본 발명은 높은 트리펩타이드 생산능 및/또는 알코올 분해 효소 활성을 갖는 칸디다 유틸리스(Candida utilis) 균주 및 이의 용도에 관한 것이다.
알코올은 다른 식품과는 달리 체내에 축적되지 못하는 xenobiotics의 한 종류로 acetaldehyde와 같은 분해산물로 인해 산화스트레스에 의한 세포손상과 숙취증상의 주요 원인으로 알려져 있다. 알코올은 섭취량에 따라 간 대사에 여러 가지 영향을 미치게 되는데 알코올 그 자체보다는 산화분해 과정에서 생성된 nicotinamide adenine dinucleotide phosphate (NADP) 및 acetaldehyde가 생체 내 활성 아민류와 축합 반응을 거쳐 간 손상의 주요 매개체로 작용하는 동시에 뇌로 전해진 acetaldehyde는 많은 유해 화합물로 변화되어 안면홍조, 맥박증가, 오심, 구토 등의 숙취 증상이 나타나는 것으로 알려져 있다.
체내에 들어온 알코올의 80 내지 90%는 간 세포에 존재하는 alcohol dehydrogenase (ADH)에 의해 먼저 아세트알데히드로 분해되고, 다시 aldehyde dehydrogenase (ALDH) 효소에 의해 대사되어 아세트산을 형성한 후 이산화탄소와 물로 가수분해 되어 완전히 분해과정을 거치게 된다. 간에서의 에탄올 분해는 ADH의 산화반응을 통한 아세트알데히드로의 전환되는 단계가 중요한 단계가 된다.
글루타치온 (L-γ-glutamyl-L-cysteinylglycine, GSH)은 세포내 존재하는 생리활성물질로서 글루타메이트 (glutamate), 시스테인(cystein) 및 글라이신(glycine) 의 세가지 아미노산으로 구성된 트리펩타이드 형태이다. 체내에서는 환원형 글루타치온(GSH)과 산화형 글루타치온(GSSG) 두 가지 형태로 존재한다. 글루타치온은 동물, 식물 및 미생물의 세포 내에서 0.1~10mM의 농도로 존재하며, 세포의 총 비단백질성 활성분의 90% 이상을 차지하고 있다. 글루타치온의 다양한 기능은 농업 분야뿐 아니라 효소학, 수송, 약물학, 치료, 독물학, 내분비학 및 미생물학을 포함하는 많은 의학 분야에서 중요하다.
글루타치온은 주로 미생물을 이용한 발효 또는 효소 합성 공정으로 생산 가능한 데 현재 높은 생산 단가로 인해 효소 합성 공정은 아직 상용화가 되어 있지 않은 반면 산업적으로 미생물을 배양하여 균체로부터 추출하는 방법이 널리 이용되고 있다. 글루타치온 생산을 위한 미생물 균주는 세포내 높은 글루타치온을 함유하며 식품 생산에 안전한 미생물로 인식되는 효모가 널리 이용되어 왔는데, 특히 Saccharomyces 속 균주와 Candida 속 균주가 대표적이다. 이 효모 종들의 야생형 균주들의 경우 글루타치온 농도가 건조 중량의 0.1-1%정도로 이미 상당히 높으며, 저가의 배지에서 고밀도 세포 배양 및 빠른 성장이 가능하다는 장점은 효모를 이용한 발효적 생산이 경쟁력을 갖게 한다.
본 발명의 일 예는 높은 ADH활성을 갖는 칸디다 유틸리스(Candida utilis) 균주를 제공하는 것이다.

본 발명의 추가 일 예는 상기 칸디다 유틸리스(Candida utilis) 균주의 균체, 상기 균체의 추출물, 상기 균체의 용해물, 및 상기 균체의 파쇄물로 이루어지는 군에서 선택된 1종 이상을 포함하는 숙취 해소, 숙취 개선, 또는 숙취 예방용 조성물을 제공하는 것이다.
본 발명의 추가 일 예는 상기 칸디다 유틸리스(Candida utilis) 균주의 균체, 상기 균체의 추출물, 상기 균체의 용해물, 및 상기 균체의 파쇄물로 이루어지는 군에서 선택된 1종 이상을 포함하는 산화 스트레스 경감용 조성물 또는 항산화 조성물을 제공하는 것이다.
본 발명의 추가 예는 상기 칸디다 유틸리스(Candida utilis) 균주의 균체, 상기 균체의 추출물, 상기 균체의 용해물, 및 상기 균체의 파쇄물로 이루어지는 군에서 선택된 1종 이상을 포함하는 식품 조성물을 제공하는 것이다.
본 발명에 따른 칸디다 유틸리스(Candida utilis) 균주는 글루타치온 생성능및/또는 알코올 분해 효소(alcohol dehydrogenase, ADH) 활성을 가지고 있으며, 더욱 자세하게는 글루타치온 생성능이 높을 뿐만 아니라 알코올 분해 효소 활성을 가지고 있어, 체내 알코올을 해독하고 배출하는데 더 원활하게 작용하는 장점이 있다. 본 발명에 따른 효모 균주는 재조합 방법이 아닌 자연에서 분리한 non-GMO 균주로서 활용성이 높다는 장점이 있다.
본 발명에 따른 칸디다 유틸리스(Candida utilis) 균주는 균체 건조 중량(g) 당 글루타치온 생성량이 0.8 중량%이상 및/또는 ADH활성이 0.18 mU/ml이상인 것일 수 있으며, 바람직하게는 균체의 건조 중량(g) 당 글루타치온 약 1.5 중량%이상이고 ADH 활성이 0.2 mU/ml 이상일 수 있다.
본 발명에 따른 칸디다 유틸리스(Candida utilis) 균주는 알코올 분해능(Alcohol dehydrogenase, ADH) 및 알코올 내성을 가지며, 자세하게는 에탄올 농도가 2 내지 15%(v/v)인 조건에서 생육할 수 있는 알코올 내성을 가지며, 더욱 자세하게는 알코올 농도가 6 내지 15%(v/v)인 조건에서 배양한 경우 기탁번호 KCCM 11355을 갖는 Candida. utilis의 균체 광학밀도(optical density, OD)값 100%를 기준으로 120 내지 내지 200%을 갖는 것일 수 있다.
