Abstract
말초혈액 단핵구에서 NK 세포를 배양함에 있어서, NK 세포를 IL-15, IL-18, 및 IL-27를 포함하는 NK 세포 배양용 조성물을 함유하는 배지에서 배양하여, 대량으로 증식시키고 NK 세포의 활성화를 촉진시킬 수 있다. 따라서, 이를 이용하는 경우, 암세포의 세포사멸 또는 살상능이 촉진될 수 있어, 암의 예방 또는 치료에 효과적인 입양 면역세포 치료제로 사용할 수 있다.
Description
NK 배양용 조성물 및 이를 이용하여 NK 세포를 배양하는 방법에 관한 것이다.
현재 암의 일반적인 치료법은 외과적 수술(surgery), 방사선 요법(radiation therapy), 항암화학요법(chemotherapy) 등이 있으며, 암의 종류 및 병기에 따라 단독 혹은 복합적으로 사용되고 있지만, 환자들에게 심한 부작용과 고통을 수반하는 실정이다. 완벽한 암 치료법은 정상 세포는 손상시키지 않고 암세포만 골라서 죽이는 방법일 것이다. 하지만, 기존 치료법은 정상 세포에 어느 정도 손상을 미친다. 최근 부각되고 있는 암 면역치료법(cancer immunotherapy)은 인체 고유의 면역 시스템을 활용하여 정상 세포의 손상을 최소화하여, 암세포를 보다 특이적으로 제거하는 치료법으로, 여러 세부 분야(항체 치료법, 면역세포 치료법, 바이러스 면역치료법, 나노 기술 면역치료법 등)의 연구가 활발히 진행되고 있다. 이 중 면역세포 치료는 환자의 혈액에서 얻어진 림프구(lymphocyte) 중, 자연살해세포(natural killer cell), 자연살생 T 세포(natural killer T cell), T 세포, B 세포, 수지상 세포(dendritic cell) 등의 세포 수를 늘리고, 체외(in vitro) 에서 기능을 강화하여, 이를 다시 환자 몸에 되돌림으로써, 암을 치료하는 방법이다. 이러한 면역세포를 이용한 치료법은 면역 반응 조절 치료에 좋은 효과를 보이고, 독성 및 안전성 면에서 우수하다고 평가되고 있다. 이 중 NK 세포는 선천면역(innate immunity)을 담당하는 중요한 세포로써, T 세포와 달리 간(liver), 골수(bone marrow)에서 성숙된다. 특히, 바이러스(virus)에 감염된 세포나 종양세포(tumor cells) 등 비정상세포를 스스로 식별하여 사멸시키는 기능을 가진다. 지난 10 여 년 동안 환자들의 면역 시스템을 이용한 종양 면역치료가 꾸준히 발전되어 왔고, 이를 이용한 '세포 치료제(cell therapy product)'도 상업화되고 있다. 세포 치료제는 자가(autologous), 동종(allogeneic), 혹은 이종(xenogeneic)의 세포를 체외(in vitro)에서 증식, 선별하는 방법을 통해서, 세포의 생물학적 특성을 변화시키는 일련의 행위를 통하여 치료, 진단, 예방의 목적으로 사용하는 의약품으로 정의된다(식품의약안전청 고시 제2003-26호 제2조).
암과 같은 난치성 질환의 치료를 위한 세포 치료제로서 면역 거부 반응을 해소할 수 있으며, 환자 맞춤형 세포 치료를 제공할 수 있는, NK 세포를 대량으로 증식하는 방법 및 활성이 강화된 NK 세포를 배양하는 방법이 요구된다.
일 양상은 NK 세포 배양용 조성물을 제공한다.
다른 양상은 상기 NK 세포 배양용 조성물을 이용하여 NK 세포를 배양하는 방법을 제공한다.
일 양상은 IL-15, IL-18, IL-27 또는 이들의 조합을 포함하는 NK 세포 배양용 조성물을 제공한다.
NK 세포는 림프구의 일종인 거대 과립형 림프구(Large granular lymphocytes: LGL)로서, 감염된 바이러스 및 종양세포를 죽이는 능력이 뛰어나고, 대부분의 정상 세포는 죽이지 않는 특성을 가진다. 따라서, NK 세포는 활성 세포 독성 T 림프구가 다량으로 생성되기 전에 바이러스 감염 혹은 종양 형성 초기 단계에서 중요한 역할을 한다. 예를 들면, NK 세포가 표적 세포와 접촉하는 경우, 몇몇 분자는 표적 세포의 막에 구멍을 형성하여 세포를 용해시키며, 또 다른 분자는 표적 세포에 들어가서 핵 DNA의 분절을 증가시켜, 괴사(necrosis), 세포사멸(Apotosis) 또는 세포 예정사(Programmed cell death)를 야기한다.
