Description

[0001] Cosmetic composition comprising aged chrysanthemum [

The present invention relates to a cosmetic composition containing chrysanthemums.

The extent to which aging of cells affects the aging of individuals (humans, animals) is not yet known. It is well known that the telomere at the end of the chromosome is shortened each time it breaks down and the cell dies when it reaches its limit. Recently, there has been research that collagen, which is the main component of cell epilepsy, is reduced to 65% of the 20s when it is in their 80s and is involved in aging. There are also studies that abnormal protein accumulation due to intracellular ER stress is involved in aging.

In skin aging, the three major components of connective tissue, collagen and proteoglycan, are catabolized by intermolecular cross-linking or branching, resulting in changes in cell-extracellular matrix interactions. In addition, the decrease of the polar cells and the turnover of the epidermal cells (turn over, usually 28 days) are reduced from 70 to 40% of their 30s, causing skin dullness. And collagen, ceramide reduction is the main cause of skin wrinkles due to aging. It has been reported that the administration of estrogen improves the moisturization by increasing the synthesis of hyaluronic acid as well as the collagen increase of the skin (EL-Domyati M et al .: Exp Dermatol 11, 2002)

Oxidative stress is known to be an important cause of aging. Cellular stressors include reactive oxygen species called reactive oxygen species (ROS), such as peroxides, hydroxy radicals, and hydrogen peroxide. RNS (reactive nitrogen species) such as nitric oxide and peroxynitrite are known, and lipid peroxides are also known as oxidative stress inducers. These oxides are known to bind to guanine, a DNA base, to induce mutagenesis, leading to skin aging and cancer. It has been reported that it participates in the arachidonic acid cascade and causes vasoconstriction and platelet aggregation to develop as a risk factor for arteriosclerosis or Alzheimer's disease. Vitamin C and B12 are known to be water soluble and lycopene and beta-carotene are known to be lipid soluble.

Chrysanthemum is a perennial herb belonging to the Compositae, mainly edible or medicinal. It is also called as "Ganja" and "Nakyanghwa". Chrysanthemums can be used in a variety of ways, including chrysanthemums, chrysanthemums, and tea. The pharmacological action of chrysanthemum has the functions of promoting blood circulation, warming, and pain. It can be used for fever, fever, headache, dizziness, earache, eye disease, tumor pain, fever, detoxification, analgesia and inflammation.

The effect of chrysanthemum on the primordial gangbok is' good for blood, take a long time and lighten the body, and do not grow old. It comforts the stomach, helps the five, and regulates the limbs. It is also effective for colds, headaches and dizziness. " In addition, references to the efficacy of chrysanthemum tea are also mentioned in the literature such as the Enhanced Forest Economy, Chungdang Shoten, and Divine Materia Medica. Chrysanthemum tea has these effects because it contains vitamin A, vitamin B1, choline, stachydrin, and adenine which are good for restoring eye and liver function among the components of chrysanthemum. Especially, chrysanthemum flowers contain trimethylcyclohexane, carbosilicic acid and acacetin, leaves and stems of adenine, and cholinestatidrine.

Although the usefulness of the above-mentioned chrysanthemum is found, the chrysanthemum has many problems to be used as it is, so it is a level of extracting useful components of chrysanthemum flower and chrysanthemum using the chrysanthemum flower as an additive. Korean Patent Publication No. 0278203 discloses a stress-relieved beverage produced by mixing ethanolic extracts of chrysanthemums with 12 kinds of food ingredients such as soybean embryo and bamboo shoots. However, this is merely the use of ethanol extract of chrysanthemum, and when chrysanthemum itself is used as it is, most of it remains in a state of concern with hydrothermal fluids, so that the functional materials contained in chrysanthemum can not be utilized sufficiently.

Korean Patent Registration No. 0278203 (October 17, 2000). Korean Patent Publication No. 2009-0042450 (2009.04.01.).

In order to solve the above-mentioned problems, the present invention provides a method for producing a chrysanthemum comprising the steps of mixing a small amount of saccharides and minerals in a sterilized environment to activate the enzyme, Respectively.

Thus, the effect of improving skin aging can be remarkably improved.

In the present invention, the saccharide may be, but not limited to, one or more selected from the group consisting of glucose, lactose, fructose, sugar, trehalose, xylitol, mannitol, sorbitol, erythritol and galacto-oligosaccharide.