본 발명에 따른 칸디다 유틸리스(Candida utilis) 균주는 (i) 30℃에서 진탕배양시 글루타치온 생산량 증가, (ii)진탕배양하는 조건에서 포도당 함유 배지에서 배양하는 경우 균체 농도(600nm OD값) 100%를 기준으로, 수크로오스 포함 배지에서 균체 농도가 105 내지 120%이고 또는 진탕배양하는 조건에서 포도당 함유 배지에서 배양하는 경우 글루타치온 함량(mg/L) 100%를 기준으로, 수크로오스 포함 배지에서 글루타치온 함량이 105 내지 150%, 또는 110 내지 140%, (iii) Cysteine 첨가 배양 시 글루타치온 생산량 증가 및 (iv) Feeding rate 6g/L.h-1으로 공급한 fed-batch배양조건에서 글루타치온 생성량을 유지함으로 이루어지는 배양 특성으로 이루어지는 군에서 선택된 1종 이상을 갖는 것인 균주일 수 있다.
본 발명에 따른 칸디다 유틸리스(Candida utilis) 균주는 알코올에 대한 내성과 알코올 분해능을 가지고 있어, 음주 전 또는 후 알코올 분해에 따른 숙취해소 효과와 간의 해독작용을 도와 향후 알코올성 간 질환을 예방할 수 있는 식품 등의 제조에 광범위하게 활용될 수 있는 알코올 분해능이 있는 건강 기능성 식품 및 식품 첨가제에 관한 것이다.
본 발명에 따른 균주는 알코올(에탄올) 분해효소 활성이 우수하기 때문에 섭취한 알코올(에탄올)의 흡수를 효과적으로 억제함으로써 과다한 알코올 대사에 의한 간기능 장애와 알코올성 간질환 및 장 질환 발생을 예방 할 수 있게 된다.
구체적으로, 알코올은 체내에 저장되지 못하기 때문에 대사가 되어야 하는데, 이 과정은 대부분 간에서 이루어지며 이러한 대사는 간에 존재하는 알코올 분해 효소(alcohol dehydrogenase)들에 의해 이루어진다. 알코올은 이러한 효소들에 의해 아세트알데하이드란 물질을 거쳐 분해되는데, 상기 생성된 아세트알데하이드는 독성이 있어 간세포에 손상을 주고, 알코올 대사 결과 지방산이 많이 만들어져 간에 지방이 축적되어 알코올성 간질환을 일으킨다. 이러한 알코올성 간질환은 크게 알코올성 지방간, 알코올성 간염, 알코올성 간섬유증, 알코올성 간경변으로 분류되는데 간 손상 기전은 알코올 자체에 의한 경우, 아세트알데하이드와 같은 대사산물에 의한 경우, 면역반응에 의한 경우 등이 있으며, 특히 아세트알데하이드는 지방의 페록시데이션(Peroxidation), 세포질과의 결합, 미토콘드리아의 전자전달계 교란, 마이크로터블(Microtuble)의 기능방해, 단백질과 결합물질 형성, 콜라겐 합성의 증가 등 간 독성의 주범으로 작용한다.
또한, 본 발명에 따른 균주는 글루타치온 생성 활성이 우수하기 때문에 섭취한 알코올(에탄올)의 대사에 의한 독성물질의 배출을 촉진하고 유해 활성 산소 등에 의한 간 세포의 손상을 방어하여, 간기능 장애와 알코올성 간질환 및 장 질환 발생을 예방할 수 있게 한다.
구체적으로, 글루타치온은 독성물질이 소변이나 담즙으로 배설되도록 촉진하는 항산화제이며, 특히 알코올의 대사산물이 간에서 해독하는 과정에 작용하여 독성물질의 배설을 촉진하는 기능을 가진다. 또한 글루타치온은 유해 활성 산소 등에 의한 간 세포의 손상을 방어한다. 따라서, 본 발명에 따른 균주는 알코올에 대한 내성과 알코올 분해능을 가지고 있어 알코올의 흡수를 방지 및 분해하는 기능뿐만 아니라, 글루타치온 함량이 높아 알코올의 분해에 따른 독성 물질의 배출을 촉진하며, 간손상 및 간질환을 방지할 수 있다.
본 발명은 혈중 알코올 수준 감소 및 체내 항산화능 증진에 유효한 성분을 함하는 식품, 식품 첨가제, 음료 또는 건강보조식품에 관한 것이다.
본 발명에 따른 균주는 탄소원, 질소원 및 무기염 등을 포함하는 배지에서 호기적으로 배양할 수 있다.
이들 균주의 배지 조성으로서는, 탄소원으로서 통상의 미생물의 배양에 이용되는 덱스트린, 글루코오스, 수크로오스(sucrose), 아세트산, 에탄올, 당밀(molasses) 및 아황산펄프 폐액 등으로 이루어지는 군에서 선택되는 1종 또는 2종 이상이 이용될 수 있으며, 글루타치온 생산을 고려하면 수크로오스가 바람직하다. 더욱 자세하게는, 본 발명에 따른 칸디다 유틸리스(Candida utilis) 균주는 (ii)진탕배양하는 조건에서 포도당 함유 배지에서 배양하는 경우 균체 농도(600nm OD값) 100%를 기준으로, 수크로오스 포함 배지에서 균체 농도가 105 내지 120%이고 또는 진탕배양하는 조건에서 포도당 함유 배지에서 배양하는 경우 글루타치온 함량(mg/L) 100%를 기준으로, 수크로오스 포함 배지에서 글루타치온 함량이 105 내지 150%, 또는 110 내지 140%, (iii) Cysteine 첨가 배양 시 글루타치온 생산량 증가 및 (iv) Feeding rate 6g/L.h-1으로 공급한 fed-batch배양조건에서 글루타치온 생성량을 유지함으로 이루어지는 배양 특성으로 이루어지는 군에서 선택된 1종 이상을 갖는 것인 균주일 수 있다.
질소원으로서는 요소, 암모니아, 황산암모늄, 염화암모늄 혹은 인산암모늄 등의 무기염, 및 옥수수 침지액(corn steep liquor)(CSL), 카세인, 효모 엑기스 혹은 펩톤 등의 질소함유 유기물 등으로 이루어지는 군에서 선택되는 1종 또는 2종 이상이 사용된다. 또한, 인산 성분, 칼륨 성분, 마그네슘 성분을 배지에 첨가하여도 좋고, 이들로서는, 과인산석회, 인산암모늄, 염화칼륨, 수산화칼륨, 황산마그네슘, 염산마그네슘 등의 통상의 공업용원료이면 된다. 그 외에 아연, 동, 망간, 철이온 등의 무기염을 사용하여도 된다. 그 외에 비타민, 핵산 관련물질 등을 첨가하여도 된다.
