(Test Example 1) Antioxidant activity measurement (DPPH)
The antioxidative activity of the solution isolated from chrysanthemum was measured as follows.
50 uL of each solution of each test group was dispensed into a 96-well plate and 150 uL of 100 uM DPPH (1,1-diphenyl-2-picrylhydrazyl, Sigma, USA) solution dissolved in ethanol was added. After incubation at room temperature for 30 minutes, the absorbance was measured with an ultraviolet / visible spectrophotometer (SPECORD 200, Analytic jeca) at 530 nm using an ELISA reader (DYNEX, USA). As a control, 50 uM L-ascorbic acid (Sigma, USA) was used. The DPPH radical scavenging activity of each sample was calculated in comparison with the absorbance of the control group to which no sample was added. The absorbance was measured and the degree of antioxidant activity was measured according to the following equation.
Antioxidant activity (%) = [1- (sample absorbance 530 nm) / (absorbance 530 nm)] x 100
(Test Example 2) Measurement of cell activity
The macrophage cell line (RAW 264.7) used in the experiment was purchased from Korean cell line bank and the morphology was observed. Cells subcultured 10 times or less were used for the experiment. RAW 264.7 cells were subcultured in a 5% CO2 incubator at 37 ° C in DMEM (Invitrogen, USA) supplemented with 10% (v / v) FBS Respectively.
Cell safety (cytotoxicity)
Cytotoxicity was performed by modifying Mosmann's method of measuring cell viability using 3- (4,5-dimethylthiazol-2-yl) -2-5-diphenyltetrazolium bromide (MTT) reagent. RAW 264.7 cells were seeded at a density of 1 × 10 5 cells / well in a 96-well plate and cultured at 37 ° C and 5% CO 2 for 24 hours. After the culture medium was removed, the sample was incubated in the treated medium for 24 hours, and the medium was removed and washed twice with PBS (phosphate buffered saline). MTT was dissolved in PBS at 300 μg / mL and 100 μL was added and incubated at 37 ° C and 5% CO 2 for 4 hours. DMSO was added to each well of 150 μL and stirred for 30 minutes. Absorbance was measured at 570 nm (reference 600 nm) using an ELISA reader (DYNEX, USA). The degree of cell proliferation was expressed as a percentage of the absorbance of the control.
Promoting collagen biosynthesis
Collagen aggregation performance was assessed by the use of antibodies that recognize the C-terminus of the collagen precursor (PIP, Procollagen Type 1 C-peptide). Cells were treated with 100 ug / mL of each sample at a concentration of 200 ug / mL, and the culture solution was collected to measure the amount of the C-terminus of the collagen precursor.
According to the above results, it was confirmed that the chrysanthemum matured according to the present invention had statistically significant activity against collagen biosynthesis, and thus it was confirmed that the chrysanthemum matured according to the present invention had a remarkably excellent effect for wrinkle improvement .
NF-kB activity inhibitory activity
To determine whether the activity of inhibiting the production of inflammatory cytokines (IL-1β, IL-6 and TNF-α) produced by LPS stimulation in RAW 264.7 cells was related to nuclear factor-kappaB (NF-κB), RAW 264.7 Cells were subcultured in 96-well plates at a density of 5 x 10 cells / well and stabilized for 24 hours. Samples were diluted to 0-0.5 mg / mL and stimulated with 1 ug / mL LPS (Lipopolysaccharide, Sigma, USA) 2 hours later. The expression of NF-κB was measured using a mouse nuclear factor-kappaB p65 ELISA kit (MyBioSource, USA) and a mouse inhibitory subunit of NF-κB (IKBa) ELISA kit (MyBioSource, USA) . The concentration of NF-κB p65 was calculated to 12 ng / mL, and the concentration of IKBα was diluted by 2 fold, compared with the standard curve obtained.
According to the above results, it was confirmed that the chrysanthemums of the present invention had a statistically significant effect on inhibition of NF-kB activity. Thus, the chrysanthemum matured according to the present invention has a remarkably excellent effect for relieving or improving inflammation .
The results of Test Example 1 and Test Example 2 are summarized in Table 1 below.

(Test Example 3) Evaluation of effect of cosmetic composition through sensory evaluation
The hue and flavor of the chrysanthemums prepared in Examples 1 to 3 and Comparative Examples 1 to 16 were evaluated by a sample of 40 adult women between 20 and 40 years old.
The evaluation was made using a conventionally used blind-test so that each sample experimenter was unaware of the sample numbers of Examples 1 to 3 and Comparative Examples 1 to 16 to prevent the prejudice of the samples from intervening.
In the evaluation process, each sample was simultaneously supplied to the experimenter. To prevent the influence of the results on the previous sample from affecting the evaluation of the next sample, wash the hands to treat the sample before evaluation of each sample. And the following samples were evaluated.
The evaluation score was 5 points of 5 points for each evaluation item, and the score was divided into 5 points, 5 points for the best, 4 points for the normal, 3 points for the normal, 2 points for the bad, and 1 point for the worse. The mean value for the sample was derived. The results are shown in Table 2 below.

As shown in Table 2, when Examples and Comparative Examples were compared, in Examples 1 to 3, both color tone and flavor were evaluated. From the above results, it was found that the chrysanthemums obtained by the present invention had excellent antioxidative ability, collagen production ability and NF-kB inhibitory ability, and remarkably decreased cytotoxicity. Also, in the evaluation of the blinded method, And thus the present invention has been completed.
