Abstract
본 발명은 B형 간염 예방 또는 치료용 약학 조성물에 관한 것이다. 본 발명의 약학 조성물은 생체 이용률이 우수하고, 면역억제 및 신독성 부작용이 없어 B형 간염 예방 또는 치료에 유용하게 사용될 수 있으며, 다른 B형 간염 치료제와 병용 투여함으로써 그 효과를 더 증가시킬 수 있다.
Description
본 발명은 B형 간염 예방 또는 치료용 약학 조성물에 관한 것이다.
B형 간염 바이러스(HBV)는 급성 또는 만성 간 질환을 유발하는 헤파드나바이러스과에 속하는 바이러스다.
만성 B형 간염(Chronic hepatitis B, CHB)은 B형 간염 바이러스(HBV)에 의해 유발되는 전 세계적으로 확산되는 전염성 간 질환으로, HBV가 간세포에 침입하여 자신을 복제하고 감염된 간세포에 염증 손상, 더 나아가 간세포 섬유증, 간경변증 및 암종을 유발한다. 약 2억 5천만 명의 사람들이 HBV 감염으로 인한 간 기능 장애로 고통받고 있다. 바이러스의 역전사를 억제하는 라미부딘, 엔테카비르, 테노포비르 등의 뉴클레오시드 유사체 항바이러스제가 치료제로 사용되고 있다.
그러나, B형 간염 바이러스의 역전사를 억제하는 항바이러스제들은 B형 간염 바이러스의 간세포 내 진입을 억제하지 못하며, 간세포 내 침투한 바이러스를 제거하지 못하는 문제점이 있다.
본 발명은 B형 간염 예방 또는 치료용 약학 조성물을 제공하는 것을 목적으로 한다.
1. 하기 화학식 1로 표시되는 화합물 또는 이의 약학적으로 허용되는 염을 포함하는 B형 간염의 예방 또는 치료용 약학 조성물:

본 발명의 약학 조성물은 B형 간염 예방 또는 치료 효과가 뛰어나다. 또한, 본 발명의 약학 조성물은 신독성 및 면역억제 부작용이 없는 효과적인 B형 간염치료제로 활용될 수 있다.

본 명세서의 구조식(화학식)에서 치환기가 필요한 자리이나, 어떠한 치환기도 기재되지 않은 경우, 수소 치환기를 생략한 것이며, 이는 본 발명의 모든 구조식에서 동일하게 적용된다.
상기 B형 간염은 급성 B형 간염 및 만성 B형 간염을 모두 포함한다.
상기 화학식 1로 표시되는 화합물은 B형 간염 바이러스의 간 세포 내 진입을 억제할 수 있으며, 이를 통해 B형 간염을 예방 또는 치료할 수 있다.
상기 화학식 1로 표시되는 화합물은 생체 내 투여 시 높은 혈장 농도를 보이며, 경구 생체 이용률이 뛰어나 경구 투여시에도 B형 간염 바이러스(HBV)의 간 세포 내 진입을 효과적으로 억제할 수 있다.
상기 화학식 1로 표시되는 화합물은 낮은 신독성(nephrotoxicity)을 나타내며, CypA와의 낮은 결합력으로 인해 체내에서 면역억제 작용을 일으키지 않는다. 따라서, 상기 화학식 1로 표시되는 화합물을 포함하는 약학 조성물은 신독성 및 면역억제 부작용이 없는 B형 간염 치료제로 활용될 수 있다.
