Abstract
본 발명은, 천식 진단용 조성물을 제공함으로서, 천식환자에게서 발현이 변화하는 단백질 또는 그 유전자의 발현 수준을 측정 및 비교함으로서, 천식의 조기진단 및 질병 정도를 유의적으로 예측 또는 파악할 수 있다.
아울러, 본 발명의 천식 진단용 조성물은 비침습성 진단을 가능하게 하여 혈액, 뇨 검사 등으로 간단하고 유효성 있는, 천식의 초기 진단방법이다.
Description
도 2는 면역조직화학 염색법에 의해 오브알부민으로 감작 유발된 마우스 폐 조직의 넥틴-4와 아파딘 발현을 분석한 것이다.
도 3은 대조군과 천식 환자의 혈장 넥틴-4 수치를 나타낸다. *p<0.05, 대조군 vs. 천식 환자. 본 연구에 사용된 생체표본과 데이터는 한국 바이오뱅크 네트워크 회원인 순천향대학교 부천 병원의 바이오뱅크에서 얻었다.
도 4는 혈장 넥틴-4와 상기 임상변수와의 상관관계를 나타낸다.
Claims (15)
- 넥틴-4 (nectin-4)의 단백질 또는 이의 유전자의 mRNA의 수준을 측정하는 제제를 포함하는, 천식 진단용 조성물.
- 제1항에 있어서, 아파딘(afadin), Src, Rac1 또는 JNK(c-Jun N-terminal kinase)의 단백질 또는 이의 유전자의 mRNA의 수준을 측정하는 제제를 추가로 포함하는 것인, 천식 진단용 조성물.
- 제1항에 있어서, 상기 유전자의 mRNA 수준을 측정하는 제제는 상기 유전자에 특이적으로 결합하는 프라이머 쌍, 프로브 또는 안티센스 올리고뉴클레오티드인 것인, 천식 진단용 조성물.
- 제1항에 있어서, 상기 단백질 수준을 측정하는 제제는 상기 단백질에 특이적으로 결합하는 항체를 포함하는 것인, 천식 진단용 조성물.
- 제1항에 따른 조성물을 포함하는, 천식 진단용 키트.
- 제5항에 있어서, 상기 키트는 RT-PCR(Reverse transcription polymerase chain reaction) 키트, DNA 칩 키트, ELISA(Enzyme-linked immunosorbent assay) 키트, 단백질 칩 키트 또는 래피드(rapid) 키트인 것인, 천식 진단용 키트.
- 생물학적 시료로부터 넥틴-4 (nectin-4)의 단백질 또는 이의 유전자의 발현 수준을 측정하는 단계; 및
상기 단계에서 측정된 단백질의 발현 수준 또는 유전자의 발현 수준을 정상 대조구 시료와 비교하는 단계를 포함하는, 천식 진단을 위한 정보의 제공 방법. - 제7항에 있어서, 아파딘(afadin), Src, Rac1 또는 JNK(c-Jun N-terminal kinase)의 단백질 또는 이의 유전자의 mRNA의 수준을 추가로 측정하는 것인, 천식 진단을 위한 정보의 제공 방법.
- 제7항 또는 제8항에 있어서, 천식 의심 개체의 분리된 시료의 상기 단백질의 발현 수준 또는 유전자의 발현 수준이 정상 대조군 시료의 단백질의 발현 수준 또는 유전자의 발현 수준보다 높을 경우, 천식으로 판단하는 것을 특징으로 하는, 천식 진단을 위한 정보의 제공 방법.
- 제7항에 있어서, 상기 유전자의 발현 수준은 상기 유전자의 mRNA 발현 수준인, 천식 진단을 위한 정보의 제공 방법.
- 제10항에 있어서, 상기 mRNA 발현 수준 측정은 역전사 중합효소반응, 경쟁적 역전사 중합효소반응, 실시간 역전사 중합효소반응, RNase 보호 분석법, 노던 블랏팅 또는 DNA 칩에 의한 것인, 천식 진단을 위한 정보의 제공 방법.
- 제7항에 있어서, 상기 단백질 발현 수준 측정은 해당 단백질에 특이적으로 결합하는 항체를 이용하는 것인, 천식 진단을 위한 정보의 제공 방법.
- 제7항에 있어서, 상기 단백질 발현 수준 측정 또는 비교는 단백질 칩 분석, 면역측정법, 리간드 바인딩 어세이, MALDI-TOF(Matrix Desorption/Ionization Time of Flight Mass Spectrometry) 분석, SELDI-TOF(Sulface Enhanced Laser Desorption/Ionization Time of Flight Mass Spectrometry) 분석, 방사선 면역분석, 방사 면역 확산법, 오우크테로니 면역 확산법, 로케트 면역전기영동, 면역조직화학(immunohistochemistry) 분석, 면역세포화학(immunocytochemistry) 분석, 보체 고정 분석법, 2차원 전기영동 분석, 액상 크로마토그래피-질량분석(liquid chromatography-Mass Spectrometry, LC-MS), LC-MS/MS(liquid chromatography-Mass Spectrometry/ Mass Spectrometry), 웨스턴 블랏, 및 ELISA(enzyme linked immunosorbentassay)로 구성된 군으로부터 선택되는 것을 이용하여 수행하는 것을 특징으로 하는, 천식 진단을 위한 정보의 제공 방법.
- 분리된 세포, 분리된 조직 또는 인간을 제외한 동물에 천식 치료제 후보물질을 처리하는 단계;
상기 후보물질 처리군에서 넥틴-4 (nectin-4)의 단백질의 발현수준을 측정하는 단계; 및
상기 측정된 넥틴-4 (nectin-4)의 단백질의 발현수준이 후보물질을 처리하지 않은 대조군보다 감소하면, 상기 후보물질을 천식치료제로 선택하는 단계를 포함하는, 천식치료제의 스크리닝 방법. - 제14항에 있어서, 아파딘(afadin), Src, Rac1 또는 JNK(c-Jun N-terminal kinase)의 단백질의 발현 수준을 추가로 측정하고, 상기 측정된 아파딘(afadin), Src, Rac1, 또는 JNK(c-Jun N-terminal kinase)의 단백질의 발현수준이 후보물질을 처리하지 않은 대조군보다 감소하면, 상기 후보물질을 천식치료제로 선택하는 단계를 더 포함하는 것인, 천식치료제의 스크리닝 방법.
