실험예 1 : 염증 개선(NO 생성 억제) 효과 평가Experimental Example 1: Evaluation of the effect of improving inflammation (inhibiting NO production)
RAW264.7 대식세포는 세포배양 플레이트에 2.5 × 105cells/mL로 분주하고 37℃, 5% CO2 인큐베이터 조건 하에 18시간 배양하였다. 배양된 세포는 NO의 생성을 유도하기 위하여 리포폴리사카라이드 (LPS) 1μg/mL가 포함된 배지로 교환한 후, 상기 실시예에서 제조된 한방복합추출물을 첨가하여 24시간 재배양하였다. 이후 생성된 NO의 양은 세포배양 상등액 100 μL와 Griess 시약 (0.1% 나프틸에틸렌디아민 디하이드로클로라이드 및 2.5% 인산 내 1% 설파닐아민, 1:1) 100 μL를 혼합하여 96-웰 플레이트에서 10분 동안 반응시킨 후 ELISA 리더(Xmark microplate absorbance spectrophotometer)를 이용하여 540 nm에서 흡광도를 측정하였다. 생성된 NO의 양은 질산나트륨 (NaNO2)의 농도별 표준곡선과 비교하여 산출하였다. 그 결과는 하기 표에 나타내었다. 과도하게 형성된 NO는 염증을 유발하는 것으로 알려져 있다.RAW264.7 macrophages were seeded on a cell culture plate at 2.5 × 10 5 cells/mL and cultured at 37° C., 5% CO 2 in an incubator condition for 18 hours.