본 발명에 있어 "약학적으로 허용되는 염"은 여기서 언급한 화합물들에서 발견되는 특정 치환체에 의존하는 비교적 비독성 산 및 염기로 제조된 활성 화합물의 염들을 포함한다. 본 발명의 화합물들은 상대적으로 산성 기능성을 포함할 때, 염기(base) 부가 염들은 충분한 양의 원하는 염기, 순수한 또는 적당한 비활성(inert) 용매로 그러한 화합물들의 중성 형태를 접촉하여 얻을 수 있다. 약학적으로 허용 가능한 염기 부가 염의 예들은 나트륨, 칼륨, 칼슘, 암모늄, 유기 아미노 또는 마그네슘 염 또는 유사한 염을 포함한다. 본 발명의 화합물들은 상대적으로 염기성 기능성을 포함할 때, 산성 부가 염들은 충분한 양의 원하는 산, 순수한 또는 적당한 비활성(inert) 용매로 그러한 화합물들의 중성 형태를 접촉하여 얻을 수 있다. 약학적으로 허용 가능한 산성 부가 염의 예들은 초산, 프로피온산, 이소부틸산, 옥살릭산(oxalic), 마레익(maleic), 말로닉(malonic), 안식향산, 숙신산, 수버릭(suberic), 푸마릭(fumaric), 만데릭(mandelic), 프탈릭(phthalic), 벤젠설포닉(benzenesulfonic), p-토릴설포닉(tolylsulfonic), 구연산, 주석산, 메탄솔포닉(methanesulfonic), 및 그 유사체를 포함하는 상대적으로 비독성 유기산에서 유래한 염들 뿐만 아니라, 염화수소, 브롬화 수소, 질산, 탄산, 일수소탄산(monohydrogencarbonic), 인산(phosphoric), 일수소인산, 이수소인산, 황산, 일수소황산, 요오드화수소 또는 아인산(phosphorous acid) 및 그 유사체를 포함한다. 또한 알긴네이트(arginate)와 그 유사체와 같은 아미노산의 염 및 글루쿠로닉(glucuronic) 또는 갈락투노릭(galactunoric) 산들과 그 유사체와 같은 유기산의 유사체를 포함한다(예를 들어, Berge 등 (1977) J. Pharm. Sci. 66: 1-19). 본 발명의 일부 특정한 화합물들은 화합물들을 염기성 또는 산성 부가(addition) 염들로 전환하게 하는 염기성 및 산성 기능성 모두를 갖는다. 염들의 다른 예들은 본 발명이 속한 분야에서 공지된 문헌들, 예를 들면, Remington's Pharmaceutical Sciences, l8theds., Mack Publishing, Easton PA (1990) 또는 Remington: The Science and Practice of Pharmacy, 19th eds., Mack Publishing, Easton PA (1995)에 개시되어 있다.
본 발명의 조성물에 따라 치료될 적합한 개체는 포유동물 개체를 포함한다. 본 발명에 따른 포유동물은, 이에 한정되는 것은 아니지만, 인간, 개(canine), 고양잇과동물(feline), 소(bovine), 염소(caprine), 말(equine), 양(ovine), 돼지(porcine), 설치류(rodents), 토끼목(lagomorphs), 영장류(primates) 등을 포함하고, 자궁 내의(in utero) 포유동물을 포함하고, 구체적으로는 인간일 수 있다. 개체는 양쪽 성(性) 모두 일 수 있고 발생(development)의 임의의 단계일 수 있다.
본 발명에 따른 화합물은 임의의 적합한 경로에 의하여 이러한 경로에 적당한 약학 조성물의 형태, 그리고 의도된 치료를 위하여 효과적인 투여량으로 투여될 수 있다. 효과적인 투여량은 단일 또는 분할 투여로 일반적으로 약 0.001 내지 약 100 mg/체중kg/일이고, 바람직하게는 약 0.01 내지 약 30 mg/kg/일이다. 나이, 종, 및 치료될 질병 또는 상태(condition)에 따라 이 범위의 하한 미만의 투여량 수준이 적합할 수 있다. 다른 경우에는, 여전히 더 큰 투여량이 해로운 부작용없이 사용될 수 있다. 더 큰 투여량은 하루 동안 투여를 위하여, 여러 작은 투여량으로 분할될 수 있다. 적절한 투여량을 결정하기 위한 방법들이 본 발명이 속한 분야에 잘 알려져 있으며, 예를 들어, 문헌 Remington: The Science and Practice of Pharmacy, Mack Publishing Co., 20th ed., 2000이 이용될 수 있다.