Glucose is widely contained in fruits, honey and vegetables in a small amount, and most of the commercially available crystalline glucose is prepared by hydrolyzing corn starch. Lactose is contained in the milk of all animals. Galactose and glucose are combined to form lactose. Fructose is widely distributed in plants along with glucose. Powdered honey is made by pulverizing honey. Honey is made up of 20% water and 80% carbohydrate. Most of them are glucose and fructose. Honey is in a liquid state, but because it contains a large amount of fructose, it absorbs moisture in the air and dissolves itself. In the case of high glucose, it also forms crystals. Sugar is made from raw sugar obtained by heating, precipitation and centrifugation of squeezed juice by squeezing sugar cane and sugar beet with high sugar content or sugar content. The brown sugar and the yellow sugar are produced by refining the raw sugar with little or no purification. And trehalous (trehalous) is widely found in nature such as plants and microorganisms, as well as protection against drying and freezing of food, as well as anti-aging starch. It is known that it has various excellent functions such as prevention of fatty acid corrosion, stabilization of food texture, improvement of taste and flavor. Powdered alcohol is powdered by evaporating water by partial dehydration in short time by adding dextrin to alcohol, suppressing degeneration of starch, inhibiting denaturation of protein, suppressing decomposition of lipid, etc. And sugar alcohols are xylitol, mannitol, sorbitol, erythritol, and galacto-oligosaccharides as non-cyclic linear polyhydric alcohols.

In the present invention, the minerals may be any one or more selected from the group consisting of nephrite, talc, muscovite, sericite, kaolin, seamstone and organic germanium.

Nephrite (nephrite, CaO 4 3 MgO SiO 2) is intertwined with the fine fibrous crystals refers to a compact-to-seomseok or tunok seomseok achieved the gyeonggoe, it said corresponding to jade. The quality is strong and does not break well, and when it is polished, it becomes glossy. The hardness is 5.5 ~ 6.0, the specific gravity is 2.9 ~ 3.0, the color is white and dark green. "It is known that ogal is tasteless and poisonous, it releases the heat of the stomach and intestines, relaxes the osmanthus, and expels waste from the body." Okamu promotes blood circulation and brightens the eyes. " It is known that the clinical effect of nephrite has been shown to be therapeutic effect such as headache, insomnia, burning of mouth and limb and electroacoustic adsorption of anion by Ca 2+ , which is excellent in sterilization action and waste adsorption ability and can prevent acidification of body fluids . The content of nephrite in the cosmetic composition of the present invention is preferably 1 to 10% by weight based on the total weight of the composition. If it is less than 1% by weight, the adsorption force by Ca 2+ becomes weak. If it exceeds 10% by weight, absorption of zinc, magnesium and iron components is interfered.

Talc (talc, Mg 3 Si 4 O 10 (OH) 2) is a rock belonging to the monoclinic system having a crystal structure such as mica, hardness of 1, specific gravity, and can be cut with a knife to 2.7 to 2.8, the color is white, silver white And pale green. Magnesium in talc is known to promote skin metabolism, contribute to the structural stabilization of ATP energy, and promote protein synthesis. Magnesium also has the ability to control contraction and relaxation of muscle contrary to calcium. Talc is mainly used for infant perspiration. It is used to treat eczema because it has the function of eliminating the sausage and reducing the heat. It is used as a base in cosmetics and as an additive in foods and pharmaceuticals. The content of talc in the cosmetic composition of the present invention is preferably 10 to 20% by weight based on the total weight of the composition. If it is less than 10% by weight, the skin metabolism and muscles become too shrunk, and if it exceeds 20% by weight, skin metabolism and muscles become too relaxed.

Muscovite (K (OHF 2 ) 2 Al 3 Si 3 O 10 ) is a monoclinic mineral that contains a small amount of iron and magnesium. The hardness is 2 ~ 2.4, the specific gravity is 2.7 ~ 3, colorless, white, yellow, gray, green, The fluoride component of muscovite is known to inhibit the activation of melanin pigment, which has a whitening effect and moisturizes the skin. The content of muscovite in the cosmetic composition of the present invention is preferably 20 to 35% by weight based on the total weight of the composition. If it is less than 20% by weight, skin whitening and moisturizing effect hardly appear, and if it exceeds 35% by weight, skin whitening and moisturizing effect become excessively excessive.