본 발명에 적용 가능한 배양 온도는, 효모의 배양 조건일 수 있으며, 예컨대 20~40℃, 바람직하게는 25~35℃가 좋으며, pH는 3.5~8.0, 특히 4.0~6.0이 바람직하다.
본 발명에 적용 가능한 배양 형식으로서는 회분배양, 유가배양(fed-batch) 또는 연속배양 중 어느 것이라도 좋지만, 공업적으로는 유가배양 또는 연속배양이 채용된다.
본 발명은 상기 칸디다 유틸리스(Candida utilis) 균주를 배양하여, 글루타치온 생성량을 증가시키는 방법을 제공한다. 상기 균주의 배양은 탄소원을 설탕 또는 포도당으로 사용한 것일 수 있다. 상기 균주의 배양은, 발효액 내 에탄올 함량을 측정하여 탄소원 공급 속도를 조절하여 수행하는 것일 수 있다. 예를 들면 상기 발효액 내 에탄올 함량이 5g/L 이하일 경우, 탄소원을 연속적으로 공급할 수 있으며, 탄소원 공급 속도는 5 내지 10 g/L.h-1, 5.5 내지 8 g/L. h-1, 5.5 내지 7 g/L. h-1, 또는 5.5 내지 6.5 g/L. h-1일 수 있다.
더욱 자세하게는, 본 발명에 따른 칸디다 유틸리스(Candida utilis) 균주는 글루타치온 생산을 고려하면 유가배양 바람직하며, 더욱 바람직하게는 Feeding rate 6g/L.h-1으로 공급한 fed-batch배양조건에서 글루타치온 생성량을 유지할 수 있다.
본 발명의 방법에 따라, 고농도 글루타티온을 효모 균체 내에 함유하는 배양물이 얻어지지만, 배양물로부터 글루타티온을 함유하는 분획물을 얻어도 좋다. 배양물로부터 글루타티온을 함유하는 분획물을 분획하는 방법으로서는, 통상 행해지고 있는 방법이라면 어떤 방법이어도 좋지만, 열수(熱水)추출, 균체 파쇄에 의한 추출 등을 들 수 있다. 또한, 얻어진 추출물을 담체(carrier)에 담지(support)시킴으로써, 글루타티온을 고농도로 포함하는 분획물에 농축도 가능하다. 또한, 상기 방법에 의해 배양한 배양물로부터 효모 추출물을 제조할 수도 있다. 효모 추출물을 제조하는 방법으로서는, 통상 행해지고 있는 방법이라면 어떤 방법이어도 좋지만, 자기소화법, 효소분해법 혹은 알칼리추출법 등이 공업적으로 채용된다.
또한, 상기 방법에 의해 배양한 배양물로부터 건조 균체를 제조할 수 있다. 건조 효모 균체를 제조하는 방법으로서는, 통상 행해지고 있는 방법이라면 어떤 방법이어도 좋지만, 공업적으로는 동결건조법, 스프레이드라이법, 드럼드라이법 등이 채용된다.
본 발명의 일 예는, 칸디다 유틸리스(Candida utilis) 균체, 상기 균주의 배양물, 상기 균체의 파쇄물, 상기 균체의 용해물, 및 상기 균체, 균체 배양물, 균체의 파쇄물, 균체의 용해물 및 이들의 추출물로 이루어지는 군에서 선택된 1종 이상을 포함하는 산화 스트레스 경감용 조성물, 항산화 조성물, 또는 산화 스트레스 관련 질병의 예방, 개선, 경감 또는 치료용 조성물에 관한 것이다. 상기 조성물은 약학 조성물, 식품 조성물 또는 화장료 조성물일 수 있다.
본 발명은 신체의 세포, 조직 및 기관에서 산화성 스트레스의 역효과 및/또는 자유 라디칼의 생성으로부터 초래되었거나 관련된 질환, 증상, 병리학 및 질병을 치료하기 위한 새로운 적용에서 항산화제로서의 칸디다 유틸리스(Candida utilis) 균주의 용도를 제공한다.
산화성 스트레스는 단백질, DNA 및 지질에 손상을 일으키는 신경퇴행성 및 노화-관련 질병의 진행에 중대한 역할을 한다. 저분자량의 소수성 항산화제 화합물은 예컨대 급성 호흡곤란증후군(respiratory distress syndrome), 근육위축가쪽경화증(amyotrophic lateral sclerosis), 죽상경화성 심장혈관병(atherosclerotic cardiovascular disease), 다기관 기능부전(multiple organ dysfunctions) 및 예컨대 파킨슨 병, 알츠하이머 병 및 크루츠펠트-야콥 병(Creutzfeldt- Jakob's disease)과 같은 중추신경계 신경퇴행성 질환 등의 말초조직의 증상을 치료하는데에 유용하다. 산화성 스트레스는 다른 타입의 질환들뿐만 아니라 파킨슨 병, 알츠 하이머 병 및 크루츠펠트-야콥 병의 발병기전에 원인적으로 관련되어 있는 것으로 알려져 있다.
세포 내에 항산화제가 결핍되면 과량의 자유 라디칼이 발생하여 거대세포가 파괴되고 지질이 과산화되고 독소가 축적되어 결국 세포가 사멸하게 될 수 있다. 세포의 산화를 방지하는데 있어서 항산화제 화합물의 중요성 때문에, 예컨대 글루타치온(glutathione (GSH) ([γ]-글루타밀 시스테이닐 글리신))과 같은 천연 항산화제가 조직에 계속적으로 제공된다. GSH는 거의 모든 세포에서 합성되며, 체내에 적당한 산화상태를 유지하는 것을 담당하는 기본적 세포 항산화제중의 하나이다. 산화되었을 때, GSH는 GSSG라는 2합체를 형성하는데, 이것은 글루타치온 환원효소를 생산하는 기관에서 재순환하게 된다. 인간 성인에게서, 환원된 GSH는 기본적으로 간에서 GSSG로부터 생산되고, 골격근과 적혈구 및 백혈구에서도 비교적 소량 합성되며, 혈류를 타고 체내의 다른 조직으로 보내지기도 한다.