그러므로, NK 세포는 특정 바이러스에 감염된 세포와 암세포를 선 자극 없이 용해 시킬 수 있다. TCR의 발현을 통해 항원-특이적으로 인식하는 세포독성 T 림프구와 다르게, NK 세포는 항원-특이적 수용체가 결여되어 있다. NK 세포는 정상 세포의 MHC 유형 I과 결합하는 살해 세포 억제 수용체(killer cell immunoglobulin-like receptor: KIR)를 발현한다. 살해 세포 억제 수용체는 MHC 유형 I과 결합하면 특정 전사 인자의 억제를 유도하는 세포 내 신호가 생성된다. 이 결과 NK 세포의 활성화, 표적 세포의 분해 및 파괴가 억제된다. 바이러스 감염 세포 또는 암 세포는 그들의 표면에 MHC 유형 I 분자가 크게 감소되어 있다. 따라서 이런 세포가 NK 세포를 만나면, 효과적으로 살해 세포 억제 수용체와 결합하지 못하기 때문에 NK 세포-매개 세포 독성에 노출되어 용해된다.
상기 NK 세포는, 예를 들면, 포유동물, 인간, 원숭이, 돼지, 말, 소, 양, 개, 고양이, 마우스 또는 토끼 등으로부터 유래한 것일 수 있다. 상기 NK 세포는 정상인 또는 암환자로부터 수득한 것일 수 있다. 상기 NK 세포는 혈액, 말초혈액 단핵구(Peripheral blood mononuclear cell: PBMC)로부터 분리된 것일 수 있다. 혈액을 분리하는 방법, 이로부터 PBMC를 분리하는 방법, 이로부터 NK 세포를 분리하는 방법은 공지된 방법으로 수행되는 것 일 수 있다.
상기 조성물은 IL-15, IL-18, IL-27 또는 이들의 조합을 포함할 수 있는데, IL-15는 NK 세포에서 발현되는 IL-15 수용체를 통해서 NK 세포의 성장과 분화를 직접적으로 증진시키는 것일 수 있고, IL-18은 NK 세포의 인터페론 감마 생산을 유도하고 세포 독성을 증진시키는 것일 수 있으며, IL-27은 면역 반응을 조절하고, NK 세포의 생존율을 증진시키며, 인터페론 감마의 발현을 증진시키는 것일 수 있다.
NK 세포 배양 배지 중에 상기 IL-15의 농도는 약 0.1 ng/ml 내지 약 1000 ng/ml, 약 0.2 ng/ml 내지 약 500 ng/ml, 약 0.5 ng/ml 내지 약 200 ng/ml, 약 1.0 ng/ml 내지 약 100 ng/ml, 약 2.0 ng/ml 내지 약 50 ng/ml, 또는 약 5.0 ng/ml 내지 약 20 ng/ml인 것일 수 있고, IL-18의 농도는 약 0.25 ng/ml 내지 약 2500 ng/ml, 약 0.5 ng/ml 내지 약 1250 ng/ml, 약 1.25 ng/ml 내지 약 500 ng/ml, 약 2.5 ng/ml 내지 약 250 ng/ml, 약 5.0 ng/ml 내지 약 125 ng/ml, 약 12.5 ng/ml 내지 약 50ng/ml인 것일 수 있으며, IL-27의 농도는 약 0.20 ng/ml 내지 약 2000 ng/ml, 약 0.40 ng/ml 내지 약 1000 ng/ml, 약 1.0 ng/ml 내지 약 400 ng/ml, 약 2.0 ng/ml 내지 약 200 ng/ml, 약 4.0 ng/ml 내지 약 100 ng/ml, 약 10.0 ng/ml 내지 약 40 ng/ml인 것일 수 있다.
상기 조성물은 IL-7, IL-21 또는 이들의 조합을 더 포함하는 것일 수 있다. IL-21은 골수로부터의 NK 세포 전구체의 성숙을 유도하고, 특히 NK 세포의 사이토카인 생성능 및 세포사멸능과 같은 효과기 기능(effector functions)을 증가시키는 것일 수 있다. IL-7은 NK 세포 전구체의 성숙을 유도하고, T 세포의 증식을 조절하는 것일 수 있다. 다만, IL-7, IL-21 또는 이들의 조합을 포함하지 않더라도, NK 세포의 증식 또는 활성화를 촉진할 수 있다.
상기 IL-7의 농도는 약 0.05 ng/ml 내지 약 500 ng/ml, 약 0.10 ng/ml 내지 약 250 ng/ml, 약 0.25 ng/ml 내지 약 100 ng/ml, 약 0.5 ng/ml 내지 약 50 ng/ml, 약 1.0 ng/ml 내지 약 25 ng/ml, 약 2.5 ng/ml 내지 약 10 ng/ml인 것일 수 있고, IL-21의 농도는 약 0.05 내지 약 500 ng/ml, 약 0.10 내지 약 250 ng/ml, 약 0.25 내지 약 100 ng/ml, 약 0.50 내지 약 50 ng/ml, 약 2.5 내지 약 10 ng/ml인 것일 수 있다.