본 발명에 따른 약학 조성물은 다음과 같이 투여될 수 있다.
본 발명에 따른 약학 조성물은 구강으로 투여(경구 투여)될 수 있으며, 구강은 연하(swallowing)를 포함하는 개념이다. 구강 투여에 의하여 본 발명의 화합물이 위장관(gastrointestinal tract)에 들어가거나, 예를 들어, 구강(buccal) 또는 설하(sublingual) 투여와 같이, 입으로부터 혈류로 직접적으로 흡수될 수 있다.
구강 투여를 위한 적합한 조성물은 고형상, 액상, 겔(gel), 또는 파우더 형상일 수 있으며, 정제(tablet), 로젠지(lozenge), 캡슐(capsule), 과립제, 산제 등의 제형을 가질 수 있다.
구강 투여를 위한 조성물은 선택적으로 장용 코팅(enteric coating)될 수 있으며, 장용 코팅을 통하여 지연된(delayed) 또는 지속된(sustained) 방출을 나타낼 수 있다. 즉, 본 발명에 따른 구강 투여를 위한 조성물은 즉시 또는 변형된(modified) 방출 패턴을 가진 제형일 수 있다.
액체 제형은 용액, 시럽 및 현탁액을 포함할 수 있으며, 이러한 액상 조성물은 연질 또는 경질 캡슐 내에 함유된 형태일 수 있다. 이러한 제형은 약학적으로 허용 가능한 담체, 예를 들어, 물, 에탄올, 폴리에틸렌글리콜, 셀룰로오스, 또는 오일(oil)을 포함할 수 있다. 상기 제형은 또한 하나 이상의 유화제 및/또는 현탁제를 포함할 수 있다.
정제(tablet) 제형에서, 활성 성분인 약물의 양은 정제 총 중량 대비 약 0.05 중량% 내지 약 95 중량%, 더욱 일반적으로 제형의 약 2 중량% 내지 약 50 중량%로 존재할 수 있다. 또한, 정제는 약 0.5 중량% 내지 약 35 중량%, 더욱 일반적으로 제형의 약 2 중량% 내지 약 25 중량%를 포함하는 붕해제를 함유할 수 있다. 붕해제의 예로는 유당, 전분, 소디움스타치글리콜레이트, 크로스포비돈, 크로스카멜로스소디움(croscarmellose sodium), 말토덱스트린 또는 이들의 혼합물이 사용될 수 있으나 이에 한정되는 것은 아니다.
정제로 제조하기 위해 포함되는 적합한 활택제는 약 0.1 중량% 내지 약 5 중량% 양으로 존재할 수 있고, 탈크(talc), 이산화규소, 스테아린산, 칼슘, 아연 또는 마그네슘 스테아레이트, 소듐 스테아릴 푸마레이트 등이 활택제로 사용될 수 있으나, 본 발명은 이러한 첨가제들의 종류에 한정되는 것은 아니다.
정제로 제조하기 위한 결합제(binder)로는 젤라틴, 폴리에틸렌글리콜, 당(sugar), 검(gum), 녹말(starch), 폴리비닐피롤리돈, 하이드록시프로필셀룰로오스, 하이드록시프로필메틸셀룰로오스 등이 사용될 수 있으며, 정제로 제조하기 위한 적합한 희석제로는 만니톨, 자일리톨, 락토오스, 덱스트로오스, 수크로오스, 솔비톨, 녹말(starch), 미결정셀룰로오스 등이 사용될 수 있으나, 본 발명은 이러한 첨가제들의 종류에 한정되는 것은 아니다.