그러나, 어떤 조건에서는 GSH의 정상적인 생리적 공급이 불충분하고, 그 분배가 부적절하거나 국소적 산화요구도가 너무 높아서 세포의 산화를 방지할 수 없게 된다. 다른 조건에서는, 예컨대 GSH와 같은 세포 항산화제의 생산과 요구도가 불일치되어 체내에서 상기 분자들이 불충분한 농도로 된다. 다른 경우에는, 어떤 조직이나 생물학적 경로에 의해 항산화제가 소진되어, 세포간 항산화제 농도가 억제된다. 이러한 어느 경우에, 예를 들어 글루타치온과 같은 항산화제의 혈청 농도가 증가하면 세포내로 들어갈 수 있는 항산화제의 양이 증가하게 된다. 세포 흡수를 위해 촉진된 수송시스템에 있어서는, 흡수를 유도하는 농도구배가 증가된다.
산화제의 과잉생산과 결부된 증상, 질병, 질환 또는 병리학적 증세를 예방, 개선 또는 치료하기 위한 것으로서, 상기 증상, 질병, 질환 또는 병리학적 증세가 AIDS, 당뇨병, 황반변성, 울혈성심장기능상실(congestive heart failure), 심장혈관질환(cardiovascular disease), 심장동맥재협착(coronary artery restenosis), 폐질환(lung disease), 염증성 질환(inflammatory disease), 천식(asthma), RNA 바이러스 감염, DNA 바이러스 감염, 패혈증(sepsis), 골다공증(osteoporosis), 골질환(bone disease), 미생물 감염, 독소노출(toxin exposure), 방사선노출(radiation exposure), 화상(burn trauma), 프라이온(prion)병, 신경질환(neurological disease), 혈액질환(blood disease), 혈구 질환(blood cell disease), 동맥질환(arterial disease) 및 근육질환(muscle disease)으로 이루어지는 군으로부터 선택될 수 있다.
상기 글루타치온 함유 효모 추출물는 균체의 추출물을 의미하는 것이며, 글루타민산, 시스테인, 및 글리신의 3 개의 아미노산으로 구성된 트리펩티드인 글루타치온이 풍부하게 함유되어 있다. 본 발명에 따른 숙취의 예방 또는 치료용 조성물에서는 글루타치온이 5% 이상 함유되어 있 는 글루타치온 함유 효모 추출물을 이용하는 것이 바람직하다.
본 발명에 따른 식품 조성물은, 통상 건조 효모, 효모 엑기스를 첨가할 수 있는 음식품이라면 어떤 것이어도 좋지만, 예컨대 알코올음료, 청량음료, 발효식품조미료, 스프류, 일반식품, 과자류 등을 들 수 있다. 따라서, 본 발명에 의해, 글루타티온을 고농도로 포함하는 음식품을 효율적으로 제조할 수 있다.
본 발명의 일 예는, 칸디다 유틸리스(Candida utilis) 균체, 상기 균주의 배양물, 상기 균체의 파쇄물, 상기 균체의 용해물, 및 상기 균체, 균체 배양물, 균체의 파쇄물, 균체의 용해물 및 이들의 추출물로 이루어지는 군에서 선택된 1종 이상을 포함하는 알코올 분해용 조성물; 숙취 해소, 숙취 개선, 숙취 경감, 또는 숙취 예방용 조성물; 또는 알코올성 간질환의 예방, 개선, 경감 또는 치료용 조성물 관한 것이다. 상기 알코올성 간질환은 알코올성 지방간, 알코올성 간염, 알코올성 간섬유증, 및 알코올성 간경변을 포함하나 이에 한정되지 않는다. 상기 조성물은 약학 조성물 또는 식품 조성물일 수 있다.
본 발명에 따른 칸디다 유틸리스(Candida utilis) 균주는 알코올에 대한 내성과 알코올 분해능을 가지고 있어, 음주 전 또는 후 알코올 분해에 따른 숙취해소 효과와 간의 해독작용을 도와, 상기 균주의 균체 등을 이용하여 알코올성 간 질환을 예방, 경감, 개선 또는 치료할 수 있는 약학 조성물, 식품 조성물, 건강 기능성 식품 및 식품 첨가제에 관한 것이다.
본 발명의 숙취 관련 조성물은 상기 주요성분 이외에도 숙취 효과를 더욱 증대시키기 위한 보조성분으로서 비타민 B군, 비타민 C, 비타민 E, 또는 베타카로틴과 같은 비타민, Ca, Mg, 또는 Zn 등의 미네랄 성분, 레시틴 등의 인지질, 알라닌 또는 타우린 등의 아미노산, 과당, 올리고당, 영지, 또는 이들의 혼합물을 더욱 포함할 수 있다. 상기 효모 추출물에 포함된 아미노산 등의 성분들이 알코올대사를 촉진하는 기능을 가져 숙취 해소, 개선, 또는 예방 기능을 가진다.
본 발명에 따른 칸디다 유틸리스(Candida utilis) 균주는 알코올에 대한 내성과 알코올 분해능을 가지고 있어, 음주 전 또는 후 알코올 분해에 따른 숙취해소 효과와 간의 해독작용을 도와 향후 알코올성 간 질환을 예방할 수 있다. 본 발명에 따른 균주는 알코올(에탄올) 분해효소 활성이 우수하기 때문에 섭취한 알코올(에탄올)의 흡수를 효과적으로 억제함으로써 과다한 알코올 대사에 의한 간기능 장애와 알코올성 간질환 및 장 질환 발생을 예방 할 수 있게 있으며, 더욱이 본 발명에 따른 균주는 글루타치온을 생산하므로 알코올 대사 과정에서 발생되는 독성물질로 인한 간 독성을 예방, 개선 또는 경감할 뿐만 아니라 알코올 대사산물의 배출을 촉진할 수 있다.
본 발명에 따른 약학 조성물은 유효성분 이외에, 약제학적으로 허용 가능한 담체 및 첨가제를 선택하여 첨가함으로써 통상적인 제형으로 제조할 수 있다. 상기 의약 조성물은 액제, 현탁제, 산제, 과립제, 정제, 캡슐제, 환제, 또는 엑스제의 제형으로 제제화 하여 경구 복용할 수 있으나, 이에 한정되는 것은 아니다. 상기 약제학적으로 허용 가능한 담체로는 희석제, 활택제, 결합제, 붕해제, 감미제, 안정제, 방부제 중에서 1 종 이상을 선택하여 사용할 수 있으며, 약제학적으로 허용 가능한 첨가제로는 향료, 색소, 감마제, 및 산미제 중에서 1 종 이상을 선택하여 사용할 수 있다.