상기 배지는 인 비트로(in vitro)에서 세포의 성장 및 생존을 지지할 수 있게 하는 물질을 의미한다. 상기 배지는 세포 배양에 이용될 수 있는 것이라면, 특별히 제한되는 것은 아니며, 예를 들면, DMEM (Dulbecco's Modified Eagle's Medium), MEM (Minimal Essential Medium), BME (Basal Medium Eagle), RPMI 1640, F-10, F-12, DMEM/F12, MEM-α (Minimal Essential Medium-α), G-MEM (Glasgow's Minimal Essential Medium), IMDM (Iscove's Modified Dulbecco's Medium), MacCoy's 5A 배지, AmnioMax complete 배지, AminoMax Ⅱ complete 배지, EBM (Endothelial Basal Medium) 배지, 및 Chang's Medium 배지로 이루어진 군으로부터 선택된 하나 이상을 포함하는 것일 수 있다.
상기 NK 세포는 CD3- 및 CD56+의 표면 항원 특성을 갖는 것일 수 있다.
상기 배양은 NK 세포의 증식 또는 활성화를 위한 것일 수 있다. 상기 NK 세포의 증식은 세포 수의 증가를 의미하며, 성장과 혼용될 수 있다. 상기 NK 세포의 활성화는 전술한 NK 세포가 제 기능을 수행하는 것을 의미하며, CD226+, CD69+, CD14-, CD19-, CD16+ 또는 이들의 조합의 표면 항원 특성을 갖는 것일 수 있다.
상기 표면 항원 특성은 면역학적 특성과 동일한 의미이며, 유세포 분석 또는 면역세포화학과 같은 기법을 사용하여 세포 표면 표지 (예를 들면, 조직-특이적 또는 세포-표지 특이적 항체로 세포를 염색함), 또는 광학 현미경 또는 공초점 현미경을 사용하여 세포 표면 표지를 관찰함으로써, 또는 중합효소 연쇄 반응 (polymerase chain reaction: PCR), 또는 유전자-발현 프로파일과 같은 당해 분야에 익히 공지된 기법을 사용하여 유전자 발현상의 변화를 측정함으로써 확인될 수 있다.
상기 "양성 또는 +"는 세포 표지(마커)와 관련하여, 그 표지가 기준이 되는 다른 세포와 비교하였을 때 더 많은 양 또는 더 높은 농도로 존재하는 것을 의미하는 것일 수 있다. 세포는 어느 표지가 세포 내부 또는 표면에 존재하기 때문에 그 표지를 이용하여 그 세포를 하나 이상의 다른 세포 유형과 구별할 수 있으면, 그 표지에 대하여 양성이 될 수 있다. 또한, 세포가 배경 값보다 더 큰 값으로 신호, 예를 들면, 세포 측정 장치의 신호를 낼 수 있는 만큼의 양으로 그 표지를 가지고 있다는 것을 의미하는 것일 수 있다. 예를 들면, 세포를 CD56에 특이적인 항체로 검출 가능하게 표지 할 수 있고, 이 항체로부터의 신호가 대조군(예를 들면, 배경 값)보다 검출 가능하게 더 크면, 그 세포는 "CD56에 대해 양성" 또는 "CD56+"으로 나타낼 수 있다. 용어, "음성 또는 -"는 특정 세포 표면 표지에 특이적인 항체를 사용하여도 배경 값에 비교하여 그 표지를 검출할 수 없음을 의미하는 것일 수 있다. 예를 들면, CD3에 특이적인 항체로 세포를 검출 가능하게 표지할 수 없으면. 그 세포는 "CD3에 대해 음성" 또는 "CD3-"으로 나타낼 수 있다.
다른 양상은, NK 세포를 IL-15, IL-18, 및 IL-27를 포함하는 NK 세포 배양용 조성물을 함유하는 배지에서 배양하는 단계를 포함하는 NK 세포(natural killer cell)를 배양하는 방법을 제공한다.
상기 배양하는 방법은 NK 세포를 대량 증식하는 방법 또는 NK 세포를 활성화시키는 방법인 것일 수 있다.
상기 방법은, 상기 배양하는 단계 전에, 말초혈액으로부터 말초혈액 단핵구(Peripheral blood mononuclear cell: PBMC)를 수득하는 단계; 및 수득된 PBMC로부터 NK 세포를 분리하는 단계;를 더 포함하는 것일 수 있다.
상기 단계는 CD3+의 표면 항원 특성을 갖는 세포를 제거하는 단계를 더 포함하는 것일 수 있다.
혈액을 분리하는 방법, 이로부터 PBMC를 분리하는 방법, 이로부터 NK 세포를 분리하는 방법, 및 특정 표현 항원 특성을 갖는 세포를 분리 또는 제거하는 방법은 특정 항체를 이용하는 등의 공지된 방법으로 수행되는 것 일 수 있다.
상기 방법에 따르면, NK 세포를 장기간 배양하거나 또는 수회 계대 배양하는 경우에도, NK 세포의 증식능이 다른 사이토카인을 투여하여 배양한 군에 비하여 우수하여, NK 세포를 대량으로 수득할 수 있다.
상기 방법에 따르면, 상기 조성물을 투여하고 배양된 NK 세포는, 다른 사이토카인을 투여하여 배양한 군에 비하여, IFN-감마 (interferon-gamma)의 분비가 증가된 것일 수 있다. 또한, 상기 조성물을 투여하고 배양된 NK 세포는, 다른 사이토카인을 투여하여 배양한 군에 비하여, 암세포에 대한 세포 독성이 증가한 것일 수 있고, 암세포의 세포사멸(apoptosis)을 촉진하는 것일 수 있다.