선택적으로 정제에 포함될 수 있는 가용화제는 정제 총 중량 대비 약 0.1 중량% 내지 약 3 중량% 양이 사용될 수 있고, 예를 들어, 폴리소르베이트, 소디움 라우릴설페이트, 소디움 도데실설페이트, 프로필렌 카보네이트, 디에틸렌글리콜모노에틸에테르, 디메틸이소소르비드, 폴리옥시에틸렌글리콜화된 천연 또는 수소화 피마자유, HCORTM(Nikkol), 올레일에스테르, 젤루시어(GelucireTM), 카프릴릭/카프릴산 모노/디글리세리드, 소르비탄지방산에스테르, 솔루톨HSTM 등이 본 발명에 따른 약학 조성물에 사용될 수 있으나, 본 발명은 이러한 가용화제의 구체적 종류에 한정되는 것은 아니다.
본 발명에 따른 약학 조성물은 피하조직, 혈류, 근육 또는 내장 내로 직접 투여될 수 있다. 비경구 투여를 위한 적합한 방법은 정맥내(intravenous), 근육내(intra-muscular), 피하 동맥내(subcutaneous intraarterial), 복강내(intraperitoneal), 척추강내(intrathecal), 두개내(intracranial) 주사 등을 포함한다. 비경구 투여를 위한 적합한 장치는 (바늘 및 바늘 없는 주사기를 포함하는) 주사기(injector) 및 주입 방법(infusion method)을 포함한다.
비경구 투여를 위한 조성물은 즉시 또는 변형된 방출 패턴을 가진 제형일 수 있으며, 변형된 방출 패턴은 지연된(delayed) 또는 지속된(sustained) 방출 패턴일 수 있다.
대부분의 비경구 제형은 액상 조성물이며, 이러한 액상 조성물은 본 발명에 따른 약효 성분, 염, 완충제, 등장화제 등을 포함하는 수용액이다.
비경구 제형은 또한 건조된 형태(예를 들어, 동결 건조) 또는 멸균 비-수용액으로서 제조될 수 있다. 이들 제형은 멸균수(sterile water)와 같은 적합한 비히클(vehicle)과 함께 사용될 수 있다. 용해도 증강제(solubility-enhancing agents) 또한 비경구 용액의 제조에 사용될 수 있다.
본 발명에 따른 약학 조성물은 피부 또는 경피로 국소적으로 투여될 수 있다. 이 국소 투여를 위한 제형은 로션, 용액, 크림, 젤, 하이드로젤, 연고, 폼(foam), 임플란트(implant), 패치 등을 포함한다. 국소 투여 제형을 위한 약학적으로 허용 가능한 담체는 물, 알코올, 미네랄 오일, 글리세린, 폴리에틸렌글리콜 등을 포함할 수 있다. 국소 투여는 또한 전기천공법(electroporation), 이온도입법(iontophoresis), 음파영동(phonophoresis) 등에 의하여 수행될 수 있다.
국소 투여를 위한 조성물은 즉시 또는 변형된 방출 패턴을 가진 제형일 수 있으며, 변형된 방출 패턴은 지연된(delayed) 또는 지속된(sustained) 방출 패턴일 수 있다.
질병 또는 상태의 적절한 치료 또는 예방을 위한 약학 조성물의 제조 방법은 본 발명이 속한 분야에서 통상의 지식을 가진 자에게 잘 알려져 있다. 예를 들어, Handbook of Pharmaceutical Excipients (7th ed.), Remington: The Science and Practice of Pharmacy (20th ed.), Encyclopedia of Pharmaceutical Technology (3rd ed.), Sustained and Controlled Release Drug Delivery Systems (1978) 등에 기재된 바에 따라, 약학적으로 허용 가능한 담체, 운반체, 첨가제들 등을 본 발명에 따른 화합물과 적절히 혼합하여 본 발명의 목적을 위한 약학 조성물을 제조할 수 있다.