또한, 첨가제로서 미각을 돋구기 위하여, 매실향, 레몬향, 파인애플향, 허브향 등의 천연향료, 천연과즙, 클로로필린, 플라보노이드 등의 천연색소, 과당, 벌꿀, 당알코올, 설탕과 같은 감미성분, 또는 구연산, 구연산 나트륨과 같은 산미제를 혼합하여 사용할 수도 있다.
상기 약학 조성물은 숙취 예방 또는 치료 효과를 얻기 위하여, 유효성분으로서 체중 60 kg 성인을 기준으로 1 일 총량이 0.3~10g, 바람직하게는 0.7~4.2g이 되도록 임의로 수회 경구투여할 수 있으나, 이에 한정되지 않고 목적 하는 효과를 위해 적절한 함량으로 사용될 수 있다.
아울러 본 발명의 조성물은 건강보조식품으로서 사용될 수도 있다. 이러한 건강보조식품은 상기 약용식물의 추출물을 유효성분으로 한 차, 젤리, 엑스, 음료 등으로 제조될 수 있다.
본 발명은 트리펩타이드 생산능 및/또는 높은 ADH활성을 갖는 칸디다 유틸리스(Candida utilis) 균주, 이를 이용한 알코올 분해용 조성물, 숙취 해소, 숙취 개선, 또는 숙취 예방용 조성물, 또는 산화 스트레스 경감용 조성물 또는 항산화 조성물을 제공한다.

하기 실시예를 들어 본 발명을 더욱 자세히 설명할 것이나, 본 발명이 하기 실시예로 한정되는 의도는 아니다.
실시예 1: 글루타치온 생성 균주의 분리
1-1:균주 배양
글루타치온을 생산하는 미생물을 탐색하기 위해 전국의 전통시장에서 판매하는 막걸리, 누룩, 전통 장류 등을 구입하여 시료로 사용하였다. 시료 1g을 0.85% NaCl 10mL에 현탁하고, 현탁액 100μl를 YPD (Yeast extract 10 g/L, Peptone 20 g/L, Dextrose 20 g/L) agar plate에 도말한 후 30℃에서 2일간 고체 배양을 실시하였다. 고체배지에서 자란 콜로니 중에서 모양과 크기가 각기 다른 것을 선별하여 150개의 콜로니를 분리한 후 YPD broth (yeast extract 10 g/L, Peptone 20g/L, Dextrose 20g/L)를 이용하여 Test tube에서 30℃에서 2일간 진탕 배양하여 균주 배양물을 얻었다.
1-2:균체 증식정도(흡광도) 측정
상기 균주 배양물에 대해 600nm 에서 흡광도를 측정하여 균체 농도를 측정하여, 그 결과를 cell OD 값을 측정하였다.
1-3: 배양물에 포함된 글루타치온 함량 측정
상기 균주 배양액을 원심분리하여 상등액을 제거하고 증류수로 1회 세척하여 균체를 회수하였다. 회수한 균체에 40~70% 에탄올을 투입하고 fine mixer를 사용하여 10~30분간 세포 내 글루타치온을 추출하였다. 상기 추출용액을 원심분리 후 상등액을 취하여 0.5M potassium phosphate pH8.0 버퍼에 용해시킨 10mM DTNB (5,5'-Dithiobis-(2-Nitrobenzoic Acid))와 40℃에서 20분간 반응하여 412nm에서 흡광도를 측정하여 글루타치온 함량을 확인하였고 상기 글루타치온 함량(GSH mg/L)을 측정하였다. 글루타치온 분석에 주로 사용되는 DTNB는 Ellman’s reagent로 알려져 있고 thiol 화합물을 검출하기 위해 고안된 시약이다. DTNB와 GSH의 반응으로 노란색의 2-nitro-5-benzoic acid 와 GSSG가 생성되는데 412nm에서 OD값을 측정함으로서 GSH의 농도를 계산할 수 있다. GSSG는 glutathione reductase에 의해 GSH로 환원되어 다시 DTNB와 반응하는 recycling system을 이루게 된다.
1-4: 균체 건조 중량(g) 당 글루타치온 함량 측정
상기 균주 배양액을 원심분리하여 상등액을 제거하고 증류수로 1회 세척하여 균체를 회수하였다. 상기 회수된 균체에 대해 흡광도를 측정하고, 흡광도에 따른 건조균체농도를 계산하였다. 구체적으로, 균체 건조 중량(g)은 상기 배양액을 원심분리 후 상등액을 제거하고 균체만 회수하고, 회수한 균체를 0.9% NaCl로 세척하고 증류수로 희석하여 흡광도 0.1~1 사이가 되도록 샘플을 준비하였다. 상기 희석한 균체 시료에 대해 600nm에서 흡광도를 측정하고 흡광도에 따른 건조균체 중량을 계산하였다. 상기 흡광도를 측정한 후에, 상기 균체를 0.2㎛의 여과지로 감압하에서 여과하였다. 균체가 걸러진 여과지는 60℃에서 12시간 이상 건조하고, 실리카겔이 들어있는 데시케이터에서 6시간 이상 방치한 후 무게를 측정하였다. 빈 여과지와 균체가 걸러진 여과지의 무게 차이를 계산하여 건조 균체량을 확인하였다. 따라서, 흡광도 값에 따른 건조균체농도(g/L)을 확인할 수 있었다.
상기 흡광도 측정 후 균체에 대해 상기 실시예 1-3과 실질적으로 동일한 방법으로 글루타치온을 추출하고 상기 추출용액을 원심분리 후 상등액을 취하여 글루타치온 생성량(g/L)를 측정하였다. 측정된 글루타치온 생성량을 상기 계산된 건조 균체 농도(g/L)로 나눈 뒤 100을 곱하여 건조 균체 중량(g)당 GSH %를 계산하였다. 상기 균체 건조 중량(g) 당 글루타치온 함량을 측정하여 하기 표 1에 건조균체 중량(g)당 GSH 함량(%)을 GSH(%)/g-cell으로 나타낸다.
상기 방법으로 150개 균주의 글루타치온 생산성을 확인한 결과로서 균체 건조 중량(g) 당 글루타치온 함량(GSH(%)/g-cell)은 약 0.3~2 중량% 범위의 분포를 나타내었고, 균체 건조 중량(g) 당 글루타치온의 함량을 기준으로 상위 7개 후보 균주를 분리하였다. 상기 선별된 7개 균주의 분석 결과로서, 실시예 1-2의 cell OD, 실시예 1-3의 글루타치온 함량, 및 균체 건조 중량(g) 당 글루타치온 함량을 표 1에 나타냈으며, 균체 건조 중량(g) 당 글루타치온 함량을 도 1에 나타냈다.