상기 배양은 약 7일 내지 약 2달, 7일 내지 약 30일, 약 14일 내지 약 30일, 약 15일 내지 약 27일, 약 16일 내지 약 26일, 약 17일 내지 약 25일, 약 18일 내지 약 24일, 약 19일 내지 약 23일, 또는 약 20일 내지 약 22일 동안 수행되는 것일 수 있다.
다른 양상은 상기 NK 세포를 배양하는 방법에 의하여 제조된 NK 세포를 제공한다.
다른 양상은 상기 NK 세포를 배양하는 방법에 의하여 제조된 NK 세포를 포함하는 암 예방 또는 치료용 조성물을 제공한다.
상기 암은 고형암, 폐암, 간암, 유방암, 자궁암, 혈액암 등인 것일 수 있으나 이에 제한되는 것은 아니다. 암환자에서 이러한 NK 세포가 폐암(Carrega P, et al., Cancer, 112, 863-875, 2008), 간암(Jinushi M, et al., J Hepatol., 43, 1013-1020, 2005), 유방암(Bauernhofer T, et al., Eur J Immunol., 33, 119-124, 2003.), 자궁암(Mocchegiani E., et al., Br j Cancer., 79, 244-250, 1999), 혈액암(Tajima F., et al, Lekemia, 10, 478-482, 1996) 등의 질환의 발생과 깊은 연관이 있는 것으로 보고되어 있다.
상기 조성물은 약학적으로 허용가능한 담체를 포함하는 것일 수 있다. 상기 조성물에 있어서, "허용가능한 담체"는 활성 성분의 적용을 돕기 위하여 활성 성분과 조합되어 사용되는 물질, 일반적으로 불활성 물질을 나타낸다. 상기 담체는 부형제, 붕해제, 결합제, 활택제, 희석제, 또는 그 조합일 수 있다. 상기 부형제는 미결정 셀룰로오즈, 유당, 저치환도 히드록시셀룰로오즈, 또는 그 조합일 수 있다. 상기 붕해제는 전분글리콜산 나트륨, 무수인산일수소 칼슘, 또는 그 조합일 수 있다. 상기 결합제는 폴리비닐피롤리돈, 저치환도 히드록시프로필셀룰로오즈, 히드록시프로필셀룰로오즈, 또는 그 조합일 수 있다. 상기 활택제는 스테아린산 마그네슘, 이산화규소, 탈크, 또는 그 조합일 수 있다.
다른 양상은 상기 NK 세포를 배양하는 방법에 의하여 제조된 NK 세포의 치료학적 또는 약제학적 유효량을 개체에 투여하는 단계를 포함하는 암을 치료하는 방법을 제공한다.
상기 "투여"는 어떠한 적절한 방법으로 개체에게 소정의 물질을 도입하는 것을 의미하며, 물질의 투여 경로는 목적 조직에 도달할 수 있는 한 어떠한 일반적인 경로를 통하여 투여될 수 있다. 복강내 투여, 정맥내 투여, 근육내 투여, 피하 투여, 피내 투여, 경구 투여, 국소 투여, 비내 투여, 폐내 투여, 또는 직장내 투여인 것일 수 있으나, 이에 제한되지는 않는다. 또한, 투여는 표적 세포로 이동할 수 있는 임의의 장치에 의해 수행될 수 있다. 투여량은 암의 유형, 투여 경로, 환자의 나이 및 성별, 및 질병의 정도에 따라 적절히 선택될 수 있으나, 평균 성인의 경우, 약 1x106 내지 약 1x1011 세포로 투여할 수 있다.
상기 "치료학적 유효량"은 치료를 필요로 하는 개체 또는 세포에게 투여되는 경우 치료 효과를 나타내기에 충분한 양을 의미한다. "치료"는 개체, 예를 들면 사람을 포함한 포유동물에서 질환 또는 의학적 증상을 치료함을 의미하고, 이는 다음을 포함한다: (a) 질환 또는 의학적 증상의 발생을 예방, 즉, 환자의 예방적 치료; (b) 질환 또는 의학적 증상의 완화, 즉, 환자에서 질환 또는 의학적 증상의 제거 또는 회복 야기; (c) 질환 또는 의학적 증상의 억제, 즉, 개체에서 질환 또는 의학적 증상의 진행을 늦춤 또는 정지; 또는 (d) 개체에서 질환 또는 의학적 증상을 경감.
말초혈액 단핵구에서 NK 세포를 배양함에 있어서, NK 세포를 IL-15, IL-18, 및 IL-27를 포함하는 NK 세포 배양용 조성물을 함유하는 배지에서 배양하여, 대량으로 증식시키고 NK 세포의 활성화를 촉진시킬 수 있다. 따라서, 이를 이용하는 경우, 암세포의 세포사멸 또는 살상능이 촉진될 수 있어, 암의 예방 또는 치료에 효과적인 입양 면역세포 치료제로 사용할 수 있다.