본 발명에 따른 약학 조성물은 단독 또는 다른 약학적 활성 화합물(pharmaceutically active compound)과 조합하여 B형 간염의 예방 또는 치료를 위하여 사용될 수 있다. 본 발명에 따른 화합물 및 다른 약학적 활성 화합물(pharmaceutically active compound)(들)은 동시에(동일한 제형 또는 분리된 제형으로) 또는 연속적으로 투여될 수 있다.
상기 하나 이상의 추가적인 약학적 활성 화합물(pharmaceutically active compound)은 공지된 B형 간염 바이러스(HBV)에 대한 항바이러스제 또는 면역조절제일 수 있다. 항바이러스제로는 예컨대 HBV의 캡시드 조립 억제제(Capsid assembly inhibitors), 표면항원 분비 억제제(HbsAg secretion inhibiotrs), 리보핵산 저해제(RNA interference and antisense oligonucleotides), 뉴클레오사이드 유사체(nucleoside analog) 등이 있고, 면역조절제로는 예컨대 페길화 인터페론, 인터페론 알파 등이 있다.
본 발명의 약학 조성물은 본 발명에 따른 화합물 이외의 하나 이상의 추가적인 약학적 활성 화합물(pharmaceutically active compound)을 더 포함할 수 있다.
하나 이상의 추가적인 약학적 활성 화합물은 전술한 바와 같다.
이하, 본 발명을 실시예로 보다 상세하게 설명한다.
실시예 1. PAMPA를 이용한 투과성 평가
병렬 인공 막 투과성 분석(Parallel artificial membrane permeability assay, PAMPA)을 수행하여 수동확산 하에서의 Carbamazepine(Cz), 사이클로스포린 A(CsA) 및 사이클로스포린 O(본 발명의 화학식 1로 표시되는 화합물, CsO)의 투과성을 평가하였으며, CsO가 CsA보다 7배 낮은 투과성을 보임을 확인하였다(표 1).

실시예 2. 세포막 투과성 평가
Caco-2 세포를 사용하여 CsO의 in vitro 막 투과성을 평가하였다. Caco-2 세포 단층은 약물의 겉보기 투과성을 평가하기 위해 장 상피를 나타내는 데 사용되는 세포주로, 약물은 세포간 경로, 세포주위경로 또는 활성 수송체에 의해 Caco-2 세포 단층을 투과할 수 있다.
Caco-2 투과성 분석은 3회 수행되었다. 10μM 시료를 1%(v/v) DMSO/HBSS 용액에 녹이고 Hank의 HBSS(Balanced Salt Solution) 완충액에 100μM verapamil을 P-당단백질 억제제로 사용하였다. Caco-2 세포가 있는 삽입물을 37℃에서 30분 동안 verapamil과 함께 사전 인큐베이션한 다음 37℃에서 흔들지 않고 2시간 동안 인큐베이션했다. Efflux ratio는 A to B permeability / B to A permeability 로 계산되었다. 각 시점(0.5, 1, 1.5, 2시간)에 aliquots(20μL)를 채취했다. *실험은 이중으로 수행되었다.
CsA 및 CsO의 Caco-2 막 투과성은 PAMPA에 의한 막 투과성 결과와 일치했다. 이러한 결과는 CsA 및 CsO가 동일한 메커니즘, 즉 수동 확산 하에서 세포간 경로로 세포막을 투과함을 나타낸다(표 2).

실시예 3. In Vivo 약동학 연구
In vivo 약동학적 프로파일 평가를 위해 수컷 ICR 마우스에 CsA 또는 CsO를 정맥내(iv, 5mg/kg) 주사 또는 경구(po, 20mg/kg) 투여하였다(도 2).
경구 투여 후, CsO 및 CsA는 빠르게 흡수되었고 거의 동일한 Tmax를 나타내어 동일한 메커니즘에 의한 흡수를 나타낸다(도 2). CsO는 중간 정도의 경구 생체이용률을 나타냈다(F = 12 ± 0.3%). 그러나 정맥 주사 후 낮은 혈장 농도로 인해 CsA의 경구 생체 이용률은 얻을 수 없었다.