Sericite (KAl 2 (OH) 2 (AlSi 3 O 10 )) belongs to monoclinic, white or grayish white with pearly luster. Originally referred to as the main component minerals of crystalline schist, especially sericite schist, today refers to fine clay mined muscovite due to hydrothermal action. The chemical composition is almost the same as that of muscovite, but potassium (K) is generally less than muscovite and water (H 2 O) is somewhat higher. In addition, sericite contains various minerals essential for human body such as selenium (Se), zinc (Zn), magnesium (Mg), and calcium (Ca), and it shows heavy metal detoxification and antioxidant efficacy. In addition, sericite is used as a mixture of ceramics and refractory bricks, and there are many other uses such as paints, electric insulators, active materials and cosmetics. The content of sericite in the cosmetic composition of the present invention is preferably 10 to 20% by weight based on the total weight of the composition. If the amount is less than 10% by weight, the detoxification of heavy metals and the antioxidant effect are weakened. If the amount is more than 20% by weight, the intracellular total antioxidant system is destroyed and the detoxification of heavy metals and the antioxidant effect are more weakened.

China clay (kaolin, Al 2 Si 2 O 5 (OH) 4) is kaolinite (kaolinite) and halroyi the site is the main component, feldspar, in the rock o orthoclase o Soda Feldspar o as chairman seats jangseokryu is chemically by acid or water It is produced by decomposition weathering, and is pure white or slightly gray. Kaolin powder cleanses the skin and removes dirt and toxins from the surface of the skin, sebum from the pores, and dead skin cells, and is good for oily and acne skin. In terms of the term, it is called Baekseokji and Hwangseokji according to the color, and they are all used for skin troubles. The content of the kaolin in the cosmetic composition of the present invention is preferably 35 to 45% by weight based on the total weight of the composition. If the content is less than 35% by weight, the affinity between the constituents decreases, and the adsorption force is weakened. If the content exceeds 45% by weight, the affinity becomes too high and the adsorption force becomes too strong.

Pumice is a pore-rich mineral formed by solidification of volcanic rocks. The anatomic rocks are silicate minerals ([SiO 2 ] x ) including rhyolite, trachyte and phonolite, which are essential minerals such as calcium, iron, zinc, copper, nickel, chromium, cobalt, vanadium, . It is also known to treat the symptoms of blurred vision in the eye. In the cosmetic composition of the present invention, the content of the hydrous stones is preferably 1 to 3% by weight based on the total weight of the composition. If it is less than 1% by weight, it hardly acts on the skin. If it is more than 3% by weight, heavy metals are harmful to the skin.

Organic germanium (Ge) exists in a trace amount, and has an oxygen supplying action, a semiconductor action (an electron flow regulating action, an anion effect), an immunity enhancing action, an interferon production, , The action of administra- tion, the excretion of heavy metals in the body, and the enhancement of natural healing power. Therefore, cosmetics using high purity germanium; Health tools such as bracelets, necklaces, and coats; Soap, and so on, and it is a substance that shows a strong growth rate in demand around the world. In the cosmetic composition of the present invention, the content of the anatomic stones is preferably 0.1 to 0.3% by weight based on the total weight of the composition. If it is less than 0.1% by weight, the effect of germanium does not appear. If it exceeds 0.3% by weight, it shows toxicity to the skin.

Minerals other than organic germanium in the above minerals include essential elements such as Ca, Mg, P and K and elements such as Fe, Zn, Se, Mn, Cu, Mo, Co, Cr, F, Ni, V, B, Se and other micro-elements, which can contribute to the cell's life activity. In particular, selenium among the above elements is a representative biodegradable enzyme that acts to remove hydrogen peroxide generated in the body as a coenzyme of glutathione peroxidase enzyme. Therefore, the elements such as selenium contained in the mineral complex component are known to promote the action of glutathione peroxidase, thereby removing the by-products of intracellular metabolism, and participating in cell division and differentiation.

Silicon (Si, silicon), which is commonly contained in the minerals, is a natural substance contained in uncooked grains. It is an important element accounting for 0.5% of body weight and plays an important role in protein synthesis as a connective tissue So that the connective tissue is an essential component for maintaining elasticity. Silicone also has research on immunity-enhancing functions. In order to use the silicon ingested by cells, various essential minerals such as boron, calcium, magnesium, manganese, and potassium are needed at the same time.

Antioxidant activity (%) = [1- (sample absorbance 530 nm) / (absorbance 530 nm)] x 100

(Test Example 2) Measurement of cell activity

The macrophage cell line (RAW 264.7) used in the experiment was purchased from Korean cell line bank and the morphology was observed. Cells subcultured 10 times or less were used for the experiment. RAW 264.7 cells were subcultured in a 5% CO2 incubator at 37 ° C in DMEM (Invitrogen, USA) supplemented with 10% (v / v) FBS Respectively.