(1.1) 연구 대상 선정, 혈액 및

만 20세 내지 65세 건강한 남성 및 여성으로서, 본 연구를 위한 채혈에 동의한 대상자를 대상으로 하여 문진, 체중, 활력 징후를 측정하여 연구 대상 적합자를 선정하였다. 적합자는 하기와 같은 기준으로 선정하였다.
1) 문진을 통해 아래의 제외 조건이 없는 자
- 고혈압 등의 심혈관계 질환, 신장 질환, 당뇨, 암을 진단받은 과거력이 있는 자
- 종교적인 이유 등으로 수혈을 할 수 없는 자
- 임산부
2) 체중은 남자 50kg 이상, 및 여자 45kg 이상인 자
3) 활력 징후는 하기의 조건을 만족하는 자
- 혈압(mmHg) : 수축기 90~179, 이완기 100 미만
- 체온(℃) : 37.5℃ 이하
- 맥박(회/분) : 50~100
연구 대상 적합자로 선정된 대상에 대하여, 방문 당일, 1회 총 100 ml를 채혈하여, 헤파린(heparin)이 들어있는 튜브에 수집하였다.
채혈된 혈액에서 Ficoll-Paque(GE Healthcare, 17-1440-02)를 이용하여 다음과 같은 방법으로 말초혈액 단핵구(Peripheral blood mononuclear cell: PBMC)를 분리하였다. 채혈된 전혈을 인산염 완충 식염수(phosphate buffered saline: PBS)(pH 7.4, Thermo Fisher Scientific)와 1:1로 희석하고, Ficoll 상층에 조심스럽게 희석된 혈액을 첨가하였다. 이어서, 2500rpm으로 22분 동안 25℃에서 원심 분리하여, PBMC를 분리하였다. 분리된 PBMC를 PBS로 세척하였다. 그 후, NK 세포 분리 전까지 PBMC 상태로 보관하였다. 또한, 분리 과정에서 나오는 혈장(Plasma)을 수집하여 함께 보관하였다.
분리된 PBMC에서 NK 세포 분리 키트(NK Cell isolation kit, Miltenyi Biotec, 130-092-657) 및 CD3+ 자기 비드(Miltenyi Biotec)를 이용하여 다음과 같은 방법으로 NK 세포를 분리하고, CD3+ 세포를 제거하였다. 분리된 PBMC를 1x107개의 세포 당 40 ul의 MACS 러닝 버퍼(MACS 러닝 버퍼, PBS, 2mM EDTA, 0.5% BSA)를 첨가하여 PBMC 세포 펠렛을 풀어준 뒤, 1x107개의 세포 당 20 ul의 CD3+ 자기 비드를 첨가하여 4℃에서 10분 동안 반응시켰다. 이어서, MACS 버퍼로 세척한 후, MACS 세포 분리기(Miltenyi Biotec)를 이용하여 CD3-CD56+의 NK 세포를 회수하였다.
도 1 및 표 1은 건강한 정상인으로부터 분리된 PBMC 및 NK 세포를 계수한 결과이다. 도 1에서 A는 건강한 정상인으로부터 분리된 PBMC를 계수한 결과이고, 도 1에서 B는 건강한 정상인으로부터 분리된 NK 세포를 계수한 결과이다. 도 1 및 표 2에 나타낸 바와 같이, 혈액 속 PBMC에서 NK 세포는 평균 약 8 내지 20%로 분포하였다.

이어서, 분리된 PBMC 중의 NK 세포 및 회수된 NK 세포가 CD3-CD56+의 특성을 가지는 것을 FACS(형광표지세포분류기, Fluorescence-activated cell sorting, BD FACSCalibur™)를 이용하여 확인하였다.
도 2는 건강한 정상인으로부터 분리된 PBMC 중의 NK 세포 및 회수된 NK 세포의 표현형을 분석한 FACS 결과이다. 도 2에서 A는 NK 세포를 분리한 직 후(D0)와 배양 21일 후(D21), NK 세포의 표현형을 분석한 결과이다. 도 2에서 B는 PMBC, NK 세포를 분리한 직 후(D0)와 배양 21일 후(D21), CD3-CD56+인 NK의 분포를 분석한 결과이다. 도 2를 참조하면, 건강한 정상인(대상 #8)으로부터 회수된 NK 세포는 95% 이상의 순도를 가지며, T 세포, B 세포, 및 단핵구(monocyte) 등은 약 1 내지 2 %로 분포하였다.

(1)에서 수득한 NK 세포를 1x105 세포/ml 농도로 12 또는 24-웰 티슈 배양 플레이트(tissue culture plate)의 각 웰에 넣은 다음, CellGro® 무혈청 배지(Serum-free medium)(CellGenix, USA), 10 % 인간 혈청(human serum)(Sigma Aldrich, USA), 10,000 U/mL 페니실린/스트렙토마이신(Pen/Strep)(Gibco/Life Technologies, Carlsbad, CA), 사이토카인(Cytokine)(IL-15, IL-27, 1-100ng/ml,; Peperotech, Inc. NJ, USA; IL-18, 1-100ng/ml; R&D Systems, Inc., MN, USA)을 첨가하여 37℃, 5 %의 CO2 항온기에서 21일 동안 배양하였다.