CsO는 정맥 주사 후 CsA보다 더 높은 혈장 농도를 보였다. 또한, CsA의 혈장 농도는 15분 후에 정량 하한선 아래로 떨어졌다.
Blood to plasma partitioning assay(BPP) 에서 CsA는 1μM 용량에서 CsO보다 5배 더 높은 혈액 대 혈장 분할비(blood-to-plasma partitioning ratio,BPR)를 보여주었다(CsA = 3.12, CsO = 0.62, 도 3 참조). 용량이 증가함에 따라 CsO에 대한 BPR이 증가했다(도 3). CsA의 높은 BPR은 CsA가 적혈구에 우선적으로 축적되어 더 낮은 혈장 농도를 유발함을 나타내고, CsO의 낮은 BPR(모든 용량에서 BPR <1)은 CsO가 혈장에 우선적으로 축적되어 높은 혈장 농도를 유발함을 나타낸다.
실시예 4. CsO의 사이클로필린 A과의 결합력
펩티딜-프롤린 이성질체화 효소 작용 어세이(uncoupled peptidyl-prolyl isomerase functional assay )를 이용하여 CypA에 대한 저해상수(inhibitory constant, Ki)를 측정하였다.
표 3은 CsA 및 CsO의 사이클로필린 A(Cyclophilin A, CypA)에 대한 inhibitory constant(Ki)를 나타낸다.

CsO는 CsA에 비해 CypA에 대한 결합 결합저해상수가 300배 이상 높은 것으로 나타났다. CypA는 특히 적혈구에서 과발현되는 물질로, CsA는 CypA와의 결합으로 적혈구에 우선적으로 축적되어 낮은 혈장 농도를 나타내며 CsO는 CypA와의 낮은 결합력으로 인해 혈장에 우선적으로 축적되어 높은 혈장 농도를 나타냄을 알 수 있고, 이는 임상 약동학적 프로파일 결과와도 일치한다.
실시예 5. 신장 상피 세포주에 대한 CsA 및 CsO의 세포독성
MDCK 및 BCS-1 세포는 Korean Cell Link Bank에서 구입하였다. MDCK 및 BCS-1 세포는 5% CO2와 37℃에서 10% FBS가 포함된 DMEM 배지에서 성장했다. 세포 성장에 대한 고리형 펩티드의 영향을 평가하기 위해 MTS cell-viability assay가 수행되었다. 1 x 105개 세포를 포함하는 배지의 aliquots(100μL)를 96웰 플레이트(SPL)의 각 웰에 부었다. 세포는 37℃의 공기 중 5% CO2의 가습 대기에서 성장하고 부착되도록 24시간 동안 두었다.
세포 밀도가 대략 70% confluency에 도달했을 때, 배지는 고리형 펩타이드 스톡 용액(DMSO 중 10mM)의 연속 희석액으로 교체하고 24시간 동안 배양했다. 배지를 흡입한 후, tetrazolium compound [3-(4,5-dimethylthiazol-2-yl)-5-(3- carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium, inner salt; MTS] (20 μL)가 포함된 CellTiter 96 aqueous non-radioactive cell proliferation assay reagent(Promega, Madison, WI, USA)를 포함하는 Opti-MEM 용액(200 μL)을 각 웰에 첨가하였다. 플레이트는 대사를 위해 37℃에서 4시간 동안 배양되었다. 마이크로플레이트 리더를 사용하여 490 nm에서 생존 세포의 MTS-포르마잔 생성물을 측정하고 세포 생존율(%)을 측정하여 미처리 세포와 비교하였다. 세포 생존율의 백분율 = (A-A blank)/(A control-A blank)×100, 여기서 A는 사이클로스포린을 처리한 세포를 포함하는 웰의 흡광도, A control은 사이클로스포린으로 처리되지 않은 세포를 포함하는 웰의 평균 흡광도, A blank는 세포가 없는 상태의 background absorbance이다.
