 L-AHG 존재 하에서는 IFN-γ 농도는 거의 영향을 받지 않았으나 IL-4 농도는 L-AHG 무처리 대조구의 38,5% 수준으로 현저하게 감소되는 것으로 확인되었다.
결론적으로, immobilized anti-CD3/ant-CD28의 자극으로 휴지기 TH 세포의 세포주기가 개시되도록 활성화시켜서 세포 증식에 따른 클론확장 및 효과 TH 세포 아형으로의 분화가 일어날 때, L-AHG의 존재는 CD4 T 세포의 세포주기진행을 저해함으로써 TH1 아형 및 TH2 아형으로의 분화를 억제할 수 있으며, TH1 아형에 비해 TH2 아형으로의 분화를 더 강력하게 억제할 수 있음을 시사한다.