구체적으로,사이토카인 혼합처리군 4 세트, 혼합처리군 1-1: IL-7(5 ng/ml), IL-15(10 ng/ml), IL-18(25 ng/ml), IL-21(5 ng/ml), 및 IL-27(20 ng/ml), 혼합처리군 1-2: IL-15(10 ng/ml), IL-18(25 ng/ml), IL-21(5ng/ml), 및 IL-27(20 ng/ml), 및 혼합처리군 1-3: IL-15(10 ng/ml), IL-18(25 ng/ml), 및 IL-27(20 ng/ml), 혼합처리군 1-4: IL-15(10 ng/ml) 및 IL-18(25 ng/ml), 각각을 새로운 배지에 혼합한 후, 배지를 교체하는 시기인 2 내지 3일에 한번 씩, 배양 중인 NK 세포에 첨가하여 배양하였다. 사이토카인을 제외하고, 배지의 다른 조성은 전술한 바와 동일하다.
도 2를 참조하면, 건강한 정상인(대상 #8)으로부터 회수된 NK 세포는, 혼합처리군 1-3의 NK 세포 배양용 조성물을 첨가하여 배양된 후에도, 약 95% 이상의 순도를 가지는 것을 알 수 있다.

(1.2)과 같이 배양하는 중에, 증식된 NK 세포의 수는, 7일 간격으로 트립판 블루(trypan blue) 염색제(Thermo Fisher Scientific, USA)를 이용해서 증식된 NK 세포를 염색한 뒤, 헤모사이토미터(Hemocytometer)를 이용하여 측정하였다.
도 3은 본 발명의 NK 세포 배양용 조성물이 NK 세포의 증식 및 생존율(viability) 미치는 영향을 확인한 결과이다. 도 3에서 A에 나타낸 바와 같이, 본 발명의 NK 세포 배양용 조성물 중 혼합처리군 1-3을 NK 세포에 가하여 배양한 경우, 일차(primary) NK 세포는 클러스터를 형성하면서 지속적으로 성장하였으며, 배양하는 21일 동안, 세포의 생존율이 평균 약 85 내지 90%로 높았다. 도 3에서 B에 나타낸 바와 같이, 건강한 정상인(대상 #12)의 NK 세포는, 배양 전에 비하여, 혼합처리군 1-3을 NK 세포에 가하여 배양한 21일 후, NK 세포의 수가 약 6 x 106 에서 1.58 x 108로, 약 26배 증가하였다. 또한, 건강한 정상인(대상 #2)의 NK 세포는, 세포의 수가, 혼합처리군 1-2를 첨가하여 배양한 경우 약 40배, 혼합처리군 1-3을 첨가하여 배양한 경우 약 53.3배 증가하였다. 도 3에서 C에 나타낸 바와 같이, NK 세포가 증식한 정도는, 세포의 수가, 혼합처리군 1-3을 첨가하여 21동안 배양한 경우 유의하게 높았다. 정상인 간의 차이는 조금씩 관찰되었다.

(1.2)과 같이 배양하는 중에, NK 세포의 활성도(activity)와 수용체(receptor)의 발현 정도의 차이를 확인하였다.
NK 세포를 배양하는 과정 중에, NK 세포를 회수하여, 1x105의 NK 세포를 FACS 튜브(Falcon® 5mL Round Bottom Polystyrene Test Tube)에 분주하였다. 상기 FACS 튜브에 타겟 인자(Target gene)의 항체(antibody)를 넣어준 뒤, 30분 동안 반응시켰다. 이 후, FACS 버퍼를 이용하여 세척하고, FACS(형광표지세포분류기, Fluorescence-activated cell sorting, BD FACSCalibur™)를 이용하여 표현 항원 특성을 분석하였다. 데이터 분석은 FlowJo 프로그램을 이용하였다.
도 4는 본 발명의 NK 세포 배양용 조성물이 NK 세포의 활성도(activity) 미치는 영향을 확인한 결과이다. 도 4를 참조하면, NK 세포의 활성 수용체(activating receptor) 중 하나인 CD226의 발현 수준은, 배양 전에 비하여, 혼합처리군 1-3을 첨가하여 배양한 후, 약 12.3%에서 약 95.7%로 증가하였다. 또한, 활성화의 지표인 CD69의 발현 수준은, 배양 전에 비하여, 혼합처리군 1-3을 첨가하여 배양한 후, 약 3.3%에서 약 91.4%로 증가하였다. 피더(feeder)를 사용하지 않고, 적은 수의 사이토카인(cytokine)만을 사용하는 것으로도, 많은 수의 NK 세포를 유도, 증식할 수 있다는 것을 알 수 있다.