Cell safety (cytotoxicity)

Cytotoxicity was performed by modifying Mosmann's method of measuring cell viability using 3- (4,5-dimethylthiazol-2-yl) -2-5-diphenyltetrazolium bromide (MTT) reagent. RAW 264.7 cells were seeded at a density of 1 × 10 5 cells / well in a 96-well plate and cultured at 37 ° C and 5% CO 2 for 24 hours. After the culture medium was removed, the sample was incubated in the treated medium for 24 hours, and the medium was removed and washed twice with PBS (phosphate buffered saline). MTT was dissolved in PBS at 300 μg / mL and 100 μL was added and incubated at 37 ° C and 5% CO 2 for 4 hours. DMSO was added to each well of 150 μL and stirred for 30 minutes. Absorbance was measured at 570 nm (reference 600 nm) using an ELISA reader (DYNEX, USA). The degree of cell proliferation was expressed as a percentage of the absorbance of the control.

Promoting collagen biosynthesis

Collagen aggregation performance was assessed by the use of antibodies that recognize the C-terminus of the collagen precursor (PIP, Procollagen Type 1 C-peptide). Cells were treated with 100 ug / mL of each sample at a concentration of 200 ug / mL, and the culture solution was collected to measure the amount of the C-terminus of the collagen precursor.

According to the above results, it was confirmed that the chrysanthemum matured according to the present invention had statistically significant activity against collagen biosynthesis, and thus it was confirmed that the chrysanthemum matured according to the present invention had a remarkably excellent effect for wrinkle improvement .

NF-kB activity inhibitory activity

To determine whether the activity of inhibiting the production of inflammatory cytokines (IL-1β, IL-6 and TNF-α) produced by LPS stimulation in RAW 264.7 cells was related to nuclear factor-kappaB (NF-κB), RAW 264.7 Cells were subcultured in 96-well plates at a density of 5 x 10 cells / well and stabilized for 24 hours. Samples were diluted to 0-0.5 mg / mL and stimulated with 1 ug / mL LPS (Lipopolysaccharide, Sigma, USA) 2 hours later. The expression of NF-κB was measured using a mouse nuclear factor-kappaB p65 ELISA kit (MyBioSource, USA) and a mouse inhibitory subunit of NF-κB (IKBa) ELISA kit (MyBioSource, USA) . The concentration of NF-κB p65 was calculated to 12 ng / mL, and the concentration of IKBα was diluted by 2 fold, compared with the standard curve obtained.

According to the above results, it was confirmed that the chrysanthemums of the present invention had a statistically significant effect on inhibition of NF-kB activity. Thus, the chrysanthemum matured according to the present invention has a remarkably excellent effect for relieving or improving inflammation .

The results of Test Example 1 and Test Example 2 are summarized in Table 1 below.

(Test Example 3) Evaluation of effect of cosmetic composition through sensory evaluation

The hue and flavor of the chrysanthemums prepared in Examples 1 to 3 and Comparative Examples 1 to 16 were evaluated by a sample of 40 adult women between 20 and 40 years old.

The evaluation was made using a conventionally used blind-test so that each sample experimenter was unaware of the sample numbers of Examples 1 to 3 and Comparative Examples 1 to 16 to prevent the prejudice of the samples from intervening.

In the evaluation process, each sample was simultaneously supplied to the experimenter. To prevent the influence of the results on the previous sample from affecting the evaluation of the next sample, wash the hands to treat the sample before evaluation of each sample. And the following samples were evaluated.

The evaluation score was 5 points of 5 points for each evaluation item, and the score was divided into 5 points, 5 points for the best, 4 points for the normal, 3 points for the normal, 2 points for the bad, and 1 point for the worse. The mean value for the sample was derived. The results are shown in Table 2 below.

[Table 2] Hue, incense

Figure pat00002

As shown in Table 2, when Examples and Comparative Examples were compared, in Examples 1 to 3, both color tone and flavor were evaluated. However, in the case of Comparative Examples 12 to 16, only the extracts were extracted with hot water or alcohol only. Experiments were sensitive to chrysanthemum, and the color tone was not so good. In addition, in the case of Comparative Examples 1 to 11, the chrysanthemum fermentation solution obtained in a non-sterilized condition, and in this case also, the evaluation of the hue and flavor of the subject is better than that of the chrysanthemum obtained through the examples of the present invention I did not receive it.

From the above results, it was found that the chrysanthemums obtained by the present invention had excellent antioxidative ability, collagen production ability and NF-kB inhibitory ability, and remarkably decreased cytotoxicity. Also, in the evaluation of the blinded method, And thus the present invention has been completed.