대조군으로서, 표준균주 C. utilis KCCM 11355에 대해서는 동일하게 실험을 수행하여 결과를 표 1 및 도 1에 나타냈다.

상기 표 1에 나타낸 바와 같이, 에탄올 무함유 배지에서 균체 성장과 글루타치온 생성량을 분석한 결과, 선정된 7종 균주의 글루타치온 함량은 표준균주 C. utilis KCCM 11355보다 높으며, 구체적으로 약 0.7 내지 2 중량%의 글루타치온 함량 및 생산량을 나타냄을 확인하였으며, SYC-7D 균주를 제외한 6개 균주가 표준 균주의 균체 성장 정도가 동등하거나 높아 더욱 바람직한 특성을 가짐을 확인하였다.
따라서, 균주 선정 기준에서 글루타치온 생산량 (mg/L) 및 건조균체 중량당 글루타치온 생산량 (GSH(%)/g-cell)를 고려하면 SYC-7D, SYC-JH, SYC-P1, SYC-P3, SYC-PR9, SYC-PR19, SYC-PR20을 선정할 수 있으며, 바람직하게는 건조균체 중량당 생산량 (GSH(%)/g-cell)이 0.8 중량%이상인 SYC-7D, SYC-JH, SYC-P1, SYC-P3, SYC-PR9, SYC-PR19, SYC-PR20가 선정될 수 있으며, 더욱 바람직하게는 1.5 중량%이상인 SYC-PR9, SYC-PR19, SYC-PR20이 선정될 수 있다.
특히, 산업적 생산을 고려하는 경우 균체 OD값과 건조균체 중량당 생산량 (GSH(%)/g-cell)를 함께 고려함이 더욱 바람직함을 알 수 있었다. 이러한 과점에서, 표준 균주보다 균체 OD값이 높으면서도 글루타치온 생산량이 높은 균주는 SYC-P1, SYC-PR9, SYC-PR19, SYC-PR20이 더욱 바람직함을 확인하였다.
실시예 2: 알코올 분해 효소 (ADH) 활성 분석
상기 실시예 1에서 선정된 글루타치온 함량이 높은 7종 균주를 배양하여 ADH 활성을 확인하였다.
구체적으로 ADH 활성은 ADH Activity Assay Kit (Abcam)를 사용하였다. 온도 30℃, YPD 배지에서 24~48시간동안 배양한 후 cell이 1*106 CFU/ml이 되도록 회수하였다. 증류수로 회수한 cell을 세척한 후 ADH assay buffer를 첨가하고 bead beater를 사용하여 세포벽을 파쇄하였다. 반응액 조성은 ADH assay buffer 82 μl, Developer 8 μl, Isopropanol 10 μl에 샘플 또는 NADH 표준물질을 농도별로 50 μl 혼합하였다. 37℃에서 3분간 반응 후 450nm에서 실험구(A0)와 대조구 흡광도를 측정하였다. 37 ℃에서 30분간 추가 반응 후 450nm에서 흡광도의 변화를 측정한 후 단위 시간당 생성된 NADH 농도를 계산하여 균주별 ADH 활성을 비교하였다. 상기 측정된 균주별 NADH 생성량을 도 2에 나타낸다. 7종 균주가 24~160nmol 범위의 분포를 갖는 NADH 생성량를 갖는 것을 확인하였다. 대조군으로서, 표준균주 C. utilis KCCM 11355에 대해서는 동일하게 실험을 수행하여 결과를 표 2에 나타낸다
효모 균체의 파쇄 후 얻은 상등액으로 7종 균주의 ADH 활성을 비교한 결과를 하기 표 2에 나타낸다. 표 2에 기재된 수치는 반응액(ml)당 Alcohol dehydrogenase(ADH)의 역가(mUnit, mU)의 의미이며 단위는 mU/ml이며, 단위 시간(분) 당 생성되는 NADH 농도를 기준으로 ADH 활성을 계산하여 얻는 결과값을 나타낸다. 또한, 단위 시간(분) 당 생성되는 NADH 농도를 도 2에 나타냈다.

상기 표 2에 나타낸 바와 같이, 7종 선정 균주의 ADH 활성은 0.03~0.26 mu/ml의 분포를 나타내었으며, 표준균주 C. utilis KCCM 11355 (대조군)의 ADH활성보다 높은 균주는 SYC-P1, SYC-PR9, SYC-PR19, SYC-PR20 이었고, 가장 높은 활성을 나타낸 균주는 SYC-PR20으로 확인되었다. 이에, ADH 활성 측면에서 SYC-P1, SYC-PR9, SYC-PR19, SYC-PR20를 2차로 선정할 수 있었다. 바람직하게는 ADH 활성이 0.20 (mU/ml)이상인 SYC-P1, SYC-PR9, SYC-PR20를 선정할 수 있었다.
상기 실시예 1의 표 1에 나타낸 균체 중량과 균체 건조 중량(g) 당 글루타치온 함량과, 표 2의 균주의 ADH 활성 분석 결과를 종합하면, 글루타치온 생성량과 ADH 활성이 모두 높은 균주가 바람직하다. 이에 따라 SYC-PR9, SYC-PR19, SYC-PR20이 2차 후보 균주로 선정될 수 있다.
실시예 3: 배양시간에 따른 균체 성장 및 GSH생산 평가
실시예 1에서 선정된 SYC-PR20에 대해 배양시간에 따른 균체 성장 및 GSH생산 평가하였다.
SYC-PR20 균주 배양을 위한 배지는 YPD (yeast extract 10 g/L, Peptone 20g/L, Dextrose 20 g/L)를 제조하고. Test tube에 배지 3 ml을 분주하고, 30℃에서에서 24 내지 60시간까지의 배양시간을 진탕배양을 수행하였다. 상기 배양물을 배양시간 24시간, 36시간, 48시간 및 60시간에 각각 취하여, 실시예 1-2 내지 실시예 1-4와 실질적으로 동일한 방법으로 cell OD, 글루타치온 함량, 및 균체 건조 중량(g) 당 글루타치온 함량을 각각 측정하여 그 결과를 하기 표 3에 나타낸다.