세포 독성은 NK 세포에 대한 감수성이 높아 NK 세포의 활성 측정에 주로 사용되고 있는 K562 세포(human chronic myelogenous leukemia cell line)를 대상으로, Promega의 CytotTox-GloTM 세포 독성 분석 키트를 이용하여, 세포 독성 분석(cytotoxicity assay)을 수행하였다. 이는 손상된 세포막으로부터 유리된 효소를 측정하는 방법으로, 루미노제닉 펩티드 기질(luminogenic peptide substrate)(alanyl-alanyl-phenylalanyl-aminoluciferin, AAF-Glo Substrate)을 측정함으로써, 죽은 세포의 효소 반응을 측정할 수 있는 방법이다.
폴리-D-라이신(poly-D-lysine)이 코팅된 96-웰 플레이트의 각 웰에 K562 세포를 1x104 세포로 분주하고, 세포 독성을 측정하고자 하는 NK92와 NK 세포를 0:1, 1.25:1, 2.5:1, 5:1, 10:1의 E:T 비율(ratio)로 넣어주고, 4시간 동안 반응시켰다. 그 뒤, 50 uL의 CytotTox-GloTM 세포 독성 분석 시약(Cytotoxicity Assay reagent)를 넣고 상온에서 15분 동안 반응시킨 후, 루미노미터(luminometer)를 사용하여 죽은 세포의 효소량을 측정하였다. 이어서, 50 uL의 용해 버퍼(lysis buffer)를 첨가하고, 15분 동안 반응시켜, 전체 세포의 수를 확인한 뒤, 전체 세포에 대한 죽은 세포의 비율을 산출하여, 세포 독성을 분석하였다. 이 때, 수득된 데이터는 Microsoft Excel과 GraphPad Prism 5.0을 이용하여 분석하였다.
도 5는 본 발명의 NK 세포 배양용 조성물을 첨가하여 배양된 NK 세포의 암세포에 대한 세포 독성 확인한 결과이다. 도 5를 참조하면, 건강한 정상인(대상 #2)의 NK 세포는, 세포 독성이, 혼합처리군 1-3을 첨가하여 배양한 경우, 혼합처리군 1-4를 첨가하여 배양한 경우에 비하여 유의하게 높았다. 건강한 정상인(대상 #2)의 NK 세포는, 세포 독성이, 혼합처리군 1-3을 첨가하여 배양한 경우, 대조군인 NK92 세포에 비하여, 약 2배 이상 높았다. 건강한 정상인(대상 #1)의 NK 세포는, 세포 독성이, 혼합처리군 1-3 및 혼합처리군 1-4 각각을 첨가하여 배양한 경우, 대조군인 NK92 세포에 비하여, 유의하게 높았고, 건강한 정상인 (대상 #1 및 대상 #2) 모두에서 혼합처리군 1-3을 첨가한 경우에 세포 독성이 가장 높았다. 즉, NK 세포를 IL-15, IL-18, 및 IL-27을 포함하는 조성물을 처리하여 배양한 경우, 암세포에 대한 세포 독성이 유의하게 증가한 것을 알 수 있다.

폴리-D-라이신(poly-D-lysine)이 코팅된 6-웰 플레이트의 각 웰에 Calcein AM(Thermo Fisher Scientific)이 염색된 K562 세포를 1x105 세포로 분주하고, 세포 독성을 측정하고자 하는 NK92와 NK 세포를 0:1, 1.25:1, 2.5:1, 5:1, 10:1의 E:T 비율(ratio)로 넣어주고, 21일 동안 공동 배양하였다. 이 후, Calcein AM 방출 분석(release assay)를 통해서, K562 세포가 NK 세포에 의해서 용해(Lysis)되는 정도를 형광현미경(zeiss microscope)을 이용하여 관찰하였다.
도 6은 본 발명의 NK 세포 배양용 조성물을 첨가하여 배양된 NK 세포의 암세포에 대한 세포 독성을 확인한 결과이다. 녹색으로 염색된 영역은 K562 세포를 나타낸다. 도 6에 나타난 바와 같이, NK 세포(Effector)의 비율이 높은수록, 살아 있는 K562 세포의 수가 감소하였다. 이는 NK 세포와 K562 세포가 상호작용(Interaction)함에 따라, K562 세포가 세포사멸(apoptosis)하는 것으로 추측된다.

NK 세포 배양용 조성물을 첨가하여 배양된 NK 세포의 인비트로 독성을 평가하기 위하여, 자궁암 세포와 NK 세포를 배양한 후, 자궁암 세포에서의 케스파제-3(caspase-3)의 활성 수준 및 발현 수준을 확인하였다.