그 결과 CsO는 신장 상피 세포주에 대해 CsA 보다 낮은 독성을 보임을 확인하였다(표 4 및 도 5 참조).
표 4는 진핵 세포주에 대한 CsA 및 CsO의 LC50 값을 나타낸다. BSC-1 및 MDCK는 각각 원숭이 및 개 신장 상피 세포, HeLa는 인간의 자궁경부 종양 세포이다. a: 200 μM에서 세포 생존율 백분율, b: 100 μM에서 세포 생존율 백분율.

실시예 5. In vitro HBV 숙주침투 억제 효과 확인
HepG2 세포는 사람 간세포를 불멸화시킨 대표적인 세포이고, HepG2-NTCP 세포는 이 세포에 나트륨-타우로콜산 동시수송 단백질 (sodium-taurocholate cotransporting polypeptide, NTCP)를 과발현 시킨 세포를 지칭한다. HepG2-NTCP 세포는 HBV 숙주침투 억제 효과를 확인하기 위해 사용하는 대표적인 세포이다.
HepG2-NTCP 세포는 5% CO2와 37℃에서 10% FBS, 페니실린 (100 U), 그리고 스트렙토마이신 (0.1 mg/mL)이 포함된 DMEM 배지에서 성장했다. 이 세포에 대한 유전자증폭검사를 실시한 결과 미코플라스마균에 대해 음성이 확인되었다. HepG2-NTCP 세포는 12-웰 플레이트에서 고리형 펩타이드와 함께 2시간 동안 배양되었다. 이후 HBV 입자(2 Х 106 바이러스/mL)와 고리형 펩타이드를 같이 처리하여 세포를 감염시켰고 3시간 동안 배양하였다. 바이러스가 포함된 배지를 제거한 후, 세포를 16시간동안 배양하였다. 세포는 고리형 펩타이드로 처리한 후, 세포 내 HBV의 DNA를 추출하고 정제하고 유전자증폭검사를 이용해 정량화하였다. 숙주세포 내 HBV의 DNA를 분리하기위해 세포는 세포 용해 완충제 (50 mM 트리스-염산, pH 7.5, 1 mM EDTA, 1% NP40)에 의해 얼음에서 10분 동안 용해되었다. 세포의 잔해와 핵을 15,000배 중력의 원심분리를 통해 제거한 후, 상층액에 마이크로코칼 뉴클레아제(0.25 U/μL)를 37℃에서 1시간동안 처리하였다. 최종농도가 10 mM이 되도록 EGTA를 첨가하여 마이크로코칼 뉴클레아제를 비활성화시켰다. 그 후에 G-spinTM 세트를 이용해 HBV의 세포 내 DNA를 추출하였다. 추출된 HBV 세포 내 DNA는 HBV DNA 프라이머 (포워드: TCCTCTTCATCCTGCTGCTATG(서열번호 1), 리버스: CGTGCTGGTAGTTGATGTTCCT(서열번호 2))와 프로브 (TATTGGTTCTTCTGGACTA(서열번호 3))를 이용해 증폭시켰다.
HBV의 세포 내 DNA의 정량화에 기초한 in vitro HBV 진입 억제 분석을 수행하였다. CsA 및 CsO는 1μM 농도에서 대등한 진입 억제율을 보였다(표 5). IC50 측정 결과에서도 CsO는 CsA와 유사한 IC50 값을 나타내어 HBV 감염을 억제하는데 큰 효능(IC50 = 0.51 ± 0.05 μM)을 보였다(표 6).
표 5는 In vitro HBV 진입 억제 스크리닝 결과를 나타낸다. 각 사이클로스포린 A 또는 O를 1μM 농도 사용하여 HBV 진입 억제 활성을 평가했다. 실험은 삼중으로 수행되었다(n = 3).