균주의 배양 시간 24시간에서 60시간까지 균체 O.D 값과 GSH 생산량을 측정한 결과, 균체 O.D는 24시간에 12에서, 36시간에 15, 48시간에 18, 60시간에 19까지 증가하였다. GSH 생산량은 24시간에 69, 36시간에 82, 48시간에111, 60시간에 120mg/L로 생산됨을 확인되었다. 따라서 건조균체량 g 당 GSH %는 24시간에 1.4%에서 60시간 까지 1.6%로 증가함을 확인할 수 있었다.
실시예 4: 미생물의 에탄올 내성 평가
실시예 1에서 선정된 SYC-PR20에 대해 에탄올 함유 배지에서 균체 성장을 평가하였다.
구체적으로, 배지는 5X YPD (yeast extract 50 g/L, Peptone 100 g/L, Dextrose 100 g/L)를 제조하여 100% 에탄올과 혼합하여 배지 내 최종 에탄올 농도를 0, 2, 4, 6, 8, 10, 또는 15 v/v%로 조정하였다. Test tube에 농도별 에탄올이 함유된 배지 3 ml을 분주하고 30℃에서 60시간 동안 진탕배양하여 배양물을 얻었다. 상기 배양물에 대해, 실시예 1-2와 실질적으로 동일한 방법으로 600nm 에서 흡광도를 측정하여 균체 농도를 측정하고, 그 결과를 하기 표 4에 나타낸다.
대조군으로서, 표준균주 C. utilis KCCM 11355 (대조군)에 대해서는 동일하게 실험을 수행하여 결과를 표 4에 나타낸다.

내알코올성이 높은 균주로 알려진 Saccharomyces serevisiase가 에탄올7~11%(v/v)농도 에서 생육 가능하며, 실시예 2 및 3에서 2차 선정된 SYC-PR20 균주가 높은 내알코올성을 가지며, 특히 표준균주 C. utilis KCCM 11355(대조군)에 비해 SYC-PR20 균주는 알코올 함유 배지에서 높은 균체 성장을 나타내며, 특히 에탄올 농도 0.5 내지 15 v/v%에서 110%이상의 높은 균체 성장을 보임을 확인하였다.
상기 표 4에서 ADH activity가 상대적으로 높은 SYC-PR20 균주가 대조군에 비해 동일한 에탄올 농도에서 균체 농도가 높은 것을 확인하였다. 이에, SYC-PR20 균주가 글루타치온 생성능, ADH 활성 및 알코올 내성이 우수한 것을 확인하였다.
실시예 5: 균주 동정
실시예 4에서 SYC-PR20 균주는, 글루타치온 수율 1.6%, ADH activity 0.26 mU/ml로 선별된 7종의 균주 가운데 가장 우수한 특성을 나타내었다.
상기 SYC-PR20 균주를 18S rRNA 서열 분석을 진행하였으며 universal primer ITS1 (서열번호 2: 5'-TCCGTAGGTGAACCTGCGG-3'), ITS4(서열번호 3: 5'-TCCGTAGGTGAACCTGCGG-3')를 사용하였다. 상기 SYC-PR20 균주의 18S rDNA 서열은 서열번호 1에 나타냈다. 상기 18S rDNA 서열정보를 바탕으로 균주 동정을 실시한 결과 Candida utilis (Pichia jadinii)로 확인되었다.
상기 Candida utilis SYC-PR20 균주를 서울시 서대문구 홍제내 2가길 45에 위치한 한국미생물보존센터에 2020년 8월 7일에 기탁하여 수탁번호 KCCM 12777P을 받았다.
실시예 6: 발효조 배양에 따른 글루타치온 생산량 평가
미생물의 분리 및 특성 조사를 위한 실시예 1-4과 같은 플라스크 배양 조건에서는 기계적인 한계로 일정 이상 교반속도 증가가 어려우며, 일정한 배양 환경 (공기량, pH 등) 조절이 어렵다. 이와 달리, 산업적 배양은 배양액에 에어를 공급하고 교반을 수행함으로써 배지성분 및 산소 등이 모든 공간에서 일정하게 분포되고, 온도, pH 등의 조절이 가능하여 최적화 조건에서 배양이 가능하다. 이에, 발효조 배양은 flask 대비 높은 균체 성장율을 나타내며 그에 따라 투입된 당의 소비도 빠르게 일어나고 생산성도 높아지게 된다. 이에, 산업적 균주 적용을 위한 발효조 배양 실험이 필요하다.
구체적으로 실시예 5에 따른 Candida utilis SYC-PR20 균주가 접종된 YPD agar plate에서 형성된 colony를 YPD (Yeast extract 10 g/L, Peptone 20 g/L, Dextrose 20 g/L) broth 3mL에 colony를 접종하여 온도 30 ℃, 240rpm의 조건하에서 24시간 동안 배양한 종균을 다시 동일한 YPD broth 100mL에 앞서 배양한 종균 3mL을 접종하여 동일한 조건 하에 배양을 실시하여 5L 발효조용 종균을 준비하였다.
탄소원에 따른 균체 성장 및 글루타치온 생산량을 평가하기 위해, 5L 발효조 배양을 수행하였으며, 표 5의 배지 조성으로 5L 발효조에서 최종 2L 배양 부피로 포도당 기반 배양 배지 및 설탕 기반 배양 배지를 각각 준비하였다. 상기 준비된 종균을 5L 발효조에 접종하여 표 6과 같은 조건으로 교반하면서 배양을 수행하였다.

상기 얻어진 배양액에 대해 균체 농도 및 글루타치온 생성량을 실시예 1의 방법과 실질적으로 동일한 방법으로 측정하였으며, 그 측정 결과를 하기 표 7에 나타냈다.

표 7의 결과에 나타낸 바와 같이, 초기 투입된 당이 거의 소모된 시점은 포도당 및 설탕 기반 배양 모두 29시간에 배양이 종료 되었으며, 그 때의 균체 농도는 포도당 기반 균체의 600 nm OD값이 28.4, 설탕 기반 균체의 600 nm OD값이 OD 31.9로 비교적 설탕 기반의 대당 균체 수율이 112% 증가한 것으로 나타났다. 상기 글루타치온 함량은 설탕 기반 223.3mg/L로 동일 시간 배양 에서 포도당 기반 대비 약 122% 증가한 결과를 나타냈다.