폴리-D-라이신(poly-D-lysine)이 코팅된 6-웰 플레이트의 각 웰에 난소암 세포(A2780, SKOV3)를 1x105 세포로 분주하고, 다음날 세포 독성을 측정하고자 하는 NK92 세포와 NK 세포 1x105 개를 상기 난소암 세포와 3시간 동안 배양한 후, 배양액과 NK 세포는 걷어내고, 남아있는 난소암 세포만을 회수하고, 용해 버퍼를 이용하여 세포를 용해시켰다. 세포 용해물을 95℃에서 10분간 가열한 후, 13,000rpm에서 20분간 원심분리하여, 단백질을 분리하였다. 수득한 단백질을 SDS-PAGE(SDS-polyacrylamide gel electrophoresis)젤을 이용하여 분리하고, 분리된 단백질을 PVDF(polyvinylidene fluoride) 멤브레인(EMD Millipore, Billerica, MA, USA)으로 이동시켰다. 상기 PVDF 멤브레인을 5%의 탈지유(skim milk)로 인큐베이션하여, 비특이적인 항체 결합을 차단(blocking) 하였다. 이어서, PVDF 멤브레인을 1차 항체, 항 케스파제-3(anti-caspase-3) 및 항-액틴(anti-actin) 각각과 4℃에서 하룻밤 동안 반응시켰다. 다음날, 1차 항체가 결합된 PVDF 멤브레인을 페록시다제(peroxidase)가 결합된 2차 항체와 상온에서 1시간 동안 반응시켰다. 단백질 밴드를 증가된 화학 발광(Enhanced chemiluminescence: ECL) 키트 시스템(EMD Millipore)을 사용하여 가시화하고 정량하였다.
도 7은 본 발명의 NK 세포 배양용 조성물을 첨가하여 배양된 NK 세포와 공동 배양된 암세포에서 케스파제-3(caspase-3)의 활성, 발현 양상, 및 발현 수준을 확인한 결과이다. 케스파제-3는 세포사멸(apoptosis)이 진행됨에 따라 whole form이 감소되고 cleaved form이 증가한다. 도 7에 나타낸 바와 같이, 본 발명의 NK 세포 배양용 조성물을 첨가하여 배양된 NK 세포와 공동 배양된 암세포(대상 # 9)는 cleaved form의 케스파제-3의 단백질 양이 증가하여, 암세포에 대한 세포 독성이 유의하게 증가한 것을 알 수 있다. 또한, 본 발명의 NK 세포 배양용 조성물을 첨가하여 배양된 NK 세포와 공동 배양된 암세포에서의 cleaved form의 케스파제-3의 발현 수준은, NK92 세포와 공동 배양된 암세포에서의 케스파제-3(caspase-3)의 발현 수준보다, 약 5% 내지 10% 증가하였다.

이어서, 암세포 내 세포사멸 진행 여부를 확인하였다.
폴리-D-라이신(poly-D-lysine)이 코팅된 챔버 슬라이드(chamber slide)에 난소암 세포(A2780, SKOV3)를 분주하고, 다음날 동일한 수의 NK92 세포와 NK 세포를 각각 상기 난소암 세포와 3시간 동안 배양한 후, 배양액과 NK 세포는 걷어내고, 남아있는 난소암 세포에 면역형광염색(immunofluorescence)을 수행하였다. 1차 항체로는 항-케스파제-3(anti-caspase3)과 항-액틴(anti-actin)을 사용하였고, 2차 항체로는 Alexa 488 고트 항-래빗(goat anti-rabbit)과 Alexa 546 고트 항-마우스(goat anti-mouse)를 사용하였다. 액틴과 DAPI는 대조군(control)으로 사용하였다. 염색 후, 슬라이드(slide)를 마운팅하고, 공초점 레이저-주사 현미경(confocal laser-scanning microscope)으로 관찰하였다.
도 8은 본 발명의 NK 세포 배양용 조성물을 첨가하여 배양된 NK 세포와 공동 배양된 암세포에서의 케스파제-3(caspase-3)의 발현 수준 및 발현 위치를 확인한 이미지이다. 도 8에 나타낸 바와 같이, 본 발명의 NK 세포 배양용 조성물을 첨가하여 배양된 NK 세포와 공동 배양된 암세포에서, 케스파제-3의 활성도가, 대조군(control)에 비해 약 50% 이상 높았으며, 다른 대조군인 NK92 세포와 공동 배양된 암세포에 비해 약 10% 이상 높았다.
Claims (7)
- IL-15, IL-18, IL-27 또는 이들의 조합을 포함하는 NK 세포(natural killer cell) 배양용 조성물.
- 청구항 1에 있어서, 상기 배양은 NK 세포의 증식 또는 활성화를 위한 것인 조성물.
- 청구항 1에 있어서, 세포 배양 배지 중에 상기 IL-15의 농도는 0.1 ng/ml 내지 1000 ng/ml, IL-18의 농도는 0.25 ng/ml 내지 2500 ng/ml, 및 IL-27의 농도는 0.20 ng/ml 내지 2000 ng/ml인 것인 조성물.
- 청구항 1에 있어서, 상기 NK 세포는 CD3- 및 CD56+의 표면 항원 특성을 갖는 것인 조성물.
- NK 세포를 IL-15, IL-18, 및 IL-27를 포함하는 NK 세포 배양용 조성물을 함유하는 배지에서 배양하는 단계;를 포함하는 NK 세포(natural killer cell)를 배양하는 방법.
- 청구항 5에 있어서,
상기 배양하는 단계 전에,
말초혈액으로부터 말초혈액 단핵구(Peripheral blood mononuclear cell: PBMC)를 수득하는 단계; 및
수득된 PBMC로부터 NK 세포를 분리하는 단계;를 더 포함하는 방법. - 청구항 5에 있어서, 상기 배양은 7일 내지 30일 동안 수행되는 것인 방법.