실시예 7: 배양조건에 따른 글루타치온 생산량 평가
상기 선별된 Candida utilis SYC-PR20 균주가 접종된 YPD agar plate에서 형성된 colony를 YPD (Yeast extract 10 g/L, Peptone 20 g/L, Dextrose 20 g/L) broth 3mL에 colony를 접종하여 온도 30 ℃, 240rpm의 조건하에서 24시간 동안 배양한 종균을 다시 동일한 YPD broth 100mL에 앞서 배양한 종균 3mL을 접종하여 동일한 조건 하에 배양을 실시하여 5L 발효조용 종균을 준비하였다.
고농도 균체 및 글루타치온 생산량을 평가하기 위해, 실시예 5와 같이 종균 및 5L 발효조 배양을 준비하였으며, batch 배양에서 초기 당 소비 이후 축적된 에탄올 농도가 5g/L 이하일 때 Sucrose 600g/L인 당액을 연속적으로 공급하는 방법으로 Fed-batch 배양을 수행하여 고농도의 균체 농도 배양과 글루타치온 생산성을 유지 하는 방법에 대한 실험을 수행하였다.
그 결과로 sucrose feeding rate에 따른 배양액 내 글루타치온 함량의 변화량을 배양 시간에 따라 측정하여 도 3에 나타내었으며, 배양액 내 에탄올 함량을 도 4에 나타내었다. 또한, sucrose feeding rate에 따른 시간별 균체 농도 변화를 도 5에 나타내었다. 도 3, 도 4 및 도 5에서 흰구는 Sucrose feeding rate 4.5g/L.h-1을 의미하고, 검은 구는 Sucrose feeding rate 6g/L.h-1를 의미한다.
Sucrose feeding rate 6g/L.h-1 공급 시 배양액에 함유된 에탄올 함량 3g/L 이상 유지되며, 그에 따라 글루타치온 함량 변화율이 거의 일정하게 나타났다. 반면, 낮은 Feeding rate (4.5g/L.h-1)에서는 에탄올 함량이 낮게 유지되며 세포 내 글루타치온 함량이 감소함을 나타났다. 이러한 사실은, sucrose feeding rate가 높은 경우 배양액 내 충분한 당 공급에 의해 글루타치온 생합성에 필요한 에너지가 충분히 공급되나, sucrose feeding rate가 낮은 경우 에너지 공급이 충분하지 않아 균체가 제한적으로 성장하고 그 결과 글루타치온 함량이 감소하게 되는 것으로 나타났다
따라서, 본 발명에 따른 Candida utilis SYC-PR20 균주는 Feeding rate 6g/L.h-1으로 공급 시, 충분한 당 공급에 의한 균체 대사산물로서 배양액 내 에탄올이 축적되며, 고농도 배양 구간에서 batch 수준의 글루타치온 함량을 유지할 수 있어, 더욱 바람직하다.

Claims (13)
- 알코올 분해능(Alcohol dehydrogenase, ADH) 및 알코올 내성을 가지며, 글루타치온 생산능을 갖는 칸디다 유틸리스(Candida utilis) 균주.
- 제1항에 있어서, 상기 균주는 균체의 건조 중량(g) 당 ADH활성이 0.18 mU/ml이상이고, 균체의 건조 중량(g) 당 글루타치온 생성량이 0.8 중량%이상인 균주.
- 제1항에 있어서, 상기 균주는 에탄올 농도가 2 내지 15%(v/v)인 조건에서 생육할 수 있는 알코올 내성을 갖는 균주.
- 제3항에 있어서, 상기 균주는, 알코올 농도가 6 내지 15%(v/v)인 조건에서 배양한 경우 기탁번호 KCCM 11355을 갖는 Candida utilis의 균체 광학밀도(optical density, OD)값 100%를 기준으로 120 내지 내지 200%을 갖는 것인 균주.
- 제1항에 있어서, 상기 칸디다 유틸리스는 수탁번호 KCCM 12777P을 갖는 것인 균주.
- 제1항 내지 제5항 중 어느 한 항에 따른 칸디다 유틸리스(Candida utilis) 균주의 균체, 상기 균주의 배양물, 상기 균체의 파쇄물, 상기 균체의 용해물, 및 이들의 추출물로 이루어지는 군에서 선택된 1종 이상을 포함하는, 산화 스트레스 관련 질환의 에방, 개선 또는 치료용 조성물.
- 제6항에 있어서, 상기 조성물은 AIDS, 당뇨병, 황반변성, 울혈성심장기능상실(congestive heart failure), 심장혈관질환(cardiovascular disease), 심장동맥재협착(coronary artery restenosis), 폐질환(lung disease), 염증성 질환(inflammatory disease), 천식(asthma), RNA 바이러스 감염, DNA 바이러스 감염, 패혈증(sepsis), 골다공증(osteoporosis), 골질환(bone disease), 미생물 감염, 독소노출(toxin exposure), 방사선노출(radiation exposure), 화상(burn trauma), 프라이온(prion)병, 신경질환(neurological disease), 혈액질환(blood disease), 혈구 질환(blood cell disease), 동맥질 환(arterial disease), 근육질환(muscle disease) 및 피부질환으로 이루어지는 군으로부터 선택되는 것인 조성물.
- 제1항 내지 제5항 중 어느 한 항에 따른 칸디다 유틸리스(Candida utilis) 균주의 균체, 상기 균주의 배양물, 상기 균체의 파쇄물, 상기 균체의 용해물, 및 이들의 추출물로 이루어지는 군에서 선택된 1종 이상을 포함하는, 알코올 분해용 조성물.
- 제8항에 있어서, 상기 균주는, 균체의 건조 중량(g) 당 알코올 탈수소화효소(alcohol dehydrogenase, ADH)활성이 0.18 mU/ml이상인 알코올 분해용 조성물.
- 제1항 내지 제5항 중 어느 한 항에 따른 칸디다 유틸리스(Candida utilis) 균주의 균체, 상기 균주의 배양물, 상기 균체의 파쇄물, 상기 균체의 용해물, 및 이들의 추출물로 이루어지는 군에서 선택된 1종 이상을 포함하는, 숙취 해소, 개선, 경감, 또는 예방용 조성물.
- 제1항 내지 제5항 중 어느 한 항에 따른 칸디다 유틸리스(Candida utilis) 균주를 배양하여, 글루타치온 생성량을 증가시키는 방법.
- 제11항에 있어서, 상기 균주의 배양은 탄소원을 설탕 또는 포도당으로 사용한 것인 방법.
- 제11항에 있어서, 상기 균주의 배양은, 발효액 내 에탄올 함량을 측정하여 탄소원 공급 속도를 조절하여 수행하는 것인 방법.
